1.The role of estrogen in the pathogenesis of lupus in NZB/w F1 mice by modulating the function of den dritic cells

Objective To investigate the effect of estrogen on dendritic cells (DCs) and its role in disease progression by comparing the effects of 17β-estradiol (E2) on spleen DCs in systemic lupus erythe matosus (SLE) murine modeI-(NZBxNZW) F1 (NZB/w F1 ) female mice before and after the disease onset.Methods Anti-CD11c antibody labeled magnetic bead was used to purify spleen DCs.Flow cytometry was utilized to detect the co-stimulatory molecules and intracellular cytokines of DCs.Mixed lymphocytes reaction (MLR) was used to measure the stimulatory activity of DCs to T iymphocytes.And semi-quantitative RT-PCR was employed to check the expression of estrogen receptor (ER).Results E2 could modulate the expression of surface molecule CD40,the production of cytokines IL-6,IL-10,IL-12 and TNFa,and the stimulatory a bility of spleen DCs in SLE model mice.Tamoxifen (TAM) could antagonize E2 effects and E2 could affect the estrogen receptor (ERα) level of DCs.These changes of DCs varied with age.Conclusion E2 may be in volved in the pathogenesis of SLE by modulating DCs by binding ERa.The effects of E2 vary in different stages of SLE progression.
2.17β-estradiol modulates bone-marrow derived dendritic cells in(NZB×NZW)F1 female mice

Objective To investigate the mechanisms of estrogen in the pathogenesis of systemic lupus erythematosus(SLE),we compared the effects of 17β-estradiol(E2)on bone marrow(BM)-derived dendritic cells (BMDCs)in(NZBxNZW)F1(NZB/w F1)female mice befbre and after the disease onset.Methods Immature,BMDCs were differentiated from BM monocytes cultured with rmGM-CSF,rmlL-4,E2 and estrogen receptor (ER)modulator-tamoxifen(TAM)for 7 days.Then immature DCs were stimulated by LPS for 24h to get mature BMDCs.Flow cytometry was utilized to detect the production of costimulatory molecule CD40 and intracellular IL-6,IL-10,IL-12 and TNFα of DCs.Mixed lymphocytes reaction was used to measure the stim-ulatory activity of DCs on spleen T lymphocytes.Results E2 mainly increased the expression of CD40 on im-mature BMDCs but reduced its expression on mature BMDCs.E2 enhanced the stimulatory activity of immature BMDCs,but weakened the activity of mature BMDCs.E2 decreased the production of cytokines of BMDCs in young mice,but increased them in old mice.Conclusion E2 modulates the functions of BMDCs of lupus model mice by binding with ER.The modulation varies depending on disease progression and cell maturation status.
3.Clinical observation of the safety of dental treatment in pediatric patients with leukemia

Objective:To evaluate the safety of dental treatment in pediatric patients with leukemia. Methods:Eighty-nine patients were included and dental treatment was conducted under the condition of neutrophil counts ranging from 3.0?109/L to 10?109/L and platelet counts of more than 50?109/L. Supportive medical care was continued, the patients were monitored for at least one month after dental treatment for bleeding,pain,local and/or systemic infection,fever and other complications. Results:Dental treatments were performed successfully.All patients, except one with delayed wound healing after tooth extraction, recovered uneventfully. Conclusions:As long as the hematological values are within the suggested range, preoperative preparation and supportive medical care are well conducted,dental treatment can be safely performed in pediatric patients with leukemia.
5.Simultaneous determination of six Salvia miltiorrhiza gradients in rat plasma and brain by LC-MS/MS.

To develop a LC-MS/MS method for the determination of protocatechuic acid, protocatechuic aldehyde, salvianolic acid A, salvianolic acid B, cryptotanshinone and tanshinone II(A) in rat plasma and brain. The plasma and brain samples were precipitated with ethyl acetate, then were separated on an Agilent eclipse plus-C18 column (2.1 mm x 50 mm, 3.5 microm) using acetonitrile (consisting of 0.1% formic acid) and water (consisting of 0.1% formic acid) as mobile phase in gradient elution mode. The mass spectrometer was operated under both positive and negative ion mode with the ESI source, and the detection was performed by MRM. The transition of 154.3/153.1 m/z for protocatechuic acid, 137.3/108 m/z for protocatechuic aldehyde, 493.0/295.2 m/z for Salvianolic acid A, 718.0/520.0 m/z for salvianolic acid B, 321.4/152.3 m/z for chloramphenicol, 297.4/254.3 m/z for cryptotanshinone, 295.5/249.3 m/z for tanshinone II(A) and 285.2/154.0 m/z for Diazepam. The calibration curves in the range of 0.625-1 000 microg x L(-1) for protocatechuic acid and protocatechuic aldehyde, 1.25-1 000 microg x L(-1) for salvianolic acid A, 2.5-1 000 microg x L(-1) for salvianolic acid B, 0.15-1 000 microg x L(-1) for cryptotanshinone, 0.625-1 000 microg x L(-1) for tanshinone II(A) are with good linearityin rat plasma and brain. The analysis method is sensitive, simple, and suitable enough to be applied in the pharmacokinetic study of the 6 main components. Animal testing gives the lgBB of the drugs and further studies of the 6 components cross the blood-brain barrier can be carried out.

6.Fisetin attenuates diabetic nephropathy by regulating transcriptional coactivator p300 and matrix metalloproteinase-2

Rat models of diabetes were established by injecting streptozocin intraperitoneally.According to random number table,three groups were divided:normal group,diabetic group,and fisetin-treated group.After 24 weeks,all rats were sacrificed.Biochemical parameters of blood and urine samples were tested.The pathological changes were observed by paraffin sections staining with HE.The expression of extracellular matrix proteins was analyzed via PAS and Masson staining.Location of p300 protein expression was analyzed by immunohistochemistry.The protein expressions of p300 and MMP-2 were determined by Western blotting.The mRNA expressions of MMP-2 were analyzed via real-time PCR.The biochemical parameters and kidney pathological images in fisetin-treated group were better than those in diabetic group.The expression of extraeellular matrix proteins was lower than that in diabetic group.Immunohistochemistry analysis showed that among three groups the expression of p300 was mainly in glomeruli,and was also expressed in cell nucleus and cytoplasm and the coloration of fisetin-treated group was weakened as compared with diabetic group.Western blotting analysis showed that the expression of p300 protein in fisetin-treated group was lower than that in diabetic group(P<O.05).The expressions of MMP-2 mRNA and MMP-2 protein were higher than those in diabetic group (P < 0.05).It is suggested that fisetin may attenuate diabetes associated abnormalities in the kidney of rats,owing probably to inhibiting the expressions of p300 and enhancing the expressions of MMP-2.
7.A New Ionizing-radiation Resistant Strain WGR702 Isolated,Identified, and Radioresistant Character

A new ionizing-radiation resistant strain WGR702 was isolated from arid soils which had been ra-diated. The strain WGR702 was Gram-positive and coccus, the diameter of the cell was 1.5 ?m~2.5?m.The strain WGR702 was pink-pigmented, motile, facultative anaerobe and non-spore forming. The range tem-perature and pH for strain WGR702 growth were 10℃~35℃ and pH 5.0~10.0 respectively. The strain WGR702 had a G+C content of 60.5 mol%. UV and gamma radiation survival curves showed the strain WGR702 had highly ionizing-radiation resistant. Phylogenetic analysis of the 16S rDNA gene sequences (EU315117) showed 94.79%~98.53% similarities with other recognized Serratia species. Primary charac-teristics that distinguish isolate WGR702 from the species of genus Serratia include the cells are spherical and Gram-positive. Based on the phenotypic, biochemical and physiological characteristics differences it is proposed that the new isolated strain WGR702 might be classified as a novel species of Serratia.
8.A clinical study for radiotherapy positioning with references images on CT simulator

Objective To explore a new method of comparing the references images first to enhance the precision of the central point of the radiation treatment planning(RTP), try to establish a reference standard for this method in the nasopharyngeal cancer(NPC)and carcinoma of utercin cervix in the work of position verification. Methods For 50 RTPs of NPC and 20 RTPs of carcinoma of utercin cervix, the reference-CT-images in set-up and in position verification were compared, and to measure the difference between the two images. Then, in the same way, compare and measure the difference in the central-pointimages. Results For NPC, there was over 90% RTPs in which every difference measured was less than 2 mm;for carcinoma of utercin cervix, over 80% RTPs meet the criterion:the value of △MU1 ' or △MU2' was less than 5 mm and the others are less than 3 mm. Conclusions By comparing the references-CT-images in set-up and in position verification, the precision of the central point of the RTP is enhanced. The marks on the skin become more credible. So, it is feasible to perform the criterions in the work of position verification:for NPC every difference measured is less than 2 mm;for carcinoma of utercin cervix the value of △MU1 ' or △MU2 ' is less than 5 mm and the others are less than 3 mm.
9.Clinicopathological study of deep angiomyxoma

Objective To study the clinicopathologic features and differential diagnosis of deep angiomyxoma(DAM). Methods Seven cases of DAM were collected from 2000 to 2008. All the patients were examined by microscopy and immunohistochemistry. Results 6 patients with DAM were females and 1 was male, with median age of 48.5 years. Median maximum dimension was 5.9cm, with invasive growth pattern. The tumor cells of DAM were infantile, spindle or stellate,diffuse and nodular arrangement. A distinctive histologic feature of DAM was its vascularity. Non-arborizing, thin-wall, ectastic capillaries or more commonly, small thick-wall vessels were dispersed throughout the tumor. Mast cells and extravasated red blood cells were frequently found in the stroma, immunohistochemical study showed that 7 cases were positive for vimentin, desmin, ER and PR, 5 for CD34 and SMA, and negative for S-100 and CK. Conclusion DAM is a rare soft tumor that occurs principally in the vulval and vagina region of woman. Misdiagnosis has happened frequently. Immunohistochemical staining are helpful to diagnosis for DAM, but no significance to distinguish it.
10.The effect of Prunellu vulgaris L.extracts on isolated rabbit artery

Objective To investigate the effect of Prunellu vulgaris L.extracts on isolated rabbit artery. Methods The effect of Prunellu vulgaris L.extracts on the contraction curve caused by norepinephrine (NE) was recorded on isolated rabbit artery. Results The contraction curve moved to the right and the maximum contraction range was decreased by chloroform extract, butanol extract and ethyl acetate extract; the contraction curve had no move by aquatic extract. Conclusion Prunellu vulgaris L. has the effect of dilating artery and reducing blood pressure.
