1.New advances in the treatment of multisystem Langerhans cell histiocytosis

Langerhans cell histiocytosis (LCH) is a rare proliferative disorder of dendritic cells and the reticular cells. It is divided into single-system langerhans cell histiocytosis (SS-LCH) and multi-system langerhans cell system (MS-LCH). MS-LCH has a very poor prognosis. The key of survival is timely and effective treatment. The treatment protocols include chemotherapy, immunotherapy and stem cells transplantation. In this review, the new advances in the treatment of MS-LCH were systemically reviewed.
2.Immunological tolerance of rat pancreatic islet transplantation induced by transfusion of donor apoptotic splenocytes

Objective To investigate the effect of transfusion of allogeneic apoptotic splenocytes on immunological tolerance to pancreatic islets transplantation in rat. Method Recipient SD rats were rendered diabetic mellitus by a single intraperitoneal injection of streptozotocin (STZ). The apoptosis of donor Wistar rat splenocytes was induced by X-irradiation with electron linear accelerator. Pancreatic islets were isolated by intraductal collagenase P, and were purified by Ficoll-400 discontinuous density gradient after culturing for 1-2 days from donor Wistar rat. The diabetic SD rats were randomly divided into 4 groups, and respectively received infusion of Hanks solution, donor normal splenocytes, donor apoptotic splenocytes, and donor necrotic splenocytes via dorsal veins of penis, and then they received pancreatic islets from the same donor rat underneath their renal subcapsular space 7 days later. Mixed lymphocyte reaction (MLR) was used as immunological parameter of transplantation tolerance besides the observation of blood glucose level and allograft survival time. Islet graft failure was defined as the presence of a plasma glucose concentration of at least 11.0mmol/L on 2 successive days. Results The pre-infusion of donor apoptotic splenocytes significantly prolonged the survival time of allograft as shown by the median survival time (MST) of graft of 31days, and the longest survival time of 42 day, and it apparently decreased the rejection response in recipient SD rat in one-way mixed lymphocyte culture. Conclusion Transfusion of donor apoptotic splenocytes could induce immunological tolerance of allogenic islets transplantation in rat.
3.Association of depression with dietary fatty acids and the progress of mechanism research

Depression is one of the diseases with the highest disability rate in the world. A large number of studies have shown that the intake of unsaturated fatty acids can deal with depression while chronic overconsumption of saturated fatty acids is a risk factor for depression. It was suggested that the mechanism of saturated fatty acids inducing depression is related to the following four aspects: regulating the function which links to depression in whole brain and specific brain regions, including the hippocampus, the hypothalamic-pituitary-adrenal axis, the striatum, and the prefrontal cortex; stimulating the secretion of inflammatory factors; affecting the balance and function of metabolic regulatory hormones, including leptin, adiponectin, glucocorticoid, and insulin; inducing the disturbance of intestinal flora. This article reviews the relationship between dietary fatty acids and depression, and the possible mechanisms by which saturated fatty acids induce depression from the four aspects mentioned above.

5.Studies on the etiologies and classification of uveitis

Objective To study the clinical classification and etiologies of uveitis based on 1 214 uveitis patients reffered to Zhongshan Ophthalmic Center. Methods A retrospective analysis was made on the patients with uveitis, coming from all over China between January 1996 and December 2001. All kinds of uveitis were classified according to the anatomical criteria and etiological criteria. The relevant data of these patients, such as the age at uveitis onset and sex were also analyzed. Results The total number of the patients is 1 214 (male 698, female 516), with the average age at disease onset being 34.43. Anterior uveitis, the most common type, was seen in 546 cases, accounting for 44.98% of all the patients, followed in descending order by panuveitis (530 cases, 43.66%), intermediate uveitis(78 cases, 6.43%) and posterior uveitis(60 cases, 4.94%). Etiological factors and clinical entities were identified in 703 patients, accounting for 57.91% of all the patients, and the other 511 patients were idiopathic ones. The most common types of anterior uveitis were idiopathic uveitis(316 cases, 57.88%), followed by Fuchs syndrome(85 cases) and ankylosing spondylitis(45 cases). Behcet ) disease(218 cases, 41.13%) and Vogt-Koyanagi-Harada syndrome(196 cases, 36.98%) were the most common entities in panuveitis. Neither etiological factors nor clinical entities could be identified in the patients with intermediate uveitis and those with posterior uveitis. Conclusions Uveitis occurs mostly in young and middle-aged adults. In general, a predilection was seen in the male as compared with the female in the development of uveitis. Idiopathic anterior uveitis, Behcet ) disease and Vogt-Koyanagi-Harada syndrome are the most common entities of uveitis seen in China. Classification based on etiological and anatomical factors may provide a reasonable system for the study of uveitis.

7.Study of Anti-Tumor Effect of NK Cells on Oral Tongue Squamous Cell Carcinoma

Objective To explore the treatment effect and mechanism of A-NK cells on the subcutaneous trans?planted tumor of oral tongue squamous cell carcinoma in nude mice through observing the external growth and hyperplasia of A-NK cells and to provide theoretical evidence for squamous cell carcinoma treatment. Methods A-NK cells and NA-NK cells were both derived from healthy human peripheral blood and cultured in vitro. Cell growth was observed under micro?scope. The squamous cell carcinoma model in nude mice was established through subcutaneous implanting of Tca8113 cells. Then they were randomly assigned into three groups who were injected with either saline solution, or A-NK cells or NA-NK cells paraneoplastically. All animals were sacrificed after 33 days when tumor were isolated then weight and change in tumor size were assessed. Finally curve of tumor growth was drawn. Results Under the microscope, the proliferation of A-NK cells peak in 15 days and NA-NK cells peak in 12 days. After 3 weeks, the number of A-NK cells increased by 39.33 times while the number of NA-NK cells increased by 16.33 times. The Volume of tongue squamous cell carcinoma in saline solu?tion group was larger than that in A-NK cells and NA-NK cells groups, and volume in the NA-NK cells group was larger than that in A-NK cells group. The volume of tongue neoplasms in different groups, time, and interaction effects are statisti?cally significant (P<0.01). The tongue neoplasms weight in the saline solution group was greater than that in the A-NK cells and NA-NK cells group, and the weight in NA-NK cells group was greater than that in A-NK cells group, and the difference are statistically significant (P<0.05). Conclusion A-NK cells and NA-NK cells can significantly inhibit the subcutane?ous transplanted tumors in nude mice and the anti tumor effect of A-NK group is stronger than NA-NK.
10.Design, synthesis and cholinesterase inhibitory activity of quinoline-polyamine conjugates.

A series of quinoline-polyamine conjugates (8a-8n) were designed, synthesized and evaluated as inhibitors of cholinesterases (ChEs). Some of these compounds had potent ChEs inhibitory activity with IC50 values at micromolar range. Compound 8n exhibited the strongest inhibition on acetylcholinesterase (AChE) with an IC50 value of 8.78 micromol x L(-1), and compound 8i showed the most potent inhibition on butyrylcholinesterase (BChE) with IC50 value of 1.60 micromol x L(-1) which was slightly better than rivastigmine. The structure-activity relationship revealed that the chain length of polyamine and linker played important roles for inhibitory activity. Molecular modeling studies showed that 8i targeted both the catalytic active site (CAS) and the peripheral anionic site (PAS) of cholinesterases.
