1.Effects of fuling shengmai yin on superoxide dismutase, malondialdehyde and lipofuscin level of rat hippocampal neurons in vitro

BACKGROUND: Deposition of lipofuscin (LF), decrease of superoxide dismutase (SOD) level and increase of malondialdehyde (MDA) level are important manifestations of senility. Some Chinese medicinal herbs have obvious advantage in delaying senility.OBJECTIVE: To explore the effects of fuling shengmai yin (FLSMY)serum on SOD, MDA and LF level of rat hippocampal neurons in vitro with vitamin E serum and blank serum as controls.DESIGN: Randomized controlled study.SETTING: Experimental Animal Department of Tongji Medical College,Huazhong Science and Technology University; Cell Culture Center of Anatomy Teaching and Research Section of Tongji Medical College,Huazhong Science and Technology University; Scientific Research Department of Wuhan Municipal Hospital of Traditional Chinese Medicine.MATERIALS: From October 2000 to February 2001, the experiment was conducted at the Experimental Animal Department of Tongji Medical College as well as Cell Culture Center of Anatomy Teaching and Research Section of Tongji Medical College, Huazhong Science and Technology University. Six Japanese male white rabbits with big ears, fed normally and weighting (2.5±0.1)kg, were divided into three groups with 2 in each group.Thirty newly born SD rats aged 1-2 days, of clean grade and either gender,were selected.METHODS: [1] Hippocampal neurons were inoculated to 24-well culture plates with the density of 2.5×105/L. Cells in each well were randomized into three groups: A. FLSMY group (fuling shengmaiyin composed of Poria,Radix Ginseng, Radix Ophiopogonis and Fructus Schisandrae; each milliliter contained 1.5 gram of the herbs); B. drug control group (serum containing vitamin E); C. blank control group (blank serum). Each group had 8 wells. After 24 hours, 1 mL serum containing FLSMY, 1ml serum containing vitamin E, or 1 mL blank serum was added into each plate well of corresponding groups. [2] On the 6th day of cell culture, XO-HPX reaction system was added to induce cell injury, in which senility model was established by producing hyperoxide anion free radicals. [3] In the process of cell culture different drug was added to the corresponding different group plate well during replacement of culture fluid each time. [4] The level of SOD, MDA, and LF was detected after 15 days of culture.MAIN OUTCOME MEASURES: Level of SOD, MDA and LF in rat hippocampal neurons.RESULTS: SOD level was higher in FLSMY group than in blank control group (P < 0.01), whereas MDA level in FLSMY group was lower than that in vitamin E group (P < 0.05); LF level was lower in FLSMY group than in blank group (P > 0.05).CONCLUSION: FLSMY as a drink is composed of traditional Chinese medicinal herbs: Poria, Radix Ginseng, Radix Ophiopogonis and Fructus Schisandrae; each has the properties of anti-oxidation and eliminating free radicals. FLSMY can strengthen SOD activity, lower MDA level, and inhibit LF formation. Therefore, FLSMY can delay senility through protecting neurons.
2.Sterilization effect of Er:Yag laser irradiation combined with 2% chlorhexidine gel on single tube premolar Enterococcus faecalis infection model

Objective To evaluate the sterilization effect of Er:Yag laser irradiation combined with 2%chlorhexidine gel on single tube premolar enterococcus faecalis infection model.Methods After disinfection test was performed without molars bacterial infection,200 single rooted premolar root canal were selected.10μL enterococcus faecalis( ATCC 29212) bacterium suspension were inoculated into single intratracheal with high pressure sterilization and and then were cultured for 2 weeks.200 infected premolar model were divided into four groups:control group (n=50),A group (n=50),B group (n=50) and C group (n=50).The control group did not receive any disinfec-tion treatment,group A received Er:Yag laser irradiation and 2%chlorhexidine gel disinfection,B group received only Er:Yag laser irradiation,the C group received only 2% chlorhexidine gel disinfection.After disinfection,premolars were put into 10mL sterile water,mixed thoroughly,and then 1mL liquid were taken and diluted by10 times.Then the 200μL dilutions lipuid were coated on BHI agar plate,and plate could be selected for counting colony count.Results The average number of colonies was(127 ±12) in the control group,average number of group A(10 ±3) and which was lower than that group C (48 ±6)(t=12.005,P=0.000) and group C (43 ±5) (t=19.277,P=0.000). There were only 2 cases,which had obvious ablation phenomena in group A.Conclusion Disinfection effect of Er:Yag laser irradiation combined with 2%chlorhexidine gel is better than that of Er:Yag laser irradiation or 2% chlo-rhexidine gel disinfection.The incidence rate of the ablation phenomenon was low.
3.Efficacy of Venlafaxine and Reboxitine in treatment of selective serotonin reuptake inhibitors non-responder with major depressive disorder

Objective The purpose of this study was to access the efficacy of Venlafaxine and Reboxitine on treatment of depressive patients who had not responded to selective serotonin reuptake inhibitors for eight weeks. Methods 117 cases depressive patients who had not responded to selective serotonin reuptake inhibitors for eight weeks were divided into three Venlafaxine group(n =41),Reboxitine group(n =39),Original medicine group(n =37) by randomly and were treated by corresponding drugs for 8 weeks. The therapeutic response and safety were evaluated by scale of HAMD and TESS at before and after treatment the end of 1,2,4,8 week. Results The score of HAMD in Venlafaxine and Reboxitine groups were significantly lower than Original medicine group at the end of 4,8 weeks after treatment and the score of HAMD in Venlafaxine group were significantly lower than Reboxitine group at the end of 8 weeks after treatment. The score of HAMD in Venlafaxine and Original medicine groups were marked descendanter than before treatment from after treatment second weekend(P <0.01),while reboxitine group did not marked descent until the 4th weekend after treatment. The reduce score of factors in cognition,retardition and desperation of Venlafaxine and Reboxitine groups were significantly higher than Original medicine group at the end of treatment. The curative effect of Venlafaxine group were supper than Original medicine group. The side reaction of three groups were not difference. Conclusions There are obviously curative effect using Venlafaxine to treat the depressive patients who had not responded to selective serotonin reuptake inhibitors. The time of initial effect in Venlafaxine were rapider than Reboxitine group.
4.Preliminary study of the establishment of the system of statistic index of health examination information

Objective To establish the system of statistic index of health examination information. Methods The items of health examination were screened, the valuable indexes for guiding the health condition were selected and standardized, and then the system of statistic index of health examination information was constructed. Results The 42 items for male clients and 74 items for female clients were selected as the statistic index of health examination information system. Standardized the statistic index with international classification of diseases system, including the criterion of name, diagnosis and data collection.The standard statistical tables were constituted. Conclusions Health examination information system can be statistically added up by standard index system.
5.The clinical research about the relationship between serum levels of 8-hydroxy-deoxyguanosine (8-OHdG) of type 2 diabetes patients and its macrovascular complications

Objective To investigate the role of oxidative stress in type 2 diabetes patients with macrovascular complications. Methods Serum levels of 8-hydroxy-deoxygnan osine (8-OHdG) and hemoglobin Alc (HbAlc) in 32 cases of type 2 diabetic patients with macrovascular complications and 46 cases of patients without complications were determined. 8-OHdG was detected by enzyme linked immu-nosorbent assay (ELISA). Results The level of 8-OHdG (2.93±1.37) ng/mL in type 2 diabetic patients with macrovascular complica-tions was higher than those without complications, whose 8-OHdG level was (2.67±1.30)ng/mL, but the difference was not significant(P>0.05). Compared with all kinds of macrovascular complications in patients, the 8-OHdG levels were increased, but the difference were not significant (P>0.05). Compared with patients without complications, the duration and age of the patients with macrovascular compli-cations also increased significantly (P<0.05), but the HbAlc levels were not statistical difference between them (P>0.05). Spearman regression analysis showed that 8-OHdG level was positively correlated with the duration (r=0.33, P<0.05), while the age and HbAlc level had no obvious correlation (P>0.05). Conclusion The role of oxidative stress may not be the important reason in type 2 diabetes with macrovascular complications, but the duration and age of the patients may be closely related with the disease.
6.Clinical features and long-term follow-ups of juxtapapillary retinal capillary hemangioma

Objective:To evaluate the clinical features and follow-up effect of treatment for retinal capillary hemangioma (RCH) of myopic disc.Methods:Retrospective case series studies. A total of 14 patients (15 eyes) diagnosed with juxtapapillary RCH (JRCH) at Peking Union Medical College Hospital and followed up for more than 12 months from January 2007 to December 2019 were included. Among these patients, there were 7 males (7 eyes) and 7 females (8 eyes) with the average age of 29.8±12.7 years. There were 6 isolated JRCH cases, all of which were monocular patients, including 1 case with a single peripheral RCH; 8 cases with von Hippel-Lindau disease, including 5 cases of binocular RCH. Out of 15 eyes, 6 eyes were merged with peripheral RCH. Three eyes were simply observed, and 12 eyes were treated with photodynamic therapy (PDT) alone or PDT combined with other treatments. The average follow-up time was 55.2 months. Its clinical characteristics and long-term treatment effects were analyzed.Results:At the first visit, among the 15 eyes, 3 eyes had visual acuity <0.1, 5 eyes were 0.1-0.4, and 7 eyes were> 0.5. The endogenous and fixed exogenous JRCH were 12 (80%, 12/15) and 3 (20%, 3/15) eyes, respectively. The size of the tumor was 1/4 to 4 optic disc diameters (DD); the combined macular edema, epiretinal membrane, and macular hole were 11, 5, and 1 eyes, respectively. There were 3 eyes with tumor diameter less than 1 DD. The visual acuity at the first diagnosis was 0.6-1.0, and no treatment was given. The follow-up time was 12 months in 2 eyes and 120 months in 1 eye. At the last follow-up, his vision remained stable and the tumor did not enlarge. Six eyes with tumor diameter ranging from 1.0 to 2.5 DD, visual acuity was 0.06 to 0.6 at first diagnosis, accompanied by retinal exudation. 5 eyes were treated with PDT alone, and 1 eye was treated with PDT combined with anti-vascular endothelial growth factor drugs. The follow-up time was 12 to 84 months. At the last follow-up, the visual acuity improved or stabilized in 5 eyes and decreased in 1 eye. There were 6 eyes with peripheral RCH, including 4 eyes with retinal detachment, 2 eyes with epiretinal membrane, and 1 eye with vitreous hemorrhage. The visual acuity at the first visit was 0.02-0.6. All patients were treated with PDT combined with transpupillary thermotherapy, extrascleral drainage, and vitrectomy. The follow-up time was 12 to 132 months. At the last follow-up, all eyesight decreased.Conclusions:Visual decrease cause by JRCH often occurs at young adults. Bilateral JRCH are more common in patients with von Hippel-Lindau's disease. The tumor can cause retinal exudation, macular edema, epiretinal membrane and macular hole, resulting in gradual vision loss. PDT and/or combined anti-vascular endothelial growth factor drugs for medium sized JRCH with retinal exudation can maintain long-term vision stability. Patients with large JRCH or severe complications such as retinal detachment tend to have poor prognosis.
7.Analysis of Arsenic Metabolites of Realgar in Rat Viscera by HPLC-ICP-MS

HPLC-ICP-MS method was established to separate AsB, As(Ⅲ), monmethyl arsenic ( MMA ) , dimethyl arsenic ( DMA ) and As(V) in rat viscera. Ultrasonic water bath extraction was used for sample pretreatment. Dionex IonPac AS19 anion-exchange column and 20 mmol/L (NH4)2CO3(pH=9. 7) were used to analyze arsenic species in rat liver and kidney after realgar infected. The results showed that the method was not interfered by 40 Ar35 Cl+ peak, the linear ranges for five arsenic species were all between 1 and 300 μg/L with linear coefficients more than 0. 9990, and the detection limits were between 0. 3 and 0. 5 μg/L. The relative standard deviations ( RSDs) for the determination of five species were less than 5% and the recoveries were between 83. 8% and 111. 7%. After speciation analysis, DMA, As(Ⅲ) and unknown compound 2 were found in liver;DMA, MMA, As(Ⅲ), unknown compound 1, unknown compound 2 and unknown compound 3 were found in the kidney. The results indicated that this method could be used for arsenic species analysis of realgar metabolism.
8.Optimal concentration of sodium taurocholate inducing acute necrosis pancreatitis in a rat

Objective To explore the dose-effect relationship and optimal concentration of sodium taurocholate (NaTc) to establish acute necrosis pancreatitis (ANP) complicating MODS in a rat. Methods 280 SD rats were randomly divided into control group (n =40) and 3.5%, 4%, 4.5%, 5% ANP group (n = 60, respectively). ANP groups were induced by retrograde injection of NaTc solution into the pancreatic duct of rats. Rats in each model group were sacrificed at 1, 3, 6, 12 and 24 h after the model induction. Serum amylase, serum total bilirubin, creatinine, blood gas analysis, and pathological changes of pancreas were determined. Survival rate at 24 h after the model induction was observed in other 20 ANP rats in each group. Results In each groups, the serum amylase level, creatinine, blood gas analysis and pathological changes all increased corresponding to the increase of NaTc, and there was significant dose-effect relationship and time-effect. In 4.5% group, the dysfunction of liver and kidney reached the peak at 6 h,while the respiratory dysfunction reached its peak at 12 h. Its incidence of MODS was 52.5%, which was significantly higher than those in 3.5%, 4.0% group (15.0% and 37.5%) ;and 24 h survival rate of 4.5% group was 65%, which was significantly higher than that in 5% group (30%). Conclusions 4.5% NaTc solution may be the optimal concentration to establish the ANP model, which had higher incidence of MODS and 24 h survival rate.
9.Time-course changes of intra-abdominal pressure and plasma TNF-α in rats with acute necrotizing pancreatitis

Objective To investigate the time-course change of intra-abdominal pressure (IAP), TNF-α in rat with acute necrotizing pancreatitis (ANP) and its mechanism. Methods 80 SD rats were randomly divided into ANP group and control group. ANP group was induced by retrograde injection of 4.5% sodium taurocholate into the pancreatic duct in SD rats, while control group was induced by injection of saline at the same dosage. Eight rats in each group were killed at 1 h, 3 h, 6 h, 12 h and 24 h, respectively. The serum levels of amylase, TNF-α were determined;IAP, amount of abdominal ascites, pancreatic pathologic changes were evaluated. Results The levels of amylase in ANP group increased progressively with time, which were 32-folds higher than those in control group at 24 h. The serum levels of TNF-α in ANP group reached the peak at 6 h, and were significantly higher than those in the control group. The amount of abdominal ascites increased post-ANP induction and reached the peak at 24 h, which was 4.7-folds higher than that in the control group. The IAP significantly increased 1 h after induction, which was 3 times as high as the control group in 3 h group and 9 times in 12 h group. Pathologic injuries deteriorated progressively and reached the peak at 24 h. Conclusions Significant IAP occurred in ANP rats and reached the peak at 12 h, TNF-α reached the peak at 6 h, therefore IAP may be the secondary response to inflammatory reaction.
