4.Influence on mechanical properties and microstructure of nano-zirconia toughened alumina ceramics with nano-zirconia content.

OBJECTIVETo investigate the mechanical properties and microstructure of nano -zirconia toughened alumina ceramics with variety of nano-zirconia content in centrifugal infiltrate casting processing of dental all-ceramic.
METHODSComposite powder with different ethanol-water ratio, obtained serosity from ball milling and centrifugal infiltrate cast processing of green, then sintered at 1 450 degrees C for 8 h. The physical and mechanical properties of the sintered sample after milling and polishing were tested. Microstructures of the surface and fracture of the sintered sample were investigated by SEM.
RESULTSThe experimental results showed that there had statistical significience (P < 0.01) on static three-point flexure strength and Vickers Hardness in three kinds of different nano-zirconia content sintered sample. Fracture toughness of 20% group was different from other two groups, while 10% group had not difference from 30% group (P < 0.05). The mechanical properties of this ceramic with 20% nano-zirconia was the best of the three, the static three-point flexure strength was (433 +/- 19) MPa and fracture toughness was (7.50 +/- 0.56) MPa x min 1/2. The intra/inter structure, fracture of intragranular and intergranular on the surface and fracture of sintered sample in microstrucre was also found.
CONCLUSIONIntra/inter structure has strengthen toughness in ceramics. It has better toughness with 20% nano-zirconia, is suitable dental all-ceramic restoratives.

5.Observation of Dynamic Changes in Ultra-Micro-Structure of Pulmonary Arteries and Endogenous Hydrogen Sulfide in Rats with Left-Right Shunt

Objective To explore the relationship between dynamic changes in ultra-micro-structural of pulmonary arteries and endogenous hydrogen sulfide in rats with left-right shunt.Methods Rats in shunt group were subjected to an abdominal aorta-inferior vena cava shunt to create an animal model of pulmonary artery structural remodeling. After 1 day, 3 days, 1 week, 4 weeks and 8 weeks of experiment, the ultra-micro-morphologic changes of pulmonary arteries of rats were observed under electronic microscope and H_2S concentration in serum was evaluated by modified sulfide electrode method.Results The changes of ultra-micro-structure of pulmonary arteries were progressively exacerbated, endothelial cells became swollen and large in size on 3 days, smooth muscular cells increased in size as well as the change of endothelial cells in 1 week, and they changed from contractile phenotype to synthetic phenotype in 4 weeks.Conclusions Shunt exhibited changes of ultra-micro-structure of pulmonary arteries are accompanied by the changes of endogenous H_2S. It is suggested that endogenous H_2S might play a protective role in changes of ultra-micro-structure of pulmonary artery.
6.Expression of cyclooxygenase-2 in bone marrow cells of chronic leukemia and its significance.

The study was aimed to investigate the expression of COX-2 in bone marrow cells of chronic leukemia patients and its potential pathogenetic implications. Western blot was applied for detecting COX-2 expression levels in bone marrow cells of 67 chronic leukemia patients and beta-actin expression levels. Bone marrow aspirations from 14 healthy donors were used as negative controls. The results showed that the positive rates of COX-2 in chronic-phase group of chronic myeloid leukemia (CML-CP) and in group of chronic lymphocytic leukemia (CLL) were 76.32% (29/38) and 75.86% (22/29) respectively. Both CML-CP and CLL group showed a higher expression than control group (p = 0.0000, p = 0.0000 respectively). The expression of LDH in Cox-2 positive group was higher than that in Cox-2 negative group, and the difference between the two groups was statistically significant (p < 0.001). It is concluded that the expression of COX-2 protein can be detected in bone marrow cells of CML-CP and CLL and the expression level of LDH were higher in cells of CML-CP and CLL. The expression of COX-2 may be correlated with prognosis of CML-CP and CLL.

8.Non-equilibrium properties of drug permeation through stratum corneum in vitro.

AIMTo illustrate the non-equilibrium properties of drug permeation through stratum corneum (SC) to provide theory and method for transdermal drug delivery.
METHODSThe system of side-by-side permeation chambers in vitro was isolated, thus conditions for equilibrium state were decided. And a network thermodynamic model was established. Non-equilibrium and leakage experiments were completed with which tinidazole was pattern drug.
RESULTSThe properties of non-equilibrium including: long term to reach equilibrium state; delay-start of the permeation; uncertainty of initial drug permeable flux. The properties of leakage including: degression of drug permeable flux; changeability of permeation time constant.
CONCLUSIONThe permeation cell is believed to be a non-linear and time variable system.

9.Study on inhibitory effect of water extract of Polygonum multiflorum on CYP1A2 and CYP2E1 enzymatic activities and mRNA expressions in rat liver.

Rats were continuously given different doses of water extract of Polygonum multiflorum (1, 10 g x kg(-1)) for 7 days to prepare liver microsomes. Cocktail in vitro incubation approach and Real-time quantitative PCR technology were used to observe the effect of water extract of P. multiflorum on CYP450 enzymatic activities and mRNA expressions in rat liver. Compared with the blank control group, both 1, 10 g x kg(-1) water extract of P. multiflorum treated groups showed significant inhibitions in CYP2E1 enzymatic activities and mRNA expressions (enzymatic activities of CYP2E1, P < 0.01; mRNA expression of CYP2E1, P < 0.05 in 1 g x kg(-1) group, P < 0.01 in 10 g x kg(-1) group). They revealed a significant increase in the enzymatic activity of CYP3A1 (P < 0.01), but without significant change in mRNA expressions. The 10 g x kg(-1) group showed a significant inhibition in CYP1A2 enzymatic activities and mRNA expressions in rat livers (P < 0.01).

10.Efficacy and safety of the thrombolytic therapy with urokinase after systemic-pulmonary shunt.

OBJECTIVETo evaluate the efficacy and safety of thrombolytic therapy with urokinase after systemic-pulmonary shunt.
METHODSSix patients who had thrombosis after systemic-pulmonary shunt were enrolled in this study. At the background of administration of the heparin at a dose of 0.2-0.3 U x kg(-1) min(-1), urokinase was intravenously administered with a loading dose of 15-20 U x kg(-1) x min(-1) and a locked time period of 30 minutes, and then the dose was incessantly decreased to 4-10 U x kg(-1) x min(-1). In addition to echocardiography (ECG), arterial partial pressure of oxygen/inspired oxygen fraction (PaO2/FiO2), fibrinogen, activated partial thromboplastin time, and prothrombin time were determined to assess the clinical efficacy and side effects.
RESULTSThe thrombolytic therapy with urokinase showed clinical effectiveness within 1 or 2 hours in all 6 patients. Efficiency of this therapy reached 100% during 12 to 24 hours. In 5 patients, the PaO2/FiO2 were over 50% higher than the early postoperative values. One patient received a second operation due to the excessively increased pulmonary blood flow. In 2 patients, pleural and mediastinal drainages increased when the thrombolytic therapy with urokinase began; however, they decreased after the urokinase dosages were adjusted.
CONCLUSIONIt is feasible to use the thrombolytic therapy with proper dosage of urokinase after systemic-pulmonary shunt.
