5.Effects of polysaccharide from spirulina platensis on the growth of HeLa cells
Chinese Journal of Marine Drugs 1994;0(01):-
Objective To investigate the effects of polysaccharide from Spirulina platensis (PSP) on the growth of HeLa cells. Methods The antiproliferation activities of PSP on HeLa cells were evaluated by MTT assay. The morphologic changes of HeLa cells were observed under light microscope. Cells division cycle was measured by flow cytometry. Results It was showed that with the increase of PSP concentration, the inhibited rate on HeLa cells increased and the survived rate declined; The HeLa cells exposed to PSP for 24~48h displayed significant morphologic changes; PSP had a specific effect on the HeLa cell division cycle, increased the number of cells in phase G 1. Conclusion These results indicated that the effects of PSP on HeLa cells may probably be attributed to increase of the number of cells in phase G 1.
6.Collagen matrix compared with mitomycin C for treatment of primary open-angle glaucoma with trabeculectomy performed
International Eye Science 2017;17(9):1712-1715
AIM:To evaluate the effectiveness and safety between trabeculectomy with collagen matrix versus trabeculectomy with mitomycin C(MMC) for patients with primary open-angle glaucoma(POAG).METHODS:In this prospective randomized comparative study from January 2015 to December 2016.Thirty-two eyes presented with POAG were included in this study, 14 eyes treated by trabeculectomy with subconjunctival implant of collagen matrix (study group) and the other 18 eyes treated by trabeculectomy with mitomycin C.Postoperative IOP, the success rate of operation, number of postoperative glaucoma medications and postoperative complications were recorded. Each patient was followed up at least 6mo.RESULTS:The mean postoperative IOP was statistically different between the study group and the control group after 1d (P<0.05), while not statistically different at 1 and 1mo follow-up (P>0.05), and the mean postoperative IOP was statistically different between the two groups (P<0.05) at 3 and 6mo after surgery. There was no significant difference in the success rate of operation between the two groups at 6mo after operation (P>0.05).The IOP decreased at 1d after openations compared with before, kept stable at 1wk to 6mo.IOP of study group was lowen than control.IOP was controlled by glaucoma medications in the study group by 28% compared to control group by 33% at 6mo after operation, but there was no significant difference. There was no significant difference between the study group and the control group in complications (P<0.05).CONCLUSION:Trabeculectomy with collagen matrix implant is comparable to the use of MMC with a similar success rate in open-angle glaucoma and the range in reducing intraocular pressure was significantly higher than that of MMC and it can significantly avoid the occurrence of low IOP postoperatively, transient anterior chamber, conjunctival wound leakage complications has no advantages compared with the use of MMC.
7.Study on expression and functions of staphylococcal enterotoxin B mutants
Chinese Journal of Microbiology and Immunology 2001;21(2):200-203
Objective To obtain staphylococcal enterotoxin B (SEB) mutant with normal antigenicity but low toxicity. Methods Using PCR technique, normal SEB (SEB-N) gene which was amplified from S. aureus S6B. SEB mutant gene (SEB-M) was prepared from the same strain, but one nucleotide in SEB gene was changed from asparagine (N23) to serine (S23). SEB-N and SEB-M were cloned into procaryotic expression vector pTrc99A then and transferred into E. coli JM109. SEB-N and SEB-M which were cloned into plasmid were sequenced directly by dideoxynucleotide method. The crude expressed proteins were identified by double agar immunodiffusion. The level of IL-2 in supernatants of mouse splenocytes stimulated by crude expressed proteins was determined by ELISA. Results SEB-N and SEB-M were obtained through PCR. The sequence of SEB-N was changed with non site-directed mutagenesis, threonine at the residue 150 of SEB-N was replaced with alanine (ACT→GCT, T150A). As being expected, at the residue 23 of SEB-M, serine substituted for asparagine (AAT→AGT, N23S) with site-directed mutagenesis. Double agar immunodiffusion showed obvious precipitin line with anti-SEB by both crude SEB-N and SEB-M mutant proteins could produce, but not by non-recombinant strain. ELISA demonstrated that the level of IL-2 in supernatant of mouse splenocytes stimulated by natural SEB protein (containing equal amount of JM109P crude protein) was 40 times as much as that stimulated by SEB-M and 12.5 times as much as that stimulated by SEB-N. Conclusions We obtained two recombinant strains which produced T150A and N23S mutant SEB protein. The mutant proteins showed binding ability to anti-SEB as the normal protein. However, their biological activity as superantigen decreased sharply. We consider that it is promising for further study of molecular adjuvant or superantigen vaccine.
8.Assessment of the ability of the new software SonoAVC to measure follicular volume
Chinese Journal of Ultrasonography 2009;18(9):780-782
Objective To assess the ability of the new software SonoAVC to measure follicular volume and to compare these volume calculations with those made by conventional methods. Methods Three-dimensional ultrasound imaging was used to acquire volumetric data from the ovaries of 50 women undergoing controlled ovarian stimulation as part of in-vitro fertilization treatment. SonoAVC was used to automatically measure the volume of follicles and virtual organ computer-aided analysis(VOCAL) was also applied to measure volume, which was used as the standard of volume measurement. And the three perpendicular diameters (xyz diameters) of follicles were manually measured by conventional two-dimensional(2D) ultrasound, which were used to estimate volume using the sphere formula. And the validity of each technique was compared using limits of agreement. Results One hundred and seventy-six follicles were studied. SonoAVC and VOCAL provided highly accurate and repeatable follicular volume measurements in all cases. Volumes estimated from manually derived follicular diameter measurements were less accurate and repeatable. VOCAL took more time in follicular volume of measurement than SonoAVC and conventional 2D displays. Conclusions SonoAVC provides highly accurate, repeatable, and efficient measurements of follicular volume.
9.Effect of water-soluble polymers on the inhibition of osthole crystallization.
Acta Pharmaceutica Sinica 2010;45(12):1559-64
This paper is to study the inhibitory effect of water soluble polymers--methyl cellulose (MC), hydroxypropyl methyl cellulose (HPMC), hydroxypropyl cellulose (HPC-M), poloxamer (F68) and polyvidon (PVP) on osthole (OST) crystallization and investigate the impact of polymer concentration and viscosity on crystallization behavior. Also, UV spectrophotometry method was used to determine the drug concentration at different time point to draw the OST concentration-time curve. Results show that HPMC has the most significant inhibition effect on OST crystallization, and drug concentration level is 1.61 times higher than that in control solution within 8 h followed by PVP (1.54) and MC (1.45) respectively. The kinetics of OST recrystallization can be described using first-order reaction, and the crystallization rate constants obtained by analyzing the regression equation indicate that HPMC-60SH-4000 and HPMC-60SH-10000 can greatly influence OST crystal formation. The dissolution rate of drugs precipitated from water-soluble polymer solutions is faster compared with controls in pH 1.2 HCl and pH 6.8 phosphate buffers, which demonstrated that water-soluble polymers can not only change the behavior of drug crystallization but markedly improve the dissolution rate of water insoluble drugs.
