1.Photoprotection by the selective COX-2 inhibitor NS398 against UV-mediated damage in keratinocytes

Objective To investigate photo-protective effect of NS398 on above cells damaged from UV irradiation.Methods HaCaT cell and HaCaT keratinocytes were incubated in the culture medium supplemented with different concentration of NS398 for 2h before diffierent dosages of UV irradiation.MTT assay was used to detect cell proliferation and cellular activity:Fluorospectrophotometer assay was used to measure the change of ROS level in the cells which were UV-mediated.Resuits After irradiation with UV,proliferation and cellular activity of HaCaT cells were decreased(P<0.05);NS398 reduced the generation of ROS in cells induced by UV irradiation apparently and the cell apoptotic rate was dependent on the concentration of NS398. Conclusion NS398 might play an important role in the treatment of UV irradiation injury by decreasing the generation 0f ROS in cells after UV irradiation and increasing the survival ability of HaCaT cells.

3.Transfection of embryonic stem cells with green fluorescent protein gene and their differentiation into neural cells

Objective To establish embryonic stem cells (ESC) that can express green fluorescent protein (CFP) and differentiate them into neurons. It would provide tagging neurons for clinical transplantation to cure neural system diseases. Methods ESC (R1) was transfeeted with a plasmid containing the GFP by electroporation. A transgeuic cell line was obtained after selection with G418. The ESCs were characterized by AKP staining. Monolayer differentiation method was used to induce neural differentiation derived from GFP-ESC and immunofluorescence method was used to identify Tuj1 positive cells. Results There was no significant difference(X2=3.14,P0.05) in transfect rates between liposome and electroporation (65% vs 79%). The AKP staining of GFP-ESC was positive. GFP-ESC could be differentiated into neural cells. Conclusions These results show that ESC expressing GFP has been estabhshed, which can be differemiated into neurons.

Hyperthermia is a means of adjuvant therapy, which have a sensitizing effect to radiotherapy and chemotherapy. In recent years, the molecular biology, cell and animal experimental research of tumor thermochemotherapy progressed very quickly, which provide theoretical foundation and guidance for us to further develop hyperthermia combined with chemotherapy in clinical trials. In this paper, the studies with the mechanism of thermo-chemotherapy treatment of tumor, different ways of thermochemotherapy and commonly used drugs in thermochemotherapy are reviewed.

7.Case-control study on risk factors of maternal adverse pregnancy outcomes

ABSTRACT:Objective To explore the risk factors for maternal adverse pregnancy outcomes.Methods An unmatched case-control study based on hospital was performed.Univariate and multivariate Logistic regression were used to analyze the related factors of maternal adverse pregnancy outcomes,including general condition,history of fertility,abnormal symptoms and diseases during pregnancy.Results Univariate analysis results showed that high education level of gravida might be the protective factors of adverse pregnancy outcomes.The risk factors for adverse pregnancy outcome might include advanced maternal age,intensive workload,frequent pregnancy,history of spontaneous abortion,severe morning sickness,and sickness during pregnancy or progestation.Multivariate Logistic regression analysis showed that high education level of gravida (OR=0.63,95% CI:0.50-0.80)was the protective factor of adverse pregnancy outcomes;severe morning sickness (OR=2 .1 3 ,9 5% CI:1 .6 3-2 .7 9 )and sickness during progestation (OR=2.25,95% CI:1.06-4.77)were the risk factors for maternal adverse pregnancy outcomes.Conclusion The level of maternal education should be improved.We should attach great importance to preventive education and thorough treatment of severe morning sickness. Couples should be encouraged to have physical examination before marriage and pregnancy.Corresponding pregnancy care guidance should be given to pregnant women with different physical conditions so as to effectively reduce the occurrence of adverse pregnancy outcomes.
8.Simulation research on gastric emptying measurement experiment by electrical impedance tomography with three-layer electrodes

Objective To verify the effectiveness of electrical impedance tomography (EIT) with multi-layer electrodes in gastric emptying measurement by simulation experiments using EIT with three-layer electrodes.Methods Firstly,EIT system with three-layer electrodes and gastric emptying measurement simulation equipment was designed and constructed to simulate gastric emptying process and obtain EIT reconstruction images.Secondly,the region where the electrical characteristic of gastric changes was selected as region of interest.Finally,the relative impedance change rate in region of interest for every image obtained from all three layers was consecutively recorded,then the gastric emptying curve was formed and gastric emptying time was calculated.Results There are obvious differences among the sharp of impedance change curves of interested region for three layers,and the gastric emptying time and process of each layer is also different from one another.It is suggested that the passing path and emptying manner of food in gastric emptying process reflected by EIT images varied from layer to layer.Therefore,the necessity and feasibility of using EIT with three-layer electrodes in gastric emptying measurement study is verified.Conclusions EIT with three-layer electrodes can provide more abundant information of gastric motility function associated with gastric emptying pathology and physiology state.It is likely to be a noninvasive,convenient and effective imaging method for the detection and evaluation research on gastric emptying and gastric motility.
9.Emergency Management of Infection Control of Mass Earthquake Wounded Victims

OBJECTIVE To explore the infection control measures in operation of mass earthquake wounded victims,and improve nursing quality.METHODS By setting up infection control group,strengthening nurse' awareness of infection control,strict division of region for operation,paying great attention to early first-aid treatment and strengthening the key link in the quality control.RESULTS Infection control were effectively improved the salvage rate of mass earthquake victims.There was non-cross infection among victims.CONCLUSIONS According to the infection trait of earthquake wounded victims,perfecting the system of measures of infection control is an important way to control hospital infection of mass earthquake wounded victims.
10.Effect of blocking Wnt-?-catenin signaling pathway on activation of hepatic stellate cells

Objective To investigate whether functional Wnt-?-catenin signaling is present in activated hepatic stellate cells (HSC),and the effect of blocking this signaling on activation of HSC. Methods?-catenin expression in HSC was examined by immunocytochemistry.Wnt signalings in HSC-T6 were assessed using a T cell factor (TCF)-dependent luciferase reporter gene (pTOP- FLASH) assay.Wnt signalings in HSC-T6 were blocked by transfecting with a dominant negative TCF (dnTCF) expression plasmid,then the expression of alpha smooth muscle actin (?-SMA) and collagen typeⅠwere examined by Western blot.Results?-catenin staining was positive in the nuclei of HSC-T6.Luciferase activity in the cells transfected with pTOPFLASH was significantly higher than that in the cells transfected with pFOPFLASH (P
