Objective To detect DNA lesions induced by complex pollutants existing in environment using the technique of randomized terminal linker,dependent PCR (RDPCR). Methods SD rats were treated with organic concentrate by injection i.p., DNA were extracted from rat's liver, kidney, lung and leucocytes, then RDPCR was used to detect DNA damage of exon 7 of p53 gene. Results Organic concentrate of drinking water could make rat's genomic DNA broken. Hybridization bands were found in liver and kidney tissues after RDPCR, none in blood and lung tissues. Conclusion Organic concentrate of drinking water can cause the DNA lesions of exon 7 of p53 gene and the major target organs are liver and kidney. The technique of RDPCR can be used to detect DNA damage induced by complex environmental pollutants.

OBJECTIVE:To analyze the general patterns and characteristics of drug-induced lupus (DIL) induced by inflix-imab,and to provide reference for the safety of drug use. METHODS:Usinginfiximab,lupusas searching words,inflix-imab-induced DIL literatures included in CJFD,VIP and PubMed from Jan. 2002 to Jun. 2016 were retrieved and analyzed. RE-SULTS:Totally 30 effective literatures were retrieved,involving 31 DIL patients,there were 8 male cases (25.81%) and 23 fe-male cases(74.19%);17 patients aged more than 50(54.84%). 20 cases occurred within 5-24 weeks(64.51%). 16 patients re-ceived infliximab alone (51.61%). Main clinical manifestations were skin rash or photosensitivie enteritig (15 cases),joint pain (16 cases,multiple arthritis and synovitis)and fever(12 cases). 5 patients suffered from 3 above symptoms simultaneously. Labo-ratory examination mainly manifested as antinuclear antibody,double stranded DNA antibody and anti-histone antibody positive. Thirty patients with DIL received drug withdrawal or steroid hormones treatment,among which the symptoms of 26 patients disap-peared after drug withdrawal or treatment within 6 months,and 1 patient deteriorated. CONCLUSIONS:DIL-induced by inflix-imab invovle patients'age and gender,disease types,family history and other factors. Clinicians should be aware of rules and char-acteristics of DIL induced by infliximab,and tighten drug use monitoring to reduce ADR.

Objective To study the relationship between hepatic resection,hepatic regeneration and hepatic function.Methods Totally 8 cases undergone partial hepatic resection were undergone CT scan pre-and pastoperation.The volumes of the whole liver and the lesions were measured on CT images,and the hepatic resectable rate was calculated before operation.The volume of liver was measured again after operation,and the hepatic regenerative rate at different time was also calculated after operation.The hepatic function was tested before and after operation.Results The negative relation was showed between hepatic resectable rate and hepatic regenerative rate.Conclusion There is postiive relation between hepatic resectable rate and the function of liver.

OBJECTIVE: To discuss the mechanism of dioscin against apoptosis of cardiomyocytes induced by hydrogen peroxide through Serum Pharmacologic Method. METHODS: The drug-containing serum [0.6 g (crude drug) · kg-1] was prepared by serum pharmacologic method. Cardiomyocytes from neonatal SD rats were cultured in Dulbecco Modified Eagle Medium (DMEM). The primary cultured cardiomyocytes were injured by hydrogen peroxide before giving the drug-containing serum with different concentrations. The cardiomyocyte viability was determined by MTT method. Morphological changes of cardiomyocytes were observed by fluorescence microscope after treating with the drug-containing serum at different concentrations. The anti-apoptotic effect of dioscin was indicated by detecting the activity of caspase-3. The protein expression of Bcl-2 and Bax were semi-quantified by Western-blot method after treating with the drug-containing serum. RESULTS: The cardiomyocyte viability was elevated (P < 0.01) after being treated with different concentrates of drug-containing serum (0.4, 0.8, 1.2 g (crude drug) · kg-1, prepared from the original drug-containing serum). Typical apoptotic features of the cardiomyocytes such as membrane blebbing, cell shrinkage and detachment, and nucleus condensation and fragmentation were dose-dependently improved after being treated with drug-containing serum. The activity of caspase-3 decreased with the increasing concentration of drug. Western blot approach showed that the Bcl-2 level increased meanwhile the Bax decreased. CONCLUSION: Dioscin could increase the viability of the primary cultured cardiomyocytes in hydrogen peroxide induced injury. It could also decrease the activity of caspase-3, improve nucleus condensation and fragmentation, increase Bcl-2 level and decrease the Bax level. BACKGROUND: Threatened abortion is a high risk pathological change during gestation. Fetus usually could be protected after the application of all kinds of medical interventions; however, whether the healthy development of the newborn could be assured simultaneously when a new life is protected?Especially the long-term impact on the normal development of the intelligence is an important issue deserving investigation.OBJECTIVE: To investigate the differences of intelligence level between protected fetus after threatened abortion and normal mature fetus, and the influencing factors.DESIGN: A case-controlled observational comparative study introducing children as the subjects.SETTINGS: Psychological department of a normal university; Department of gynecology and pediatrics of a district women and children' s health care hospital.PARTICIPANTS: Totally 170 children(mature and natural delivery after protection) of 5 to 7 years old, whose mothers experienced threatened abortions during gestation, and 182 children born in the same period(normal mature and natural delivery) of 5 to 7 years old, whose mothers did not have threatened abortion history were selected as the subjects. No other factors would affect the intellectual development in the subjects of both groups.INTERVENTIONS: The Chinese Version of Wechsler young children scale of intelligence(C-WYCSI) for children below 6 years old or Wechsler intelligence scale for children-Chinese revision( WISC-CR ) for children no younger than 6 years old were introduced in the individual test performed by trained psychological junior students for every testee.children in both groups [intelligence quotient(IQ) gained in the intelligence of mother on the intellectual development of the children in both groups.was found in the testees, the IQ of all tested children was within the normal dren was sinificantly lower in the threatened abortion group than in the normal were significant interactions between threatened abortion and gender, body mass at birth and the pregnant age of mother, etc.: if the mother experienced threatened abortion, the newborn would be even more easily to suffer from relative low body mass at birth(low body mass fetus accounted for 28% of threatened abortion group, which accounted for about 66% of the total low body mass at birth fetuses in our study); the male newborn would be even more easily to have relative low intellectual development(the difference of IQ between the male children of two groups was significant) (F = 2. 809, P < 0.05). Especially, the negative impact on children' s intellectual development would be even greater if the pregnant woman were older than 35 years old and had threatened abortion experience(the average IQ of 32 tested children was 86).CONCLUSION: Threatened abortion might be a potential innate factor that would affect the intellectual development of the children. For those high-risk individual cases with this kind of experience, early intervention should be prepared as early as possible to create a favorable substantial and culture environment for avoiding the actual occurrence of delayed intelligence.

Objective To study the effects of rehabilitation training on the regeneration of nerve cells in rats after intracerebral hemorrhage (ICH).Methods A total of 75 male SD rats were randomized into a training group,a control group and a sham operated group,25 rats/group.The ICH models were induced by stereotactical injection of collagenase type Ⅶ into the globus pallidus.The training group was trained with grasp,balancing and rotating exercise every day,the control group was restricted to their cages,and the sham operated group received normal saline injections.Each group was further subdivided into 1,4,7,14 and 28 day subgroups.Neurological function was measured in each group.Bromodeoxyuridine (BrdU) was used to label S phase cells,immunohistochemical single and double staining with antibodies against BrdU,microtubal-associated protein (MAP) and neuronal nuclei (NeuN) were used to determine neuronal proliferation,migration and differentiation in the subventricular zone ( SVZ ) and subgranular zone (SGZ) in the training and control groups.Results The motor function scores of the animals in the rehabilitation group were significantly lower than those of the control group.Proliferative BrdU + cells of the SVZ and SGZ in the control group rats were clearly less than those in the rehabilitation training rats at all time points.The results of the immunohistochemical double staining indicated that one week after ICH BrdU +/MAP + cells in the SVZ had increased significantly in the training group compared to the control group,and then decreased two weeks later.At the same time,BrdU +/MAP cells were found in the striatal boundary on the hemorrhage side,in numbers up to 8 times that in the control group.In the rehabilitation group striatal neuron differentiation on the hemorrhage side was 2 to 3 times that in the control group.Conclusion Rehabilitative training can enhance nerve cell proliferation,regeneration and neuron migration after ICH.

BACKGROUND:Thyroid cancer stem cells are one of the reasons for tumor resistance that promotes tumor development. The research of thyroid cancer stem cells has provides a new clinic means for the diagnosis and treatment of thyroid cancer. OBJECTIVE:To overview the discovery, identification, and correlation of thyroid cancer stem cells with thyroid cancer. METHODS:A computer-based online search of PubMed database and Wan Fang database between 1995-01/2014-01 was performed to search related articles with the key words of“thyroid cancer, cancer stem cell, stem cell, cancer suppressor gene”in English and Chinese, respectively. Literatures related to thyroid cancer stem cells were selected;in the same field, the articles published lately in authoritative journals were preferred. RESULTS AND CONCLUSION:A total of 561 literatures were primarily selected, and 57 documents were involved for result analysis according to inclusion criteria. Cancer stem cells have become a focus in the study of carcinogenesis. Stem cells also exist in the thyroid gland and its tumor. At present, there are several ways to isolate and identify thyroid cancer stem cells. The tumor thyroid stem cells are closely related to the occurrence, transfer and treatment of tumors. But whether we can cure thyroid cancer through restraining or eliminating thyroid cancer stem cells is stil unknown that needs further studies.
