1.Exhaled volatile organic compounds detection in cancer diagnosis

Volatile organic compounds (VOCs) are organic chemicals which can exist in the form of vapor at room temperature.Endogenous VOCs can be produced by different biochemical processes in the human body.They can be transfered to lung through bloodstream and released through breath.Many researches show that exhaled VOCs can be significantly changed when people suffering cancers or inflammation. Detecting exhaled VOCs through instrument with high sensitivity make it possible to detect cancers in early phase in a noninvasive and rapid way,which reduce the suffering of patients.In conclusion,exhaled VOCs detection is a promising method for early diagnosis of cancers.

8.The effect of isosteviol on growth of human osteosarcoma cell lines U-2OS

Background and purpose:Osteosarcoma, a highly malignant bone tumor, develops rapidly. The current medicines for osteosarcoma present some limitations with serious side effects of long-term use. Isosteviol has the structure of tetracyclic diterpene which is the starting material of many anti-cancer drugs. However, its anti-tumor activity has been rarely reported. This study investigated the effect of isosteviol on proliferation of human osteosarcoma cell line U-2OS.Methods:The effect of isosteviol on U-2OS cell proliferation was assayed by MTT method. Cellular morphologic changes were observed under an inverted phase contrast microscope. The cell condition was observed with Hoechst 33342 and PI staining. Generation of reactive oxygen species and cell membrane potential were detected as well. The cell cycles were analyzed with lfow cytometry. The expressions of apoptosis-related proteins Bcl-2 and Bax were measured by Western blot assay.Results:The result indicates that isosteviol suppressed the growth of U-2OS cells in time- and concentration-dependent manner. Isosteviol could cause S phase cell cycle arrest at 24 h and apoptosis at 48 h. With the increased drug concentration, reactive oxygen species increased significantly, and the membrane potential gradually reduced. In addition, isosteviol treatment enhanced the expression of Bax but reduced that of Bcl-2.Conclusion:The inhibition of isosteviol on cell growth of U-2OS cells was possibly caused by promoting apoptosis through regulating the apoptosis-related protein expressions, such as the enhancement of Bax and reduction of Bcl-2 expression.
9.Effect of fibroblast growth factor receptors 1-dominant negative strategy on alkaline phosphatase activity of bone marrow stromal cells after osteogenic induction

Objective To explore the effect of fibroblast growth factor receptors 1-dominant negative strategy (FGFR1-DN) on alkaline phosphatase (ALP) activity of bone marrow stromal stem cells (BMSCs) after osteogenic induction.Methods BMSCs were transfected with eukaryotic expression plasmid pcDNA 3.1 (+)-DN FGFR1 and pcDNA3.1 (+)-FGFR1.The experiment was conducted in 4 groups:FGFR1-DN transfection group,FGFR1 transfection group,pcDNA3.1(+) empty vector transfection group and non-transfection group.The ALP activity of BMSCs was detected in logarithmic growth phase after osteogenic culture.The qualitative detection of ALP activity was carried out immunohistochemically while the quantitative detection by cALP kit.The ALP activity was compared between the 4 groups at 7 and 14 days after osteogenic induction.Results Compared with 7 days,the ALP activity at 14 days was significantly increased in the 4 groups,and the increase in FGFR1-DN transfection group was significantly higher than in the other 3 groups (P < 0.05).At both 7 and 14 days,the ALP activity in FGFR1-DN transfection group was the highest while that in FGFR1 transfection group was the lowest (P < 0.05).Conclusions FGFR1-DN can promote the ALP activity of BMSCs during osteogenesis.This may provide an experimental basis for the joint application of local gene therapy and tissue engineering and for construction of tissue engineered bone with better biocompatibility.
10.Application of nurse-administered psychological assessment scale for patients early after trauma in the wounded after earthquake

Objective To explore the application of nurse-administered psychological assessment scale for patients early after trauma in the wounded after Wenchuan earthquake.Methods The clinically commonly used self-rating scales (integrated SCL-90,SDS and SAS,and mental status scale in non-psychiatric settings),clinician-administered scale (nurse-administered psychological assessment scale for patients early after trauma)and diagnostic criteria in psychiatric settings(CCMD-3)were employed to assess the mental status of 21 patients after Wenchuan earthquake,and the recovery rates of these scales were analysed.Results All the 21 patients suffered from psychological disturbance to some extent, while none met the criteria of mental diseases.The recovery rate of the integrated SCL-90,SDS and SAS was only 9.5%, while those of the nurse-administered psychological assessment scale for patients early after trauma and mental status scale in non-psychiatric settings reached 100%.Conclusion The nurse-administered psychological assessment scale for patients early after trauma and mental status scale in non-psychiatric settings with fewer items are more suitable for the wounded early after earthquake,with a favourable confidence and efficacy.The diagnostic criteria in psychiatric settings can not be universally used in the wounded after earthquake.
