Objective To investigate the effect of Doxycycline on gastric cancer cell line BGC-823 proliferation and Notch1, Matrix metalloproteinase-9(MMP-9)expression in order to shine a light on new anticancer drugs. Methods Final concentration of 0, 5, 10, 20, 40 mg/L doxycycline were added into human gastric cancer cell line BGC-823. Cell pro-liferation was detected by MTT;Cell cycle distribution was assessed by flow cytometry;Notch1, MMP-9 protein expression was revealed by Immunoblot. Results Doxycycline can inhibit proliferation of human gastric cancer cells line BGC-823, and its effect is dose and time-dependent(P＜0.01). Doxycycline alters distribution of gastric cancer cell line BGC-823. With increasing drug concentration, the proportion of cells in S phase dropped(P＜0.01). Notch1 expression rose and MMP-9 expression decreased(P＜0.01). Conclusion Doxycyclinecan inhibited gastric cancer cell line BGC-823 prolif-eration and up-regulating Notch1 might be one mechanisms.

The structure and characteristics of the soluble exopolysaccharide(PS-238) produced by Alcaligenes sp.NX-3 were studied.The analytic sample was prepared by ethanol precipitation then proteins were removed by the Sevag method.The components of polysaccharide were identified by TLC、 GC and chemical method after acid hydrolysis as: glucose,mannose,rhamnose and glucuronic acid.By means of IR and periodate oxidation,it showed that the polysaccharide was mainly consisting of(1→3)and(1→4) glucosidic linkages.The relation between viscosity and shear rate、concentration、temperature、pH and salts was investigated.The results showed that the solution of PS-238 is a typical pseudoplastic liquid,having high viscosity at low concentrations.The viscosity of 1%PS-238 water solution could reach 3300cp at 25℃.It has the excellent stability towards pH2~13,also has outstanding property of viscosity retention even at temperatures up to 150℃.Some kinds of salts were cooperated with PS-238 as a thickening agent.

Objective: To investigate the relationship between the gene polymorphisms of cholecystokinin (CCK) and cholecystokinin A receptor (CCKAR) and genetic susceptibilities of biliary tract cancer and cholelithiasis in a Chinese population from Shanghai. Methods: A population-based case-control study was carried out in 253 cases of gallbladder cancer, 133 cases of extrahepatic bile duct cancer, 53 cases of cancer of Vater's ampulla, 440 cases of biliary stones, and 445 population controls. The genotypings of CCK rs747455 and CCKAR rs1800856 were performed by Real-time PCR. Results: Non-stone cases carrying the CCK rs747455 CT genotype reduced the risk of ampulla cancer, compared with the carriers of CC genotype [odds ratio (OR)=0.31, 95% confidence interval (CI)=0.10-0.96], and this association was still significant after Bonferroni correction (P=0.042). The drinkers who carrying TT or CT genotype increased the risk of cholelithiasis, compared with the carriers of CC genotype (OR=2.81, 95% CI=1.08-7.29; OR=2.93, 95% CI=1.03-8.33), but this association was not significant after Bonferroni correction (P=0.061). The genotyping of CCKAR rs1800856 was not associated with the risk of biliary tract cancer and cholelithiasis. Conclusion: CCK rs74745 gene polymorphism may be correlated to the susceptibilities of biliary tract cancer and cholelithiasis in a Chinese population form Shanghai, China. Objective To evaluate the clinical effect of fluoride agents in prevention of caries in preschool 4 to 5 years old children.Methods 700 kindergartens 4 -5 years old children were selected to conduct prevention study with the agree of guardian.The children were randomly divided into observation group and control group. 350 preschool children in the control group received regular oral health measures and fluoride foam treatment every six months.350 preschool children in the observation group received fluoride products every six months on the basis of the control group.After two years study,the deciduous caries and caries rate of the two groups were analyzed.Results After prevention of dental caries 24 months,the prevalence of dental caries and caries of deciduous teeth surface in the observation group significantly reduced,and were significantly lower than the control group ( observation group:caries prevalence 11.3%,caries mean 0.85;control group:caries prevalence 36.1%,caries mean 1.77),the deciduous caries rate and the number of surface caries between the two groups had statistically significant differences (χ2 =4.339 1,3.016 8,all P<0.05).Conclusion Child deciduous dental caries in China is still at a relatively high inci-dence of disease,regular use of fluoride foam and fluoride products namely fluor protector can significantly prevent dental caries,oral health measures than on a regular basis is more effective,and significantly reduce deciduous caries among children,protection of children's oral health,and it is worth further clinical application.

Objective To study the pathological features of two huge spontaneous tumors in Wistar and GK rats. Methods Forty Wistar rats and 40 GK rats were included in this study. Among those rats, two huge spontaneous tumors were observed in a Wistar rat at 14 months of age and in a GK rat at 22 months of age. The growth and survival status of the tumor?bearing rats were recorded. The tumors were surgically removed, and their pathological features were examined using HE and immunohistochemical staining (vimentin, CK19, α?SMA, CD31, CD34, S?100, NF及Ki?67). Results Both the two tumors were completely resected by surgery without much difficulties, and both host rats survived well after the operation. The weight of the two huge tumors was 502 g and 119 g, which corresponding to 64% and 24% of the body weight of their host rats, respectively. The tumors surface had a complete capsule, with a clear boundary separating from the normal surrounding tissues, and no vascular pedicle structure was found. According to the results of immunohistochemical staining, both the two tumors were diagnosed as benign fibroma. Conclusion This type of huge spontaneous tumors is benign fibroma. Besides the impact on the activities of the rats, the tumors have no significant impact on the living conditions in the hosts.

Objective To explore the effect of fibroblast growth factor receptors 1-dominant negative strategy (FGFR1-DN) on alkaline phosphatase (ALP) activity of bone marrow stromal stem cells (BMSCs) after osteogenic induction.Methods BMSCs were transfected with eukaryotic expression plasmid pcDNA 3.1 (+)-DN FGFR1 and pcDNA3.1 (+)-FGFR1.The experiment was conducted in 4 groups:FGFR1-DN transfection group,FGFR1 transfection group,pcDNA3.1(+) empty vector transfection group and non-transfection group.The ALP activity of BMSCs was detected in logarithmic growth phase after osteogenic culture.The qualitative detection of ALP activity was carried out immunohistochemically while the quantitative detection by cALP kit.The ALP activity was compared between the 4 groups at 7 and 14 days after osteogenic induction.Results Compared with 7 days,the ALP activity at 14 days was significantly increased in the 4 groups,and the increase in FGFR1-DN transfection group was significantly higher than in the other 3 groups (P ＜ 0.05).At both 7 and 14 days,the ALP activity in FGFR1-DN transfection group was the highest while that in FGFR1 transfection group was the lowest (P ＜ 0.05).Conclusions FGFR1-DN can promote the ALP activity of BMSCs during osteogenesis.This may provide an experimental basis for the joint application of local gene therapy and tissue engineering and for construction of tissue engineered bone with better biocompatibility.

Objective To investigate the mental health status of middle school students in Three-Gorge area in Chongqing, and to explore the relationship with their life events and coping styles. Methods The Compi-ling of mental health scales for middle school students(MSSMHS),adolescent self-rating Life events checklist and trait coping style questionnaire(TCSQ) were adopted in the study to test the randomly sampled 1 632 middle school students in Three-Gorges area in Chongqing. Results The rate of mental disorder in the samples was 27.5%. The total average scores and each factor of the mental health were positively correlated with the factors of life events(r=0. 29～0.55). Negative coping styles were positively correlated with the total average scores and each factor of the mental health (r=0.38～0.56). Positive coping styles was negatively correlated with anxiety, depression, study pressure(r=-0.07～-0.05). The factors of life events and coping styles explained 27%～49% variation of scores of the mental health total average scores and each factor of the mental health. Conclusion Both life events and the coping styles are the important factors for the mental health of middle school students in Three-Gorge area in Chongqing. Reducing negative life events and coping styles benefits the mental health of middle school students.

Objective To evaluate the impact of human papillomavirus(HPV)infection on the risk of HIV acquisition in the female group.Methods Searched PubMed,EMbase,Coehrane Library,CNKI,wanfang database and Chinese Biomedical Lit-erature Database etc for articles about HIV-acquisition in HPV infected female patients.The quality of the literature were e-valuated according to standards of inclusion and exclusion.Data was extracted and methodologically quality evaluated by two independent investigators.Meta-analysis was accomplished using RevMan4.2 software.Results 6 articles were included,all of them were randomized controlled trials,a total of 9 606 cases studied.HIV infection risk was doubled in women with any HPV infection(OR= 2.02,95% CI:1.48 ~ 2.77),demonstrating High-risk HPV-positive (OR= 2.50,95% CI:1.73 ~3.61)and low-risk HPV-positive (OR=2.10,95% CI:1.48~2.96)respectively.Funnel plot analysis revealed no signifi-cant publication bias on HPV genotypes.Conclusion The analysis of selected research suggests the risk of HIV infection was increased in HPV-positive women.HPV vaccine may have some preventive effect on HIV infection.

Objective To evaluate the clinical esthetical effect in the cervical margin of the porcelain fused to metal crown restoration.Methods 225 cases with the porcelain fused to metal crown were observed.The evaluator examined these restorations for cervical color,marginal fit,and gingival state.(Results 53) cases of restoration changed in the cervical color.The method and the tooth preparation volume,the procedure and the material,which were all closely relevant to the cervical color change.(Conclusion Using)the correct method,improving the procedure and choosing the proper material would reduce the cervical color change.
