Objective To study the clinical significance and changes of plasma nitric oxide (NO) and cyclic adernosine monophosphate(cAMP)/cyclic guanosine monophosphate(cGMP) ratio in children with asthma.Methods NO,cAMP,cGMP levels and changes of cAMP/cGMP in 40 asthmatic children in acute attack stage and restoration stage and in 23 health children were measured by nitric acid reductase method and radioimmunoassay.Results 1.the amount of NO 3-/NO 2-in the plasma of asthmatic children in acute attack stage was obviously higher than that in restoration stage and in normal control group(P0.05);6.the results of linear correlation analysis showed that there were positive correlations between plasma NO 3-/NO 2- and plasma cGMP levels in acute attack stage (r=0.401 P

Background The select of supporter is critical for the construction of tissue engineering cornea.Many carrier carl be utilized in the construction of tissue engineering cornea,but de-cellular corneal matrix is known to be one of optimal supporters.Objective Present study was to investigate the characteristics of de-cellular corneal matrix of porcine of structure and biocompatibility for rabbit cornea stroma and limbal epithelial ceHs. Methods The porcine cornea was prepared as de-cellular corneal matrix of porcine by the application of detergent enzyme combined process.The corneal epithelial cells,keratocyte and endothelial cells of porcine were removed completely and stored in -20℃ refrigerator after sterilization.The morphology of de-cellular corneal matrix of porcine was examined by hematoxylin-eosin staining under the light microscope.The structure characteristics of de.cellular corneal matrix of porcine under the scan electron microscope,and its physics features were investigated by the evaluation of water content,strength,expansion and transparency.The de-cellular corneal matrix of porcine were implanted to cornea stroma of rabbit and co-cultured with rabbit corneal epithelial cells for 4 weeks in vitro to assess the keracyte compatibility. Results The epithelial cells,keratocyte and endothelial cells of porcine were removed completely by trypsogen digestion.The collagen fibril network and collagen plates paralleled to corneal surface under the light microscope.The water content,strength,expansion。Ratio of light transparency of de-cellular corneal matrix of porcine were similar to normal porcine cornea.After implantation of de.cellular comeal matrix of porcine into rabbits corneal stroma,the edema of tissue was found in one week,and edema disappeared on two weeks and became clear on four weeks after surgery.The de-cellular eorneal matrix attached to rabbit cornea stroma well.No inflammatory eell and new vessel were found after surgery.The co-cultured rabbit corneal epithelial cells differentiated and proliferated on the surfaee of de-cellular corneal matrix and showed positive response for CK3.No statistically significant differences were found in the water content,strength,expansion of de-cellular cornea matrix of porcine among the normal,before dehydration,2 and 4 hours after dehydration cornea matrix(P>0.05).However,the transparency was much better in the corneal matrix with 2-hour,4-hour dehydration in comparison with non-dehydration one(P<0.05). Conclusion The structure features of de-cellular cornea matrix of porcine are similar to normal porcine cornea.Good biocompatibility is proved for xenogenesis of rabbit cornea.
3.Assessing the effect of pit and fissure sealing on 733 six-year-teeth four years later

Objective:
Assessing the effect of pit and fissure sealing on children’mandibular first molar in four years follow-up.
Methods:
The second grade pupils of five primary schools in Guangzhou were given pit and fissure sealing, 733 teeth were performed pit and fissure sealant. The retention rate of the sealant and caries incidence of the mandibular first molar were analysed in four years follow-up.
Results :
In four years follow-up, the retention rate of mandibular first molar was 57.16%. In terms of the caries incidence, the sealing group was 9.82%, while the unsealing group was 15.77%(χ2 = 7.045, P < 0.05). The decrease rate of caries in the sealing group was 37.73%, the caries incidence for the shallow pit and fissure group was 7.76%, no statistical differentce could be found two groups (χ2 = 0.844,P = 0.358).
Conclusion
Caries incidence of mandibular first molar obviously decreased 4 years later after the pit and fissure sealing.

5.Studies on in vitro culture of adventitious root in Salvia miltiorrhiza.

OBJECTIVETo study the culture of adventitious root of Salvia miltiorrhiza in vitro systemically.
METHODEffects of sucrose concentrations, medium pH, inoculum size and plant growth regulators on adventitious root growth and secondary metabolites production in S. miltiorrhiza were investigated.
RESULTWith the increase of initial sucrose concentration, adventitious root growth rates increased and tanshinone II A content decreased, while content of protocatechuic aldehyde showed a broken line change and its highest production was obtained under 30 g x L(-1) sucrose in the medium. As for the effect of medium pH, medium pH of 6.5, 5.5 (or 6.0) and 5.8 was favorable for adventitious root growth, tanshinone II A and protocatechuic aldehyde synthesis respectively. Furthermore, adventitious root growth, rate was greatly increased when inoculum size was 2.5%. MS medium added with 0.5 mg x L(-1) KT was much favorable for tanshinone II A and protocatechuic aldehyde accumulation.
CONCLUSIONParameters including sucrose concentrations, medium pH, inoculum size and plant growth regulators have distinct effects on the in vitro culture of adventitious root growth and secondary metabolites synthesis of S. miltiorrhiza.

There are dispute about the status of taxonomy among Astragalus membranaceus (Fisch.) Bge, A. membranaceus (Fisch.) Bge. var. mongholicus (Bge.) Hsiao. and A. pallidipurpureus stat. nov. The varieties and taxa of the complex are still in need of revision. With molecular biology study used trnH-psbA intergenic region, the taxonomic revision of Radix Astragali has been made. A. pallidipurpureus stat. nov is suggested as a new species.

In this paper, based on analyzing the status quo and key problem of research on nature of Chinese materia medica, we propose the thinking of research on nature of Chinese materia medica in view of genetic relationship. Namely, according to the cue of documents arrangement and data excavation, in certern scope of genetic relationship, difinite the common pharmaco-effect of the same nature of drug, and analyze the chemical composition correlated the nature of drug, further more, reveal the related regularity of nature of Chinese materia medica-pharmaco-effect- chemical composition. Linking the nature of Chinese materia medica, pharmaco-effect and chemical composition will provide original thinking and view angle to break through the key problem of research on nature of Chinese materia medica.

8.Clinical effects of Supplemented Buzhong Yiqi Decoction on patients with HP-positive chronic atrophic gastritis of Spleen-Stomach Deficiency Pattern

AIM To explore the clinical effects of Supplemented Buzhong Yiqi Decoction on patients with HP-positive chronic atrophic gastritis of Spleen-Stomach Deficiency Pattern.METHODS One hundred and thirty-two patients were randomly assigned into control group(66 cases)for 12-week intervention of conventional treatment,and observation group(66 cases)for 12-week intervention of both Supplemented Buzhong Yiqi Decoction and conventional treatment.The changes in clinical effects,HP clearance rate,miR-32,TGF-β1,IL-6,PGⅠ,PGⅡ,EGF,somatostatin,gastrin,motilin,gastroscopy pathological score and TCM symptom score were detected.RESULTS The observation group demonstrated higher total effective rate and HP clearance rate than the control group(P<0.05).After the treatment,the two groups displayed decreased miR-32,TGF-β1,IL-6,gastroscopy pathological score,TCM symptom score(P<0.05),and increased PGⅠ,PGⅡ,EGF,somatostatin,gastrin,motilin(P<0.05),especially for the observation group(P<0.05).CONCLUSION For the patients with HP-positive chronic atrophic gastritis of Spleen-Stomach Deficiency Pattern,Supplemented Buzhong Yiqi Decoction can alleviate inflammation,regulate gastrointestinal hormone levels,improve symptoms,and enhance efficacy.
9.Development of a novel protein carrier inducing immune response and binding DNA in gene therapy.

OBJECTIVETo develop a novel protein carrier which can not only regulate the immune system but also deliver DNA into the tumor cell as an effective non-viral gene delivery system.
METHODSBy using gene engineering techniques, we constructed a fusion protein containing the -COOH end of human hepatitis B virus core antigen (HBcAg), small home-to-cancer peptide ligand RGD and Glutathione S-transferase (GST), which was expressed in E. coli and purified by size exclusion chromatography and affinity chromatography. We labeled it with FITC to observe whether it could bind prostate cancer PC-3 cell lines, and meanwhile used it as a non-viral gene delivery carrier with the plasmid pEGFP-N1 that could express GFP in PC-3 cells. Furthermore, we observed the regulatory function of this fusion protein to the mouse immune system.
RESULTSThe results of SDS-PAGE showed that the new protein carrier was obtained, which It could enter PC-3 cells with DNA in vitro and induce the mouse immune system to produce IgG1 and IgG2alpha simultaneously.
CONCLUSIONThe new protein carrier can be used as a target protein, especially in positive cells and the immune system. It promises to be a good novel carrier for the gene therapy of cancer.

10.The Effects of IFN-γ on AKT activated 32D Cells and its Mechanisms.

OBJECTIVETo investigate the effects of activated AKT on murine myeloid precursor cells (32D cells), and the effects of IFN-γ on 32D cells and its mechanisms.
METHODSPlasmid transduction was used to enhance the expression of AKT on 32D cells. After the transfected cells treated with IFN-γ for 24 hours, proliferation rate was tested by WST-1, apoptosis by flow cytometry, expression of phosphorylated Erk1/2, Stat3 and phosphorylated Stat3 was determined by Western blot.
RESULTS(1) IFN-γ at low concentration (100 U/ml) enhanced the growth and proliferation of 32D cells, while at high concentration (1000 U/ml) suppressed them. (2) Compared with control groups, low concentration IFN-γ increased (1124 ± 13) Stat3 phosphorylation in 32D-cell, while it high concentration IFN-γ decreased (601 ± 13). 32D cells transfected with activated Akt grew rapidly (0.287 ± 0.010) and had a low apoptotic rate [(9.57 ± 0.17)% (P < 0.05)]. (3) The expression of p-Erk1/2 in transfected 32D-cell was significantly reduced (P < 0.05). (4) Apoptosis rate of IFN-γ treated group was significantly decreased in transfected 32D cells (P < 0.05).
CONCLUSIONSIFN-γ has dual effects on 32D cells, namely, at low concentration enhanced the growth and proliferation of 32D cells, while at high concentration suppressed them. Its mechanisims is possibly through Stat3 pathway. Activated Akt can significantly promote the growth and proliferation of 32D cell and significantly inhibit apoptosis and IFN-γ can regulate cell proliferation and apoptosis through AKT. AKT activation can inhibit the Erk signal pathway, which may be affected by inhibition the modificaton of Raf1.
