1.Effects of domestic recombinant adenovirus p53 injection combined with cisplatin on growth and apoptosis of ovarian carcinoma cell lines

Objective:To evaluate the effects of domestic recombinant adenovirus p53 injection (rAd-p53,Gendicine) combined with cisplatin (DDP) on growth and apoptosis of ovarian carcinoma cell lines. Methods:MTT assay, flow cytometry, and Western blotting were used to compare the effects of rAd-p53, DDP and their combination on the growth, cell cycle, apoptotic rate, and p53 protein expression of ovarian carcinoma cell lines. Results:rAd-p53 inhibited ovarian carcinoma cells proliferation in dosE- and timE-dependent manners. After combined treatment, the growth inhibition rate was significantly increased (P<0.01). There was no significant diffE-rence in the inhibition potency induced by combined treatment with different sequences (P>0.05). Combined treatment significantly arrested ovarian carcinoma cell lines in G0/G1 phase at 72 h, the cell number in S phase was significantly decreased and the apototic rate was significantly increased (P<0.01). High expression of p53 protein was detected in ovarian carcinoma cell lines at 72 h after administeration of rAd-p53. Conclusion:rAd-p53 tranfects the exogenous widE-type p53 into the genome group of human ovarian tumor cells, induces p53 protein expression, thereby arresting tumor cells at G0/G1 phase, inhibiting tumor growth, and inducing apoptosis.

Objective:To study the effects of parathyroid hormone(PTH) upon dentinogenesis by investigating the effects of hPTH1-34 on the development and calcification of parathyroidectomy rats' dentin. Methods: hPTH1-34 were injected into SD rats at different dosages after parathyroidectomy. Six weeks later lower incisors were dissected and prepared for light microscopy and image analysis. Results:The concentration of serum calcium decreased after parathyroidectomy, while increased after being injected hPTH1-34. Background Sustained abnormal tear secretion in dry eye patients may lead to ocular surface and lacrimal glands in the state of long-term inflammatory cell infiltration.Lacrimal gland suffers immune attack by lymphoproliferative,and inflammation interferes normal gland secretion,in which chemokines and their receptors on lymphocytes play a key role.Objective This study was to investigate the inflammation mechanism of delayed allergy induced by Th1 cell in the development of dry eye.Methods Sixty eyes of 30 patients with dry eye and matched 30 healthy volunteers were enrolled in Affiliated First Hospital of Xinjiang Medical University from June to December in 2012.Schirmer Ⅰ test (S Ⅰ t),break up time (BUT) of tear and corneal fluorescence stain (FLS) were performed on the subjects,and conjunctiva epithelial cells were obtained using cytological method of conjunctiva imprinting.Positive cell rates of CC chemokine receptor 5 (CCR5) and CXC chemokine receptor 3 (CXCR3) were detected by flow cytology,and the relative expression levels of regulated upon activation of normal T cells exp ressed and secreted (RANTES),macrophage inflammatory protein (MIP)-lα and MIP-1β,monokine induced by interferon-γ (IFN-γ) (MIG),interferon-γ inducible protein 10 (IP10) and interferon-inducible T-cell α chemoattractant (I-TAC) mRNA were quantitatively assayed by real-time PCR.Differences of the positive rates of CCR5,CXCR3 and lignds were compared between dry eye group and normal control group.Relationship between the positive rates of CCR5,CXCR3 and BUT,S Ⅰ t,FLS scores was analyzed.Results The values of BUT,S Ⅰ t were (2.90±1.37) seconds and (4.00±2.49) mm/5 minutes in the dry eye group,which were significantly lower than (8.56±4.69) seconds and (11.31 ±5.23) mm/5 minutes in the normal control group (t =3.172,2.186,both at P<0.05).FLS scores,positive rates of CCR5 and CXCR3 were0.90±0.57,(3.38±0.66) % and (2.64±0.47)% in the dry eye group,showing significant elevations in comparison with 0.14±0.06,(2.12±0.21) % and (1.12±0.11) % in the normal control group (t=2.297,3.151,5.454,all at P<0.05).In the dry eye group,the masculine rate of CCR5 was negatively correlated with BUT and S Ⅰ t (r=-0.473,-0.385,both at P<0.05).The masculine rate of CXCR3 was negatively correlated with BUT and S Ⅰ t (r =-0.753,-0.684,both at P<0.05).No considerable correlations between the positive rate of CCR5 with the positive rate of CXCR3 or FLS scores (r =0.231,0.336,both at P<0.05).The relative expression levels of RANTES,MIP-1α,MIG and IP10 mRNA in the dry eye group were significantly higher than those in the normal control group (t =3.091,2.894,2.688,2.245,all at P<0.05),but there were no significant differences in the relative expression levels of MIP-1β and I-TAC mRNA between the two groups (t =0.512,1.979,both at P>0.05).The positive correlations were seen between the masculine rate of CCR5 with the relative expression of RANTES or MIP-1α mRNA (r=0.473,0.285,both at P<0.05),but there was no obvious correlation between the masculine rate of CCR5 and the MIP-1 β mRNA expression(r=0.214,P>0.05).In addition,the masculine rate of CXCR3 was positive correlated with the expressions of MIG and IP10 mRNA (r=0.553,0.314,both at P<0.05),whereas the masculine rate of CXCR3 was not related to the expression of I-TAC mRNA (r=0.364,P>0.05).Conclusions Dry eye is probably along with the long-term infiltration of inflammatory cells.The delayed allergy induced by Th1 cells and the nature killed cells is probably the primary cause to xerophthalmia.CCR5,CXCR3 and their ligands might be the regulative targets in the inflammation mechanism of dry eye.

The tongue and its surrounding structures have special anatomical and functional characters and how to restore its shape and function after glossectomy (especially total glossectomy) still remains a difficult problem. The authors devised a new method of repair by using a forearm free flap with results in 16 out of 17 patients. The selection and shaping of the flap, the evaluation of the recipient site and its blood vessels, the prevention of the oral infection,the monitoring of the circulatory status of the flap and the evalution of the operative result were discussed.
7.Accuracy of end-tidal carbon dioxide in reflecting arterial carbon dioxide during mechanical ventilation via laryngeal mask airway in children

Objective To investigate the accuracy of end-tidal carbon dioxide (PETCO2) in reflecting arterial carbon dioxide (PaCO2) during mechanical ventilation via laryngeal mask airway (LMA) in children. Methods Fifty-two ASA Ⅰ patients, aged 2-9 yr, weighing 10-30 kg, undergoing orthopaedic surgery under general anesthesia, were randomized into 2 groups (n = 26 each) : LMA group and endotracheal tube (ETT) group. After anesthesia was induced with fentanyl, propofol and succinycholine, LMA or ETT was inserted and the children were mechanically ventilated. After the hemodynamics was stable, arterial blood samples were obtained to detect PaCO2, and PETCO2 was recorded simultaneously. Results There was no significant difference in PaCO2 and PCT CO, between groups LMA and ETT ( P > 0.05) . There was no significant difference between PaCO2 and PETCO2 in LMA group (P > 0.05). Conclusion When mechanical ventilation is performed via LMA in children, PETCO2 can reliably reflect PaCO2 and guide the regulation of ventilatory parameters.
8.The effect of the expression and influence of MMP-9 on prognosisof nasal polyps

Objective:To study the expression of MMP-9 and eosinophils in nasal polyps and its influence on the prognosis. Method:Study the expression of MMP-9 and EOS on the tissue of nasal polyps of patients who were cured or recurrent and normal nasal mucosa(NM) by immunohistochemisty and HE staining. Result:In the patients of recurrent group (Group B) , the expression of MMP-9 was much stronger than that of good prognosis group (Group A) and NM. The MMP-9 was stronger in the eosinophil nasal polyps than that of non-eosinophil groups. The percentage of EOS in Group A was 16.67% , while 62.50% in the Group B. Conclusion:The stronger expres-sion of MMP-9 on Group B and eosinophil group indicated that MMP-9 might affect the prognosis of nasal polyps through inducing the number of EOS in the mucosa. MMP-9 may be one of the risk factors for the relapse of nasal polyps.

Glaucoma,especially common primary open-angle glaucoma and primary angle-closure glaucoma,shows high genetic heterogeneity. The causing-disease genes known are difficult to explain some glaucomatous cases,and the study on the susceptibial gene of glaucoma do not achieve new breakthroughs. Combined with the newest progression in genetic study of glaucoma,some views were proposed in this paper in order to better understand the updating study of the pathogenesis mechanism.
10.Health self-management club for hypertensive patients in Gushan town of Fuzhou

Objective To evaluate the impacts of health self-management club on hypertensive patients who lived in Gushan town of Fuzhou.Methods Thirty-three community hypertensive patients who partitipated in health self-management club during July and October 2010 were enrolled in this investigation.Control and awareness of high blood pressure and patients' health condition were compared before and after the intervention.Results Normal blood pressure (systolic pressure < 140 mm Hg (1 mm Hg =0.133 kPa),diastolic pressure <90 mm Hg) was found in 19 patients after intervention ((19/31 vs 16/33),x2 =1.058,P > 0.05).Meanwhile,awearness rate of normal blood pressure and mediciation order-guided drug taking was increased to 27/33 and 26/31,respectively (x2 vales were 0.047 and 1.242,respectively ; both P > 0.05).Other information inluding annual blood pressure measurement in > 35 years old populations was well known by more participants (x2 vales were 10.810,4.280,4.039,14.249,10.573and 14.539,respectively; all P< 0.05).Since intervention,blood pressure was measured at least once a month in all the subjects,and 28/31 patients measured blood pressure weekly (x2 =11.532,P < 0.05).Health condition of the patients were significantly improved after intervention (Z =-2.034,P < 0.05).Conclusions Community health education could raise patients' awareness of hypertension and improve their health condition.Health self-management club may be feasible to hypertensive patients living at rural-urban fringe.
