1.Association between single nucleotide polymorphisms of BRCA1-A complex genes and susceptibility ;of triple-negative breast cancer

Background and purpose:The mutation of BRCA1 gene is widely acknowledged to be related to the incidence of triple-negative breast cancer (TNBC). The aim of this study was to investigate the association between TNBC and single nucleotide polymorphisms (SNPs) of BRCA1-associated genes. Methods:This study investigated the associations between the BRCA1-A complex genes and risk of developing TNBC in a case-control study of Chinese Han Women population including 414 patients with TNBC and 354 cancer-free controls diagnosed in the Fudan University Shanghai Cancer Center during 2008-2011. This study also detected 37 common variants in Abraxas, BRE, Rap80, NBA1 and BRCC36 genes encoding the BRCA1-A complex and evaluated their genetic susceptibility to the risk of TNBC. An additional cohort with 652 other types of breast cancer (non-TNBC) cases and 890 controls were used to investigate the associations between TNBC-speciifc SNPs genotype and non-TNBCs susceptibility. Results:This study found that rs7250266 in the promoter region of NBA1 confers a decreased risk to TNBC (P<0.01). Compared with CC genotype, women with the GC genotype (OR=0.70, 95%CI:0.51-0.97) and GG genotype (OR=0.48, 95%CI:0.21-1.07) had a lower risk of developing TNBC (P=0.03). In addition, the haplotypes containing two polymorphisms rs7250266 and rs2278256 were associated with a lower chance of TNBC development. In the second part of the study, the result showed that there was no difference in rs7250266 expression between non-TNBC and normal people (0.19 vs 0.18, P=0.85).Conclusion:Genetic variants in NBA1 may be an important genetic determinant of TNBC susceptibility in Chinese women.

The research of interleukin-13 receptor ?2(IL-13R?2) chain is a rising pop these years.Previous studies have shown that many human tumors overexpress IL-13R?2 chain,while normal cells do not express this receptor or express very low level.This difference of express level is significant to diagnose and cure tumors by the IL-13R?2-directed toxin fusion protein.During the past decade,the structure and the function of IL-13R?2 together with the relationship between this receptor and tumors has been further developed.Therefore new therapies and theories can be proposed as the clinical tumor treatments.In this case,the expression in various tumor cell lines was not only focused on but also on the IL-13 and toxin fusion protein killing effect in both the cell level and the in vivo level.Besides,an overview of the mechanism in the treatment of IL-13R?2-directed toxin fusion protein together with the improved methods for fusion protein purification and other relative tumor therapy was given.In conclusion,the current status and progress of IL-13R?2 as well as IL-13R?2-directed toxin fusion protein in tumor therapy were represented.

5.Demands on the technology of prevention and treatment of hypertension and evaluation on training effect among doctors in county and countryside hospitals of Xinjiang

Objective To analyze the demands and to evaluate training effect on the knowledge and technology of prevention and treatment about hypertension among doctors in county and countryside hospitals of Xinjiang.Methods 336 doctors were trained about knowledge of prevention and treatment of hypertension and technology of useful therapeutic regimen,and were investigated for demands of the knowledge and technology and self-evaluation of training effect by questionnaire.Results The knowledge of prevention and treatment about hypertension and the technology of useful therapeutic regimen were needed urgently by 94.8% subjects.91.3% trained doctors thought that the technology suited practically in county and countryside hospitals,and 96.7% hospitals had condition of carrying out the technology.97.9% trained doctors expected this kind of technology on hypertension prevention and treatment to be trained again.74.1% trained doctors comprehended the technology,and the percent of doctor practically using the technology increased from pre-training 12.3% to post training 62.1%.Conclusion The knowledge of prevention and treatment about hypertension and technology of useful therapeutic regimen were needed urgently and were comprehended with training for the most doctors in county and countryside hospitals of Xinjiang.
6.Therapic effect of lauromacrogol sclerotherapy on infantile hemangioma

Objective To investigate the effect of intralesional lauromacrogol therapy on infantile hemangioma.Methods 30 cases all received intralesional injection of lauromacrogol.The changes of tumor size,texture and color were monitored and recorded.The adverse effects after medication were observed and managed accordingly.The adverse effects after medication were observed.The times of treatment were determined according to degree of reduction of tumor volume.To assess the efficacy,a 4 scales system was adopted.Results All patients had completed the treatment and followup.The overall response was scale Ⅰ in 2 cases (6.7 %),scale Ⅱ efficacy in 4 patients (13.3 %),scale Ⅲ efficacy in 5 cases (16.7 %) and scale Ⅳ in 19 cases (63.3 %).The smaller the tumor,the fewer the times of treatment,and the better the results.The effects of treatment were better if patient could be treated by intralesional lauromacrogol therapy in early stage.No severe adverse effects were observed during 6 months of follow-up.Conclusions Intralesional injection of lauromacrogol sclerotherapy is a safe and effective way to treat infantile hemangioma.
8.Observations on the Efficacy of Mallet Needle plus Acupuncture in Treating Stiff Neck

Objective To investigate the clinical efficacy of mallet needle plus acupuncture in treating stiff neck. Method Eighty patients with stiff neck were randomly allocated to treatment and control groups, 40 cases each. The treatment group received mallet needle therapy plus acupuncture and the control group, acupuncture alone, for three consecutive days. In the two groups, the Pain Rating Index (PRI) score and the Visual Analogue Scale (VAS) score were recorded before and after treatment and the clinical therapeutic effects were evaluated after treatment. Result The PRI score and the VAS score decreased in both groups after treatment compared with before (P<0.05) and decreased more in the treatment group than in the control group at the end of treatment (P<0.05). The total efficacy rate was 95.0% in the treatment group and 87.5% in the control group; there was a statistically significant difference between the two groups (P<0.05). Conclusion Mallet needle therapy plus acupuncture is more effective than acupuncture alone in treating stiff neck.
9.Detection of BRCA mutations for breast cancer in southern China

Objective To investigate the prevalence of BRCA1/2 mutations in breast cancer patients of Han ethnic group in southcrn China,and to provide the genetic basis for early diagnosis and treatment of breast cancer.Methods 70 samples of breast cancer patients of Han ethnic group in southern China and 70 normal control samples were tested.The whole coding exons of BRCA1 and BRCA2 genes were analyzed using hybridization based enrichment and next-generation sequencing (NGS).According to the results of NGS,we detected pathogenic mutations of BRCA genes.Results In 70 breast cancer patients,a total of 5 deleterious mutations were identified,4 of these were novel mutations,and no pathogenic variation was found in normal control group.Furthermore,we detected 3 low frequency mutations which are likely to relate to cancer susceptibility.Conclusions 4 novel deleterious mutations were reported,as well as some variants of unknown clinical significance for the prevalence of BRCA1/2 mutations of 70 breast cancer patients of Han ethnic group in southern China.
10.Significance of Serum Interleukin-6 in Attack or Stable Stage of Asthmatic Children

Objective To explore the role and clinical significance of interleukin-6(IL-6) in children with bronchial asthma.Methods Sera were collected from 29 cases with asthmatic attacks,32 asthmatic children who in stable conditions,and 20 health children.Serum IL-6 concentrations were measured by radioimmunoassay(RIA).Results 1.Asthmatic children appeared significantly higher levels of serum IL-6 during asthmatic attacks as compared to those in stable conditions and healthy ones respectively(P3 years old had significantly higher serum IL6 concentrations than ≤3 years old children in remission.Conclusion IL-6 may participate pathogenesis of asthma,and it may play different biological roles during asthmatic attacksas or stable conditions.
