The influence of inner cell mass (ICM) and trophectoderm (TE) score on pregnancy outcomes in frozen-thawed blastocyst transfer cycles was analyzed. A retrospective analysis of 741 cycles of frozen-thawed blastosysts transfer was performed. All cycles were divided into four groups based on the number and morphological score of blastocysts: S-ICM B/TE B group (n=91), the single blastocyst transfer of ICM B and TE B; D-ICM B/TE B group (n=579), double blastocysts transfer of ICM B/TE B; D-ICM B/TE C group (n=35), double blastocysts transfer of ICM B/TE C; and D-ICM C/TE B group (n=36), double blastocysts transfer of TE B/ICM C. The pregnancy outcomes were compared among the four groups. As compared with D-ICM B/TE C group, the clinical pregnancy rate, implantation rate and multiple pregnancy rate were increased in D-ICM B/TE B group (74.96% vs. 57.14%, 57.43% vs. 37.14%, and 48.62% vs. 25%, respectively, P<0.05 for all). Clinical pregnancy rate and implantation rate in D-ICM B/TE B group were also higher than in D-ICM C/TE B group (74.96% vs. 50%, and 57.43% vs. 33.33%, both P<0.05). Multivariable Logistic regression analysis indicated that ICM score was a better predictive parameter for clinical pregnancy (OR=3.05, CI 1.70-5.46, P<0.001), while the trophectoderm score was a better one for early abortion (OR=0.074, CI 0.03-0.19, P<0.001). Clinical pregnancy rate and multiple pregnancy rate in S-ICM B/TE B group were significantly lower than those in D-ICM B/TE B group (46.15% vs. 74.96%, and 2.38% vs. 48.62%, both P<0.05), but there was no significant difference in the implantation rate between the two groups. It was suggested that the higher score of ICM and TE may be indicative of the better pregnancy outcomes. The ICM score is a better predictor of clinical pregnancy than TE, while TE score is a better one in predicting early abortion. Single ICM B/TE B blastocyst transfer in frozen-thawed cycles can also get satisfactory pregnancy outcomes.

The activity of nano carbon fullerene lipidosome (NCFL) against influenza virus HINI in vitro was studied by observing the cytotoxicities and its activity rendered by different intensities of lighting with various periods of time. Rimantadine hydrochloride was used as the positive control drug. By using microcultural technique, the morphological changes of cells were observed and by using the gentian violet staining, antiviral activity of the NCFL against influenza virus was assayed. The results showed that: (1) The maximal concentration of the NCFL was 7μg/mL and the 50% toxic concentration (TC50) was 13.54μg/mL respectively; (2) NCFL had a significant activity of directly killing the influenza virus, while the activities in antiadsorption and antireplication were not obvious; (3) There was a dose-activity relationship between the dosages of NCFL and the direct killing effect against the influenza virus, and the periods of lighting-time could influence the activity partly. It was concluded that NCFL had a significant activity of directly killing the influenza virus.

The latest findings of our laboratory showed that Angelica sinensis polysaccharide (ASP) showed a definite effect in regulating the aging of hematopoietic stem cells. Leukemia is a type of malignant hematopoietic tumor in hematopoietic stem cells. There have been no relevant reports about ASP's effect in regulating the aging of leukemia cells. In this study, human acute myeloid leukemia (AML) KG1alpha cell lines in logarithmic growth phase were taken as the study object, and were divided into the ASP group, the cytarabine (Ara-C) group, the ASP + Ara-C group and the control group. The groups were respectively treated with different concentration of ASP, Ara-C and ASP + Ara-C for different periods, with the aim to study the effect of ASP combined with Ara-C in regulating the aging of human acute myeloid leukemia KG1alpha cell lines and its relevant mechanism. The results showed that ASP, Ara-C and ASP + Ara-C could obviously inhibit KG1alpha cell proliferation in vitro, block the cells in G0/G1 phase. The cells showed the aging morphological feature. The percentage of positive stained aging cells was dramatically increased, and could significantly up-regulate the expression of aging-related proteins P16 and RB, which were more obvious in the ASP + Ara-C group. In conclusion, the aging mechanism of KG1alpha cell induced by ASP and Ara-C may be related to the regulation of the expression of aging-related proteins, suggesting that the combined administration of ASP and anticancer drugs plays a better role in the treatment of leukemia .

OBJECTIVETo evaluate the therapeutic effect of Longjintonglin Capsules on type IIIA prostatitis accompanied by abnormal semen liquefaction.

METHODSWe selected 140 patients with type IIIA prostatitis accompanied by abnormal semen liquefaction according to the diagnostic standards of the American Institutes of Health (NIH) and treated them with Longjintonglin Capsules orally 3 capsules once tid for 12 weeks. We obtained the NIH Chronic Prostatitis Symptom Indexes (NIH-CPSI), traditional Chinese medicine (TCM) syndrome scores, leukocyte count in the expressed prostatic secretion (EPS), semen liquefaction time, and the results of semen analysis and compared these indicators before and after the treatment.

RESULTSOf the 140 cases, 132 were included in this study, excluding 8 due to their incomplete case histories. Before and after 4, 8 and 12 weeks of medication, the total NIH-CPSI scores were 24.52 ± 5.43, 21.28 ± 4.85, 18.01 ± 4.28, and 14.49 ± 3.65 (P < 0.01), the TCM syndrome scores were 35.63 ± 6.07, 26.66 ± 5.03, 17.37 ± 4.18, and 11.11 ± 3.96 (P < 0.01), and the leukocyte counts (/HP) were 27.50 ± 7.01, 22.38 ± 5.22, 16:76 ± 4.10, and 11.40 ± 4.74 (P < 0.01), respectively. After 12 weeks of treatment, 31 of the patients with type IIIA prostatitis were cured and another 72 well responded, with an overall response rate of 78.0%. Of those with abnormal semen liquefaction, 61 were cured, 39 well responded, and 32 failed to respond, with an overall effectiveness rate of 75.8%. Semen analysis showed significantly increased percentage of progressively motile sperm after 4, 8 and 12 weeks of medication as compared with the baseline (P < 0.01). No abnormal liver or renal function or other adverse reactions were observed during the treatment.

CONCLUSIONLongjintonglin Capsules, with its advantages of safety, effectiveness and no obvious adverse effects, deserve to be recommended for the treatment of type IIIA prostatitis accompanied by abnormal semen liquefaction.

Objective To study the effect mechanism of momordica charantia total saponin ( MCTS)′s ameliorating insulin resistance through discussing MCTS′s influence of protein kinase B ( Akt -2 ) and adenosine monophosphate activated protein kinase ( AMPK ) in type 2 diabetes ( T2DM ) rats.Methods T2DM models were built with the method of higher lipid and sugar diets and injection of a low dose of streptozotocin.Rats are divided into normal group, model group, control group and three dosage test groups.The positive control group and three dosage test groups rats were administrated with 2 mL metformin (50 mg· kg-1) and MCTS (100, 200, 400 mg· kg -1) per day for 8 weeks.Fasting serum glucose and insulin were detected with biochemical analyzer and ELISA method.The Akt-2 and AMPK′s mRNA and the activated protein were measured by RT-PCR and Western blot method.Results The homeostasis model assessment of insulin resistance ( HOMA-IR) reduced and the mRNA and protein expression amount of Akt-2 and AMPK increased obviously in all test groups compared to the model group.Conclusion The ameliorating insulin resistance mecha-nism of MCTS maybe caused by up-regulating the mRNA and protein of Akt-2 and AMPK.

Objective To investigate the evidence of the knowledge, attitudes and behaviors of nurses to prevent falls of inpatients and to analyze the correlation among them. Methods The self-designed questionnaire about knowledge, attitudes and behaviors of nurses to prevent falls of inpatients was used to investigate nurses from a hospital of Grade Ⅲ Level A in Beijing. Results Scores of related knowledge to prevent falls of these nurses were in a better level(standard score 88.25 ± 8.97); the scores of knowledge to prevent falls without walking aids were relatively higher ( standard score 98.08± 6. 44), and the scores of walking aids-using-knowledge were relatively lower(standard score 69.98 ± 25.29), attitude towards prevention of falls were neutral, and scores of behaviors to prevent falls of these nurses were in a better level(standard score 87.49 ±7.82). Positive correlation was shown among attitudes, knowledge and behaviors to prevent falls. (r =0.179、0. 200, P ＜0. 05), however,there were no correlation between scores of knowledge and behaviors to prevent falls. Work experience and scores of knowledge to prevent falls were negatively correlated (r= -0.191, P＜0.05 ). Scores of attitude to prevent falls showed significant difference between the group of nurses who had learnt knowledge to prevent falls and group of those who had never encountered falls ( P ＜ 0. 05 ). Good department atmosphere had a significant impact to nurses' knowledge and behaviors to prevent falls (P＜ 0.05 ). Conclusions Nurses had inadequate cognition of walking aids-using-knowledge; attitudes towards prevention of fall were neutral; they had appropriate acts to prevent the falls; good department atmosphere can enhance the behaviors of nurses to prevent falls.

BACKGROUNDIt is known that excessive release of glutamate can induce excitotoxicity in neurons and lead to seizure. Dexamethasone has anti-seizure function. The aim of this study was to investigate glutamate-dexamethasone interaction in the pathogenesis of epilepsy, identify differentially expressed genes in the hippocampus of glutamate-induced epileptic rats by mRNA differential display, and observe the effects of dexamethasone on these genes expression.

METHODSSeizure models were established by injecting 5 microl (250 microg/microl) monosodium glutamate (MSG) into the lateral cerebral ventricle in rats. Dexamethasone (5 mg/kg) was injected intraperitoneally at 30 minutes after MSG inducing convulsion. The rats' behavior and electroencephalogram (EEG) were then recorded for 1 hour. The effects of dexamethasone on gene expression were observed in MSG-induced epileptic rats at 1 hour and 6 hours after the onset of seizure by mRNA differential display. The differentially expressed genes were confirmed by Dot blot.

RESULTSEEG and behaviors showed that MSG did induce seizure, and dexamethasone could clearly alleviate the symptom. mRNA differential display showed that MSG increased the expression of some genes in epileptic rats and dexamethasone could downregulate their expression. From more than 10 differentially expressed cDNA fragments, we identified a 226 bp cDNA fragment that was expressed higher in the hippocampus of epileptic rats than that in the control group. Its expression was reduced after the administration of dexamethasone. Sequence analysis and protein alignment showed that the predicted amino acid sequence of this cDNA fragment kept 43% identity to agmatinase, a member of the ureohydrolase superfamily.

CONCLUSIONSThe results of the current study suggest that the product of the 226 bp cDNA has a function similar to agmatinase. Dexamethasone might relax alleviate seizure by inhibiting expression of the gene.

OBJECTIVETo evaluate in vitro anti-tumor effect of chemotherapeutic drugs on human gastric cancer cells, and investigate the relationship with Bcl-2 expression.

METHODSSingle cell suspension was prepared from fresh gastric cancer tissue and exposed to taxol (Tax), 5-fluorouracil (5-FU), cisplatin (CDDP), adriamycin (ADM), mitomycin (MMC) respectively for 48 hours. Metabolic activity and inhibitory rate of cells were detected by MTT assay. Expression of Bcl-2 was examined with immunohistochemistry.

RESULTSThe inhibitory rates of cancer cells exposed to chemotherapeutic drugs were different and Tax, 5-FU, CDDP had remarkably higher rates than ADM and MMC. The lower differentiated gastric cancer cells were more sensitive than the higher ones. Positive expression rate of Bcl-2 was 80% and the positive cells showed resistance to 5-FU, ADM and MMC.

CONCLUSIONSChemosensitive testing by MTT assay can constitute the prediction for the application of chemotherapeutic drugs individually. Overexpression of Bcl-2 may contribute to multiple drug-resistance of tumors.

Objective:To describe the prevalence of anxiety disorders and distribution in Chifeng City,and explore the related risk factors and health service seeking.Methods:Individuals aged 18 years and over were sampled by stratified,probability-proportional-to-size sampling,randomized cluster sampling in Chifeng City through November,2010 to April,2011.All respondents were interviewed by trained interviewers face-to-face.The Composite International Diagnostic Interview-3.0 Computer Assisted Personal Interview (CIDI-3.0-CAPI) was used to diagnose anxiety disorders according to the criteria and definition of the Diagnostic and Statistical Manual of Mental Disorders,Fourth Edition (DSM-Ⅳ).Single and multivariate analysis were applied to explore risk factors of anxiety disorder.Results:Totally 4528 subjects were interviewed with CIDI-3.0-CAPI.The 12-month prevalence rate of anxiety disorders was significantly higher in rural area than in urban area (4.62％ vs.2.61％,P ＜0.01).The rate was significantly higher in female than in male (4.92％ vs.2.82％,P ＜0.01).The rate was significantly higher in the unmarried,the divorced and the widowed than in the married (5.91％ vs.3.70％,P ＜0.01).Logistic regression analysis indicated that dwelling in urban areas and education level of 7-9 years were the protective factors of anxiety disorders,while being female,unmarried,divorced or widowed were the risk factors of anxiety disorders.It was found that only 3.1％ of the patients with anxiety disorders reported to seek help from psychiatrists.Conclusion:The anxiety disorder is a common mental disorder in Chifeng City,while few people with anxiety disorders seek mental health professionals.Therefore,it should be paid attentions to anxiety disorder,especially in rural area,women,and low educated people for enhancing mental health service and knowledge popularization.
