China Pharmacy 2016;27(19):2654-2656
OBJECTIVE:To study the preventive effects of arhinhor-11 on acute alcoholism in mice. METHODS:48 mice were randomly divided into model group (sodium carboxymethyl cellulose),arhinhor-11 low-dose,medium-dose and high-dose groups [1.23,2.46,4.92 g(crude drug)/kg],with 12 mice in each group. Each group was given relevant medicine intragastrically, and 30 min later given 56% liquor intragastrically to induce acute alcoholism model. The disappearance time and recovery time of righting reflex were determined. 160 mice were grouped, given medicine and modeled with the same method, with 40 mice in each group. The alcohol concentration and alcohol dehydrogenase(ADH)activity in liver of 10 mice were determined 0.5,1,2,3 h af-ter giving alcohol. Other 10 mice were included in normal control (ig administration of sodium carboxymethyl cellulose). RE-SULTS:Compared with model group, the disappearance time of righting reflex were prolonged while recovery time shortened in arhinhor-11 groups, and alcohol concentration and ADH activity increased 0.5,1,2,3 h after giving alcohol (P<0.05 or P<0.01),especially 1 h after giving alcohol. 3 h after giving alcohol, alcohol concentration in blood of arhinhor-11 high-dose group decreased while ADH activity increased (P<0.05);there was no statistical significance among other arhinhor-11 groups (P>0.05). CONCLUSIONS:Arhinhor-11 has preventive effect on acute alcoholism in mice. It has a positive control effects on the alco-hol concentration in blood and ADH activity in liver.

Objective To determine the incidence of clinically common acute avulsions of the popliteus tendon (PT) and the lateral collateral ligament (LCL) of the femur (peel-off lesion) in knees with multiple injured ligaments. Methods A retrospective review of 48 acute grade 3 posterolateral corner (PLC) knee surgeries was conducted. All office evaluations were reviewed to determine the demographic data, tear description, preoperative MRI examination, physical examination under anesthesia, intraoperative arthroscopic findings and surgical techniques. Results In total,19 patients were surgically verified as having femoral peel-off lesions as a portion of a series of multiple pathologic findings.Tear patterns were typically categorized into 3 patterns:isolated PT tear (pattern 1,4 patients),combined PT and LCL tears (pattern 2,8 patients) and complex tears (associated intrasubstance and/or fibular-based injuries, pattern 3,7 patients).Seventeen of the 19 patients underwent preoperative MRI examination, which demonstrated conclusive signs of femoral insertion separation and discontinuity in 13 patients (76%) and inconclusive signs of femoral insertion separation.Examination under anesthesia (EUA) revealed that for the 4 patients with pattern 1 injuries, the tibial external rotation increases were all greater than 10°.For the 15 patients with pattern 2 and 3 injuries, the yarus instability was as follows:grade 3 in 8 patients, grade 2 in 2,grade 1 in 1,and negative in 4.External rotation increases were greater than 10° in 12 patients and less than 10° in 3.Seventeen of the 19 patients were inspected with an arthroscope prior to PLC surgery:of these patients,94% had acute avulsion findings and positive lateral gutter drive-through sign;8 patients were repaired with recess or reattachment procedures;6 patients were primarily reconstructed;4 patients were treated with combined repair and reconstruction;and 1 patient was left untreated for secondary reconstruction due to priority repair for concurrent patellar tendon rupture. Conclusion For adequate clinical identification,we recommend a comprehensive diagnostic protocol, including MRI,physical examination and arthroscopic inspection,to avoid underdiagnosis of the lesion in multiple-ligament injured knees. The lateral gutter arthroscopic evaluation is particularly sensitive for detecting the lesion.

Specific primers 1CaA/1CaB for full cry1C gene in Bacillus thuringiensis subsp.colmeri strain 15A3 were designed. The 4.0kb PCR product included the whole ORF and regulation region of cry1C gene. This PCR product was linked with shuttle vector pHT315 by two cloning steps. The recombined plasmid pHT-1C was electroporated into Bacillus cereus 9509, a kind of bacteria that beneficial to crops. The transformant could produce bipyramidal-shaped parasporal inclusions. The 60kD protein band was detected by SDS-PAGE. The bioassay result showed that the cry1C gene transformant of Bc 9509 had insecticidal activity to Spodoptera exigua.

China Journal of Chinese Materia Medica 2014;39(1):83-88
Chemical constituents of Inula japonica were isolated and purified by repeated column chromatographies, over silica gel, and Toyopearl HW-40, and preparative HPLC. On the basis of spectral data analysis, including NMR and MS data, the structures of the isolates were elucidated and their anti-inflammatory activities were assayed. Fifteen compounds were isolated from the ethyl acetate extract of I. japonica, and their structures were elucidated as dihydrosyringenin (1), (3S, 5R, 6S, 7E)-5,6-epoxy-3-hydroxy-7-megastigmen-9-one (2), (6R, 7E) -9-hydroxy-4,7-megastigmadien-3-one (3), arnidiol (4), taraxasterol acetate (5), 8,9,10-trihydroxythymol (6), taxifolin (7), luteolin (8), napetin (9), eupatin (10), spinacetin (11), quercetin (12), p-hydroxycinnamic acid (13), caffeic acid (14), and caffeoyl acetate (15). Compounds 1, 2, 7, 13 and 15 were isolated from the genus Inula for the first time, and compounds 3, 4, 9-11 and 14 were isolated from this plant for the first time. The anti-inflammatory activity result showed that compounds 3, 6-12 and 14 exhibited inhibition effect against leukotriene C4 (LTC4) synthesis and degranulation definitely in c-Kit Ligand (KL) induced mast cells, and compound 8 and 12 also had the suppression effect against lipopolysacharide(LPS) induced nitric oxide (NO) activity in RAW264.7 macrophages. It is firstly reported that compounds 7 and 9-11 possessed potent inhibition activities against LTC4 generation and degranulation in mast cells.

Objective To evaluate the role of oxidation reaction in the decrease in the activity of acetylcholinesterase (AChE) in the diaphragm of rats with sepsis. Methods Thirty pathogen-free healthy adult male Sprague-Dawley rats, weighing 220-260 g,were divided into 3 groups using a random number table:sham operation group (group S,n=8),sepsis group (group Sep,n =12) and antioxidant group (group Ant,n=10).Sepsis was induced by cecal ligation and puncture.Group S underwent simple laparotomy.N-acetylcystein 20 mg/kg was injected subcutaneously at 3,6,12 and 18 h after operation, and deferoxamine 20 mg/kg was injected subcutaneously at 3 h after operation in group Ant,while the equal volume of normal saline was given instead in S and Sep groups. At 24 h after operation, the surviving rats were sacrificed, and the diaphragm was removed for determination of the malondialdehyde (MDA) content and activities of AChE,myeloperoxidase (MPO),superoxide dismutase (SOD) and catalase (CAT).SOD/CAT ratio was calculated. Results Compared with group S,the AChE activity was significantly decreased, and MPO and SOD activities were increased in Sep and Ant groups, the MDA content and SOD/CAT ratio were significantly increased, and the CAT activity was decreased in group Sep,and the CAT activity was significantly increased in group Ant (P<0.05).Compared with group Sep,the AChE and CAT activities were significantly increased, and the MDA content,MPO and SOD activities and SOD/CAT ratio were decreased in group Ant (P<0.05).Conclusion Oxidation reaction is partially involved in the decrease in the activity of AChE in the diaphragm of rats with sepsis.
