China Journal of Chinese Materia Medica 2014;39(1):83-88
Chemical constituents of Inula japonica were isolated and purified by repeated column chromatographies, over silica gel, and Toyopearl HW-40, and preparative HPLC. On the basis of spectral data analysis, including NMR and MS data, the structures of the isolates were elucidated and their anti-inflammatory activities were assayed. Fifteen compounds were isolated from the ethyl acetate extract of I. japonica, and their structures were elucidated as dihydrosyringenin (1), (3S, 5R, 6S, 7E)-5,6-epoxy-3-hydroxy-7-megastigmen-9-one (2), (6R, 7E) -9-hydroxy-4,7-megastigmadien-3-one (3), arnidiol (4), taraxasterol acetate (5), 8,9,10-trihydroxythymol (6), taxifolin (7), luteolin (8), napetin (9), eupatin (10), spinacetin (11), quercetin (12), p-hydroxycinnamic acid (13), caffeic acid (14), and caffeoyl acetate (15). Compounds 1, 2, 7, 13 and 15 were isolated from the genus Inula for the first time, and compounds 3, 4, 9-11 and 14 were isolated from this plant for the first time. The anti-inflammatory activity result showed that compounds 3, 6-12 and 14 exhibited inhibition effect against leukotriene C4 (LTC4) synthesis and degranulation definitely in c-Kit Ligand (KL) induced mast cells, and compound 8 and 12 also had the suppression effect against lipopolysacharide(LPS) induced nitric oxide (NO) activity in RAW264.7 macrophages. It is firstly reported that compounds 7 and 9-11 possessed potent inhibition activities against LTC4 generation and degranulation in mast cells.

Objective To detect the microsatellite instability (MSI) of D9S166,D9S171,D9S941, D9S942 and IFNA located at chromosome 9p21 in gastric carcinoma with high-grade intraepithelial neoplasia and normal tissue, and investigate the correlation of p14,p15,p16 gene and the gastric carcinogenesis. Methods 55 cases of gastric carcinoma with high-grade intraepithelial neoplasia and normal tissue were se- lected to detect MSI by microdissection, polymerase chain reaction, denaturing polyaerylamide gel elec- trophoresis and silver nitrate staining technology of five microsatellite markers on 9p21.Results In the infor- mative cases, total ratio of MSI in gastric carcinoma was 27 % (64/233) and it was 18% (42/233) in high- grade intraepithelial neoplasia. The ratio of MSI was significantly higher in gastric carcinoma than that in high-grade intraepithelial neoplasia (P

Objective Using CT perfusion (CTP) technique,to investigate the graft perfusion changes in patients with hepatic artery stenosis (HAS) with or without ischemic-type biliary lesions (ITBL) after orthotopic liver transplantation (OLT).Methods Thirteen recipients with HAS received CTP scan of the liver, including 8 with ITBL and 5 without ITBL.For all patients, the diagnosis of HAS was made by CTA,and the diagnosis of ITBL by percutaneous transhepatic cholangiography.CT perfusion indices were obtained, including hepatic artery perfusion (HAP),portal vein perfusion (PVP),total liver perfusion (TLP) and hepatic perfusion index (HPI).Results Of the 13 patients with HAS,mean HAP in patients with and without ITBL was 59.8 and 35.1 ml·min-1 ·100 ml-1 (P =0.021,two-tailed paired Student t test) ; mean PVP was 125.4 and 166.2 ml·min-1·100 m1-1 (P =0.016) ; mean TLP was 185.2 and 201.3 ml· min-1 · 100 ml-1 (P =0.306) ; and mean HPI was 33.6 and 18.2 (P =0.005),respectively.Conclusion Using CTP technique, liver perfusion changes were reflected by measuring CTP indices noninvasively.Compared to those without ITBL in this study,HAP and HPI in patients with ITBL were higher and PVP was lower, which may be contributed to biliary inflammation.

Journal of Pharmaceutical Practice 2001;19(2):86-88
OBJECTIVE:To study the main aspects of quality control of dexamethasone liposomes.METHODS:The structure of liposomes was studied with negative staining method of TEM.K-ratio spectrophotometry was wsed to determine the contert, and the Macrosep was wsed to seperate the entrapped from free drugs.RESULTS:An excellent linear relationship was obtained with the range of 4~52μg/ml,the average recovery percent was between 98.28%~99.71%.The entrapment efficiency was one of the important parameters of liposomes quality control. It was related with the clinical effect directly, the entrapment efficiency was more than 80%.CONCLUSION:This method is accurate, quick and reproducible.

ObjectiveTo observe clinical results and histopathological changes of scleral defect repaired with acellular xenogenic dermal matrix (Xeno-ADM) in rabbits.MethodsModel of rabbit sclera defect was established, and repaired with Xeno-ADM. The rabbits were clinically examined for inflammation and eyeball healing. The animals were sacrificed at 2nd and 4th week after operation, and implants were obtained and examined histopathologyically and ultrastructully with light microscopies respectively to evaluating changes of inflammation and vascularization.ResultsThere were no obvious inflammation and eyeball deformation observed. 2 weeks after implantation, the partial inflammatory cell invasion was seen with the light microscopy, and there was an obvious borderline between the Xeno-ADM and the sclera. 4 weeks after implantation, the inflammatory cells were reduced noticeably, the Xeno-ADM and the sclera completely merged with each other.ConclusionThe acellular xenogenic dermal matrix may be an ideal materix with fast neovascularization and low immunity for replace of sclera implants.
8. Research progress on regulation of PGC-lα in obesity-related metabolic diseases
Chinese Pharmacological Bulletin 2021;37(6):741-745
Peroxisome proliferator-activated receptor γ coactivator 1-alpha (PGC-1α) is a member of superfamily of transcriptional co-regulators. It was first discovered to be related to the adaptive thermogenesis of brown adipose tissue. Later, it has been discovered that PGC-la can also regulate glucose and lipid metabolism and vascular endothelial function, thus influence the occurrence and development of obesity-related metabolic diseases such as hyperlipidemia, type II diabetes and hypertension. Drugs targeting PGC-1α are promising for the prevention and treatment of these diseases. Here we review the research progress on regulation of PGC-1α in obesity related metabolic diseases, aiming to provide theoretical basis for the development of drugs targeting PGC-1α.
