1.Effect of SIRT1 on the proliferation of ovarian cancer CAOV3 cells and its possible mechanism

Objective To observe the effect of SIRT1 on the proliferation of ovarian cancer CAOV3 cells and its possible mechanism. Methods Ovarian cancer CAOV3 cells were transfected the Lipofectamine 2000 and were divided into normal control group,negative control group and SIRT1-siRNA group.The mRNA and protein expression levels of SIRT1 were detected by RT-PCR and Western blot.MTT method was used to detect the proliferation of RNA interference at 24h,48h and 72h,respectively.The distribution of CAOV3 cell cycle phase after RNA interference of SIRT1 for 72 hours was detected by flow cytometry.SIRT1,p53,p21 protein expression and p53 acetylation status were detected by Western blot with RNA interference of SIRT1 after 72 hours.The interaction between SIRT1 and p53 protein was verified by immunoprecipitation in CAOV3 cells.Results Compared with negative control group,the expression of mRNA and protein of SIRT1 in SIRT1-siRNA group decreased after transfection of SIRT1 for 72 hours.The survival rate of CAOV3 cells in SIRT1-siRNA group decreased significantly at 48 hours and 72 hours (P<0.01).The proportion of Sphase cells in CAOV3 cells transfected with SIRT1-siRNA for 72h was significantly decreased (P<0.01),while the G1 phase cells were significantly increased (P<0.01),and the cells arrested in the G1 phase.The expression of p53 and p21 proteins were significantly increased and the p53 was in higher acetylation status after transfection with SIRT1-siRNA for 72 hours ( P<0.01 ) .SIRT1 was interacted with p53 protein in CAOV3 cells confirmed by co-immunoprecipitation. Conclusion Downregulation of SIRT1 expression can inhibit the proliferation of ovarian cancer CAOV3 cells, which maybe connected with p53 at high acetylation status,and enhancing p53-dependent p21 transcriptional activation,thus blocking the cell cycle and inhibiting the cell proliferation.
2.Application of fresh amniotic membrane transplantation combined therapeutic soft corneal contact lens in pterygium

To evaluate the clinical effect of therapeutic soft corneal contact lens in combination with fresh amniotic membrane transplantation in the treatment of pterygium and its clinical value.
●METHODS: This study was a retrospective analysis of 200 pterygium excision patients (200 cases, 220 eyes) in hospital from January 2010-June 2011. The patients were divided into two groups. Group 1 (100 cases, 110 eyes) were patients treated with fresh amniotic membrane transplantation and therapeutic soft corneal contact lens while group 2 ( control group, 100 cases, 110 eyes) did not use corneal contact lens. The clinical outcomes in these two groups were compared in a 6 - 18mo postoperative follow-up observation.
●RESULTS: ln control group using corneal contact lens the epithelium repair time ranged from 2 - 7d with an average time of 3. 8d; in group 1 the epithelial repair time ranged from 1 - 5d with an average time of 2. 5d. The patients were followed up for 6 - 18mo, 9 recurrent eyes were found in control group, the recurrence rate was 8. 2%; 5 recurrent eyes were found in experimental group the recurreat rate was 4. 5%. There were significant differences in the 2 groups comparing treatment outcomes (P< 0. 05). Early stimulative symptoms after surgery can be reduced by wearing contact lenses.
●CONCLUSlON:The therapeutic effect of fresh amniotic membrane transplantation after pterygium excision is enhanced by the therapeutic application of corneal contact lens. The epithelial repair time is shortened, the recurrence rate is reduced after pterygium excision and stimulative symptoms including photophobia, tearing, foreign body sensation and ophthalmodynia caused by corneal epithelial defect can be relieved.
3.Oncostatin M combined with dacarbazine inhibits proliferation of melanoma cell B16

Objective:To observe and identify the inhibitory effect of oncostatin M (OSM) combined with dacarbazine (DTIC) on mouse melanoma cells B16 in vitro and in vivo. Methods:The inhibitory effect of OSM combined with DTIC on the proliferation and apoptosis of B16 melanoma cell line B16 were determined through MTT assay and flow cytometry, respectively. The change in nu-cleus morphology of B16 cells was observed under a fluorescence microscope by Hoechst staining method. The effects of single agents OSM and DTIC, as well as OSM-DTIC joint treatments, on tumor in mice in vivo were observed by inoculating B16 cells into C57 BL of six mice. Results:The OSM, DTIC, and combined OSM-DTIC treatments inhibited the proliferation of B16 cells by (11.2±2.3)%, (25.3±4.6)%, and (32.5±3.8)%, respectively (P<0.05). Apoptosis of B16 occurred at (1.32±0.42)%, (10.64±2.13)%, and (15.86±2.76)%, respectively (P<0.05). Cell morphology showed a significant increase in nuclear fragmentation, as proven by OSM-DTIC combined treatment. In the in vivo experiment, DTIC caused an apparent inhibition on the growth of mouse melanoma compared with the control group, and the joint treatment showed that the addition of OSM enhanced the tumor suppression effect of DTIC. Conclusion: OSM combined with DTIC has a synergistic effect that inhibits proliferation and apoptosis of B16 in vitro. This approach suggests a new po-tential treatment for melanoma.

Objective To evaluate ischemia preconditioning (IP) and intermittent ischemia (II) in ameliorating the incidence of paraplegia. MethodCross clamping of the infrarenal aorta in New-Zealand rabbit was used to establish a spinal cord ischemia model. 25 rabbits were randomly divided into 3 groups. The continuous ischemia(CS) group (n=10) with the aorta clamped for 40 minutes. While in the IP group (n=6) the aorta was clamped for 3 sessions each lasting for 5 min, with 5 min interval between the sessions before 40 minutes clamping, in the Ⅱ group (n=9) the 40 min clamping was interposed by a 15 min interval. 6.Clinical and Autopsy Analysis of Systemic Fungal Diseases:A Report of 31 Cases

Objective To investigate the prevalence of systemic fungal infection in patients at auto-psy and its association with clinical data.Methods The materials from unselected adult patients were exa-mined at necropsy in our hospital from1971to2000,and the fungal species were identified by their mor-phologies in pathology.The clinical and pathological data were compared.Results The rate of fungal infec-tion was7.8%(31/396)among396cases.Seventeen(54.8%)of the31cases were infected with as-pergillus.The frequencies of organs involved with fungal infection were83.87%in lungs,48.39%in kid-neys,35.48%in brain and29.03%in heart,respectively.The fungal infection attributed directly to death in21cases.Conclusion Systemic fungal infection is one of the most common fatal causes among the critically ill patients,and aspergillosis becomes a major pathogenic fungus for patients with systemic fungal infection at autopsy.
7.Serum TK1 and its relation to the clinicopathological features and prognosis of patients with lung cancer

Objective:This study investigated the positive detection rate of cytoplasm thymidine kinase 1 (TK1) in lung cancer patients and the relationship of TK1 with clinicopathological features and prognosis. Methods:Sensitive chemiluminescence dot-blot assay was used to detect serum TK1 levels in 73 lung cancer patients and 56 normal control subjects. Results:The positive detection rate of TK1 was elevated in the lung cancer patients compared with the controls (P=0.006). The positive detection rate of TK1 was also correlated with distant metastases, but not with other factors, such as smoking, sex, lymph node metastasis, and pathology types. The 2 year survival of the patients with negative TK1 detection was significantly longer than that of the patients positively detected with the marker (P<0.001). Conclusion:Serum TK1, a new tumor marker, has potential applications in the diagnosis and prognosis of lung cancer.

10.Outcome of microsurgical treatment and its influencing factors in patients with aneurysmal subarachnoid hemorrhage

Objective To investigate the outcome of microsurgical treatment and its influencing factors in patients with aneurysmal subarachnoid hemorrhage.Methods The consecutive patients with aneurysmal subarachnoid hemorrhage treated with the early or ultra-early microsurgery were enrolled retrospectively.The Glasgow outcome scale (GOS) was used to assess the outcomes of patients at discharge.GOS 4-5 was defined as good outcome,and GOS 1-3 was defined as poor outcome.Results A total of 147 patients with aneurysmal subaraclnoid hemorrhage were enrolled.One hundred and twelve patients (76.2%) had good outcomes.There were significant differences in the proportions of preoperative Glasgow Coma Scale (GCS) scores (12.8 ± 2.8 vs.7.5 ± 3.8;t =7.525,P <0.001),low Hunt-Hess grade (83.0% vs.31.4%;x2 =34.318,P < 0.001),size of aneurysm (x2 =9.531,P =0.009),preoperative rebleeding (6.3% vs.25.7%;x2 =8.506,P =0.003),preoperative brain herniation (4.5% vs.40.0%;x2 =26.846,P < 0.001),initial CT scan showing intracerebral hemorrhage (19.6% vs.48.6%;x2 =11.449,P =0.002),and intraventricular hemorrhage (8.9% vs.40.0%;x2 =18.846,P <0.001) between the good outcome group and the poor outcome group.Multivariate logistic regression analysis showed that the larger aneurysm (odds ratio [OR] 3.194,95% confidence interval [CI] 1.458-6.999;P =0.004),older age (OR 1.054,95% CI 1.013-1.097;P=0.010),lower preoperative GCS score (OR 0.539,95% CI 0.410-0.724;P < 0.001),and preoperative brain herniation (OR 3.633,95% CI 1.039-12.700;P =0.043) were the independent risk factors for poor outcomes.Conclusions After active surgical treatment,most of the patients with aneurysmal subarachnoid hemorrhage have good outcomes,however,patients with older age,larger aneurysms,lower preoperative GCS scores,and preoperative brain herniation usually have poor outcomes.
