2. Cinnamaldehyde ofloxacin-3-ylhydrazone induces apoptosis of human pancreatic carcinoma cells

OBJECTIVE: To study the effect of cinnamaldehyde ofloxaein-3-ylhydrazone (FQ)16) on apoptosis of human pancreatic carcinoma cells in vitro. METHODS: Human pancreatic carcinoma cell line BxPC-3 was treated with FQ16 at different concentrations. The proliferation inhibition was examined by MTT assay. Morphological examination, TUNEF and agarose gel electrophoresis method were used to detect apoptosis. Western blotting method was used to detect the expression of apoptosis related genes caspase-3 , caspase-9, caspase-8, bcl-2, bax and cytochrome C. RESULTS: The cell proliferation was inhibited by FQ16 in a time- and a dose-dependent manner. Treatment of BxPC-3 cells with different concentrations of FQ16 for 24 h increased the percentage of the apoptotic cells obviously (P < 0.05); the morphology of BxPC-3 showed changes such as nuclear chromatin condensation and fragmentation and typical ladder DNA in apoptotic cells. FQ16 increased protein expression of bax, caspase-9 and caspase-3, and induced cytosolic accumulation of active caspase-9 and caspase-8, whereas the protein expression of bcl-2 decreased. Western blotting results reveal that FQ16 released mitochondrial cytochrome C into cytosol in a dose-dependent manner. CONCLUSION: Apoptosis in pancreatic carcinoma BxPC-3 cells is one of the key mechanisms of action of cinnamaldehyde olloxacin-3-ylhydrazone, and mitochondrial-dependent pathways are involved in the induction of apoptosis of BxPC-3 cells. 3. Increased expression of the 20S proteasome in peripheral blood mononuclear cells of type 2 diabetic patients

Objective: To investigate the dynamic expression of the 20S proteasome in peripheral blood mononuclear cells (PBMCs) of type 2 diabetic patients without vascular complications. Methods: PBMCs were prepared from 30 type 2 diabetic patients and 30 nondiabetic controls. The general indexes including weight, height and blood pressure were recorded. Fasting plasma glucose, fasting plasma insulin and glycosylated hemoglobin were measured. The protein level of the 20S proteasome was measured by Western blotting. The mRNA expression levels of the 20S proteasome β1, β2 and β5 submits were detected by real-time PCR. Results: Compared with that in the nondiabetic controls, the protein level of the 20S proteasome was significantly increased in the diabetic patients and was positively associated with glycosylated hemoglobin. Conclusion: Type 2 diabetic patients without vascular complications have an increased 20S proteasome expression, the significance of which needs to be explored by further study.

5.Effects of cyclopamine on the proliferation and apoptosis of LNCaP cells and expression of the PCA3 gene in human prostate cancer.

OBJECTIVETo explore the effects of cyclopamine on the proliferation and apoptosis of LNCaP cells and the expression of the PCA3 gene in human prostate cancer in vitro.
METHODSLNCaP cells were treated with cyclopamine at the concentrations of 1, 5, 10 and 15 micromol/L for 24, 48 and 72 hours. The inhibitory effects of cyclopamine on the proliferation and apoptosis of the LNCaP cells were detected by MTT and flow cytometry respectively, the morphological changes of the cells observed by Hoechst 33258 staining, and the expression of the PCA3 gene determined by real-time fluorescence quantitative reverse transcriptase polymerase chain reaction (FQ-RT-PCR).
RESULTSCompared with the blank control group, cyclopamine significantly inhibited the proliferation of the LNCaP cells at 5, 10 and 15 micromol/L (P <0.01), reaching IC50 at 10 micro mol/L at 48 hours. The apoptosis rates of the LNCaP cells at 24, 48 and 72 hours were 37.21%, 57.38% and 57.98% in the 10 micromol/L group and 21. 16% , 71.31% and 72.90% in the 15 micro.mol/L group, significantly different from those in the control (P <0. 01). The cell apoptosis showed a rising trend with the increase of cyclopamine concentration and acting-time, while the expression of the PCA3 gene was decreasing with the increased concentration of cyclopamine, significantly lower than that of the blank control group (P <0.01) , and extremely low in the 10 micromo/L group
CONCLUSIONCyclopamine intervention at 10 and 15 micromol/L for 48 and 72 hours could significantly inhibit the at all time points. Proliferation and induce the apoptosis of LNCaP cells and reduce the expression level of PCA3.

10.Phacofragmentation combined with vitrectomy in the treatment of traumatic dislocation lens

AlM: To explore the clinical application effect of phacofragmentation combined with vitrectomy in the treatment of traumatic lens dislocation.
METHODS:Totally 16 cases (16 eyes) of traumatic lens dislocation treated with phacofragmentation combined with vitrectomy were retrospectively analyzed, including 6 cases of high intraocular pressure ( lOP) and 10 cases of vitreous hemorrhage, and 1 case of retinal detachment. All patients were given conventional flat line standard three channel vitreous operation incision to remove the anterior, middle part and peripheral vitreous around lens dislocation. The crystalline lens were drawn to the center cavity of vitreous body and treated by ultrasonic disintegrator. ln the operation, the retina was examined and 8 of them had no retinal damage and in the first stage underwent fixation of posterior chamber intraocular lenses.
RESULTS: All crystalline lens dislocated were completely grinded and suctioned. There was no retinal detachment occurred in 3mo followed up. 16 eyes had normal lOP (12-20mmHg) at 1wk after operation. The average visual acuity was improved and with 8 cases got improved of 0. 2 or more 1wk after operation.
CONCLUSlON: Our research shows that phacofragmentation combined with vitrectomy is a safe and effective method for the treatment of traumatic lens dislocation. The patients with nondestructive retina in the operation are feasible to do first stage operation of intraocular lens suture fixation, which contributes to the best visual acuity.
