1.Effect of acute hypervolemic hemodilution on the onset and recovery of muscle relaxation induced by vecuronium

Objective To investigate the effect of acute hypervolemic hemodilution(AHHD)on the onset and recovery of muscle relaxation induced by vecuronium.Methods Thirty-two ASA Ⅰ orⅡpatients undergoing elective surgery under general anesthesia were randomly divided into 2 groups(n=16 each):control group and AHHD group.A loading dose of vecuronium 0.1 mg/kg Was given at 10 min after AHHD following tracheal intubatiom The muscular relaxation was maintained at Tl/Tc 5% to 15% by supplement with intravenous vecuroninm.Blood samples were taken at various times during AHHD for chemical analysis.The onset and recovery time of muscular relaxation were recorded.Results Compared with control group ,Hct,Hb and the concentration of TP and Alb were decreased,and the onset and recovery time of vecuronium were shortened in group AHHD(P<0.05).Conclusion Acute hypervolemic hemodihtion can shorten the onset and recovery of muscle relaxation of vecuronium in patients under general anethesia.

Objective To explore the clinical significance of the brain natriuretic peptide(BNP) in evaluating the cardiotoxicity caused by daunorubicin(DNR) through studying the changes of the plasma BNP levels in children with acute leukemia who accepted the chemotherapy with DNR.Methods Thirty-one children with acute lymphoblastic leukemia(ALL) admitted in the year of 2002-2004 underwent the chemotherapy in DVLP project.The plasma level of BNP was measured by enzyme linked immunosorbent assay(ELISA) and the left ventricular end diastolic diameter(LVEDD) by color Doppler respectively before and after the administration of DNR.Simultaneously,electrocardiography(ECG) and cardiac muscle enzymes(LDH1,CK-MB) were measured as routine.Results The plasma level of BNP increased from(3.97?2.41) ng/L to(18.25?7.63) ng/L(P
4.Correlation among apoptosis, expression of interleukin-1β and caspase-3 after spinal cord injury in rats

ObjectiveTo study the characteristic of apoptosis and the expression of interleukin-1β(IL-1β)and caspase-3, as well as the relation among them after spinal cord injury(SCI) in rats.MethodsAdult SD rats were divided into two groups randomly: sham group and spinal cord injury group. The rats were killed at 1 h, 8 h, 24 h, and 72h after surgery. The expression of IL-1β and caspase-3 was determined with immunohistochemistry and the apoptosis was labeled by TUNEL.ResultsThere was only a few cells that expressed low level IL-1β and caspase-3 in sham group, but they both enhanced in SCI group within 8 h after operation, and then decreased to the level similar to sham group 72 h after operation. The rates of TUNEL-positive cells were similar with that of IL-1β and caspase-3 after SCI. There was positive correlation among expression of IL-1β and caspase-3 and apoptosis index.ConclusionExpression of IL-1β and caspase-3 increase and a large number of TUNEL-positive cells present after SCI in rats, and there is positive correlation among them.
6.Effect of scalp acupuncture drawing method on motor function in hemiplegic patients following cerebral thrombosis at convalescent period.

OBJECTIVETo observe the effect of scalp-acupuncture drawing method (SADM) on recovering motor function in hemiplegic patients following cerebral thrombosis at convalescent period.
METHODSAdopting randomized, single-blinded, controlled method, 93 patients suffering from hemiplegia following cerebral thrombosis were randomly assigned to 3 groups, 31 in each group. All patients were treated based on neurological routine therapy, those in the treated group and the control group I were treated respectively with SADM and scalp-acupuncture twirling method (SATM) additionally, but for those in the control group II, no additional treatment was given. The treatment course for all was 4 weeks.
RESULTSBefore treatment, there were insignificant difference among 3 groups in the score of neural motor function deficits (NFDS) of limbs and activity of daily life (ADL) score (P > 0.05). After being treated for 4 weeks, the NFDS was significantly lower and in the control group II (P < 0.01, P < 0.05) and the change of the scores in the treated group was more than that in the control group I (P < 0.05).
CONCLUSIONSADM was superior to scalp-acupuncture twisting prove the ADL score and motor function of hemiplegia patients following cerebral thrombosis in the convalescent stage.

8.Simultaneous Determination of 11 Volatile Organic Compounds Metabolites in Human Urine Using High Performance Liquid Chromatography Coupled with Electrospray Ionization Tandem Mass Spectrometry

We developed and validated a method using high performance liquid chromatography ( HPLC ) coupled with electrospray ionization tandem mass spectrometry ( ESI_MS/MS ) to simultaneously quantify 11 urinary volatile organic compound ( VOC ) metabolites. The frozen urinary samples were unfrozen at room temperature and centrifuged to remove settled precipitate. 100 μL of the supernatant and 50 μL of internal standard were added and then diluted by 850μL of water so as to be analyzed by LC_MS/MS. The compounds were separated on a XSELECT HSS T3 column using acetonitrile as mobile phase A and 15 mmol/L ammonium acetate as mobile phase B at a flow rate of 250μL/min. The analytes were determined qualitatively and quantitatively under multi_reaction monitoring ( MRM) and negative polarity mode. The limits of detection ranged from 0 . 5-30 μg/L and the spiked recoveries of the analytes ranged from 89 . 8%-123 . 8% with RSDs of 3 . 1%-14 . 4%. This method was applied to 22 h urine samples collected from 16 smokers and 6 non_smokers. The results showed that the contents of the metabolites in smokerˊs urine were higher than those of nonsmokers, especially, smoking more than 25 cigarettes per day.
9.Construction and Characterization of a ppbR gene Mutant of Pseudomonas sp. M18

Pseudomonas sp. M18, one of plant-growth-promoting rhizobacteria, can produce secondary metabolites including phenazine-1-carboxylic acid (PCA) and pyoluteorin (Plt). PA2572 gene coding protein is a probable two-component response regulator in Pseudomonas according to homologous speculations. In order to investigate its genetic function, PA2572 homologous gene, ppbR, was amplified from M18 genome, inactivated by inserting a Gm cassette. The resulting reconstruct was introduced into the M18 genome using homologous recombination technique, so as to obtain the null mutant M18P. The results showed that the M18P has less flagellar swimming and swarming motility, and yielded fewer PCA. The production of PCA was only 50% of the wild type. However, there was no remarkable difference between mutant and wild type in producing pyoluteorin in KMB medium.
