1.Study on insulin sensitivity and islet p cell's function in obesity and non-obesity type 2 diabetes

Objective To study of insulin sensitivity and (3 cell's function in obesity and non-obesity type 2 diabetes for further exploring the effect of obesity on the degree of insulin resistance and islet ? cell's function in type 2 diabetes. Methods Insulin sensitivity index (S1), acute insulin response to glucose (AIRg) and disposition index (DI = S1XAIRg) were assessed by the reduced sample number of Bergman's minimal model method with frequently sampling bloodin intravenous glucose tolerance test in subjects of normal controls, obesity type 2 diabetes and non-obesity type 2 diabetes. Meanwhile the clinical parameters such as body mass index and waist to hip ratio were also evaluated. 2.High frequency ultrasound expression of abdominal wall endometriomas and diagnostic value

Objective To explore the character and clinical value of high frequency ultrasound in diagnosing abdominal wall endometriomas .Methods The characteristics of the ultrosonographic images in 32 cases with abdomi-nal wall endometriomas by operation and pathology were analyzed retrospectively .Results A total of 33 lesions, including 1 case of multiple (2 lesions),the rest of the 31 cases were single,mostly in the original abdominal incision place or incision side subcutaneous soft tissue ,1 case away from the incision ,located in the inguinal region .26 lesions was located in the subcutaneous fat and 6 lesion in the rectus abdominis muscle .All of the lesions hadn't capsule and were irregular in shape .The internal echo behaved low echo and on homogenous mostly ,one or more small anechoic lacunae could be seen on sonography .CDFI according to Adler semi -quantitative method classification ,25 lesions was 0 level (no blood flow signal),7 lesions for I level (edge had a small amount of pitting,short rod blood flow signal),Pulse doppler performance for low -speed and high-resistance characteristics.Conclusion Combining with the history and clinical performance , high frequency ultrasonography in abdominal wall endometriosis can be made relatively clear diagnosis,surgery of positioning has a guiding significance ,which can be used as the disease choice examination method.
3.Carbamylation-induced inactivation of glyceraldehydes 3-phosphate dehydrogenase and thioltransferase in bovine lens

AIM: To investigate whether potassium cyanate can inactivate glyceraldehydes 3-phosphate dehydrogenase (GAPDH) and thioltransferase (TTase) in bovine lens.METHODS: Fresh intact bovine lenses were incubated with 100mmol/L potassium cyanate (KCNO) for 7 and 12 days respectively. Then all lens were incubated in 50mmol/L DMEM solution. The proteins in the watersoluble fractions from the normal control and the cyanate-modified lens were extracted. The activity of GAPDH and TTase in the water-soluble fraction after incubation at 37℃ was measured by spectrophotometer.RESULTS: GAPDH activity was significantly lower in the cyanate-modified lens proteins than that of the normal control (P<0.01), and considerably diminished in protein incubated with 100mmol/L potassium cyanate for 12 days. There were statistically significant differences in the activity of TTase between the normal control lenses and the carbamylated lenses incubated for 7 days (P<0.05) and 12 days (P<0.01). However. there was no statistical difference between the samples incubated with 100mmol/L KCNO for 7 and 12 days (P=0.19296).CONCLUSION: This study provides evidence to show carbamylation is able to inactivate GAPDH and TTase in bovine lenses. This may have implications for the susceptibility of lenticular GAPDH and TTase to carbamylation, and also for the research on pathogenesis of cataract.

MicroRNA,which is found in recent years,is a class of small noncoding RNAs of about 18-24 nucleotides in length,that can regulate gene expression.It induces mRNA degradation or inhibit translation of tagert genes mainly by complete or incomplete base pairing to the 3' untranslated region of tagert messenger RNAs.The application of microRNA biology technology provides new methods to the research of multiple diseases,and can further elucidate its occurrence,development mechanism.In this paper,microRNA biology-related researches are introduced and miR-125 in a variety of diseases is reviewed.
5.Lysyal oxidase propeptide and tumor

Lysyal oxidase (LOX),a cooper dependent monoamine oxidase,can stabilize the extracellular matrix and is closely related to the tumor cell differentiation,vicious transformation and invasiveness.LOX is secreted into extracellular,and it is resolved into a small fragment by sol protease,which is lysyal oxidase propeptide (LOX-PP).According to research,LOX-PP has its own structure and physiological functions,which can inhibit the growth of tumor cells.LOX-PP also plays a significant role in the generation and progression of tumor,which is likely to be a new therapeutic target.

8.Early cornea healing response of PRK and free-flap Epi-LASIK under different cutting depth in rabbit

Background Free-flap Epi-LASIK surgery show a fast epithelial healing and mild trauma responses.Its mechanism is notable. Objective The aim of this study was to compare the corneal epithelial healing response following photorefractive keratectomy(PRK)and flap-free Epi-LASIK surgery. Methods Twelve experimental rabbits were randomly assigned to high cutting group(-10.00 D)and low cutting group(-3.00 D),and 2 normal matched rabbits were used as controls.PRK was performed in lateral eyes and flap-free epipolis laser in situkeratomileusis(Epi-LASIK)surgery was curried out in the fellow eyes.The corneal wound healing response in early stage of post-operation was examined under the slim lamp,and the inflammatory cells infiltration in cornea,basic fibroblast growth factor(bFGF)and nuclear transcription factor(NF-KB)expressions in corneal stroma were detected by hematine-eosin staining and immunohistochemstry in the seventh day after operation. Results The inflammatory reaction was milder in the eyes with Epi-LASIK than ones of PRK under the slim lamp.No significant difierences were found in numbers of inflammatory cells in cornea.expressions of bFGF and NF-κB in corneal stroma between two types of surgeries in low cutting group(P>0.05).In high cutting group,the numbers of inflammatory cells were(12.25±1.22)/400 field in PRK eyes,and those of free-flap Epi-LASIK eyes were(6.67±0.31)/400 field,showing obvious difference between them(t=-8.87,P<0.01).Expressions of NF-κB(A value)in cornea were 6.11±1.36 and 41.82±8.7 1 in PRK eyes of low cutting group and high cutting group.and those in free-flap Epi-LASIK were 3.01±0.81 and 11.59±4.55.indicating a drastically increase in PRK eyes(P<0.05).The expressions of bFGF(A value)in corneal stroma were 33.59±6.98 and 123.68±18.81 in PRK eyes and those in free-flap Epi-LASIK eyes were 69.20±8.85 and 123.68±1 8.81 in low cutting group and high cutting group,presenting a considerably decrease in PRK eyes (P<0.05).The corneal epithelial healing was faster in free-flap Epi-LASIK eyes than that in PRK eyes.and deeper cutting slowed the healing procedure. Conclusion In low cutting group,healing time of corneal epithelium is close between PRK and flap-free Epi-LASIK.But in high cutting group,epithelial healing is faster and inflammatory reaction is lighter in flap-free Epi-LASIK compared with PRK.
9.Association of high-sensitivity C-reactive protein with risk factors of cerebral infarction in patients with type 2 diabetic mellitus

10. Inhibitory effect of recombinant adenovirus expressing TEM8 gene against angiogenesis in mouse model of glioma

Objective: To evaluate the inhibitory effects of recombinant adenovirus carrying TEM8 (Ad-TEM8) against angiogenesis and growth of mouse glioma G422 cells in vitro and in vivo. Methods: The Ad-TEM8 was constructed and subcutaneously injected to immunize Kunming mice, and then the splenic lymphocytes were collected for co-culture with G422 cells in vitro. The growth inhibition of G422 cells was observed and IFN-γ secretion by the activated lymphocytes were examined by ELISOT method. In addition, the anti-tumor and anti-angiogenesis effects ofAd-TEM8 were observed in Kunming mice carrying G422 xeno grafts. Results: Ad-TEM8 effectively stimulated the generation of cytotoxic Tlymphocytes (CTLs)in Kunming mice and the secretion of IFN-γ. Subcutaneous injection of Ad-TEM8-induced cytotoxicity effectively inhibited in vivo tumor growth and angiogenesis. Conclusion: Ad-TEM8 can induce CTLs in mice, inhibit tumor growth in vitro and in vivo, and disrupt tumor vasculature. The anti-angiogenesis effect might be a mechanism for its in vivo anti-tumor effect; and TEM8 may serve as a novel target for anti-angiogenesis therapy of glioma.
