5.Peer Acceptance and Interpretation to Peer Rejection in Adolescence

Objective:This study investigated 376 adolescents' interpretations to peer rejection events. Methods:Measuring 376 adolescents' degrees of peer acceptance and their interpretations to peer rejection events by means of pen and paper questionnaires.Results:Peer-rejected adolescents presented a similar way of general interpretation to rejected events happened in other peers,however they interpret their own rejected events in a completely different way compared with popular adolescents:The main effects of peer acceptance types were significant on objective and subjective question scores(F(3,344)=3.212,P=0.023;F(3,344)=4.059,P=0.007, respectively);and there was a significant interaction effect of sex?age?peer acceptance types (F(9,344)=3.447, P

7.Intervention of siRNA to the effect of gelatinase on the expression of ICAM-1 in human renal tubular epithelial cells

Objective To inhibit the expression of matrix metalloproteinase-2(MMP-2) and matrix metalloproteinase-9(MMP-9) in human kidney tubular epithelial cell(HKC) with specific small interference RNA(siRNA),then to observe whether the expression of intercellular adhesion molecule-1(ICAM-1) on HKC could be influenced.Methods HKCs were transfected with MMP-2,MMP-9 siRNA or irrespective siRNA respectively,untransfected HKC was considered as blank control.Four groups of HKCs were stimulated with 100nmol/L phorbol myristate acetate(PMA) for 18h.Total RNA or protein of cells were extracted with Trizol and RIPA cell lysate respectively,the mRNA expression levels of MMP-2,MMP-9 and ICAM-1 were detected by real time PCR,the protein expression levels of above indexes were detected by western blotting and immunofluorescence combined with confocal laser microscopy.Results The specific siRNA could inhibit the expression of MMP-2 and MMP-9 in HKCs effectively.Immunofluorescence combined with confocal assay demonstrated that while HKCs was stimulated with PMA,the expression of ICAM-1 was up-regulated,which was significantly up-regulated in the group transfected with MMP-9 siRNA(P0.05).Conclusions The inhibitory expression of MMP-9 of HKC may up-regulate the expression of ICAM-1,implying that,besides taking part in the degradation of extracellular matrix,MMP-9 may throw an influence on the proceeding of renal fibrosis by inflammatory pathway.
8.New development in the research on FoxO3a and urologic neoplasm

The forkhead box O (FoxO) transcription factor family plays an important role in cell functions, including metabo-lism, apoptosis, cellular proliferation, stress reactions, DNA repair, and immune response. As a member of this family, forkhead box O3a (FoxO3a) regulates its target genes by modulating histone modifications, including phosphorylation, acetylation, and methylation. FoxO3a expression is abnormally downregulated in urologic neoplasm. Protein modifications and FoxO3a activity are mainly con-trolled by PI3K/Akt signal pathway and other signaling pathways. FoxO3a is also involved in the initiation, progression, and prognosis of urologic neoplasm. This review focuses on the function of FoxO3a in urologic neoplasm and elucidates the regulatory mechanisms involved. This article will provide novel strategies to clinical diagnosis and drug therapy of urologic neoplasms.
10.Relationship between the rehabilitative intervention of ischemic cerebrovascular diseases and the functional alteration of monomer polymerization of fibrinogen

BACKGROUND: The elevation of plasma fibrinogen(Fbg) is a key risk factor of cerebrovascular diseases. The evaluation of the monomer polymerization function of fibrin has even more important clinical merit than the detection of Fbg level.OBJECTIVE: To investigate the clinical significance of the monomer polymerization function of fibrin in patients with isehemie cerebrovascular diseases and its impacts on rehabilitative intervention.DESIGN: A case control study employing patients and healthy individual as subjects.SETTING: An Institute of Hematology and Department of Neurology of one university.PARTICIPANTS: Totally 110 patients with different ischemic cerebrovascular disease selected from the Department of Neurology, Union Hospital of Tongji Medical College, Huazhong Science and Technology University from September 2001 to March 2002, and 50 healthy individuals were included in the study.METHODS: Rehabilitative intervention was performed in 31 randomly selected cerebral infarct patients, and the parameters indicating the monomer polymerization functions of fibrin in the plasma were detected by the measurement system for the monomer polymerization function of fibrin.MAIN OUTCOME MEASURES: Abnormal condition of monomer polymerization function of fibrin in each parameter.RESULTS: Each parameter indicating the monomer polymerization functions of fibrin in plasma was significantly increased in ischemic cerebrovascular diseases patients than healthy individuals( P < 0.01 ) . The abnormal rate of Fbg leveland fibrin monomer polymerization velocity (FMPV) was significantly elevated in ischemic cerebrovascular disease patients than healthy individuals ( P < 0. 01 ) . The relative risk(RR) of ischemic cerebrovascular diseases in patients with abnormal FMP functions was 4 to 31 times more than healthy control group. In cerebral infarct group, FMPV of anterior circulation infarct subgroup was significantly elevated than that of posterior circulation infarct and lacunar cerebral infarct subgroups( P < 0.05). The FMP function of anterior cerebral infarct patients was significantly higher than that of healthy group before rehabilitative intervention. Although each FMP parameter reduced after rehabilitative intervention, the difference between was not significant compared with that of before therapy.CONCLUSION: FMP function analysis can completely and objectively reflect the coagulation status of the patients with ischemic cerebrovascular diseases, and it can also reflect the range and severity of infarct to some extent. Although common rehabilitative intervention cannot effectively improve the high-coagulation of the blood, the impacts of specific rehabilitative intervention on the coagulation mechanism deserve further investigation.
