3.Study on pure red cell aplastic lymphocyte subsets characteristics and the effect of cyclosporin A

Objective To detect and analyse acquired pure red cell aplasia (PRCA) T lymphocyte subsets distribution and to assess its condition and cyclosporine immune function of T lymphocytes subsets.Methods Flow cytometry was applicated to detect peripheral blood T lymphocyte subsets of acquired PRCA patients before and after 3 months of treatment with cyclosporine-based immunosuppressive regimen and normal controls.Results Among 22 patients,17 cases of blood returning normal (three cases of bone marrow returning normal),the total effective rate was 95.5 % (3 cases of cure,14 cases of remission,4 cases of improvement,1 case of ineffectiveness).Th (CD3+ CD4+) cells and Th/Ts ratios in acquired PRCA patient group were lower than those in the normal control group,while Ts (CD3+ CD8+) cells was higher than that in the normal control group,the differences were statistically significant (P < 0.05).After 3 months of treatment,Th cells and Th/Ts ratio were higher than those before,and Ts cells were decreased compared with the previous (P < 0.05).Conclusion Disorders of T lymphocyte subsets in acquired PRCA patients lead to immune dysfunction,however,cyclosporine can improve T lymphocyte subsets in patients with imbalance,which is an effective way to treat the disease with significant curative effect and mild adverse reactions.
4.Relationship between the Chinese Version Weehsler intelligence scale for children and the Das-Naglieri cognitive assessment system in children with attention deficit hyperactivity disorder

Objective To explore the relationship between the Chinese Version Wechsler intelligence scale for children(C-WISC) and the Das-Naglieri cognitive assessment system(DN:CAS) in children with attention deficit hyperactivity disorder (ADHD).Methods Thirty-two children with ADHD were evaluated with C-WISC and the digital remote assessment system——DN:CAS,and both results were administered for correlation analysis.Results The results showed that there was a strong correlation between the full intelligence quotient (FIQ) of C-WISC and the full score of DN:CAS,also the scores in Plan,Simultaneous,and Successive of DN-CAS,and the correlation coefficient was between 0.6 to 0.8.There was a strong correlation between the full score of DN:CAS and the verbal comprehension IQ (VCIQ),perceptual organization IQ (POIQ),memory/concentration IQ (M/CIQ),verbal IQ (VIQ),and performance IQ(PIQ) of C-WISC,as the correlation coefficient ranges from 0.6 to 0.8.There was also a strong correlation between the score in Planning and VCIQ,POIQ,M/CIQ,VIQ of C-WISC.As to the score in Simultaneous of DN:CAS and POIQ,VIQ,PIQ of C-WISC,the correlation was also strong with the correlation coefficient ranging from 0.6 to 0.8.For the rest of the sub-tests of C-WISC and DN:CAS,there was moderate correlation between them,the correlation coefficient was from 0.4 to 0.6.Conclusions These results show that the correlation between C-WISC and DN:CAS in children with ADHD is significant.The correlation coefficient shows diversity in the sub-tests of C-WISC and DN:CAS.
5.Experiment of amnion epithelial cell suspension liquid used for acute rabbit corneal alkali burn

AIM:To investigate the effects of amnion epithelial cell ( AEC) suspension liquid on the biological behavior of the rabbit's corneal epithelium, combined with the in vitro and in vivo experiments. · METHODS: The rabbit's corneal epithelium were cultured in the lower chamber of transwell, and AEC suspension liquid was dropwised in the upper chamber. There was only culture medium in the upper chamber of the control group. The proliferation of rabbit's corneal epithelium was observed with CCK-8 automated colorimetry and the expression of PCNA was detected by immunocytochemistry. We used the scratch wound assay to detect the migration of corneal epithelial cell ( CEC ) . The in vivo models were established by placing a 10mm diameter corneal trephine in the center of the cornea, within 1mol/L NaOH for 1min. We divided those into three groups: treatment group of AEC suspension liquid eye drop, AEC suspension liquid subconjunctival injection and the control group without any treatment. Using the slit- lamp biomicroscope and fluorescence staining to observe the cornea per week. After 28d we took the eyeballs with the HE staining. The expression of VEGF was detected by immunohistochemistry. ·RESULTS: The activity of CEC with AEC treatment was much higher than the control group ( P< 0. 05 ). The expression of PCNA increased in AEC group (P<0. 05). And the migration of CEC in the AEC group was faster than the control one. In vivo, the inflammation of the corneal and the CNV of the AEC group were all significantly reduced compared with the control group (P<0. 05 ). There were less invasive cells and more ordered organization arrangement in ACE group observed by the HE staining. The expression of VEGF and mcp-1 in these two AEC treatment groups all significantly decreased compared with the control group (P<0. 05). ·CONCLUSION: AEC suspension liquid can promote the proliferation and migration of the rabbit's corneal epithelium. The potential of AEC suspension liquid as a therapy for acute corneal alkali burn.
6.Study on quality assessment of Polygalae Radix based on HPLC-DAD fingerprint.

OBJECTIVETo establish an HPLC fingerprint to evaluate the quality of Polygalae Radix, root xylem, and those collected in different growth ages or harvest time.
METHODSeparation was performed at 30 °C on a Kromasil C18 column (4.6 mm x 250 mm, 5 μm); the mobile phases was acetonitrile and 0.05% H3PO4 water in the gradient elution; the flow rate was set at 1.0 mL · min(-1) and the detection wavelength at 314 nm; the quality discriminant analyses were accomplished by means of similarity analysis, cluster analysis, principal component analysis and neural network model.
RESULTIn 26 batches of Polygalae Radix, 24 batches fingerprint similarities were above 0.8. In 5 different growth or harvest time batches, 4 batches were above 0.8; in 8 batches root xylem samples, the similarities were all above 0.875. The similarity analysis was in accord with the quality discriminant analysis of cluster analysis, principal component analysis and neural network model.
CONCLUSIONFingerprint combined with chemical pattern recognition technique can effectively evaluate the quality of Polygalae Radix. The active substance species are all similar in cultivated, wild, different growth or harvest time Polygalae Radix and polygala root xylem, but the chromatography peak areas are different. The effective material contents are similar between wild and cultivated Polygalae Radix, but each chromatographic peak area of the root xylem is much smaller than that of Polygalae Radix. The chemical substance accumulation mainly depends on harvest month, but little growth time in Polygalae Radix.

7.Trabeculectomy punch combined with a novel RS tube for the treatment of punctal stenosis

AIM: To investigate the feasibility and clinical effect of punctoplasty by using trabeculectomy punch combined with a novel RS tube for the treatment of punctal stenosis.METHODS: Totally 39 patients (39 eyes) with punctual stenosis were selected from October 2013 to October 2015 in the Second People`s Hospital of Foshan.All patients underwent punctoplasty by using trabeculectomy punch combined with a novel RS tube.These tubes were removed at 3mo after operation.A follow-up of 6mo was taken for final analysis.The fluorescein dye disappearance test score was recorded before the operation and at 1,3 and 6mo after the extubation.The curative effect of the operation at 6mo after the extubation was assess.RESULTS: Fluorescein dye disappearance test: the scores at 1,3 and 6mo after the extubation all decreased compared with the preoperative ones.The difference was statistically significant(P<0.05).At the last following up, 35 eyes (90%) were cured completely, 4 eyes (10%) were improved significantly, no patients recurred.Effective rate was 100%.No serious intraoperative and postoperative complications happened.CONCLUSION: Punctoplasty by using trabeculectomy punch combined with novel RS tubes is a safe and effective method for the punctul stenosis, which is easy to perform, with high success rate.
8.Experimental study of nourishing Piyin Remedy on rat with spinal cord injury

ObjectiveTo study the therapeutic effect and mechanisms of nourishing Piyin Remedy (nPR) on experimental rat spinal cord injury.MethodsThirty-two healthy SD rats were divided into 2 equal groups randomly: nPR group and injured group. Animal model of incomplete injury of spinal cord was made by Allen's equipment on rat's T8—T9 segment. The spinal nerve function,SEP,retrograde and label technique of horse radish peroxidase,gross observation,histological and morphometric analysis were taken as the observed indices.ResultsThe value of observed indices of nPR group were improved evidently compared with injured group.ConclusionNourishing Piyin Remede can hold back the secondary spinal cord injury and accelerate the recovery of spinal nerve function.
9.Rapid determination of dopamine and its metabolites during in vivo cerebral microdialysis by routine high performance liquid chromatography with electrochemical detection.

OBJECTIVETo determine dopamine and its metabolites during in vivo cerebral microdialysis by routine high performance liquid chromatography with electrochemical detection.
METHODSMicrodialysis probes were placed into the right striatum of Wistar rat brains and perfused with Ringer's solution at a rate of 1.5 microL/min. A reverse phase HPLC with electrochemistry was used to assay DA, DOPAC, and HVA after cerebral microdialysates were collected every 20 minutes from awake and freely moving rats. In order to identify the reliability of this method, its selectivity, linear range, precision and accuracy were tested and the contents of DA, DOPAC, and HVA in rat microdialysates were determined.
RESULTSThe standard curve was in good linear at the concentration ranging from 74 nmol/L to 1.5 micromol/L for DOPAC (r2=0.9996), from 66 nmol/L to 1.3 micromol/L for DA (r2=1.0000) and from 69 nmol/L to 1.4 micromol/L for HVA (r2=0.9992). The recovery of DOPAC (0.30, 0.77, 1.49 micromol/L), DA (0.26, 0.69, 1.32 micromol/L), and HVA (0.27, 0.71, 1.37 micromol/L) was 82.00+/-1.70%, 104.00+/-4.00%, 98.70+/-3.10%; 92.30+/-1.50%, 105.30+/-2.30%, 108.00+/-2.00%; 80.00+/-7.80%, 107.69+/-8.00%, and 108.66+/-3.10%, respectively at each concentration. Their intra-day RSD was 3.3%, 3.4%, and 2.5%, and inter-day RSD was 4.2%, 2.3%, and 5.6%, respectively. The mean extracellular concentrations of DOPAC, DA, and HVA in rat brain microdialysates were 10.7, 2.4, and 9.2 micromol/L (n=6), respectively.
CONCLUSIONThe findings of our study suggested that the simple, accurate and stable method can be applied to basic researches of diseases related to monoamines neurotransmitters by cerebral microdialysis in rats.

Objective To explore the cognitive process in children with attention deficit hyperactivity disorder (ADHD) using Das-Naglieri Cognitive Assessment System (DN:CAS).Methods Sixty-one children with ADHD and sixty-one healthy controls were enrolled in this study.The cognition of all the studies individuals was evaluated by the DN:CAS.Results Compared with the control group,children with ADHD had significantly lower scores in planning[(24.18 ±5.64) scores vs (30.07 ±5.28) scores,t =-6.985,P < 0.01],simultaneous processing[(33.97 ± 7.64) scores vs (36.90 ± 7.20) scores,t =-2.211,P < 0.05],attention [(26.52 ± 6.80) scores vs (31.21 ± 5.86) scores,t =-4.909,P <0.01] in the 4 subtests of DN:CAS,and the total scores [(117.85 ±21.45) scores vs (132.48 ± 17.95) scores,t =-5.286,P < 0.01] are same.And there were significant differences in matching numbers [(8.77 ± 2.33) scores vs (10.87 ± 2.31) scores,t =-5.220,P < 0.01],planned codes [(7.87 ± 2.37) scores vs (9.44 ± 2.61) scores,t =-4.006,P < 0.01],planned connections [(7.54 ± 2.29) scores vs (9.75 ± 2.01) scores,t =-6.643,P <0.01];verbal-spatial relations in simultaneous processing [(10.05 ±2.79) scores vs (11.23 ± 3.19) scores,t =-2.382,P < 0.05] and expressive attention [(9.02 ± 2.47) scores vs (10.34 ± 3.03) scores,t =-2.938,P <0.01],number detection [(8.16 ±2.31) scores vs (10.08 ± 1.86) scores,t =-5.028,P < 0.01],receptive attention [(9.34 ± 3.68) scores vs (10.78 ± 3.20) scores,t =-3.215,P < 0.01] between ADHD group and the control group.There were significant differences of the percentile rank distribution of the scores between ADHD group and the control group in planning,attention and full score (all P < 0.05).Conclusions Children with ADHD have cognitive deficits in the cognitive processing,and planning and attention deficits are the main features.The DN:CAS could be a useful tool in evaluating cognition of children with ADHD.
