1.Effects of inoculating Nogo66 vaccine on immunoresponse of rats

Objective To study immunoresponse of SD rats to different dosages of Nogo66 vaccine,and then select the best vaccination concentration.Methods Nogo-66 peptide vaccine plus complete and imcomplete Freund's adjuvant were prepared in concentrations of 50?g/300?l,100?g/300?l,200?g/300?l,400?g/300?l and 800?g/300?l.The vaccines of different concentrations were used to immunize SD rats weekly for 8 weeks,and either complete or imcomplete Freund's adjuvant alone was used as control.Blood was drawn from caudal vein of rats weekly after the first immunization,and the titer of serum special IgG antibody was measured by ELISA.Two animals of every group were sacrificed every four weeks,and the spleens were harvested,and erythrocytes were removed with hypotonic saline.The proliferation rate of the cultured spleen T cells was determined with MTT.Meanwhile the experimental autoimmune encephalomyelitis(EAE) was evaluated for all experimental animals.Results The antibody titer of serum specific IgG of all experimental groups elevated quickly since the 4th week after vaccination,and reached the top value at the 8th week,and it then kept stable for 10 weeks.The IgG antibody titer in 200?g/300?l group was significant higher than that in other experimental groups(P
2.Efficacy and safety of anterior chamber paracentesis in treatment of acute primary angle-closure glaucoma:report of 327 cases

Objective To study the safety and effectiveness of anterior chamber paracentesis,combined with antiglaucomatous medications,in the intraocular pressure control and relief of symptoms of acute primary angle-closure glaucoma(PACG).Methods Totally 327 patients with first attack of acute PACG who were admitted in our hospital during July 2000 and October 2006 were collected in this study,and were divided into 2 groups according to whether receiving anterior chamber paracentesis or not(paracentesis group and control).Their clinical data such as sex,age,intraocular pressure(IOP) and visual acuity before hospitalization,types and times of administration of IOP drugs before and after operation,preparative time for operation and postoperative complications were retrospectively analyzed.Results There were 94 out of 327 patients receiving anterior chamber paracentesis,who had faster intraocular pressure control(2.37?1.63 d vs 2.75?2.01 d),and lesser types and dosages and shorter times of IOP drugs,shorter therapeutic time after paracentesis,shorter time to prepare operation,lesser postoperative complications [shallow anterior chamber,12 cases(12.77%) vs 57 cases(24.46%)] and better visual acuity than control.Anterior chamber paracentesis more than one time didn't increase the risk of complications.Conclusion Anterior chamber paracentesis is safe and effective in controlling the intraocular pressure and eliminating symptoms in acute PACG.
3.Advances in the studies on cytokine and chemokine gene polymorphisms associated with uveitis

·Uveitis is an inflammation of any or all parts of the uveal tract including the iris, ciliary body and the choroid. Despite current advances in diagnosis and management, visual loss occurs in 35%-45% of patients with uveitis. The etiopathogenesis of uveitis remains unknown; it may be associated with environmental and immunogenetic factors. Many studies have demonstrated polymorphisms in major histocompatibility complex (MHC) genes may determine involvement in uveitis. Recently polymor-phisms in non-MHC genes, including cytokine and che-mokine genes, have been reported to play important roles in the pathogenesis of uveitis. This article reviewed the advances in the studies on cytokine and chemokine gene polymorphisms associated with uveitis.
4.Clinical investigation on the misdiagnosis of corneal endotheliitis

· AIM: To analyze the pathogeny of corneal endotheliitis and to investigate the causes of its misdiagnosis and the key to treatment.endotheliitis presented with predisposing factors, clinical manifestations and treatment were analyzed retrospectively.junctiviis, and the other 7 were misdiagnosed as acute close-angle glaucoma, glaucomatocyclitic crisis, inflammatory reaction after cataract extracapsular extraction and IOL implantation, disciform stromal keratitis and iridocyclitis, respectively. All the 7 cases were cured through treatment, and recurrence appeared in 1 case.unclear yet. Properly grasping its features and key points for differential diagnosis and seriously and detailed examining patients will benefit to timely diagnosis. After clinical treatment, maintaining treatment for more than 4wk will lower the recurrence of corneal endotheliitis.
5.Clinical experience of glaucomatocyclitic crisis

AIM: To observe the clinical features of glaucomatocyclitic crisis and to investigate its pathogenesis, diagnosis,treatment and prognosis.METHODS: The clinical data of 15 cases (15 eyes) subjected to glaucomatocyclitic crisis were retrospectively analyzed including clinical manifestation and treatment.RESULTS: Visual field was normal in 13 eyes, and 2eyes had glaucomatous visual field defects. C/D ratio was ≤0.3 in 10 eyes and ≥0.6 in 2 eyes. Through pharmacotherapy the symptoms in 14 eyes disappeared after 2wk. One eye was diagnosed as being complicated by primary open-angle glaucoma and well treated with surgery.CONCLUSION: Glaucomatocyclitic crisis is a kind of ocular syndrome with unknown pathogenesis. It should be differentiated from primary glaucoma and uveitis clinically. Its prognosis is not absolutely benign, and the patients should be followed up regularly.

7.Clinical analysis of 35 cases of craniocerebral injury patients with intracranial infection *

Objective To investigate the characteristics of pathogenic bacteria/the drug resistance/the correlated risk fators/the prophylaxis control strategy of the severe craniocerebral injury patients combined with intracranial infection .Methods The clinical data of 35 craniocerebral injury patients with intracranial infection were retrospectively analyzed .Results 35 patients′cerebrospinal fluid were separated and 54 pathogenic bacterium had been cultured ,including G+ bacterium(61 .11% ) ,the G - bacteria(33 .33% ) , fungi(5 .56% ) .The pathogenic bacteria showed a higher resistance .The single factor analysis found that the wound itself exists in-fection factors ,the postoperative drainage of incision ,liquorrhoea ,with other basic diseases ,surgery lasted for a long time (>4 h) reoperative ,surgery is placed foreign body is severe craniocerebral trauma combined with intracranial infection were the main rele-vant factors .The total effective rate was 62 .86% ,and the mortality was 11 .43% by the positive therapy .Conclusion G+ bacteria were the main pathogenic bacterium in the severe craniocerebral injury patients combined with intracranial infection .The iatrogenic factors leaded to the increase of the proportion of intracranial infection and the resistance increased year after year .The clinical in-travenous antibiotics combined intrathecal injections were beneficial to control intracranial infection ,shorten the course of treatment and enhance the curative effect .
8.Effects of intrathecal transplantation of bone marrow stromal cells on intercellular cell adhesion molecule-1 expression and blood spinal cord barrier following spinal cord ischemia reperfusion injury

Objective To investigate the effects of intrathecal transplantation of bone marrow stromal cells (BMSCs) on inter cellular cell adhesion molecule-1 (ICAM-1) expression and blood spinal cord barrier following spinal cord ischemia reperfusion injury.Methods Ninety Sprague Dawley rats were randomly divided into three groups:sham (Sham group),ischemia-reperfusion injury (I/ R group),and BMSCs transplantation (BMSCs group).Spinal I/R injury was induced by clamping the aortic arch between left common carotid artery and left subclavian artery for 14 min in I/R group and BMSCs group.Sham group was subjected to exposure of aortic arch but without occlusion.I/R group and BMSCs group were intrathecally injected with phosphate buffered saline (PBS) or BMSCs (2 × 106) two days before injury.At 1 d,3 d,and 7 d after injury,neurological function was evaluated and damaged lumbosacral seg ment was removed for measurement of blood spinal cord barrier permeability and ICAM-1 protein expression.Results Compared with Sham group,neurological function score was significantly lower:1 d (F =38:59,P =0.001),3 d (F =31.34,P =0.001),and 7 d (F =27.71,P =0.001) ; ICAM-1 expression was increased 1 d (F =34.33,P =0.001),3 d (F =29.76,P =0.001),and 7 d (F =23.65,P =0.001),and blood spinal cord barrier permeability was higher:1 d (F =42.57,P =0.001),3 d (F =32.75,P =0.001),and 7 d (F =26.89,P =0.001) in I/R group.Compared with I/R group,neurological function score was increased:1 d (F =16.62,P =0.001),3 d (F =21.54,P =0.001),and 7 d (F =12.84,P =0.002) ; ICAM-1 expression was decreased:1 d (F =19.84,P =0.018),3 d (F =17.38,P =0.008),and 7 d (F =22.46,P =0.007),and blood spinal cord barrier permeability was lower:1 d (F =22.38,P =0.016),3 d (F =27.59,P =0.009),and 7 d (F =23.25,P =0.001) in BMSCs group.Conclusions Intrathecal transplantation of BMSCs inhibited ICAM-1 expression and decreased blood spinal cord barrier permeability,and then attenuated spinal cord ischemia-reperfusion injury.
9.Expression of bone morphogenetic protein 7 and its potential role in diabetic rats

Objective To observe the expression of bone morphogenetic protein 7 (BMP-7) in different stages of diabetic rats and investigate the potential role of BMP-7 in diabetic nephropathy (DN). Methods Wistar rats were divided into diabetes mellitus(DM) group induced by intravenous injection of streptozotocin (65 mg/kg) , and normal control group injected with citrate buffer. The rats were sacrificed at day 30,60,90,120,150 and 180 following injection, respectively. The blood glucose,Scr, urinary albumin and urinary creatinine were measured in each rat before sacrifice. The renal pathological changes were examined with hematoxylin and eosin (HE),periodic acid-silver-methenamine staining (PASM) and protein expression of collagenⅣ(ColⅣ). The mRNA and protein expression of BMP-7 and TGF-?1 in kidney were detected by reverse transcription-polymerase chain reaction (RT-RCR) and immunohistochemical staining respectively, and were quantified by computer image analysis system. 10.A new method to solve anophthalmic contracted socket in embedding orbital implant in 114 cases

AIM:To perform a new method for orbital implant and contracted socket through one time and its results. ·METHODS:Totally 114 patients 114 eyes, from January 2008 to June 2014, with contracted socket participated in this study. We incised the bulbar conjunctiva horizontally and excised scar tissue, then implanted the hydroxyapatite in the four extraocular muscles and tightly sutured the Tenon ' capsule. After that, we put the superior and inferior conjunctival petals backwards and sutured them to the Tenon ' s capsule. All the patients were divided into four groups according to the vertical diameter length of the conjunctival defect area:GroupⅠ:≤5mm; Group Ⅱ: 6-10mm; Group Ⅲ: 11-15mm; and Group Ⅳ: ≥16mm. These patients were followed up for 6mo to 3y to observe the conjunctival sac shaping and growth of conjunctiva. ·RESULTS:There were 64 cases in GroupⅠ, 31 cases in Group Ⅱ, 16 cases in Group Ⅲ and 3 cases in Group Ⅳ. All patients ' conjunctival defect was covered by new conjunctiva and scar tissue 4 to 6wk after surgeries. Ten cases had contracted socket; 2 cases had orbital implant exposure, requiring reoperation. Of the 114 cases, 8 had contracted socket and could use a smaller conformer, 106 could use a normal size conformer. ·CONCLUSION: When the conjunctival defect was ≤15mm, this new method can address the orbital implant and contracted socket at the same time. While it was ≥16mm, flap transplantation is necessary.
