Sepsis may result in a high mortality.Fish oil,which riches in ω-3 polyunsaturated fatty acids.has anti-inflammatory and immune regulatory functions.ω-3 fatty acids,which are special immune nutrient substrates,participate in the energy provision as well as improve organ function and regulate proinnammatory/anti-inflammatory cytokines.The ideal function of fish oil is associated with dosage,time of initiation and duration of application.
2.The study on the relationship of the abnormal apoptosis of lupus nephritis pathology and disease ac-tivity

Objective To explore the abnormal apoptosis and lupus nephritis pathology and disease activity relationship.Methods Forty patients with LN renal biopsy tissue, cell apoptosis with TUNEL method and counting cell apoptosis index ( AI) , application of SLE activity score ( SLEDAI) 2000 judged the disease activity, and correlation analysis.Results All patients of renal tissue apoptosis detection were positive, apoptotic cells in the glomerulus and renal tubules were scattered distribution, SLEDAI was posi-tively correlated with the AI ( r =0.38, P =0.029).Conclusions Cell apoptosis and remove obstacles exist in Patients with LN, the more the number of apoptotic cells, the more active SLE disease.
4.Effect of curcumin on the 11β-hydroxysteroid dehyarogenase 1 expression in spinal cords neuropathic pain rat model

Objective To observe the effect of stress caused by neuropathic pain on serum cortisol concentration and expres-sion of 11β-hydroxysteroid dehyarogenase 1(11βHSD1)and investigate the action mechanism of curcumin for alleviating neuropathic pain. Methods The 72 male SD rats were randomly divided into 4 groups with 18 rats in each group:sham operation group(Sham), chronic constrictive injury group(CCI),solvent contrast group(SC)and curcumin treated group(Cur100). Curcumin(100 mg/kg/d) was given after the operation in Cur100 group. Paw thermal withdrawal(PTWL)and paw mechanical withdrawal threshold(PMWT)of rats were measured on 2 pre-operative and 1,3,5,7,10,and 14 post-operative days. The animals were deeply anesthetized,the blood was taken from the heart,and the L4-L5 dorsal root ganglion(DRG)of the operated side was removed. The change of cortisol was measured by ELISA and the change of 11βHSD1 expression in spinal cord and DRG was determined by immunochemistry and Western blot. Results Compared with sham group,PTWL and PMWT decreased significantly after operation in CCI group(P<0.01),serum cortisol concentration was significantly increased(P<0.01)and the expression of 11βHSD1 was significantly increased in CCI group(P<0.01). Compared with CCI group,the PMWT and PTWL was increased in CCI group(P<0.05),the serum cortisol concentration was decreased(P<0.05)and the expression of 11βHSD1 was inhibited in Cur100 group(P<0.05). Conclusion Stress caused by neuropathic pain triggered the release of cortisol to the blood and increased the expression of 11βHSD1 at the same time. Cur-cumin could alleviate thermal hyperalgesia and mechanical hyperalgesia induced by CCI and could also inhibit the serum cortisol con-centration and the expression of 11βHSD1 in spinal cord and DRG,which was likely to be related to pathogenesy and maintenance of neuropathic pain.
5.The application of laparoscopic ultrasonography in laparoscopic cholecystectomy in 410 cases

Objective To evaluate the effect of laparoscopic ultrasonography in laparoscopic cholecystectomy. Methods Laparoscopic ultrasonography system of Sharplan Company was used for the examination of the liver, gallbladder, bile duct and pancreas in 410 patients performed by laparoscopic cholecystectomy. Results The rates of biliary duct abnormality, preoperative diagnosis change, preoperative inexpectant disease finding, bile injury and residual stone were 3.5%, 4.4%, 2.4% and 0%, respectively. Conclusion During laparoscopic cholecystectomy, laparoscopic ultrasonography is helpful for the improvement of correct diagnosis and the decrease of complications.
6.Phspho-ERK1/2 involved in propofol preconditioning on ischemia/ reperfusion injury in isolated renal hypertension rat hearts

Aim To explore the effect of propofol preconditioning on myocardial ischemia/reperfusion injury and the role of phspho-ERK1/2 during the propofol preconditioning in renal hypertension rat(RHR).Methods The isolated RHR hearts perfused on langendorff apparatus were randomly divided into 8 groups(n=8 each).After 20 min of perfusion for equilibration,the CTRL group was subsequently perfused by 148 min.The ISCH group was submitted to 35 min of ischemia and 60 min of reperfusion(I/R) to induce ischemia/reperfusion injury.The DMSO group was given by once 18 min and twice 10 min K-H solution containing 20 ?mol?L~(-1) DMSO and 5 min K-H solution reperfusion prior to I/R procedure.The three propofol preconditioning groups were preconditioned by giving 2 cycles of 10 min K-H solution containing 30 ?mol?L~(-1),100 ?mol?L~(-1) or 300 ?mol?L~(-1) propofol and 5 min K-H solution reperfusion prior to the I/R procedure.The PD group and the PD+P100 group were given K-H solution containing 20 ?mol?L~(-1) PD98059,an ERK1/2 kinase specific inhibitor,for 18 min and 5 min K-H solution washout before I/R and 100 ?mol?L~(-1) PPC respectively.Cardiac functional indices were recorded.Activity of SOD and content of MDA were measured.The level of phspho-ERK1/2 protein expression was measured using Western Blotting.Results Compared with those in the CTRL group,the recovery of cardiac functional indices in ISCH group became worse(P
7.Vascular endothelial growth factor and its effect on bone formation

OBJECTIVE: To review the outcomes of experiments about vascular endothelial growth factor(VEGF) and the effect of it on bone formation, and to make sure whether VEGF can promote bone formation or not.DATA SOURCES: Electronic data searches were performed to obtain data from the databases of http://www.ncbi. nlm. nih. gov/pubmed and http://www. zglckf.com. DATA SELECZION: Nearly 30 articles about VEGF and the role of VEGF affecting bone formation were selected, regardless of the randomized design,blind method involved in or not.DATA EXTRACTION: These researches proved the structures, biologic properties and expression of VEGF and vascular endothelial growth factor receptor(VEGFR), and showed that VEGF plays an important role in endochondral ossification by promoting angiogenesis, bone turnover and preventing apoptosis of chondrocyte. In intramembranous ossification, absence of cartilage, osteoblasts are likely producing, and responding to, VEGF. VEGF also acts to recruit and activate osteoclasts as well as stimulate osteoblast chemotaxis, differentiation and matrix mineralization. It is debatable whether local application of exogenous VEGF can promote bone repair or not. However, in certain situation such as ischemia, the increase of VEGF locally may promote bone repair.DATA SYNTHESIS: Compared with the control group, VEGF can promote osteogenesis, bone formation and remodeling through the effect of VEGF on endochondral ossification and intramembranous ossification.CONCLUSION: One possible advantage of local VEGF therapy may be its ability to couple angiogenesis with bone formation and remodeling. If we can ultimately apply VEGF to treat osteonecrosis, bone defect and nonunion, we will find a new therapy for these diseases.
8.Distribution of the Genetic Polymorphism of CYP2C9 and VKORC1 in Uighurian Patients in Xinjiang and Comparison among Different Ethnic Populations

OBJECTIVE:To provide basis for the establishment of warfarin individualized administration model that is suitable for Uighurian patients in Xinjiang. METHODS:The genotypes of CYP2C9 and VKORC1 in 200 Uighurian patients in xinjiang were detected and compared with the genotypes and allele frequency of different ethnic populations in world. RESULTS:The frequency of CYP2C9*1/*1 homozygous wild type in Xinjiang Uighurian patients was lower than South Korea,Japan,the United States and Han nationality in China,higher than Turkey;the frequency of *1/*2 heterozygous mutant was lower than Turkey,Sweden and the United Kingdom,higher than South Korea,Japan and Han nationality in China;the frequency of *1/*3 heterozygous mutant was higher than Japan,South Korea,Han nationality in China,the United Kingdom and the United States;the frequency of *3/*3 was higher than the populations above(P<0.05). *1 allele in Xinjiang Uighurian patients was lower than Han nationality in China,Ja-pan,South Korea and the United States,higher than Turkey(P<0.05). *2 allele was higher than Han nationality in China,Japan and South Korea,lower than Turkey,Sweden and the United Kingdom(P<0.05). *3 allele was higher than Han nationality in Chi-na,Japan,South Korea,Sweden,the United Kingdom and the United States(P<0.05). The frequency of VKORC1-1639AA in Xinjiang Uighurian patients was higher than the United States Sweden,the United Kingdom,lower than Singapore,Japan,Taiwan China and Han nationality in China. The frequency of type AG was higher than Singapore,Japan,Taiwan China,Han nationality in China. The frequency of type GG was higher than Singapore,Japan,Taiwan China,Han nationality in China,lower than the United States,Sweden and the United Kingdom(P<0.05). Type A allele in Xinjiang Uighurian patients was lower than Han nation-ality in China,Taiwan China,Japan and Singapore,higher than the United States,Sweden and the United Kingdom;type G was higher than Han nationality in China,Taiwan China,Japan and Singapore,lower than the United States,Sweden and the United Kingdom(P<0.05). CONCLUSIONS:The distribution of CYP2C9 and VKORC1 gene polymorphism in Uighurian patients in Xin-jiang are obviously different from Asian,European and American populations.

10. Inhibitory effects of attenuated Salmonella typhimurium-mediated TRAIL and VP3 gene against gastric cancer cells

Objective: To investigate the inhibitory effect of eukaryotic expression vector (attenuated salmonella typhimurium) carrying tumor necrosis factor-related apoptosis inducing ligand (TRAIL) and Chicken anemia virus VP3 gene on gastric cancer cells in vitro and in vivo. Methods: The cloning vectors pBud-TRAIL, pBud-VP3, and pBud-TRAIL-VP3 were transformed into attenuated Salmonella typhimurium by electric transformation technique. The S. typhimurium-based carriers were then transfected into gastric cancer cells, line SGC-7901 after stability assay. The expression of fusion green fluorescent protein was examined using fluorescent microscopy after 24 h. MTT assay was used to examine the inhibition of cell growth. Flow cytometry was used to detect cycle distribution and apoptosis rates of cells. The expression of caspase-3 and caspase-9 was assayed by immunohistochemistry method. Salmonella typhimurium carrying recombinant plasmid was administrated orally in sarcoma-bearing mice; 8 weeks later RT-PCR was used to detect the expression of cloning vectors in tumor tissue. Meanwhile, the sizes of tumors were also determined. Results: The recombinant plasmids were stably transformed into attenuated Salmonella typhimurium, and the plasmids was satisfactorily expressed in gastric cancer cells via attenuated Salmonella typhimurium. TRAIL and VP3 inhibited the proliferation of gastric cancer cells after 48 h. Flow cytometry analysis showed that the pBud-TRAIL-VP3 obviously enhanced apoptosis rates of gastric cancer cells. TRAIL and VP3 jointly increased the expression of caspase-3 and caspase-9. In vivo study showed that TRAIL and VP3 genes were expressed in tumor tissue and could inhibit the tumor growth(P<0.05). Conclusion: Attenuated Salmonella typhimurium-mediated TRAIL and VP3 transfection of gastric cancer cells can inhibit cell growth in vitro and in vivo. The joint effect of TRAIL and VP3 is correlated with the increase of caspase-3 and caspase-9 expression.
