1.Expression of autophagy related factors LC3 and Bnip-3 and apoptosis related factors Bcl-2 and Bax in brain tissue of experimental diabetic rats

Objective To evaluate the effect of autophagy-related factors LC3 and Bnip-3 and apoptosis related factors Bcl-2 and Bax on brain damage in experimental diabetic rats .Methods Thirty SD rats were randomly divided into diabetic group and control group .The diabetic group was injected with 1% streptozotocin ( 60 mg/kg body mass ) and the control group with citrate buffer .The rats were sacrificed after 12 weeks feeding and brain tissues were obtained .Pathological chan-ges were observed and the expression of LC 3, Bnip-3, Bcl-2 and Bax in brain tissues of the rats was detected by immuno-histochemical SP method .Results Compared with the control group ,the diabetic rat brain pathology showed that the cell arrangment was more disorderly and distributed more unevenly , the cell body was smaller , cytoplasm was lighter red , and the number of nerve cells of normal morphology was smaller .The positive number of LC3, Bnip-3 and Bax in brain tissues of diabetic rats was significantly larger than in the control group ,and the difference was statistically significant (P<0.05). However ,the positive number of Bcl-2 was significantly smaller than in the control group ,and the difference was statistically significant (P<0.001).In diabetic rats, LC3 and Bnip-3 showed a weak positive correlation(P<0.05), Bcl-2 and Bax were irrelevant, Bnip-3 and Bax were positively weakly-correlated(P<0.05),Bcl-2 was not correlated;and Bcl-2 and Bax were irrelevant.Conclusion LC3,Bnip-3 and Bax in the brain tissue of diabetic rats are overexpressed , while Bcl-2 shows weak expression , indicating that autophagy factors and apoptotic factors are involved in the process of brain injury in diabet -ic rats, which may be one of the mechanisms of brain tissue damage .

Standardized training of residents is a necessary way for clinical medical personnel train-ing. Residents graduating from medical colleges and universities should accept the standardized, scientific, strict pre-service training, in order to realize role conversion from medical students to hospital physicians. This paper summarizes and analyzes the pre-service training, focusing on the training object, course design, organization implementation, evaluation, questionnaire survey, and puts forward some thoughts and sugges-tions on strengthening and improving pre-service training form, expanding the content of training, and quan-tifying the evaluation of training effect.
5.Role of hypoxia-inducible factor-1α in sevoflurane postconditioning-induced reduction of myocardial ischemia-reperfusion injury in mice

Objective To evaluate the role of hypoxia-inducible factor-1α (HIF-1α) in sevoflurane postconditioning-induced reduction of myocardial ischemia-reperfusion (I/R) injury in mice.Methods Forty pathogen-free healthy male C57 mice,aged 8 weeks,weighing 20-30 g,were assigned into 4 groups (n=10 each) using a random number table:sham operation group (group Sham),myocardial I/R group (group I/R),sevoflurane postconditioning group (group SPC) and HIF-1α inhibitor 2Me2 group (group 2Me2).Myocardial I/R was induced by occlusion of the left anterior descending branch of the coronary artery for 30 min followed by 120 min of reperfusion in pentobarbital sodium-anesthetized mice.In group SPC,3% sevofiurane was inhaled for 15 min starting from the onset of reperfusion.In group 2Me2,30% 2Me2 (30 mg/kg) was intraperitoneally injected at 30 min before ischemia.The mice were sacrificed at the end of reperfusion,and the hearts were removed for determination of the myocardial infarct size (using the Image J software) and expression of HIF-1α in thc nucleus of cardiomyocytes (by Western blot).Results Compared with group Sham,the myocardial infarct size was significantly increased in I/R,SPC and 2Me2 groups,the expression of HIF-1α was significantly up-regulated in I/R and SPC groups,and the expression of HIF-1α was significantly down-regulated in group 2Me2 (P< 0.05).Compared with group I/R,the myocardial infarct size was significantly decreased,and the expression of HIF-1α was up-regulated in group SPC,and the myocardial infarct size was significantly increased,and the expression of HIF-1α was down-regulated in group 2Me2 (P<0.05).Compared with group SPC,the myocardial infarct size was significantly increased,and the expression of HIF-1α was down-regulated in group 2Me2 (P<0.05).Conclusion The mechanism by which sevoflurane postconditioning reduces myocardial I/R injury is related to up-regulation of HIF-1α expression in mice.
6.Quality of Life of Adult with Intellectual Disability

Objective To investigation the condition,expectation and support of quality of life in adult with intellectual disability.MethodsA total of 394 adult with intellectual disability,their parents and friends in Beijing were involved in the study.Their qualities of life were evaluated.ResultsAbout quality of life,the adult with intellectual disability had significant more expectation than they had gained actually;their parents and friends had significant more expectation than themselves and staff of primary rehabilitation did.ConclusionIt is necessary to provide support to improve the quality of life of adult with intellectual disability.
7.Effect of total alkaloids of Rubus alceaefolius poiron on gene expressions of CYP2E1 and CYP3A1 in rats with acute liver injury.

OBJECTIVETo explore the effects of total alkaloids of Rubus alceaefolius Poiron (RAP) on gene expressions of drug-metabolic enzymes, CYP2E1 and CYP3A1 in liver.
METHODSSixty SD rats were randomly divided into six groups (10 rats in each), the blank control group, the model control group, the bifendate group and the three RAP treated groups treated respectively with low-, middle- and high-dose of RAP. The model of acute hepatic injury was established with intra-peritoneal injection of carbon tetrachloride. Serum levels of alanine aminotransferase (ALT) and aspartate aminotransferase (AST), and severity of hepatic tissue injury were measured, and the mRNA expressions of CYP2E1 and CYP3A1 in liver tissue were detected by RT-PCR.
RESULTSAs compared with the model group, serum levels of ALT and AST were significantly lower in the high- and middle-dose ARP group (P <0.01), but in the low-dose group, only ALT was significantly lower (P<0.01); the severity of liver injury was milder in the RAP groups (P<0.01); and both CYP2E1 and CYP3A1 mRNA expressions in liver were significantly lower in the bifendate and all RAP treated groups (P<0.01 or P<0.05).
CONCLUSIONRAP could significantly reduce the ALT and AST levels, protect liver cells from injury, and inhibit the mRNA expressions of CYP2E1 and CYP3A1 in liver tissue.

8.Effects of resveratrol on the proliferation and CaN of vascular smooth muscle cell induced by angiotensin II.

AIMTo investigate the effects of resveratrol (Res) on the proliferation of VSMCs induced by Ang I and the expression of calmodulin (CaM) and calcineurin (CaN) in the proliferation of VSMCs treated by Ang 1 and to discuss the mechanism.
METHODSRabbit arterial VSMCs were cultured in vitro and VSMCs were identified with the method of immunocytochemistry. A cell proliferating model of VSMCs induced by AngII was established. VSMCs were cultured for 4-8 passages. The experiments were randomly divided into control group, AngII group (0.1 micromol/L) and AngII + Res groups with different concentrations(20, 40, 80, 160) micromol/L. VSMCs proliferation was determined with MTT colorimetric method. CaM was detected with Coomassie brilliant blue method and CaN was determined by enzyme reaction phosphorus measurement.
RESULTSRabbit VSMCs were cultured successfully and could be passaged. After immunocytochemistry staining, all the cells cytoplasm were stained and positive. Cell proliferation, CaM and CaN activities were increased significantly in VSMCs proliferation induced by AngII (P <0.05, P < 0.01). The index of AngII + Res groups were obviously reduced compared with AngII group ( P < 0.01).
CONCLUSIONThe VSMCs proliferation induced by AngII can be inhibited by Res significantly, and the inhibiting mechanism of Res may be related to inhibiting CaM and CaN activities then restraining the proliferation of VSMCs in a dose dependent manner.

9.Cytochrome C oxidase subunit 1 mutation result in mitochondrial encephaiomyopathy with lactic acidosis and stroke-like episodes

Objective To report a 33-year-old man with post-headache stroke-like episodes,with whom ischemic changes were found in basal ganglia and occipital-temporal lobe and muscle biopsy revealed abnormal mitochondrial structure and function without regular mutations detected in mtDNA.Methods Gene chip technique was used to detect the mutation of whole sequence of mtDNA,and direct sequencing technique was used to confirm the mutations.Results Three mutations were found.A new mutation in the mitochondrial cytochrome C oxidase subunit 1 (MTCO1),a T→C transition at nucleotide position 6253 resulted in conservative methionine transferring to threonine.His mother also held the mutation,which was not found in 98 control samples.So T6253C was considered the nosogenetic mutation.Conclusion This is the first time to report a mutation in MTCO1 responsible of MELAS.
10.Correlation between Level of Nitric Oxide/Nitricoxide Synthase and Ultra-Structure Changes of Placenta Exposured in Lead Gestation Period in Rats

Objective To study the correlation between level of nitric oxide/ nitricoxide synthase(NO/NOS) on placenta homogenate and ultra-structure changes of placenta in pregnancy lead exposure in rats.Methods Seventeen normal pregnant rats and 46 rats of exposured in lead which were divided into A,B,C groups were studied.The level of NO/NOS of placenta were measured by nitrate reductase and NOS kit.Placentas were randomly selected from each group to detect ultra-structure by electron-microscope.Results There were significant difference among A,B and control groups on level of NO/NOS(all P0.05).Compensation hyperplasy or minor injury were observed in lead exposure of stage groups.Lead exposure during whole gestation period,the lead level was maxmum,and decompensation were observed on placental construction.Conclusions There is a close correlations between level of lead,NO/NOS and pathological change of placental tissue,and both of them may play an important role in the pathogenesis of peripartum lead exposure.
