1.Therapeutic Effect of Reduning Injections in the Treatment of 52 Cases of Infantile Rotavirus Enteritis

Objective:To investigate the clinical efficacy and safety of Reduning injections in the treatment of infantile rotavirus enteritis. Methods:Totally 102 cases of infantile rotavirus enteritis in our hospital from June 2012 to June 2013 were randomly divided into two groups, the control group had 50 patients, and the treatment group had 52 cases. Both groups were given conventional symp-tomatic treatment, the control group was given 10 mg·kg-1 ribavirin injections in 100ml glucose injections for intravenous infusion, qd, and the treatment group was given 0. 5 ml·kg-1 Reduning injections in 100ml glucose injections for intravenous infusion, qd. Ev-ery 3-day was a course of treatment. The clinical curative effect of the two groups was observed, and the clinical symptoms, hospitaliza-tion time and adverse reactions were also studied. Results:After the treatment, the total effective rate in the control group was 70. 0%, while that in the treatment group was 92. 3%, and there was significant difference between the two groups (P<0. 05). The clinical symptom disappearance time and hospitalization time in the treatment group were significantly lower than those in the control group with statistically significant difference (P<0. 05). During the treatment, the control group had 5 cases of leucopenia and 2 cases of skin rash, and the patients all recovered after withdrawal of the drug, while the treatment group showed no obvious adverse reaction. Con-clusion:The curative effect of Reduning injections in the treatment of infantile rotavirus enteritis is promising, which can effectively improve the clinical symptoms, shorten the course of disease and reduce adverse reactions with the value of clinical application.
2.Characteristics of Removing Disinfection By-Product Precursors by Conventional Processes

Objective Characteristics of removing disinfection by-product precursors by conventional processes were studied in a certain city that lives on surface water. Methods Gas chromatography was employed to determine trihalomethane and haloacetic acids in water. Sampling time was from January to April in 2003 and there were 5 sampling sites each time. Results Trihalomethane formation potential(THMFP) in the source water was 15.00-39.83 ?g/L and in the supply drinking water it was 13.42-13.85 ?g/L. It showed that the conventional water treatment processes played a positive role in controlling THMFP. Haloacetic acids formation potential(HAAFP) in source water changed from 61.79 ?g/L to 78.72 ?g/L and the average removing rate of HAAFP by conventional processes was 44.13%. Conclusion Further studies are needed about organic matters in different kinds of source water in different areas and reactivity of DBP precursors with chlorine and DBPs generating ability. All these studies will contribute to control DBPs in drinking water.
3.Fenofibrate inhibits production of reactive oxygen species and oncofetal fibronectin induced by short-term intermittent high glucose through blockade of local aldosterone system in human mesangial cells

Objective To investigate the role of local aldosterone system in oncofetal fibronectin ( oncofetal FN) mRNA expression and reactive oxygen species (ROS) production in human mesangial cells (HMCs) exposed to short-term intermittent high glucose and the effect of Fenofibrate.Methods The HMCs were divided into 8 groups:normal glucose(NG) ;osmotic fluctuation(OF) ;mean glucose load (MGL) ;stable high glucose (SHG),short-term intermittent high glucose (IHG) ; intermittent high glucose plus eplerenone (IHGE) ; intermittent high glucose plus fenofibrate(IHGF) ; and normal glucose plus fenofibrate (NGF) groups.The mRNA expression levels of Aldosterone synthase ( CYP11 B2 ),11 β-hydroxysteroid dehydrogenase type 2 ( 11βHSD2 ) and oncofetal FN were determined by RT-PCR.The expression of CYP11B2 protein was determined by western-blot.Aldosterone level in cell culture supernatant was detected by radioimmunoassay.The expression and translocation of mineralocorticoid receptor (MR)protein were assayed with confocal laser scanning microscopy. ROS levels were determined by Fluorescence microscopy and fluorescence microplate reader.Results ( 1 ) MGL,SHG,and IHG groups showed a 2.41,3.63,and 4.45 times increase in CYP11B2 mRNA expression,and a 1.83,2.15,and 2.78 times increase in CYP11B2 protein expression,respectively,compared with NG group (P < 0.05 ).The aldosterone levels of HMCs culture supernatant in MGL,SHG,and IHG groups were also increased,being 1.49,2.04,and 2.54 times of that in NG group ( P<0.05 ),and the degree of elevation in IHG group was more marked than that in SHG group( P<0.05 ).MR was activated and translocated from cytosol to nucleus in MGL,SHG,and IHG groups.Quantitative analysis showed the ratioes of cytosol/nucleus fluorescence intensity in MGL,SHG,and IHG groups were 15%,38%,and 53% decreased as compared with that in NG group,and the decrease was more marked in IHG group ( P<0.05 ).(2) Oncofetal FN mRNA expression and ROS levels in MGL,SHG,and IHG groups were increased,being 1.54,2.31,3.65 and 1.26,1.91,2.48 times of those in NG group,respectively ( P<0.05 ),and this increase was more marked in group IHG ( P<O.05 ).Compared with IHG group,oncofetal FN mRNA expression and ROS levels in group IHGE were significantly decreased by 54% and 53%,and in group IHGF by 45% and 39%. ( 3 ) CYP11B2 mRNA,protein,and aldosterone levels in IHGF group were decreased by 74%,59%,and 50%,and the activation of MR in group IHGF was inhibited when the ratio of cytosol/nuclear fluorescence intensity was increased 1.88 fold as compared with that in group IHG ( P<0.05 ).Conclusions Increased expressions of oncofetal FN and ROS by HMCs induced by short-term intermittent high glucose were nore marked than those induced by stable high glucose.The mechanism was associated with activation of local aldosterone system.Fenofibrate may inhibit the activation of local aldosterone system and alleviate the injury to HMCs induced by intermittent high glucose.
5.Effects of physical exercise on seizure-induced cognitive deficits

Objective To explore the long-term effects of physical exercise on learning,memory and the expression of plasticity-related gene-1 (PRG-1) in the cerebral cortex of rats with penicillin-induced developmental seizures.Methods Twenty-four 21-day Sprague-Dawley rats were randomly divided into a control group (CONT1),an exercised control group (CONT2),a seizure group (EXP1) and a seizure plus exercises group (EXP2),each of 6 using a random number table.Penicillin was injected intraperitoneally to the rats in the EXP1 and EXP2 groups to induce seizures,while those in the CONT1 and CONT2 groups received saline injections.Morris water-maze tests were performed to evaluate spatial learning and memory capacity.The rats in the CONT2 and EXP2 groups were administered an aerobic exercise program 30 min per day for 6 consecutive days.The other groups were maintained on the treadmill for the same time but without exercising.Real-time polymerase chain reactions were used to quantify the expression of PRG-1 mRNA in the cerebral cortex.Results There was a decreasing trend in marginal searching and increasing taxis and linear searching in all four groups.Ridit analysis showed that in the watermaze tests on days 2 and 4 the average scores of the control groups were significantly higher than those of the EXP1 and EXP2 groups.However,significant increases in the average scores were observed in the maze tests of the EXP1 group after day 2 and with the EXP2 group from day 4 on.The average scores of the control group were significantly lower than those of the other 3 groups.In the first maze test,the average memory scores of the two seizure groups were significantly lower than those of the controls.In the second maze test,however,only the EXP1 group's average score was significantly worse than those of the other groups.That of the EXP2 group had improved significantly,and was not significantly different from that of the CONT2 group.The expression of PRG-1 was much higher in the CONT2,EXP1 and EXP2 groups than in the CONT1 group.The average expression of PRG-1 in the EXP2 group was not significantly different from that in the EXP1 group.Conclusions Physical exercise can significantly relieve the cognitive deficits induced by long-term seizures,which may be associated with the regulation of PRG-1 expression in the cerebral cortex.
6.Identification of Grain Sprouts Herbs and Their Adulterants Using ITS2 Barcode

Some grain sprouts herbs recorded in Chinese Pharmacopoeia 2010, including Oryzae Fructus Germinatus, Setariae Fructus Germinatus and Hordei Fructus Germinatus, together with their adulterants, were identified by ITS2 sequences. The genomic DNA were extracted, their ITS2 sequences were amplified, and purified PCR products were
sequenced. Sequences were assembled using the CodonCode Aligner. The genetic distances, variable sites and the neighbor-joining (NJ) phylogenetic tree were computed by MEGA 6.0 in accordance with the Kimura 2-Parameter (K2P) model. The results showed that their intraspecific genetic distances were all much lower than the interspecific ones of themselves and their adulterants. The NJ tree based on ITS2 sequence indicated that Oryza sativa, Setariaitalica, and Hordeum vulgar formed one monophyletic clade respectively, in addition any one of the grain sprouts herbs and its adulterants could be distinguished clearly. Therefore, ITS2 sequence is suitable to be as a barcode to identify the grain sprouts herbs and their adulterants.
7.Normal epithelial cell specific-1 gene hypermethylation in the carcinogenesis of hepatocellular carci-noma

Objective To probe the gene expression of normal epithelial cell specific-1 (NES1) in normal liver cells and liver cancer cell lines , and investigate the gene methylation status and its impacts on gene expression and cell biology .Methods The expression level of NES1 mRNA was detected in HepG2 and L02 cells by RT-PCR and RT-QPCR, and the level of gene methylation was examined by MSP .We de-tected cell viability by MTT , NES1 mRNA by RT-QPCR and cyclinD1, P21 and P53 level by Western blot after treating cells with 5-Aza-CdR.Results Compared with L02, NES1 mRNA expression in HepG2 was significantly reduced, and the level of NES1 exon 3 CpG island methylation in HepG2 cells was much higher than that in L02 cells.After demethylation , NES1 mRNA expression and protein level of p 21 and p53 in HepG2 cells were up-regulated , while the cell viability and the level of CyclinD 1 were decreased .Conclu-sions In hepatocellular carcinoma , low expression of NES1 mRNA is related to the gene exon 3 CpG island methylation .NES1 exon 3 methylation may be one of the molecular mechanisms for reducing NES 1 mRNA level, and 5-aza-dC could inhibit cell proliferation by inducing cell cycle arrest in HepG 2 cells.
8.Statistical analysis of articles,citations and authors in Chinese Critical Care Medicine from 2005 to 2006

Objective To study the characteristics of articles,citations and authors in Chinese Critical Care Medicine (Chin Crit Care Med).Methods The date about articles,citations and authors of Chin Crit Care Med from 2005 to 2006 were collected and analyzed in terms of bibliomertic parameters.Results There were 612 papers published in Chin Crit Care Med during 2 years and the papers with funded projects were accounted for 63.1%.The authors came from 30 provinces,autonomous regions and municipalities directly under the central government in China.The average number of articles per issue was 26;the paper density was 0.40 and the average period from the date of submission to publication was 5.2 months.Among these papers,87.6% were shared with co-authors,with cooperation rate of 4.40.The papers with citation were accounted for 85.1% and 95.9% of the citations in English came from journals.The Price's index was 58.2%.Conclusion The articles published in Chin Crit Care Med have high quality.It is one of the most important resources of information for researches in this field,and it is also a primary kernel journal in Chinese medicine field.
9.Minimally invasive percutaneous mono-segment pedicle instrumentation for thoracolumbar burst fractures

Objective To investigate the feasibility,safety and therapeutic effects of minimally invasive percutaneous mono-segment pedicle instrumentation in treating thoracolumbar burst fractures ( AO classification:A 3.1 and A 3.2 ).Methods Twenty-four inpatients with thoracolumbar burst fractures (AO classification:A 3.1 and A 3.2) treated with percutaneous mono-segment pedicle instrumentation from March 2010 to December 2010 were retrospectively studied.The operation time,blood loss,pre-and post-operative visual analogue scale ( VAS),ratio of anterior height between compressed vertebral body and normal vertebral body and vertebral kyphotic Cobb' s angle were evaluated.Results The operation lasted for (90 ± 25) minutes,with intraoperative blood loss of (20 ± 10) ml.The rate of anterior body height rose from pre-operative (56.5 ± 10.1 ) to (92.3 ± 12.2) one week post-operatively and to (90.2 ± 11.l)at the follow-up one year later.The vertebral kyphotic Cobb' s angle was pre-operative ( 16.5 ± 5.2) °,which was reduced to ( 7.3 ± 2.4 )° at oneweek after surgery and ( 7.9 ± 3.5 )° at the follow-up one year later respectively.The VAS scored ( 7.0 ± 1.2) points before surgery,( 1.2 ±0.7) points at one week after surgery and ( 1.1 ± 6..5) points at the follow-up one year later.The ratio of anterior body height at one week after surgery and at the follow-up one year later were both obviously higher than that before surgery (P < 0.05 ),but the ratio one week postoperatively showed no significant difference in comparison with that one year postoperatively (P >0.05).The kyphotic Cobb' s angle had significant decrease at one week after surgery and at the follow-up one year later,as compared with that before operation (P <0.0 5).Also,the VAS score showed marked improvement at one week after surgery and at the follow-up one year later.Conclusions Minimally invasive percutaneous mono-segmental pedicle instrumentation is effective and safe for thoracolumbar burst fractures (AO classification:A 3.1and A 3.2),but it is not suitable for thoracolumbar burst fracture with severely compressed vertebra.
10.Effect of curcumin on expression of p-ERK and p-CREB in spinal dorsal horn and dorsal root ganglion in type 2 diabetic neuropathic pain in rats

Objective To evaluate the effects of curcumin on the expression of phosphorylated extracellular signal-related kinase (p-ERK) and phosphorylated cAMP response element binding protein (p-CREB) in the spinal dorsal horn and dorsal root ganglion (DRG) in type 2 diabetic neuropathic pain (DNP) in rats.Methods Type 2 diabetes mellitus was induced by high-fat and high-sucrose diet and intraperitoneal streptozotocin (STZ) 35mg/kg,and confirmed by fasting blood glucose level≥ 16.7 mmol/L in male Sprague-Dawley rats.Type 2 DNP was confirmed by the mechanical withdrawal threshold (MWT) and thermal withdraw latency (TWL) measured on day 14 after STZ administration < 80% of the baseline value,and the rats with type 2 DNP were randomly divided into 3 groups (n =27 each):type 2 DNP group (group DNP),curcumin group (group Cur) and solvent control group (group SC).Curcumin and corn oil 100 mg/kg (25 mg/ml) were injected intraperitoneally once a day for 14consecutive days starting from 14 days after administration of streptozocin in Cur and SC groups,respectively.Another 27 normal rats were served as control group (group C) and were fed with common forage.MWT and TWL were measured at 3,7 and 14 days after curcumin injection (T1 3),and the lumbar segment 4-6 of the spinal cord and DRGs were removed at the same time for determination of the expression of p-ERK and p-CREB (by Western blot).Results Compared with group C,MWT was significantly decreased,TWL was shortened,and the expression of p-ERK and p-CREB in spinal dorsal horn and DRGs was up-regulated at T1-3 in DNP and SC groups,and at T1 in Cur group (P < 0.05).Compared with group DNP,MWT was significantly increased,TWL was prolonged,and the expression of p-ERK and p-CREB in spinal dorsal horn and DRGs was down-regulated at T2,3 in Cur group (P <0.05).There was no significant difference in the MWT,TWL and expression of p-ERK and p-CREB between DNP and SC groups (P > 0.05).Conclusion Curcumin can attenuate type 2 diabetic DNP by inhibiting up-regulation of the expression of p-ERK and p-CREB in the spinal dorsal horn and DRG in rats.
