Alzheimer's disease(AD),which is one type of senile deimentia, has three remarkable pathologic characteristics-senile plaque,neurofibrillar tangles and neuronal death. As a neurotrophic factor, basic fibroblast growth factor(bFGF)is closely related to AD. This article reviewed the interaction between bFGF and AD-associated gene products(such as amyliod ? protein,presenilin, apoloprotein E and microtubulin associated protein tau)and neuronal apoptosis. Some researchers suggested that bFGF may be put forward as potential therapeutic agent in AD and play a role in preventing or retarding the pathological process of this disease, alleviating or even eradicating the pathological insults of this disease.
3.The Antagonistic Effect of PGE_1 on TGF?_1 mRNA Expression and Rat Renal Interstitial Fibrosis Induced by Unilateral Ureleral Obstruction

Objective To explore the possible role of PGE 1 in tubulointerstitial fibrosis formation. Methods Rat model of unilateral ureteral obstruction(UUO) was established.In situ hybridation(ISH) technique, morphological observation and biochemical analysis were applied to determine TGF? 1 mRNA expression, changes of tubulointerstitium and rencortical hydroxyproline content after used PGE 1. Results 3 days after UUO,the rats renal interstitial edema, a number of mononuclear cells recruitment. On day 14,tubule atrophy, interstitial proliferative fibrosis and area enlarged,hydroxyproline content,TGF? 1 mRNA expression increased, the peak value of the latter occurred early than that of hydroxyproline peak value and interstitial area enlargement.PGE 1 could down -regulate these changes.Conclusions The antagenistic effect of PGE 1 on renal interstitial fibrosis may be partly associated with down-regulating overexpression of TGF? 1 mRNA and inhibiting mononuclear cells recruitment.
4.Hepatitis B Virus X Protein Upregulates Tumor Necrosis Factor-? Expression of Rat Mesangial Cell Line Via Extracellular Regulated Protein Kinases Pathway

Objective To investigate the extracellular regulated protein kinases (ERKs) pathway of hepatitis B virus(HBV) X protein(HBx) on glomerular mesangial cell(GMC) proliferation of rat and tumor necrosis factor(TNF)-? expression.Methods The HBV X gene was amplified by polymerase chain reaction(PCR),inserted into the eukaryotic expression vector PCI-neo and confirmed by restrict endonuclease digestion and sequence analysis.PCI-neo contained HBV X gene (PCI-neo-X) was transfected into cultured GMC via liposome.GMC proliferation,TNF-? and its mRNA expression were investigated in the condition of with or without U0126 in the culture media.HBx,ERK1/2 and phosphorynated-ERK1/2 (p-ERK1/2) expression in GMC were assessed by Western blot.TNF-? mRNA expression was assessed by semi-quantitative reverse transcriptase-PCR (RT-PCR).TNF-? level in supernatants was measured by enzymelinked immunosorbent assay(ELISA).GMC proliferation was assessed by methyl thiazolyl tetrazolium(MTT).Results HBx expression was found in transfected GMC,and became prominent at 36 and 48 hours after transfection whether with or without U0126 in culture media.TNF-? mRNA expression was significantly decreased in U0126 group compared with U0126-free group.TNF-? levels in supernatants in PCI-neo-X transfection without U0126 group were (189.0?18.1) ng/L at 36 hours and (172.3?24.3) ng/L at 48 hours after transfection,respectively.In contrast,TNF-? levels in supernatants with U0126 were (65.6?11.6) ng/L and (84.0?24.6) ng/L,respectively.The TNF-? le-vels in the latter groups were significantly lower than the formers (Pa

BACKGROUND:Bone marrow mesenchymal stem cells(BMSCs)are widely utilized as seed cells or carriers in bone tissue engineering and gene therapy.Thus,how to obtain BMSCs with high purity arose more attentions of researchers.OBJECTIVE:To observe the effects of different separation methods and culture conditions on biological characteristics of rabbit BMSCs.METHODS:BMSCs were obtained by whole bone marrow culture,density grand centrifugal and red blood cell lysis.At 48 hours after culture,the cell numbers were counted,the time of passage was recorded,in addition,the cell morphology was observed by phase contrast microscope,and the CD44 antigen expression was identified using flow cytometry.The 3rd and the 7th generation aging cells were cultured with DMEM-LG,MEM-HG,and DMEM/F12 culture medium.MTT and count cell plat were used to evaluate the growth of BMSCs.Phase contrast microscope was used to observe the morphological changes of aging cells.RESULTS AND CONCLUSION:BMSCs could be separated by each method.The adherent cells showed shuttle or multiple angle shapes,with rich cytoplasm,and positive for CD44 antigen.The more cell number and shortest primary culture time was presented in red blood cell lysis group(P < 0.05).DMEM/F12 could promote the proliferation of quiescent cells.And the cells prevented the better viability.The method of read blood cell lysis improving the efficiency of BMSCs adherent is an effective method of extraction of BMSCs.DMEM/F12 could promote the proliferation Maybe,DMEM/F12 is more suitable for BMSCs.
8.Impact of Type 2 Diabetes on Manifestations and Radiological Images of Pulmonary Tuberculosis

Objective To explore the clinical features of pulmonary tuberculosis complicated with DM and related factors. Method The manifestations of 87 newly diagnosed PTB patients with type 2 DM(DM-PTB group) at two hospitals in Tianjin during October 2003 to October 2004 were compared to 88 PTB patients without DM who were randomly choosen from the same hosipital.The factors related to sputum-positive TB or cavity were explored through unconditional logistic regression model. Results DM-PTB patients showed significanly lower frequencies of cough(66.7% vs 83.0%),toxicity symptoms(37.9% vs 62.5%) and haemoptysis(16.1% vs 33.0%) than those of PTB group.Positive-sputum(63.2% vs 34.1%),cavity(56.3% vs 13.6%),wide lesions and bilateral lesions on chest radiographs was more common in DM-PTB group than in PTB one.Introversion(OR=1.582, 95%CI:1.063 ~2.355),type 2 DM(OR =2.186,95% CI:1.082 ~4.420)and cavity(OR =2.793,95% CI:1.308 ~5.964)were factors independently associated with positive-sputum TB.The factors related to cavity included diabetes(OR=14.199,95%CI:5.376~ 37.501),cough,postive sputum and lore-up lobe lesion.Conclusion This study confirmed that clinical manifestations and chest radiographs of PTB patients complicated with type 2 DM significantly departed from the typical presentation.Type 2 DM seems to have a negative effect on postive-sputum and cavity.So it should be alarmed that DM-PTB patients who tend to have untypical clinical symptom and higher rate of positive sputum become the reservoirs of infection to cause the transmission of TB.
9.The Clinical Characteristics and Incidence Tendency Analysis on 1 628 Cases of Colorectal Carcinoma

Objective To provide the basis for clinical diagnosis and treatments of colorectal cancer by studying the clinical characteristics and incidence tendency of colorectal cancer in recent 12 years in Kunming. Methods The data of 1628 colorectal cancer patients from 2001 to 2012 in the First Affiliated Hospital of Kunming Medical University were collected. The clinic pathological data of all patients was divided into 2001-2006 (Agroup) and 2007-2012 (B group) two consecutive time periods for statistical analysis according to its time of onset. Results The incidence of colorectal cancer in the city rose from 49.2%to 56.0%, and the incidence of colorectal cancer in rural areas decreased from 50.8%to 44%( <0.05) .The location ratio of colorectal cancer in the rectum decreased from 74.8%to 73.9%, in the left half colon ratio increased from 14.8%to 19%, while the right colon ratio decreased from 10.4% to 7.1% ( < 0.05) . The change in the three parts had statistically significant difference between two consecutive time periods. The percentage of colorectal cancer patients in Dukes A stage rose from 13.4% to 18.8%, while the percentage of patients in Dukes D stage decreased from 22.3% to 14.5% ( <0.01) . Conclusions The incidence of colorectal cancer in Kunming gradually increases.The morbidity in the city is significantly higher than that in the country. The ratio of left half colon cancer is rising, but the ratio of rectal cancer and right half colon cancer is falling. The early diagnosis rate of colorectal cancer is on the rise.
10.Research on surgical methods for angle-closure glaucoma with different closure conditions accompanied with cataract

? AIM: To observe the efficacy and safety of the phacoemulsification combined with goniosynechialysis and the phacoemulsification combined with trabeculectomy for angle-closure glaucoma ( ACG ) with different closure conditions accompanied with cataract.?METHODS:A total of 65 patients (70 eyes) with primary ACG accompanied with cataract were selected from those admitted in our hospital and were given phacoemulsification with goniosynechialysis ( group A, 30 patients with 33 eyes ) and the phacoemulsification with trabeculectomy ( group B, 35 patients with 37 eyes ) according to the conditions of the closed anterior angle, respectively. The two groups of patients were observed for preoperative and postoperative intraocular pressure ( IOP ) , vision, anterior chamber depth, and complications, and were followed up for 6. 5mo (6-8mo) on average.?RESULTS: Mean postoperative IOP in either group A or group B at 1mo after operation was of statistically significant difference, compared with mean preoperative IOP (P<0. 05). Postoperative 1-week vision in each group was of statistically significant difference, compared with preoperative vision (P<0. 05). Postoperative 1-month mean anterior chamber depth in each group was of statistically significant difference, compared with preoperative anterior chamber depth (P<0. 05). There were no significant differences between the two groups on postoperative 1-month mean anterior chamber depth ( P>0. 05) and postoperative complications (P>0. 05).?CONCLUSION: Clinical doctors can choose appropriate treatment according to different conditions of the closed anterior angle in patients with primary angle- closure glaucoma.
