AIM To investigate the effect of aspirin on the proliferation and NOS expression in astrocytoma cell line, and probe into its mechanism. METHOD The effect of aspirin on the growth of astrocytoma cells was evaluated by MTT assay; NOS protein levels were determined by immunocytochemistry, NO and CEA concentration in the medium were determined by Griess assay and lepton catch immunising method respectively. RESULTS Aspirin inhibited the growth of astrocytoma cells, induced the expression of iNOS, increased the concentration of NO in the medium. The effects of these were centratoin dependent. Moreover, aspirin reduced the concentration of CEA in the medium. CONCLUSION Aspirin inhibits the growth of astrocytoma cell line. Up regulated iNOS expression resulting a increase of NO concentration are ascribed to mechanism of antiproliferation activity of aspirin. CEA is a good indicator in monitoring curative effect of astrocytoma.

Aim To probe the effect of bambuterol on experimental asthma and its pharmacodynamic and pharmacokinetic chracteristics,as well as its mechanism. Methods Experimental asthma model on guinea pigs was induced by histamine and ovalbumin in vivo and their trachea flake and pulmonary bar were emptied to the research in vitro respectively. Results Bambuterol inhibited asthma induced by histamine and ovalbumin in guinea pigs in a dose-dependent manner. Bambuterol gave no relaxation to all trachea flake, but the intragastric gavage(ig) of plasma of the bambuterol-treated guinea pigs relaxed trachea flake and pulmonary bar,and the effect on pulmonary bar was stronger than that on trachea flake. The peak value appeared about 4 h after administration, and the action continued for more than 24 h. Conclusion Bambuterol as pro-drug of terbutaline metabolized in body has a mild and permanent effect on the model of experimental asthma in guinea pigs.

AIMTo study the inhibitory effect and mechanism of nobiletin on Heps tumor bearing mice.
METHODSModels of Heps tumor bearing mice were established. The inhibitory rates of tumor growth were calculated, the apoptosis morphology of tumor tissue was observed. The T lymphocyte transformation capacity was tested by MTT assay, the TNFalpha and IL-2 production were measured by LDH kits.
RESULTSNobiletin could significantly inhibit Heps tumor growth. The inhibitory rates were 42.14% - 65.09% (P < 0.01). The morphology of tumor tissues in nobiletin group had typical characters of necrosis and apoptosis through transmission electron microscope. Nobiletin could stimulate T lymphocyte transformation and the production of TNFalpha and IL-2.
CONCLUSIONNobiletin has obvious antitumor effect on Heps, the main mechanism is to enhance the cellular immune function and induce apoptosis of tumor tissue.

OBJECTIVETo isolate and purify corynoline and acetylcorynoline from Corydalis bungeana and develop a reversed-phase HPLC method of determining the two components in C. bungeana.
METHODAlkaloids were isolated from the ethanolic extract with column gel chromatography, and identified on the basis of spectral analysis (UV, 1H-NMR, 13C-NMR) and physicochemical properties. For quantitative analysis of the two components, samples were separated on an ODS column with mobile phase of methanol-15 mmol.L-1 potassium dihydrogen phosphate/potassium phosphate dibasic (pH 6.70, 70:30). The flow rate was 0.8 mL.min-1, and the detection was set at 289 nm.
RESULTThe purity was 99.5% and 99.1% for corynoline and acetylcorynoline respectively. The calibration curves were linear in the range of 6.9-110.4 mg.L-1 corynoline and 8.7-139.5 mg.L-1 acetylcorynoline. The RSD was 2.1% and 2.7%, and the average recovery was 97.3% and 97.2% respectively.
CONCLUSIONThe method of isolating and purifying corynoline and acetylcorynoline from Corydalis bungeana and the HPLC method of simultaneous determination of the two components have been developed. The HPLC method is simple, easy to perform and applicable to the content determination of corynoline and acetylcorynoline in C. bungeana of various origins.

OBJECTIVETo explore effects and possible mechanisms of curcumin on hypoxia induced epithelial-mesenchymal transition (EMT) in hepatocellular carcinoma cell line HepG2.
METHODSHepG2 cells were divided to 3 groups, i.e., the normal control group, the CoCl2 group, and the CoCl2 plus 10 micromol/L curcumin group. The proliferation of HepG2 was determined using MTT assay. The migration of HepG2 was detected by wound healing assay. The mRNA expression of hypoxia-inducible factor-1 (HIF-1alpha) was evaluated with real-time RT-PCR. The protein expressions of HIF-1alpha, epithelial-cadherin (E-cadherin), and vimentin were determined using Western blot.
RESULTSCompared with the normal control group, the proliferation and migration of HepG2 cells under CoCl2-induced hypoxia significantly increased, the expression of HIF-1alpha was up-regulated, and the expression of E-cadherin protein was obviously down-regulated, and the expression of vimentin significantly increased (all P < 0.05). Intervention by curcumin significantly inhibited the proliferation and migration of hypoxic HepG2 cells, and expressions of HIF-1alpha and vimentin decreased, and the expression of E-cadherin was up-regulated, showing statistical difference when compared with those of the CoCl2 group (P < 0.05). There was no statistical difference in HIF-1alpha mRNA expression among the 3 groups (P > 0.05).
CONCLUSIONCurcumin could reverse the proliferation and migration of HepG2 cells under CoCl2-induced hypoxia condition, which might be associated with inhibiting up-regulated expressions of HIF-1alpha protein and EMT.

Objective To observe the clinical efficacy of warm needling plus Chinese medication for external application in treating post-stroke shoulder pain. Method Two hundred patients with post-stroke shoulder pain were randomized into a treatment group and a control group, 100 cases each. The two groups both received rehabilitation training for shoulder joint. In addition, the treatment group was given warm needling plus Chinese medication for external application, while the control group was given warm needling. Visual Analogue Scale (VAS) for pain, upper-limb Fugl-Mayer Assessment (FMA) and Modified Barthel Index (MBI) were adopted to evaluate the two groups before and after the treatment. The clinical efficacies of the two groups were also compared. Result The total effective rate was 100.0% in the treatment group versus 87.0% in the control group, and the between-group difference was statistically significant (P<0.01). The VAS, FMA and MBI scores were significantly changed after the treatment in both groups (P<0.01). After the treatment, the VAS, FMA and MBI scores of the treatment group were significantly different from those of the control group (P<0.05, P<0.01). Conclusion Warm needling plus Chinese medication for external application and rehabilitation training can obviously reduce post-stroke shoulder pain, and enhance the upper-limb motor function and activities of daily living.
