1.Analysis of monitoring data of iodine nutrition among population in Jiangxi province from 1995 to 2010

Objective To master the level of iodine nutrition among population in Jiangxi province,and to provide a scientific basis for establishing the strategy for prevention and control of iodine deficiency disorders (IDD).Methods Retrospective method was adopted to analyze the goiter rate and frequency distribution of urinary iodine of children aged 8-10,the qualified rate of iodized salt,the coverage rate of iodized salt and the consumption rate of qualified iodized salt in residents of Jiangxi province from 1995 to 2010.The method of correlation analysis was used to analyze the relationship between goiter rate of children (by palpation) and the qualified rate of iodized salt,iodized salt coverage rate and residents consumption rate of qualified iodized salt.Results The goiter rates (measured by the method of palpation) of children aged 8-10 were down from 40.17%(482/1200) in 1995 to 0.80%(16/2000) in 2010(x2 =4.864,P< 0.05).The median of urinary iodine of children was higher than 200 μg/L; the proportion of people whose urinary iodine content higher than 300 μg/L was above 25.00% and the highest propoaion was up to 58.01% (210/362) between 1995-2010.The minimum median of salt iodine was 17.77 mg/kg in 1995,and 29.30-39.10 mg/kg in other years.The qualified rates of iodized salt,the iodized salt coverage rates and the consumption rates of qualified iodized salt increased from 43.58%(452/1037),86.42%(1037/1200) and 37.67%(452/1200) in 1995 to 97.95% (1916/1956),99.95%(1956/1957) and 97.90%(1916/1957) in 2010,respectively; there was a growth trend over the years(x2 =5.240,6.118,5.631,all P < 0.05).The goiter rates of children were related to the qualified rates of iodized salt,the iodized salt coverage rates and the consumption rates of qualified iodized salt,and the correlation coefficient(r) was-0.833,-0.881 and-0.918 (all P < 0.05),respectively.Conclusions The level of iodine nutrition among residents in Jiangxi province has already gone beyond the appropriate level,and the iodine concentration in salt should be cut to ensure the appropriate iodine nutrition level among people.
2.Application of maxillary canine pulp volume

Objective To explore the clinical application value of three-dimensional reconstruction of maxillary canine pulp volume /tooth volume (PV/TV)in individual gender determination.Methods There were 103 patients (51 males and 52 females) with CBCT imaging data from September 2015 to August 2016, Shanxi Medical University, department of radiology, Mimics 17.0 software was used to measure pulp volume of maxillary canines and tooth volume, and to calculate the ratio of pulp volume/tooth volume.The data were processed by SPSS16.0 statistical software and Fisher discriminant method. The gender determination function was obtained and Cross -Validated method for performance evaluation. Results When gender determination was determined by tooth volume (TV) and pulp volume/tooth volume (PV / TV) as the study index, the gender discriminant function was Y=0.009(TV)+28.896(PV/TV) - 6.962, Cross-validated method for the effect of evaluation, to determine the compliance rate: 51 males, the correct rate of 64.7%;52 female, the correct rate of 78.8%. Conclusion The tooth volume (TV) and pulp volume/tooth volume (PV/TV)can be used as a research indicator of gender inference, it provides a new method and avenue for the gender determination in forensic medicine.
3.An analysis on the distribution and drug resistance of pathogenic bacteria in infectious diarrhea among infants

Objective To learn the main pathogenic microorganisms,and to acquire the data of pathogens composition anddrug resistance of infant diarrhea disease in Wenzhou City.Methods The diarrheal stool specimens of <5 years oldchildren were collected from outpatients and inpatients in children′s hospital and were cultured and detected.Results Atotal of 201 strains pathogenic bacteria were separated from 51 7 clinical specimens,and the rate of total detection is38.88%,of which included 74 strains intestinal pathogenic bacteria and 1 27 strains diarrhea virus,with the rate of detection1 4.31 % and 24.56%,respectively.Among intestinal pathogenic bacteria,the E.coli was the most common bacteria(36strains,48.65%),followed by salmonella(31 strains,41 .89%).Diarrhea virus included norovirus type 2 (56 strains,44.09%),norovirus type 1 (24 strains,1 8.90%),A group rotavirus (56 strains,31 .50%)and goblet virus(7 strains,5.51 %).The resistance rate of Salmonella to ampicillin was 64.52%,and that to ampicillin,tetracycline,compound newMing were more than 70.00%.The resistance rate of Rifampicin.Novobiocin in all bacteria reached 1 00%.Conclusion E.coli and salmonella epidemic strains were the common pathogenic bacteria in infectious diarrhea among infants inWenzhou City.The predominant viruses were norovirus and rotavirus group A.The drug resistance of various pathogenicbacteria was different,and active surveillance should be strengthened.
4.The Region of Interest in Boundary Calibration for Palatal Ru-gae Image of Forensic Identification

Objective To examine the method of least-square fitting for calibrating the palatal rugae boundary. Methods According to the distribution of the teeth, the feature points were selected; when they were fit, the boundary of the palatal rugae area was created, thereby constructing a mask. The mask was used to remove the part located outside the boundary and filter out the interference. Three samples were utilized for the ex-periments and analyses to come. Results To evaluate the quantitative results of the fitting curves, the correla-tion coefficients(r)of the samples and the relationship between the actual mean value and ideal mean value was obtained through six fitting processes. The differences between the actual mean and ideal mean were found to be significantly small (from 0.2857 to 2.5000) in the six fitting processes, with the range of r close to 1(from 0.9896 to 0.9995). Conclusion The effect of the cubic polynomial fitting method adopted in this study was stable.The proposed boundary calibration method can effectively locate the palatal rugae boundary and remove the interference area, further promoting the practice of forensic identification.
5.Analysis on Thinking of Warming-settling Method for Syndrome Differentiation Treatment of Malignant Tumor

In recent years, TCM treatment for malignant tumors has gotten more and more attention. This article mainly introduced the theoretical basis of the warming-settling method for the treatment of malignant tumors, and confirms the effectiveness of it through the observation of the improvement of clinical symptoms patients combined with medical cases. It showed the effectiveness of warming-settling method for the treatment of malignant tumors, and provided references for TCM treatment of malignant tumors.
6.Analysis of quality control assessment results of urine fluoride laboratories in Jiangxi Province in 2019 - 2021

Objective:To analyze the quality control test results of urine fluoride laboratories in Jiangxi Province, and support reliable data for the monitoring and prevention of fluorosis.Methods:From 2019 to 2021, the Jiangxi provincial urine fluoride laboratories used the "Determination of Fluoride in Urine-Ion Selective Electrode Method" (WS/T 89-2015) to detect quality control samples. Origin 9 software was used to analyze the data, and Youden plots and deviation bar plots were produced. The results were evaluated based on the Z-score (qualified: │Z│ ≤2, basically qualified: 2 < │Z│ < 3, unqualified: │Z│ ≥3).Results:The qualified rate of quality control assessment results of urine fluoride laboratories in Jiangxi Province was 95.00% (38/40) . Most of urine fluoride laboratories had scores of │Z│ < 2, and two laboratories had scores │Z│≥3 in 2019 and 2020.Conclusion:The quality control assessment results of urine fluoride laboratories in Jiangxi Province are relatively satisfactory, but some laboratories still need to further improve their detection capabilities.
7.Analysis on external quality control network results for iodine deficiency disorders laboratories in Jiangxi Province in 2018

Objective:To analyze the results of external quality control network of iodine deficiency disorders (IDD) laboratories in Jiangxi Province in 2018, to summarize the total detection level and capability at all levels, and to provide reliable quality assurance for monitoring and prevention of IDD.Methods:In 2018, IDD laboratories of provincial, municipal and county-level in Jiangxi Province were examined for the external quality control of salt iodine, urinary iodine and water iodine. Among them, there were 1 provincial, 11 municipal and 30 county-level laboratories participated in salt iodine assessment; there were 1 provincial, 11 municipal and 100 county-level laboratories participated in urinary iodine assessment; and there were 1 provincial and 11 municipal laboratories participated in water iodine assessment. Salt iodine was determined by direct titration method (GB/T 13025.7-2012); urinary iodine was determined by As 3+-Ce 4+ catalytic spectrophotometry method (WS/T 107.1-2016) or inductively coupled plasma mass spectrometry method (WS/T 107.2-2016); water iodine was determined by the method in the range of 0 - 100 μg/L in "Study on the Method Suitable for the Detection of Water Iodine in Areas with Iodine Deficiency and High Iodine" recommended by national IDD reference laboratory. Urinary iodine was determined with Z score method, when│Z│≤ 2, it was qualified; when 2 <│Z│ < 3, it was basically qualified; when│Z│≥ 3, it was not qualified. Water iodine and salt iodine were determined by the method of reference value ± uncertainty, and the average value of the test results within this range was judged to be qualified; otherwise, it was judged to be unqualified. Results:In 2018, the feedback rates and qualified rates of salt iodine, urinary iodine and water iodine in 1 provincial and 11 municipal laboratories in Jiangxi Province were all 100.00%. Thirty county-level laboratories participated in the salt iodine assessment, and 1 of them failed. One hundred county-level laboratories participated in the urinary iodine assessment, and 3 of them failed.Conclusion:Laboratory testing technology of IDD continues to maintain at a high level in Jiangxi Province in 2018, and the detection ability of some county-level laboratories needs to be further improved.
8.Correction of the pathogenic mutation in the G6PC3 gene by adenine base editing in mutant embryos.

Objective: To determine whether the adenine base editor (ABE7.10) can be used to fix harmful mutations in the human G6PC3 gene. Methods: To investigate the safety of base-edited embryos, off-target analysis by deep sequencing was used to examine the feasibility and editing efficiency of various sgRNA expression vectors. The human HEK293T mutation models and human embryos were also used to test the feasibility and editing efficiency of correction. Results: ①The G6PC3(C295T) mutant cell model was successfully created. ②In the G6PC3(C295T) mutant cell model, three distinct Re-sgRNAs were created and corrected, with base correction efficiency ranging from 8.79% to 19.56% . ③ ABE7.10 could successfully fix mutant bases in the human pathogenic embryo test; however, base editing events had also happened in other locations. ④ With the exception of one noncoding site, which had a high safety rate, deep sequencing analysis revealed that the detection of 32 probable off-target sites was <0.5% . Conclusion: This study proposes a new base correction strategy based on human pathogenic embryos; however, it also produces a certain nontarget site editing, which needs to be further analyzed on the PAM site or editor window.

9.Decline in semen concentration of healthy Chinese adults: evidence from 9357 participants from 2010 to 2015.

The present study aims to analyze sperm concentration trends among young and healthy Chinese adults in Wuhan, Central China, from 2010 to 2015. Semen analysis data from 9357 participants were collected and analyzed using a general linear model and the Cochran-Armitage trend test. A significant decline was observed in sperm concentration (β [standard deviation]: -1.53 [0.16]; P < 0.001). In addition, a decline in sperm density was observed by stratifying student versus nonstudent sperm donors and by analyzing the year of birth or birth year cohort of the participants. Furthermore, the percentage of participants with sperm densities of over 40 × 106 ml-1 significantly decreased with year. Notably, a dramatic decline in sperm density was recorded over the first 5 years of study. This research reported a decline in sperm concentration among young adults in Wuhan, Central China, in 2010-2015.

10.Clinical analysis of 10 cases of multi-center tumor necrosis factor receptor-associated periodic syndrome.

Objective: To summarize the clinical characteristics of tumour necrosis factor receptor-associated periodic syndrome (TRAPS) in children. Methods: The clinical manifestations, laboratory tests, genetic testing and follow-up of 10 children with TRAPS from May 2011 to May 2021 in 6 hospitals in China were retrospectively analyzed. Results: Among the 10 patients with TRAPS, including 8 boys and 2 girls. The age of onset was 2 (1, 5) years, the age of diagnosis was (8±4) years, and the time from onset to diagnosis was 3 (1, 7) years. A total of 7 types of TNFRSF1A gene variants were detected, including 5 paternal variations, 1 maternal variation and 4 de novo variations. Six children had a family history of related diseases. Clinical manifestations included recurrent fever in 10 cases, rash in 4 cases, abdominal pain in 6 cases, joint involvement in 6 cases, periorbital edema in 1 case, and myalgia in 4 cases. Two patients had hematological system involvement. The erythrocyte sedimentation rate and C-reactive protein were significantly increased in 10 cases. All patients were negative for autoantibodies. In the course of treatment, 5 cases were treated with glucocorticoids, 7 cases with immunosuppressants, and 7 cases with biological agents. Conclusions: TRAPS is clinically characterized by recurrent fever accompanied by joint, gastrointestinal, skin, and muscle involvement. Inflammatory markers are elevated, and autoantibodies are mostly negative. Treatment mainly involves glucocorticoids, immunosuppressants, and biological agents.
