1.Effect of lipopolysaccharide on activation of mouse lung fibroblasts

Objective To investigate the effect of lipopolysaccharide (LPS) on the activation of mouse lung fibroblasts. Methods Primary cultured mouse lung fibroblasts were incubated in 96 well plates and randomly divided into 2 groups: control group ( group C, n = 6) and LPS group ( n = 24). The fibroblasts were cultured for 72 h in group C. LPS 1 μg/ml was added and then the fibroblasts were incubated for 72 h in group LPS. The expression of type Ⅰ procollagen mRNA, α-smooth muscle actin (α-SMA) mRNA, Toll-like receptor 4 (TLR4)mRNA and integrin β1 mRNA was determined using real-time PCR at 3, 6, 24 and 72 h of incubation (6 wells at each time point). Results Compared with group C, there was no significant change in the expression of type Ⅰ procollagen mRNA, α-SMA mRNA, TLR4 mRNA and integrin β1 mRNA at 3, 6 and 24 h of incubation ( P >0.05), but the parameters mentioned above were significantly up-regulated at 72 h of incubation in group LPS ( P < 0.05). Conclusion In the early acute lung injury, LPS leads to pulmonary fibrosis through activating lung fibroblasts directly, and also accelerates the activation of lung fibroblasts and promotes the process of pulmonary fibrosis through up-regulating the expression of TLR4 and integrin β1 in mice.
2.Related factors analysis on complications caused by peripherally inserted central catheter puncture on low birth weight infant with different puncture sites

Objective To analyze the correlative factors that affect the main complications caused by peripherally inserted central catheter puncture (PICC) on low birth weight infant with different puncture sites,in order to reduce the incidence of complications and achieve the purpose of indwelling safely.Methods A total of 431 infants with different PICC puncture sites were recruited in this study from January 2008 to January 2013,261 cases of them were punctured in the elbow upper extremity veins, 147 cases were in the elbows downward veins,5 cases were in the superficial temporal veins, 12 cases were in the axillary veins,and 6 cases were in the femoral veins.The complications occurring in the period of indwelling catheter were analyzed, and the effective treatments or preventive nursing measures were taken.Results 431 cases were punctured successfully.The sites of puncture were mainly in the veins of the upper arm,and the main complications were phlebitis,extremities swelling,jamming pipe and infection.The incidence of phlebitis, extremities swelling in the elbows downward veins were lower than those in the elbow upper extremity veins: 10.89%(16/147) vs.3.83%(10/261), 17.69%(26/147) vs.4.98%(13/261), and the differences were significant, x2=7.840, 17.561, P<0.05.Conehusions The veins in the elbow and elbow downward are mainly selected.It is necessary to assess the complications promptly that may occur and to take effecdve measures,and good catheter maintenance consciousness also has important significance to the reduction of complications.
3.The clinical application of central venous access

Objective To advocate the importance of central venous access(CVA), and compare the technical characteristics and clinical values of central venous access constructed trans right subclavian vein (TSCV) and trans the right internal jugular vein (TIJV). Methods Between March 1999 and June 2001, we operated 348 cases of CVA, including 270 TSCV and 78 TIJV. The durations of reserving the accesses were 13~75 day, average 23 days. Results In 270 TSCV, 268 cases were successful, the successful rate was 99%. 11 cases had critical complications, including: 2 cases of pneumothorax, 1 access was constructed into subclavian artery, 3 accesses obstruction, 3 accesses of anomalous transfusion caused by catheter shift, infection of puncture site 2 cases. In 78 TIJV, 75 cases were successful, the successful rate were 96%. The critical complications occurred in 1 case.Conclusions The CVA is safe, easy, and practical, which would provide the special and immediate medical therapy for the tumorous and critical patients. In emergency, the TSCV procedure would be the choice.
4.Correlation between atherosclerotic renal artery stenosis and coronary heart disease and the inflammation related factors

Objective To investigate the correlation between atherosclerotic renal artery stenosis and the severity of coronary lesions in patients with confirmed coronary artery disease and the impact of renal artery stenosis on serum inflammation related factors.Methods 163 patients from Peking University First Hospital with confirmed coronary heart disease by coronary angiography underwent selective renal artery angiography from Dec 2002 to Sep 2003.Serum hypersensitive C reactive protein(hs-CRP)and Interleukin-6(IL-6)were measured.Results Patients combined with coronary disease and renal artery stenosis were presented with more acute coronary syndrome(ACS)and triple-vessel CAD.Serum hs-CRP and IL-6 levels were also higher in this group.Conclusion Renal artery stenosis is correlated with severe coronary disease.Inflammation state could be one of the reasons for this connection.
5.Determination of Dracohodin in Qilisan Gel by HPLC

Objective: To establish a method for determining the content of dracorhodin as an indicator in the quality control with methodological studies to provide basis for the preparation process study of Qilisan gel. Methods: An HPLC method was used with a DiamonsilR C18 (250 mm ×4. 6 mm, 5 μm) column, the detection wavelength was set at 440 nm and the column temperature was 30 ℃. Acetonitrile-0. 05 mol·L-1 sodium dihydrogen phosphate solution (45 ∶ 55) was used as the mobile phase, and the flow rate was l. 0 ml·min-1 . Results: The content of dracorhodin was in a good linear relationship (r =0. 999 6) within the range of 1. 632-64. 250 μg·ml-1 , and the average recovery was 99. 02%(RSD =0. 77%, n =6). The number of theoretical plates was calculated as 7 100 according to the dracorhodin perchlorate peak without any interference from the negative sample. Conclusion: The method is objective, accurate and sensitive with high reliability, easy operation and fast process. The measurement results can be used as the quality control basis for the preparation process study of Qilisan gel.
6.Granulosa cells model transfected by ENO1 overexpression adenovirus vector and the effect of ENO1 overexpression on progesterone secretion.

OBJECTIVETo construct primary cultured granulosa cells model of Zi Gooses tansfected by alpha-enolase (ENO1) overexpression adenovirus vector, and to detect the effect of ENO1 overexpression of granulose cells on progesterone secretion.
METHODSGranulosa cells were infected with Ad-CMV-ENO1 in gradient multiplicity of infection(MOI) levels:100, 250, 350 and 400 pfu/cell. Twenty four hours and 48 h after infection, green fluorescent protein (GDP) was respectively detected by fluorescence inverted microscopy. The effect of ENO1 overexpression of granulose cells on progesterone secretion was detected by the step double antibody sandwich enzyme-linked immunosorbent assay (ELISA).
RESULTSThe optimal infection rate (100%) was achieved when MOI was 800 pfu/cell,48h after infection. Real time RT-PCR and Western blot showed that the level of mRNA and protein expression ENO1 were increased significantly after infection (P < 0.01); The granulosa cells progesterone secretion of Ad-CMV-ENO1 group increased signigicantly (P < 0.01).
CONCLUSIONENO1 overexpression could make the primary culture follicle granulosa cells in vitro improve progesterone secretion.

7.Reliability of iWitness photogrammetry in maxillofacial application.

OBJECTIVEThis study aims to test the accuracy and precision of iWitness photogrammetry for measuring the facial tissues of mannequin head.
METHODSUnder ideal circumstances, the 3D landmark coordinates were repeatedly obtained from a mannequin head using iWitness photogrammetric system with different parameters, to examine the precision of this system. The differences between the 3D data and their true distance values of mannequin head were computed.
RESULTSOperator error of 3D system in non-zoom and zoom status were 0.20 mm and 0.09 mm, and the difference was significant (P 0.05). Image captured error of 3D system was 0.283 mm, and there was no significant difference compared with the same group of images (P>0.05). Error of 3D systen with recalibration was 0.251 mm, and the difference was not statistically significant compared with image captured error (P>0.05). Good congruence was observed between means derived from the 3D photos and direct anthropometry, with difference ranging from -0.4 mm to +0.4 mm.
CONCLUSIONThis study provides further evidence of the high reliability of iWitness photogrammetry for several craniofacial measurements, including landmarks and inter-landmark distances. The evaluated system can be recommended for the evaluation and documentation of the facial surface.

8.Monostotic fibrous dysplasia of the sphenoid sinus: two cases report and literature review

objective:To study the etiopathogenisis、clinical features、diagnosis and treatments of monostotic fibrous dysplasia of the sphenoid sinus. Method:Two cases of monostotic fibrous dysplasia of the sphenoid sinus without any symptoms was reported with relevant literature review.Result: No aggravation was found after 6 months-follow-up.Conclusion:The cranial fibrous displasia has low incidence rate with non-specific symptoms and high rate of misdiagnosis. The monostotic fibrous dysplasia of the sphenoid sinus without any symptom is rarely seen clinically. Imageological examination,for example,CT and MRI,is valuable for the diagnosis of this disease. The histopathological evidence is absolutely necessary to make definite diagnosis. The conservative treatment may be chosen for the asymptomatic cases. Nasal Endoscopic surgery can not only remove the diseased region but also make diagnosis.The long-term follow-up should be carried out in all of these patients.
9.Effect of microRNA-101 on atrial fibrosis in human chronic atrial fibrillation

Objective To investigate the effect of microRNA-101 (miRNA-101) on atrial fibrosis in human chronic atrial fibrillation (AF). Methods Right atrial appendages were obtained from 59 patients (30 with AF) undergoing cardiac surgery, including 47 patients with valve heart disease and 12 patients with congenital heart disease. The expression of miRNA-101 was determined by quantitative real-time PCR in the right atrial appendages of patients with and without AF. The cell-specific localization of miRNA-101 was detected by in situ hybridization assay. The mRNA and protein expression levels of transforming growth factor β typeⅠreceptor (TGFβRⅠ) and collagen type I (COL1) were determined by quantitative real-time PCR and Western-blot assay, respectively. Collagen in the right atrial appendages was observed by Masson staining assay. Results The expression of miRNA-101 was found to be significantly down-regulated in AF patients compared with patients with sinus rhythm (SR) (P < 0.05). The result of miRNA-ISH showed that miRNA-101, which was highly distributed within the connective tissues of heart, was down-regulated at about 24.9% in patients with AF compared with patients with SR. No significant differences at the mRNA expression level of TGFβRI was found between patients with AF and patients with SR (P > 0.05). But the protein expression of TGFβRI in patients with AF was significantly higher than that of patients with SR (P < 0.05). The mRNA and protein expressionsl of COL1 were significantly higher in patients with AF than thoset of patients with SR (P < 0.05). The collagen was significantly increased in patients with AF than that of patients with SR (P < 0.05). Conclusions Downregulation of miRNA-101 may contribute to atrial fibrosis in human atrial fibrillation by targeting TGFβRⅠ.
10.Food intolerance-related symptoms in 903 individuals

Objective To investigate the symptoms and affecting factors of food intolerance among 903 adults.Methods Food special antibody(IgG)in human serum were examined by ELISA.Food Intolerance Health Assessment Questionnaire was used to assess positive rate of food intolerance and food intake habits.Chi-square test and Mann-Whitney test were used for data analysis.Results The ratio of food intolerance was 0.33% to 28.13%.Intolerant foods include crab(28.13%),yolk(27.02%),milk (13.07%),soybean(11.96%),shrimp,tomato,corn,beef,rice,chicken,mushroom,wheat,and pork.The symptoms of nervous system(55.89%),digestive system(45.69%)and respiratory system(15.64%)were commonly seen.Prevalence of food intolerance in mental labors was lower than manual labors.Conclusion The most common intolerant food includes crab,yolk and milk.The symptoms of food intolerance are different.Manual labors are more susceptible to food intolerance.
