1.Effects of oxygen inhalation modes on retinal vessels development in neonatal rat

Objective To investigate the effects of different oxygen inhalation modes on retinal vessels development in neonatal rat in order to approach its etiopathogenesis and provide experimental evidence for proper oxygen therapy for premature.Methods Totally 150 postnatal day 7(P7)Wistar rats were randomly assigned into 5 groups according to different oxygen inhalation modes(n=30 in each group).Experimental group 1 were exposed to 75%,60%,50%,40% and 30% oxygen in turn for one day,followed by room air exposure for 5 d.Experimental group 2 were exposed to 30%,40%,50%,60% and 75% oxygen in turn for one day,then exposed in air for 5 d.Experimental group 3 were exposed to 75% oxygen for 5 d,then oxygen supply was suddenly stopped,followed by room air exposure for 5 d.Experimental group 4 were exposed to 75% oxygen for 5 d,then the oxygen supply was reduced by 10% per day till oxygen supply level equal to room air,finally room air exposure for 5 d.The control group were exposed to room air in the same experimental condition.The retinal vascular development and proliferation were evaluated by stretched preparation(ADPase stained retina)and eyeball cross-section.The nuclei of proliferative retinal vessels of 5 groups were counted and the vascular endothelial growth factor(VEGF)expressions in retina were detected by immunohistochemical staining.The dependability between the nuclei of proliferative retinal vessels and the VEGF expressions in retina were also analyzed.Results In the experimental groups 2 and 3,there existed retinal hypoxia and formed abnormal new vessels as retinopathy of prematurity(ROP).The VEGF expressions increased after hypoxia,mainly in endochylema and nucleus of capillary endothelium of ganglionic layer of retina and inner nuclear layer.Linear correlation analysis of the data indicated the positive correlation was between the nuclei of the proliferative vessels and the VEGF levels in retina(r=0.807,P
2.Phacoemulsification and implantation of artificial lens for traumatic cataract

Objective To investigate the effect of surgical management of phacoemulsification and implantation of artificial lens for traumatic cataract.Methods The surgery was performed on 19 patients.Results Post-operatively, the visual acuities were 0.5 or better in 57.9% and 1.0 or better in 15.8% in one week; 0.5 or better in 78.9% and 1.0 or better in 21.0% in one month.Conclusion It can have better effect to perform phacoemulsification and implantation of artificial lens in different conditions with surgery skill and better equipment.
3.Expressions of cyclooxygenase-2 and 5-1ipoxygenase in colonic mucosa of ulcerative colitis and the relationship between them

Objective To investigate the expressions of cyclooxygenase-2 (COX-2) and 5-lipoxygenase (5-LOX) in colonic mucosa of patients with ulcerative colitis (UC)and to analyze the relationship between them. Methods The specimens of colonic mucosa from 32 UC patients were graded according to endoscopic and histological grading standards, and specimens of colonic mucosa from 26 healthy controls were also collected. The expressions of COX-2 and 5-LOX mRNA and protein in colonic mucosa were determined using real-time fluorescent quantitative reverse transcription polymerase chain reaction(RT-PCR) and immunohistochemistry. The correlation between COX-2 and 5-LOX was analyzed. Results For UC patients,10 were class 1, 19 class 2 and 3 class 3 according to endoscopic grading, whereas 19 were class Ⅰ , 9 class Ⅱ and 4 class Ⅲ according to histological grading. The expressions of COX-2 and 5-LOX mRNA in active UC patients were 81. 25% and 53.13%, respectively, and were 11.54% and 19. 23% in healthy controls, respectively, with significant differences between the two groups (all P values<0.01). The positive expressions of COX2 and 5-LOX increased in accordance with increasing of endoscopic grading and histological grading.The levels of COX-2 and 5-LOX were 20. 08±1.17 and 37.83 ±1.48 in colonic mucosa tissues of UC patients, respectively, and 48.42 ± 1.69 and 11.28 ± 1.62 in healthy controls, respectively. There was significant difference between the two groups (all P values<0.05). A good positive correlation was found between COX-2 and 5-LOX. Conclusions The expressions of COX-2 is closely related to 5-LOX in UC patients. Both may play a pivotal role in inflammation of UC.
6.Auxiliary Diagnosis Value of Glucose-6 Phosphate Dehydrogenase Activity on Thalassemia

Objective To explore the value and applicability of the activity of glucose-6 phosphate dehydrogenase(G-6PD) for the auxiliary diagnosis of thalassemia.Methods Nine hundred and forty samples verified by the agar gel hemoglobin electrophoresis and(or) gene diagnosis,blood count measurement,serum ferritin and G-6PD activity test were divided into 3 groups [820 cases of thalassemia in group A;40 cases of iron deficiency anemia(IDA) in group B;80 cases normal control group in group C] and the G-6PD activities of them were analyzed statistically.Results The activity of G-6PD of those samples were(35.23?7.11),(34.95?10.72),(26.64?10.85),(23.86?7.68),(19.89?5.99),(18.65?6.67),(16.75?5.49) NBTU respectively in HbH disease,?-thalassemia major,?-thalassemia intermedia,IDA,?-thalassemia minor,? combine with ?-thalassemia,?-thalassemia trait,there were significant differences compared with normal control group(Pa0.05).Conclusions G-6PD activity increase in both thalassemia and IDA group,it can be used in auxiliary diagnosis of thalassemia but had its serviceable range.It′s suitable for the auxiliary diagnosis of HbH disease and ?-thalassemia major,but not for the discrimination of gene category in thalassemia.

8.Immunological regulation mechanism of (-)-epigallocatechin-3-gallate on rheumatoid arthritis

Objective To study the (-)-epigallocatechin-3-gallate (EGCG) function on the proliferation of T cells derived from the peripheral blood and synovial fluid of rheumatoid arthritis (RA) and RA-related cytokine levels and the role of EGCG on RA synovial fibroblasts (FLS) proliferation was investigated. Methods ① Mononuclear cells from RA peripheral blood (30 cases) and synovial fluid (23 cases)were isolated. Blank group, negative control group, positive control methotrexate (MTX) group and therapeutic group with three different concentrations of EGCG were set up. Incorporated isotope 3H was used to test T cell proliferation from RA-PBMC and SFMC. ELISA assay was used to test cytokine (TNF-α, IFN-γ, IL-1, IL-6 and IL-17A) levels. ② MTT assay was used to test FLS proliferation from RA synovial tissue (8 cases).Results ① The CPM value of the high-dose group of EGCG in the peripheral blood and synovial fluid of RA patients was [ ( 15 136±2910), ( 11 587±3135 ) ], which was declined significantly than the control group (42856±2127) (P<0.01). The levels of TNF-α, IFN-γ, IL-1, IL-6 and IL-17A in the high-dose group of EGCG in the peripheral blood were [(321±13), (298±20), (132±12), (197±7), (59±8) pg/ml], which were decreased significantly than those of the control group [ (458±28), (505±26), (346±28), (405±25),(109±13) pg/ml ] (P<0.05 or P<0.01 ). The levels of TNF-o, IFN-γ, IL-1, IL-6 and IL-17A in the highdose group of EGCG in the synovial fluid were [(41.4±2.9), (182±16), (56.3±11.0), (34.2±1.9), (44±8)pg/ml ], which was decre-ased significantly than the control group [ ( 388.3± 19.3 ), (469±20), ( 104.2±17.8 ),( 114.5±4.8), ( 104±11 ) pg/ml] (P<0.05 or P<0.01 ). ② The level (A) of the high-dose group of EGCG in the FLS was (0.08±0.02), which was declined significantly than the blank group (0.27±0.04) (P<0.05).Conclusion ① In vitro EGCG can inhibit T cell proliferation from peripheral blood and synovial fluid of RA patients and the TNF-α, IFN-γ, IL-1, IL-6 and IL-17A secretion are decreased. ② In vitro EGCG can inhibit the proliferation of RA FLS.
9.Cerebrospinal fluid adenosine deaminase activity for the diagnosis of tuberculous meningitis

Objective To evaluate cerebrospinal fluid adenosine deaminase(CSF-ADA)activity in the diagnosis of tuberculous meningitis(TMB), and to observe its dynamic changes. Methods A total of 160 patients were included and were divided into two groups: 76 cases of TBM and 84 cases of non-TBM.Among the cases of non-TBM, there were 36 cases of bacterial meningitis, 30 cases of viral meningitis and 18 cases of cryptocoocal meningitis. All the patients were measured with their CSF-ADA activity by Enzymecoupled assay(Trinder method)and 47 patients of TBM were measured again after 2 weeks' and 6 weeks'antitubercular therapy. Results were expressed as(-x)± s. Mann-Whitney U test and paired-samples t test were used. Results CSF-ADA activity in TBM group was(12.9 ±6.4)U/L, while that in the non-TBM group was(6.0 ± 4.1)U/L, the difference was of statistical significance(U = 7.860, P < 0.05). With the cutoff value of 9 U/L, the sensitivity and specificity to differentiate TBM from non-TBM was 84.21% and 83. 33%, respectively. CSF-ADA activity decreased in TBM patients after antitubercular treatment.Conclusions CSF-ADA activity can be an effective laboratory marker for early differential diagnosis of TMB with the cut-off value of 9 U/L. Dynamic changes of CSF-ADA activity may be a indicator for the effect of antitubercular treatment.
10.Phenolic acid derivatives from Alchornea trewioides.

To study the chemical constituents of Alchornea trewioides, silica gel column chromatography, Sephadex LH-20, reverse phase ODS column chromatography, MCI and semi-preparative HPLC were used to separate the 95% EtOH extract of the root of Alchornea trewioides. The structures were elucidated on the basis of spectroscopic studies including ESI-TOF-MS, 1H NMR, 13C NMR, HSQC and HMBC. Eight phenolic acids were obtained and identified as 1-O-galloyl-6-O-vanilloyl-beta-glucose (1), gallic acid (2), ethyl gallate (3), syringic acid (4), glucosyringic acid (5), erigeside C (6), 3, 4-dimethoxyphenyl-(6'-O-alpha-L-rhamnosyl)-beta-D-glucopyranoside (7) and 3, 4, 5-trimethoxyphenyl-(6'-O-galloyl)-O-beta-D-glucopyranoside (8). Among them, compound 1 is a new compound, compounds 4-8 are isolated from the genus Alchornea for the first time, and the others are isolated from the plant for the first time.
