1.Antisense oligonucleotide mediated inhibition on telomerase activity in gallbladder carcinoma cell

Objective:To study the antisense oligonucleotide mediated inhibition on telomerase activity and cell proliferation of GBC-SD cell.Methods:We design the antisense,sense,and random oligonucleotide with phosphoric acid modification for the hTR(Human Telomerase RNA)template sequence.MTT and PCR methods were used to observe the inhibition on telomerase activity and cell proliferation of GBC-SD cell ,and fibroblast cells were used as control group.Results:PS-ODN can lead to the reduction of cell survival rate of GBC-SD cell,wich dosage dependence.Tne experimental group cell detected by scanning electron appeared apoptotic feature.Conclusion:PS-ODN can inhibit telomerase activity of GBC-SD cell effectively and induce the cell apoptosis.
2.Effect of xanthoxylin on A23187-and calcium-induced platelet aggregation and cytosolic free-Ca2+ level

Objective To study the effects of xanthoxylin on platelet aggregation and freeCa2+ level induced by CaCl2 and A23187. Methods Platelet aggregation and free Ca2+ level in the platelet cytosol were measured by turbidimetry and fluorimetry of Fura-2/AM. Results Xanthoxylin at the concentration of 0.037, 0.185, 0.924, 9.240, 92.40 μmol/L significantly inhibited CaCl2 and A23187-induced platelet aggregation, with the inhibition rates ranging from 9.0% to 67.1% for CaCl2 and from 18.1% to 72.5% for A23187. A23187-induced free-Ca2+ level in the platelet cytosol was also significantly reduced (from 12.8% to 63.4%). Conclusion The inhibition rate of platelet aggregation was positively correlated with the reduction rate of cytosolic free-Ca2+ level by xanthoxylin.

4.The related factors of coronary CTA failure and nursing intervention of the second examination

Objective To summarize the related factors of coronary CT angiography (CTA), and to explore the nursing strategy of the second examination for patients with coronary artery disease (CTA).Methods A total of 70 patients with coronary heart disease underwent coronary CTA examination in our hospital were as research objects, and were divided into control group (coronary CTA success) with 50 cases and observation group (coronary CTA failure) with 20 cases, related factors of CTA failure were summarized and the nursing intervention of the second examination was discussed.Results There was no significant difference in two groups in gender, age, disease duration, severity of symptoms, lesions location, and manifestations(P>0.05);Blood pressure, heart rate, respiration, contrast agent injection rate showed significant difference (P<0.05);Logistic analysis results indicated that the heart rate and breathing independent were risk factors leading to coronary CTA failure (P<0.05).Conclusion Heart rate, and respiratory failure are independent risk factors for coronary CTA failure.Psychological intervention, breathing training, and heart rate controlling drugs will contribute to the successful implementation of coronary CTA for secondary examination.
