1.Expression, purification and characterization of PCP-2EC/Fc fusion protein in mammal cells

Objective: To construct the extracellular region of PCP-2(PCP-2EC) and the immunoglobin IgG Fc fusi on protein expression vector,and then express and purify the soluble PCP-2EC/Fc fusion protein for the study of its function in neuronal adhesion. Methods: PCP-2 extracellular region was amplified and cloned into an expression vector pIGplus containing human IgG Fc; PCP-2EC/Fc fusion protein was expressed by COS-7 and 293 cells transfected by the constructed plasmid and purified by protein A. The purified fusion protein was used as substrate to study its function in neuronal adhesion. Results: PCP2 extracellular region was cloned into IgG Fc expression vector successfully; PCP 2EC/Fc fusion protein was expressed and purified in mammal cells; and the purified fusion protein promoted neuronal adhesion. Conclusion:PCP 2EC/Fc fusion protein expression system is successfully constructed and the purified fusion protein can promote neuronal adhesion. These results lay a foundation for the research on the PCP-2 function in neuronal adhesion and the further functional study in the nervous system and other fields.

Indole alkaloid family has been the biggest so far among the various alkaloids,which contains pharmaceutical and effective constituents of various plants featuring diverse biological activities.Thanks to the development of metabonomics,to reveal the biosynthetic pathway of active components for the molecular mechanism of indole alkaloids and the regulation research of plant metabolism present a growing importance and significantly direct the researches of improving biological production.This paper reviewed the biosynthetic pathways of some indole alkaloids in accordance with the structure classification of indole alkaloids to lay a foundation for the further studies on the biosynthetic pathways of indole alkaloids and provide a reference for the biosynthetic pathways of other indole alkaloids.
4.Application in medicinal plants research by high-throughput metabolomics method

High-throughput metabolomics have developed very rapidly in recent years and been widely used in medicinal plants research .At present ,high-throughput metabolomics mainly applied in the following areas ,quality control of medicinal plants by fingerprints ,metabolites difference comparison before and after genetic engineering ,monitoring metabolites change in different environment and gene function study .High-throughput metabolomics have a great future ,but still have some challen-ges ,such as the requirements for more sophisticated equipment and complexity of data integration .With the advancement of science and technology ,high-throughput metabolomics will be an irreplaceable tool for the research of medicinal plants .
5.Risk factors for recurrence of lower-extremity arteriosclerosis obliterans in patients after interventional surgery

Objective:To investigate the risk factors for the recurrence of lower-extremity arteriosclerosis obliterans in patients after interventional surgery.Methods:The clinical data of 98 patients with lower-extremity arteriosclerosis obliterans who received treatment in Lishui Municipal Central Hospital from January 2021 to January 2022 were retrospectively analyzed. They were randomly divided into a recurrent group ( n = 36) and a non-recurrent group ( n = 62). The immediate efficacy of interventional therapy and the follow-up were determined in each group. Clinical data, triglyceride, high-density lipoprotein cholesterol and low-density lipoprotein cholesterol levels, neutrophil/lymphocyte ratio, and fasting blood glucose level were compared between the two groups. The risk factors for the recurrence of lower-extremity arteriosclerosis obliterans after interventional surgery were further analyzed using logistic regression analysis. Results:The success rate of interventional surgery for aortoiliac artery, superficial femoral artery, and inferior genicular artery were 95.35% (41/43), 97.73% (43/44), and 95.83% (46/48), respectively. Twelve-month follow-up results showed that the patency rate of lower-extremity vessels decreased with time ( P < 0.05). There was a significant difference in Trans-Atlantic Inter Society Consensus (TASC) classification between the two groups (11/26/18/7 vs. 4/5/10/17, χ2 = 18.33, P < 0.05). Ankle-brachial index (ABI) in the recurrence group was significantly lower than that in the non-recurrence group [(0.77 ± 0.12) vs. (0.40 ± 0.02), t = 18.31, P < 0.05]. There were no significant differences in triglyceride, high-density lipoprotein cholesterol, low-density lipoprotein cholesterol, and fasting blood glucose levels between the two groups (all P < 0.05). The neutrophil/lymphocyte ratio (NLR) in the non-recurrent group was significantly lower than that in the recurrent group [(1.98 ± 0.28) vs. (13.18 ± 0.50), t = 15.25, P < 0.05]. Logistic regression analysis results showed that ABI ( OR = 1.459, 95% CI: 1.163-1.832), TASC classification ( OR = 1.344, 95% CI: 1.090-1.658), surgical method ( OR = 1.392, 95% CI: 1.140-1.700) and NLR ( OR = 1.474, 95% CI: 1.184-1.836) were the influential factors of recurrence of lower-extremity arteriosclerosis obliterans (all P < 0.05). Conclusion:ABI, TASC classification, surgical methods, and NLR are the influential factors of lower-extremity arteriosclerosis obliterans after interventional surgery. The above indexes should be closely monitored in clinical practice to maintain the effects of interventional surgery.
6.A study of applied microanatomy by endoscope-assisted via retrolabyrinthine approach.

OBJECTIVE:
To study the microanatomy by endoscope-assisted via retrolabyrinthine approach.
METHOD:
Nineteen cadaveric heads fixed with formalin were dissected in our study. The data that endoscope could be extent and the distance between the important point were measured. By simulated the retrolabyrinthine approach, endoscope was placed to observe the nerves and vessels, the distance between nerves and the central point of the anterior edge of sigmoid sinus were measured.
RESULT:
The distance from the inferior margin of petrous ridge to the upper bound of endolymphatic sac was (9.93+/-1.52)mm; and from superior margin of petrous ridge to the inferior margin common bony crus was (4.64+/-0.91)mm;and from the intersection of posterior semicircular canal to the anterior wall of sigmoid was (7.85+/-1.47)mm on the left, and (5.69+/-1.68)mm on the right. The distance from inferior margin common bony crus to the anterior wall of sigmoid was (13.9+/-1.71)mm on the left, and (11.31+/-2.03)mm on the right. The trochlear nerve and abducent nerve could be observed under endoscope. The relationship between the trigeminal nerve, acoustic nerve and the vessels could be identified clearly. The distance from the central point of the anterior edge of sigmoid to the roots of the trigeminal nerve, facial nerve, vestibulocochlear nerve and glossopharyngeal nerve were (29.88+/-2.77) mm, (32.04+/-2.04) mm, (29.17+/-1.65) mm, (35.49+/-1.53) mm respectively.
CONCLUSION
The visual field of the cerebellopontine angle appear wider by the endoscope assisted retrolabyrinthine approach. Nerves, vessel, internal acoustic pore, jugular foramen region can be clearly seen. Some minimal invasive surgery can be done by this approach.

The resources and quality of Chinese herbal medicine is the foundation and guarantee for the sustainable development of the whole industry chain of traditional Chinese medicine.The effective component,having clinical curative effect,is the material base of Chinese herbal medicine,and is the core for the quality of medicinal material factors (quality).The accumulation of efficacy material is closely related to the genetic background of the original plant.By clarifying active ingredients biosynthesis,revealing medicinal material quality formation mechanism,regulating the content of effective components,the quality of medicine could be guarantee and ascend.The quality control technology based on active ingredients of Traditional Chinese medicine quality includes integrated analysis of high-throughput sequencing and systems biology data,revealing synthetic genetic mechanisms of active ingredients,excavating molecular markers of the quality,creating excellent germplasm,forming cultivation techniques,establishing classification standard of medicinal materials,and finally realizes the full industrial chain control of Chinese herbal medicine production.Taken all of these together could ensure the quality of medicinal materials.

Baphicacanthus cusia ,widely distributed in the Southwest ,Southern and Eastern regions of China ,is an im-portant medicinal plant of Acanthaceae family .Indigo made from stem and leaf of Baphicacanthus cusia in Fujian has the best quality in China ,and is known as “Jian Indigo naturalis” ,which is the genuine medicinal of Fujian Province .The rhizoma of Baphicacanthus cusia could be used as medicine ,called“Nanbanlangen” ,which together with indigo were included in the“Chi-nese Pharmacopoeia” .Indigo and its original plant both contain indirubin ,which has anti-cancer activity .Indirubin is an active ingredient of Huangdai Pian and Danggui Longhui Wan ,two kinds of traditional Chinese medicine ,which have been successful-ly used in the treatment of malignancies such as chronic myelogenous leukemia .The international advances in the biological characteristics ,genetic diversity ,cultivation technology and molecular biology of Baphicacanthus cusia germplasm resources were summarized .The main problems in Baphicacanthus cusia germplasm resources research are indicated ,which could pro-vide references for the further study and application of Baphicacanthus cusia germplasm resources .
9.Influence of body shape on the short-term therapeutic effects of laparoscopic distal gastrectomy: a multicentre retrospective study (A report of 506 cases)

Objective To investigate the risk factors of perioperative complications of laparoscopic radical distal gastrectomy and influence of body shape on the short-term therapeutic effects.Methods The retrospective case-control study was conducted.The clinicopathological data of 506 patients (328 males and 178 females,average age 60 years with the range of 24-85 years) who underwent laparoscopic radical distal gastrectomy+D2 lymph nodes dissection in the 8 clinical centers between March 2016 and November 2018 were collected,including 143 in the First Affiliated Hospital of Xiamen University,66 in the Affiliated Hospital of Qinghai University,66 in the Second Affiliated Hospital of Wenzhou Medical University,64 in the Zhongshan Hospital of Xiamen University,54 in the Affiliated Hangzhou First people's Hospital of Zhejiang University School of Medicine,48 in the Zhangzhou Affiliated Hospital of Fujian Medical University,35 in the Affiliated Quanzhou First Hospital of Fujian Medical University,30 in the Second Affiliated Hospital of Xiamen Medical College.The maximum thickness of subcutaneous fat at the level of umbilicus (USCF),the maximum vertical distance between the anterior abdominal skin and the back skin at the level of the umbilicus (UAPD),the maximum horizontal distance between the anterior abdominal skin and the back skin at the level of the umbilicus (UTD),the maximum verticaldistance between the anterior abdominal skin and the back skin at the level of the xiphoid bone (XAPD),the maximum horizontal distance between the.anterior abdominal skin and the back skin at the level of the xiphoid bone (XTD),the distance between the anterior abdominal skin and the root of celiac artery (CAD) and the maximum horizontal distance at a right angle to CAD (CATD) were measured using preoperative imaging examinations.Observation indicators:(1) intraoperative and postoperative situations;(2) follow-up situations;(3) risk factors analysis of perioperative complications;(4) influence of body shape related indexes on intraoperative situations and postoperative recovery:① Pearson univariate correlation analysis,② liner regression model analysis.Followup using outpatient examination and telephone interview was performed to detect the postoperative survival and tumor recurrence or metastasis up to December 2018.Measurement data with normal distribution were represented as Mean±SD.Measurement data with skewed distribution were described as M (range).Comparisons of count data were analyzed using the chi-square test.Comparisons of ordinal data were analyzed by Mann-Whitney U nonparametric test.Risk factors of perioperative complications of laparoscopic distal gastrectomy were analyzed by Logistic regression model.Influence of body shape related indexes on intraoperative situations and postoperative recovery was analyzed by Pearson univariate correlation analysis and liner regression model.Results (1) Intraoperative and postoperative situations:all the 506 patients underwent successful laparoscopic distal gastrectomy,including 103 with Billroth Ⅰ anastomosis,140 with Billroth Ⅱ anastomosis,201 with Billroth Ⅱ + Braun anastomosis,62 with Roux-en-Y anastomosis.The operation time,volume of intraoperative blood loss,number of lymph nodes dissected,time to postoperative anal exsufflation,time for initial fluid diet intake,time for initial semi-fluid diet intake and duration of postoperative hospital stay were (233±44)minutes,(102±86)mL,34±13,(3.6±1.5)days,(5.8±3.3)days,(8.3±3.8)days,(12.2±5.7)days respectively in the 506 patients.Of 506 patients,196 were defined as pathological stage Ⅰ,122 were defined as pathological stage Ⅱ and 188 were defined as pathological stage Ⅲ postoperatively.Of 506 patients,93 had 106 times of perioperative complications,including 33 times of pulmonary and upper respiratory infection,12 times of incisional infection,11 times of anastomotic leakage,11 times of abdominal infection,8 times of intestinal obstruction,8 times of gastroplegia,6 times of abdominal hemorrhage,5 times of bacteremia,3 times of anastomotic hemorrhage,3 times of lymph fluid leakage,2 times of pancreatic leakage,1 time of urinary infection,1 time of anatomotic stenosis,1 time of deep venous thrombosis and 1 time of pulmonary embolism;the same patient can merge multiple complications.Eleven patients were in the Clavien-Dindo classification ≥ Ⅲ.(2) Follow-up situations:465 of 506 patients were followed up for 1-32 months with a median time of 12 months.During the follow-up,451 had postoperative survival and 38 had tumor recurrence or metastasis.(3) Risk factors analysis of perioperative complications.① Results of univariate analysis showed that age,body mass index (BMI),preoperative hemoglobin,preoperative serum albumin and XAPD were related factors affecting perioperative complications of laparoscopic distal gastrectomy (x2 =10.289,7.427,5.269,5.481,4.285,P< 0.05).② Results of multivariate analysis showed that age,BMI,preoperative serum albumin were independent related factors affecting perioperative complications of laparoscopic distal gastrectomy (odds ratio =1.033,1.118,0.937,95% interval confidence:1.011-1.057,1.025-1.219,0.887-0.990,P<0.05).(4) Influence of body shape related indexes on intraoperative situations and postoperative recovery.① Results of Pearson univariate correlation analysis showed correlations between UAPD,XAPD,CAD,CATD and volume of intraoperative blood loss (r=0.107,0.169,0.179,0.106,P<0.05),between UAPD,XAPD,CAD and the number of lymph nodes dissected (r=-0.137,-0.143,-0.173,P<0.05),between USCF,XAPD and time to postoperative anal exsufflation (r =0.122,0.109,P<0.05),between USCF,XAPD,CAD and time for initial fluid diet intake (r=0.132,0.108,0.132,P<0.05),between USCF,XAPD and duration of postoperative hospital stay (r=0.116,0.100,P<0.05).② Results of liner regression model analysis showed a positive correlation between CAD and volume of intraoperative blood loss (r =6.776),a negative correlation between CAD and the number of lymph nodes dissected (r =-0.841),with statistically significant differences (t =2.410,-1.992,P< 0.05);a positive correlation between USCF and time to postoperative anal exsufflation (r=0.170),between USCF and time for initial fluid diet intake (r=0.365),between USCF and duration of postoperative hospital stay (r=0.636) respectively,with statisticallysignificant differences (t =2.188,1.981,2.107,P< 0.05).Conclusions Abdominal shape can influence intraoperative situations and postoperative recovery of laparoscopic distal gastrectomy,but cannot increase risks ofperioperative complications.Age,BMI and preoperative serum albumin are independent related factors affecting perioperative complications of laparoscopic distal gastrectomy.

Data standard plays an important role in the process of data collection, Integration and sharing in clinical cohort studies, and more attention have been paid to it. This paper summarizes the 5 international proven data standard model, analyze their characteristics and development status, and match their data modules with the general data set of the clinical cohorts to evaluate the international data standard models' applicability and provide reference for the development and improvement of the data standard model for clinical cohort studies in China.
