Abstract Objective:FSM-2117 and FS-5416 are two bivalent hybrid strains of S.flexneri and S.sonnei, constructed by this laboratory-The FS-5416 expresses four invassive plasmid antigens(IpaA、IpaB、IpaC and IpaD) and has a good contact heamolysis activity(CHA+ ), butFSM-2117 hasn' t. To observe the immunogenicity of Shigelk vaccines through three different mucosal administration rout, the changes of ASCin the spleen and Peyer's patch(PP) , mesenteric lymph nodes(MLN) lymphocytes are detected.Methods:BALB/c mice were divided intothree groups, 20 mice per group. Mice were immunized respectively with the Ipa+ or Ipa- vaccines(4 x 10~7 CFU) three times with an intervalof two weeks by intranasal、intragastric or intraintestinal administration routs. The spleen , PP , MLN lymphocytes were isolated of seventh dayat random after immunization. An BA-EIISASPOT was done to account the numbers of ASC.Results:The numbers of SIgA and SIgG ASC ofspleen , PP , MLN lymphocytes of intranasal and intraintestinal group were significantly increased . Significant difference were only observed inthe number of spleen lymphocytes SIgA and SIgG ASC of intranasal group between Ipa+ and Ipa- . Conclusion:Two bivalent Shigelk vaccinescan induce spleen and GALT immunity reaction by nasal or small intestinal mucosal in a low dosage compared with intragastric rout (about 1/20). Ipa can significantly increased the number of spleen lymphocytes SIgA and SIgG ASC of intranasal group.
3.Vidian canal as a landmark to endoscopic nasal skull base surgery

OBJECTIVE To analyze whether the vidian canal can be served as a landmark to endoscopic nasal skull base surgery and provide clinical reference by imaging and anatomical methods. METHODS Twenty three cadaveric heads were scanned through axial and coronal high resolution CT to observe the vidian canal and its relationship with around structures.Three cadaveric heads, in which arteries were injected with latex,were dissected by endoscopic transnasal or transmaxillary to pterygopalatine fossa and cavernous sinus approaches.RESULTS 1.The course and shape of vidian canal and its relationship with around structures can be clearly detected through high resolution CT.All the vidian canals were on the same level or below the internal carotid canal.2.Anatomic study corresponded to the results of imaging.Vidian content pointed to the anterior genu of internal carotid artery constantly.The vidian canal and foramen rotundum can serve as a surgical corridor.CONCLUSION As the landmark, vidian canal plays an important role in the endoscopic nasal skull base surgery.Being a bony canal,it can be detected through high resolution CT easily and clearly.

5.Relationship of chronic central pain and substance P in spinal dorsal horn after spinal cord injury

Objective To approach the neurobiochemical mechanism of chronic central pain (CCP) after spinal cord injury (SCI). Methods 28 SD rats were divided into four groups, the normal group (group A), the pseudosurgery group (group B), and groups with CCP (group C) and without CCP (group D) after L1 spinal cord section injured with WADE method. T13 and L2 segments of rats' spinal cord were took and concentration changes of substance P (SP) in the spinal dorsal horn between two sections were examined by immunofluorescence histochemistry staining combined with confocal laser scanning microscope. Results Concentration of SP in the group D was decreased significantly compared with groups C,A and B (P<0.05-0.01), that of the group C was less than that of group A and B (P<0.05). Conclusion The rat model established by WADE method is proper to study CCP after SCI. SP in dorsal horn of spinal cord may inhibit the CCP after SCI in some degrees.
6.Effect of Modified Hangqi Chifeng Decoction Containing Serum on the Expression of Col IV, MMP-2, and TIMP-2 in Glomerular Mesangial Cells Induced by LPS.

OBJECTIVETo explore the effect of Modified Hangqi Chifeng Decoction (MHCD) on levels of collagen type IV (Col IV), matrix metalloproteinase-2 (MMP-2), tissue inhibitor of metalloproteinase-2 (TIMP-2) in extracellular matrix (ECM) of glomerular mesangial cells (GMCs) in LPS induced mice.
METHODSNormal serum and telmisartan, high, medium, low dose MHCD containing serums were prepared by using serum pharmacology method. GMCs were cultured in vitro. The proliferation of mesangial cells were induced using LPS as stimulating factor. GMCs were divided into six groups, i.e., the normal group, the model group, the telmisartan group, high, medium and low dose MHCD groups. Col IV content in the supernatant of mesangial cells was detected using ELISA. Protein expressions of MMP-2 and TIMP-2 were detected using Western blot.
RESULTSCompared with the normal group, Col IV content obviously increased in the model group after 72-h LPS stimulation; protein expressions of MMP-2 and TIMP-2 were obviously up-regulated, and MMP-2/TIMP-2 ratio was down-regulated in the model group (P < 0.01). Compared with the model group, Col IV content obviously decreased in high and medium dose MHCD groups and the telmisartan group (P < 0.01); protein expressions of MMP-2 were obviously down-regulated in medium and low dose MHCD groups (P < 0.01, P < 0.05); the protein expression of TIMP-2 was obviously down-regulated in high, medium, low dose MHCD groups and the telmisartan group (P < 0.01). The pro- tein expression of TIMP-2 was obviously lower in the high dose MHCD group than in the low dose MHCD group (P < 0.01). MMP-2/TIMP-2 ratio was obviously up-regulated in the telmisartan group, high and medium dose MHCD groups (P < 0.01).
CONCLUSIONMHCD could regulate disordered MMP-2/TIMP-2 ratio in LPS induced ECM, inhibit excessive production of Col IV in ECM, promote the degradation of ECM, reduce the accumulation of ECM, thereby, delaying the process of glomerular sclerosis.

7.Role of mast cell and its subpopulations in simple and irradiated-wound of rats

AIM:To examine the role and dynamic changes of mast cell(MC) and its subpopulations in simple and irradiated-wound of rats.METHODS:MC and its subpopulations were estimated using alcian blue-safranin (ABS)double staining RESULTS:(1)The total number of MC in two groups decreased coincidently on day 2 after wounding, and the total MC increased rapidly and reached maximal gradually on day 5 and 7 after wounding, the increment of MC remained consistently 28 day after wounding (2)Both mucosal MC( MMC) and Mix MC decreased obviously on day 2 after wounding, hereafter,they remained the low level all the time However, the CTMC kept in the high level after wounding (3)The Mix MC on day 5 and the total MC during day 5-15 after wounding were lower in irradiated group than in simple wound group CONCLUSION: MC and its subpopulations could delay the healing process of simple and irradiated wound
8.CT in diagnosis of infantile hepatic hemangioendothelioma

Objective To explore the CT features of infantile hepatic hemangioendothelioma (IHH).Methods CT imaging features of 16 IHH patients confirmed by surgical pathology and clinical diagnosis were retrospectively reviewed.Results Of 16 patients,there were single lesion in 10 cases and multiple lesions in 6 cases.All the lesions were rich in blood supply,and the arterial phase showed obviously rings enhanced.Some linear enhancement towards to the center below the annular enhancement of solitary lesions were found.There were patchy enhancement in annular enhancement center area,and the area of enhancement obvious increasing in venous phase and delayed phase in 4 cases with multiple lesions,enhancement mode was similar to single lesion in 1 case with multiple lesions.The lesions of IHH Ⅱ were small and multiple,and showed intermittent line shaped edge enhancement;some lesions below the linear enhancement showed small nodular enhancement and the area of nodular enhancement enlarged in venous phase,the range of linear enhancement was not obvious.Conclusion CT manifestations of IHH have some characteristics,and the enhancement of CT is more important in the diagnosis of this disease.

A colistin producing strain Paenibacillus polymyxa AS1.541 was treated by N-methyl-N-nitro-N-nitrosoguanidine(NTG) for increasing yields of the antibiotic colistin.High-yield strains were obtained by selection of deregulated mutant which grow on media containing colistin,a self second metabolite,and ethionine,an analogue of methionine.Some of these mutants have higher yield of colistin than that of the parent strain.
10.Expression and correlation of ABCG2 and V-ATPase in NSCLC

Objective To investigate the expressions of ABCG2 and V-ATPase in NSCLC and their expression rates in pathological classification, TNM stages and pathological grades and the expression correlation between ABCG2 and V-ATPase. Methods Expressions of ABCG2 and V-ATPase were accessed with EnVinsion immunohistochemistry in tumor samples from 92 NSCLC patients. The corresponding data was analyzed statistically. Results Expressions of ABCG2 and V -ATPase were found both in the lung adenocarcinoma and lung squamous cell cancer, and the difference between these two kinds of tumors was significant (P =0.003,0.000). ABCG2 expression was significantly different among TNM stages of lung adenocarcinoma (P=0.004) as well as among pathological grades of lung adenocarcinoma (P =0.028) and squamous cell carcinoma (P =0.000), while no significant difference was found among TNM stages of squamous cell lung carcinoma. The level of V-ATPase expression was associated with TNM stages of lung adenocarcinoma (P =0.026) and pathological grades of lung squamous cell carcinoma (P =0.002), however, among TNM stages of lung squamous cell carcinoma and pathological grades of lung adenocarcinoma, the difference was not significant. Additionally, the significant correlation was found between expression of ABCG2 and V-ATPase in all samples, adenocarcinoma and squamous cell carcinoma (P<0.001). Conclusion The significant correlation is found between expression of ABCG2 and V-ATPase, which indicate that they may co-work to participate in the mechanism of anticancer drug resistance.
