1.The expression of survivin gene in malignant pleomorphic adenoma,relay pleomorphic adenoma and pleomorphic adenoma

Objective To study the expression of survivin in malignant pleomorphic adenoma,relay pleomorphic adenoma and pleomorphic adenoma.Methods The transcription and interpretation of urvivin mRNA in the salivary gland tumor were analyzed via immunohistochemistry and in situ hybridization methods.Results Survivin mRNA expression and protein expression were almost the same in malignant pleomorphic adenoma,relay pleomorphic adenoma and pleomorphic adenoma.But they were different in the gland around the malignant pleomorphic adenoma.There was a significant difference of Survivin mRNA and protein expression in pleomorphic adenoma and malignant pleomorphic adenoma,and relay pleomorphic adenoma.The expressions were almost the same in relay and malignant pleomorphic adenoma.Conclusion The expression of survivin mRNA and protein is increased in pleomorphic adenoma and may be involved in the onset and progression of pleomorphic adenoma.It indicates that mRNA is relatively stable in the mRNA transcription and protein interpretation in malignant pleomorphic adenoma,and it is also showed earlier in gene amplification than in protein expression of the malignant tumor in the initial stage of malignant pleomorphic adenoma.Survivin can be a new diagnostic/therapeutic target in pleomorphic adenoma.

Tumor growth depends on the tumor microenvironment(TME).Tumor-associated neutrophils(TANs)are important inflammatory cells in TME.TANs are divided intoN1type with anti-tumor effect andN2type of tumor-promoting effect.Therefore,TANs have both beneficial and harmful aspects of the body.A large number of studies have been shown that TANs affect tumor formation,metastasis,angiogenesis and immune response,regulated by the secretion of cytokines and chemokines.This review will summarize the biological characteristics of TANs,and tumor development,prognosis and treatment of tumor as well as research progress of the relationship between TANs and tumor.
3.Clinical application of homemade biodegradable drug eluting stents (EXCEL stents)

Objective To evaluate the clinical effect and safety of homemade biodegradable drug eluting stents(Excel stents)in patients with coronary heart disease.Methods Two hundred and twelve patients who received Excel stents implantation during February,2006 to March 2007 were included in the study.Procedural success rate and complications during hospitalization were monitored and all the patients were followed up clinically up to 8-21 months after discharge.Results The procedural success rate was 99.1%(210/212).A total of 420 Excel stents were implanted while direct stenting was applied in 125 patients(29.8%).Sudden cardiac death happened in 1 patient during hospitalization.Two hundred and six patients finished their clinical follow up and among them,6 patients had recurrant angina.Fifty patients received CAG examination 6-9 months after PCI and 2 patients reached the criteria of restenosis(stenosis ≥50%)and required revascularization.Conclusion Homemade biodegradable drug eluting stents(Excel stents)are safe and effective for PCI in coronary heart disease patients.
4.Experimental study on differentiation of different passages of human bone marrow mesenchymal stem cells into neurons in vitro

BACKGROUND: Bone marrow mesenchymal stem cell (MSC) is a kind of stem cell with potential of self-repair and multi-differentiation. It may differentiate into neuron, adipose cell and osteoblasts.OBJECTIVE: To observe the transforming efficacy of human bone marrow mesencymal stem cells (hMSCs) into neurons in vitro in different generations so as to provide reliable experimental data for the clinical application of MSCs.DESIGN: Single sample was designed.SETTING: Department of Neurology, Sino-Japan Friendship Hospital,Jilin UniversityPARTICIPANTS: Marrow tissue was collected from 9 cases of spinal fusion in Department of Orthopedics of First Hospital affiliated to Jilin University. Of 9 cases, all of them were in known of the experiment.METHODS: The experiment was performed in Sino-Japan Hospital affiliated to Jilin University from September 2002 to February 2003. The primary and generative culture of hMSCs was given. Experimental and the control groups were divided.-mercaptoethanol was taken as inducer. hMSCs of the 2rd, 4th, 6th and 8th generations were selected for induction in vitro for 6 h. Cytochemistry staining and immunohistochemistry staining were used to assay the expressions of neuronal and astrocytic marked proteins.MAIN OUTCOME MEASURES: ① Growth curve analysis on genera tive culture of hMSCs. ② Nissel staining. ③ Immunohistochemical staining.RESULTS: ① Common characters of generative culture: The latent phase of generative culture was 12-24 hours, exponential phase was 7-10 days and 11-13 days later, cell culture entered the platform phase. ② After induction of the 2nd, 4th and 6th generated hMSCs, deep blue granular Nissl body presented in cytoplasm. In 6 hours on the 8th induction, there was no obvious deep blue Nissl structure presented in cytoplasm.③ Except GFAP, NSE and NF-M were expressed in hMSCs of different generations after induction for 6 hours. There was no significant difference in positive rates of the 2nd, 4th and 6th generations (P > 0.05), but the significant difference presented in comparison between the 8th generation and the 2nd, 4thand 6th generations (P < 0.05). CONCLUSION:-mercaptoethanol can induce hMSCs differentiating into neuronal cells in vitro. The positive rates of the 2nd, 4th and 6th generationsare higher remarkably than the 8th generation.
5.Effect of Chinese Herbs Used in Treating Multiple Sclerosis on T Subsets Using Association Rules.

OBJECTIVETo analyze the effect of Chinese herbs used by Prof. LI Tao on peripheral blood T subsets in treating multiple sclerosis (MS) by using association rules and statistical methods, thereby providing evidence for optimizing prescriptions.
METHODSData of MS inpatients and outpatients recorded by data collecting system, Xiyuan Hospital, China Academy of Chinese Medical Sciences were resorted. The relationship between Chinese herbs and T cell subsets were analyzed using SPSS17.0 and Aprior module in SPSS Clementine 12.0.
RESULTSRadix bupleuri, Radix Paeoniae alba, Fructus Aurantii, Atractylodes, and Radix Glycyrrhizae were most commonly used herbal combinations.Radix Aconiti lateralis preparata and Rhizoma Smilacis glabrae were often added. Radix Aconiti lateralis preparata was associated with decreased Th1 cells (confidence level 83.78%, supportive level 36.26%). Decreased Th1 cell was associated with Radix Aconiti lateralis preparata (confidence level 71.26%, supportive level 36.26%).Radix Aconiti lateralis preparata was obviously associated with decreased Th1 cells. Radix Bupleuri, Radix Paeoniae alba, bitter orange, Atractylodes , Radix glycyrrhizae, and Radix Aconiti lateralis preparata could reduce peripheral blood Th1 subsets of MS patients and elevate Th2 subsets (all P < 0.01).
CONCLUSIONSThe herbal combination of Radix Bupleuri, Radix Paeoniae alba, Fructus Aurantii, Atractylodes, Radix Glycyrrhizae, Rhizoma Smilacis glabrae, and Radix Aconiti lateralis preparata could lower peripheral blood Th1 cells and elevate Th2 cells, and prevent the relapse of MS possibly by reducing Th1 cells and elevating Th2 cells. Especially Radix Aconiti lateralis preparata played important roles in aforesaid changes of Th1 and Th2.

8.Relationship between C-peptide and artherosclerosis in lower extremity in patients with type 2 diabetes

Objective To investigate the relationship between C-peptide and artherosclerosis in lower ex-tremity in patients with type 2 diabetes. Methods Two hundred and fourteen type 2 diabetic patients were included in this retrospective study. They were divided into two groups:type 2 diabetes with artherosclerosis in lower extremi-ty in 123 cases and type 2 diabetes without artherosclerosis in lower extremity in 91 cases. Their medical history and the laboratory data were collected such as fasting serum C-peptide levels and postprandial serum C-peptide levels of 1 hour, 2 hours and 3 hours, glycated hemoglobin (HbA1c) and so on. Results Compared with artherosclerosis group in lower extremity, the age, duration of diabetes, total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C), glycated hemoglobin and smoking rate were significantly higher in type 2 diabetes without artherosclero-sis in lower extremity (P < 0.05). However, the high density lipoprotein cholesterol (HDL-C) was much lower in the last group (P<0.05). There were no significant differences in the other indicators between the two groups (P>0.05). Logistic regression analysis showed that fasting and postprandial (1 hour, 2 hours and 3 hours)serum C-pep-tide levels were all negatively related to artherosclerosis in lower extremity (OR 0.524,P = 0; OR 0.440, P = 0;OR 0.688, P=0;OR 0.795, P=0). Conclusions Serum C-peptide level may be an independent associated fac-tor with artherosclerosis in lower extremity in type 2 diabetes.
9.The value of fibrin monomer combined with Wells′ score in early diagnosis of venous thromboembolic disease

Objective To investigate the change of the fibrin monomer (FM) level in the venous thromboembolic disease (VTE).To compare the diagnostic value of FM combined Wells score with the other detection methods.Methods In this case control study,121 cases were selected from the patients who were from general and orthopeadic surgery (including thrombosis group in 60 cases and non thrombosis group in 61 cases).The patients were assessed by Wells score.From one day before surgery, Plasma d-dimer (D-D) and fibrin monomer (FM) were periodic measured by CP-2000 d-dimer and fibrin monomer.Evaluation the value of d-dimer,fibrin monomer and fibrin monomer combined with Wells score in diagnosis of venous thromboembolic disease.The receiver operation cure(ROC) was drew to determine the diagnostic performance.Results The plasma FM level of patients with VTE in the thrombus group (26.11±38.34) μg/ml is higher than the non thrombus group (6.56±6.81) μg/ml and the control group (2.37±0.89) μg/ml (t=-3.82, t=-4.78,P<0.01);the sensitivity of FM was lower than the D-D (85% vs 93%);then the positive predictive value was lower than D-D (82% vs 87%) (χ2=27.01,P=0.000)but its specificity and negative predictive value (65%) are both higher than D-D (65% vs 44%)(71% vs 62%)(χ2=11.67,P=0.001);the sensitivity,the specificity,the positive predictive value and the negative predictive value of FM combined Wells score are increased (90%,85%,83%,89%)(χ2=20.95,χ2=16.65,P<0.01).The increased level of FM is earlier than imaging changes, and the elevated of plasma D-D is not obvious in a certain period of time.Conclusions The sensitivity and specificity of FM combined with Wells′ score is higher in the diagnosis of VTE, its prediction value in the diagnosis of VTE is higher.The FM level can be changed in the early stage of VTE, which has a certain value of early diagnosis.
10.Determination of 4-Hydroxy-3-Methoxy Benzoic Acid by RP-HPLC and Inhibitory Effectof Phloroglucinol on Catecholamine-O-Methyl Transferase

Objective To detect content of 4-hydroxy-3-methoxy benzoic acid by RP-HPLC and observe the inhibitory effect of phloroglucinol on catecholamine-O-methyl transferase (COMT). Methods This study used the principle of 3,4-dihydroxy benzoic acid transforming to 4-hydroxy-3-methoxy benzoic acid under COMT’ s catalytic action. COMT (20 μL) was extracted from mouse liver homogenate. In a reaction system,10 μL catecol (1í10-3 mol·L-1 ) and 10 μL phloroglucinol (5í10-3 , 1í10-3 and 2í10-4 mol·L-1 ,respectively) were added. Products were determined by RP-HPLC to analyze effects of 4-hydroxy-3-methoxy benzoic acid on COMT. Results Phloroglucinol had inhibitory effect on COMT activity at concentrations of 5í10-3 mol·L-1 ,1í10-3 mol·L-1 and 2 í10-4 mol ·L-1 ,with the inhibition rate being 25. 3% ,17. 6% and 8. 9% ,respectively. Conclusion Phloroglucinol has an inhibitory effect on COMT activity,which is weaker than the effect of catechol of the same concentration.
