1.STUDY ON SCAVENGING FREE RADICALS BY ACTIVE COMPOUNDS FROM BARLEY LEAVES

Objective: To study the scavenging capacity of extract from four kinds of barley leaves on superoxide free radical and hydroxyl free radical. Methods: Measure the content of superoxide free radical and hydroxyl free radical by nitro blue tetrazolium(NBT) photo-reduction. Results: Along with the flavonoid in the reaction liquid, the scavenging rate shows ascending trend . When the concentration of flavonoid is 12?g/ml, its scavenging rate is 95.56% and 94.12% on superoxide free radical and hydroxyl free radical respectively. Conclusion: The flavonoids of barley leave s have stronger anti-oxidative action.
2.A Review for Progress of SELDI Protein Chip Technique Applications

From genomics to proteomics, and then to miRNA, all without exception showed that proteins are the substance which directly regulate life. Comparing with genomics, the study of proteins is more difficult for its great variety, complicated modification and complicated construction. Recently, some new techniques for protein assay and research have provided us, for example, Fluorescence labeling, SELDI, SPR and optical protein chip, of which, SELDI SPR and optical protein chip are label-free. SELDI protein chip technique and its newly developed applications are briefly reviewed here.

4.Effects of deliberated hypotension and hemorrhagic hypotension on systemic and renal hemodynamics in dogs

Objective: To compare the effects of deliberate hypotension and hemorrhagic hypotension on systemic and renal hemodynamics in dogs. Method: Twelve mongrel dogs were randomly divided into nitroprusside-deliberated hypotension group and hemorrhagic hypotension group. After surgical preparation, the MAP of the dogs was decreased to 8kPa either by infusing nitroprusside or by withdrawing arterial blood and was kept for 30 min. RBF, MAP, HR, CO and CVP were monitored continuously. Result: Hypotension induced by hemorrhage was associated with significant decrease in CO, SV and CVP compared with nitroprusside. There was no significant change in HR in both groups. Hemor rhage caused more decrease in RBF than nitroprusside did (59.0% vs 37.5%). In recovery period, RBF in both groups increased, but remained significantly less than baseline in hemorrhage group. Conclusion: Hemorrhage can cause more pronounced interference in hemodynamics and more decrease in RBF with a subsequent slower recovery than with nitroprusside at the same level of hypotension.
5.Hemodynamic effects of induced hypotension with sevoflurane or nitroprusside

Objective:To compare the effects of induced hypotensions with sevoflurane and nitroprusside on hemodynamics in dogs. Method:Fourteen mongrel dogs were randomly divided into sevoflurane group and nitroprusside group. After experimental preparation,the MAP was decreased to 8kPa and was kept for 30min either with sevoflurane inhalation or with nitroprusside infusion. MAP, HR,CO,CVP,SV and SVR wrer monitored continuously. Result: During induced hypotension, CO in sevoflurane and nitroprusside groups decreased by 30.7% and 21.6% respectively; HR decreased significantly in sevoflurane group, but not in nitroprusside group; SVR in both groups showed reduction, but more pronounced in nitroprusside group. However,after the termination of sevoflurane or nitroprusside, MAP elevated over baseline with the significant increase of SVR,and CO remaining low. Conclusion:As compared with nitroprusside, sevoflurane has inhibitory effect on circulation,yet it is still efficient to induced hypotension.
6.Effect of different doses of propofol on cognitive function after chronic cerebral ischemia-induced injury in aged rats

Objective To investigate the effects of different doses of propofol on cognitive function after chronic cerebral ischemia-induced injury in aged rats. Methods Eighty male SD rats, aged 18 months, weighing 400-500 g, were randomly divided into 4 groups ( n = 20 each): shame operation group (group S), chronic cerebral ischemia group (group I), two propofol groups (groups P1 and P2 ). The chronic cerebral ischemia was induced by permanent occlusion of bilateral common carotid arteries. On 1 day after operation, intraperitoneal normal saline 2.5 ml was injected twice a day for7 consecutive days in groups S and I, and intraperitoneal propofol 10 and 50 mg/kg in 2.5 ml of normal saline were injected twice a day for 7 consecutive days in groups P1 and P2 respectively. On 3rd and 33rd days after the last injection (T1.2), 10 rats in each group underwent Morris water maze test to assess the cognitive function. After the test was completed, the rats were sacrificed and the hippocampi were removed and sliced (450-500 μm thick). Schaffer lateral branch in CA1 region was stimulated to induce long-term potentiation (LTP). Results Compared with group S, the escape latency was significantly prolonged, the number of animals' swimming across the platform, the ratio of the swimming time spent in the forth quadrant to the total swimming time, and the success rate of LTP induction were significantly decreased at T1 and T2 in groups I, P1 and P2 (P < 0.05). Compared with group I, the escape latency was significantly prolonged, the number of animals' swimming across the platform, the ratio of the swimming time spent in the forth quadrant to the total swimming time, and the success rate of LTP induction were significantly decreased at T1 in groups P1 and P2, and at T2 in group P2 ( P < 0.05). Conclusion Propofol aggravates the damage to cognitive function while it attenuates the chronic cerebral ischemia-induced injury in aged rats, especially the high dose.

9.Three-dimensional CT reconstruction for analysis of normal acetabular spatial contour and its curvature transformation

BACKGROUND: The determination of the normal acetabular spatial contour, the pattern of its curvature transformation, and adaptability between acetabulum and femoral head plays an important role in the description of spatial relationship of the hip joint the management and evaluation of hip disorders, especially developmental dysplasia of the hip.OBJECTIVE: Based on spiral threa-dimensional CT data of normal hip joints, to create isoline map of each acetabulum and femoral head by a well-developed and cooperating programmed computer software so as to determine the normal acetabular spatial contour and the pattern of its curvature transformation.METHODS: Three-dimensional CT data of 56 normal hip joints, aged from 6 to 17 years, were collected. After three-dimensional simulation and surface shade display (SSD) reconstruction by a well-developed VxWork software, the isoline map was created using contours map in Gecmorphology, and each isoline map of the normal acetabulum and femoral head was analyzed via a specific computer software (Terrain). RESULTS AND CONCLUSION: The isoline array of normal acetabulum revealed the modality of some concentric "C" or oval "O",with at least one close "O" at the central zone, indicating that there existed dome shape at the superior zone of acetabulum. The modality of sparse isolines around dome illustrated larger curvature radius and more slowgoing change of spatial contour transformation of this zone, while the dense isolines on the bottom zone Of acetabulum represented a small curvature radius,steep shape, and rapid changing profile of this zone. The isoline array of normal femoral head was multiple concentric "O" with different space between each line. The congruence isolines of acetabular dome and superior surface of femoral head emerged as the similar trend of geomorphological characteristics, revealing that both the spatial contour and the curvature pattern between acatabulum and femoral head are well-adaptable to each other. Results show that the isolines of acatabulum and femoral headcan stersoscopicly visualize the spatial contour and curvature pattern of the topographic surface of the hip joint.
10.Alleviating Japanese encephalitis virus infection with multiple receptor tyrosine kinase inhibitor sunitinib

Abstract: Objective To evaluate the effects of sunitinib on Japanese encephalitis virus (JEV) infection in vitro and vivo. Methods The 4-week-old BALB/c mice infected with JEV by intraperitoneal injection. The infected mice were treated with sunitinib for 5 days and 10 days respectively. After that, the change of weight and survival rate were evaluated continuously. The viral load variation in mice brain were detected by qRT-PCR. Indirect immunohistochemical staining assay (IFA) was used to detect the number and distribution of CD4+/CD8+T cells in mouse brain. IFA was also used to observe the expression of virus E protein in the brain of mice. Vero cells were infected with JEV in vitro and given a certain concentration of sunitinib to observe the cell survival status. The expression of virus E protein in cells was detected by IFA. Results Continuous administration of sunitinib significantly improved the survival rate of infected mice. Survival rate and body weight changes showed that the 5-day's administration strategy was significantly better than the 10-day's administration strategy. The treatment of sunitinib decreased the infiltration of CD4+/CD8+T cells in the brain and reduced the changes of vascular sleeve. However, the variation of viral load and E protein expression in brain were not obvious. The cytopathic effect (CPE) of infected Vero cells were slightly relieved after giving sunitinib, and the expression of E protein was also slightly changed. Conclusion Sunitinib can significantly reduce the mortality of infected mice, and the 5-day's administration strategy is significantly better than the 10-day's administration strategy. Sunitinib decrease T lymphocyte infiltration in brain of mice, relieve the encephalitis symptoms ,and prolonged the life of mice.
