1.Exppresion of miRNAs target regulating FASN in different invasion ability osteosarcoma cells

Objective To detect the exppresion level of miRNAs which may target fatty acid synthase(FASN) in osteosarcoma (OS) cells of diverse invasion ability ,and to select the miRNAs target regulating FASN ,acting as the basis to investigate the mech‐anism of OS metastasis .Methods The miRNA and microrna .org online software were adopted to forecast the miRNAs that might target regulate FASN(NM_004104);RT‐PCR was used to detect the expression level of the predicted miRNAs in HOS and U2‐OS cells ;the protein expression of FASN was detected by Western blot ;Transwell invasion assay was used to evaluated the invasive a‐bility of HOS and U2‐OS cells .Results Two prediction methods all showed that miR‐195/15a/15b/16/424/497 molecular might be target FASN gene;RT‐PCR result showed that the expression level of miR‐424 in HOS cells was significantly higher than that in U2‐OS cells ;the expression level of FASN was significantly higher in U2‐OS cells than that in HOS cells ;the invasive ability of U2‐OS cells is significantly higher than HOS cells .Conclusion The expression level of miR‐424 in OS cells may be negatively related to the invasive ability ,and miR‐424 likely may affect OS metastasis via targeting FASN .
2.Influencing factors for the prognosis of the patients undergoing reversion metal-on-metal hip arthroplasty for inflammatory pseudotumor

BACKGROUND: Reversion total hip arthroplasty for inflammatory pseudotumor is little reported.OBJECTIVE: To investigate the prognosis of the reversion total hip arthroplasty for inflammatory pseudotumor and the related factors.METHODS: Eighty-one patients failed metal-on-metal hip arthroplasty were recruited, the medical history, baseline data and information of primary arthroplasty were collected, and all preoperative examinations were completed prior to reversion. The postoperative joint function was evaluated by Hospital for Special Surgery score, and the morphology of prosthesis was observed by radiology. Additionally, the serum levels of cobalt and chromium ions were detected, as well as the incidence of complications and the related factors were analyzed.RESULTS AND CONCLUSION: (1) There was a significant increase in Hospital for Special Surgery hip score and significant decrease in serum levels of cobalt and chromium ions after reversion (P=0.000). (2) The postoperative incidence of complications and secondary reversion rate was 16% and 6%, respectively. (3) Preoperative prosthesis loosening (P=0.012), tissue cirrhosis revealed on MRI (P=0.000) and the injury degree of tissue (P=0.049) were found to be related to the incidence of complications. (4) Kaplan-Meier curve showed that the survival rate of prosthesis was 88% at 30 months postoperatively. (5) These results suggest that the prognosis of reversion total hip arthroplasty is poor,which is related to the preoperative aseptic loosening, tissue cirrhosis surrounding the joint and intraoperative tissue injury, thus providing guidance for surgeons to make an appropriate operative scheme.
3.Risk assessment for clinical external application of calomel.

Calomel is a common traditional Chinese medicine (TCM) containing mercury in clinical external application. Although the toxicity of calomel has attracted concern, there is no unified standard yet in clinical external application. Risk assessment is used for evaluating the potential health effects of hazardous substances. The purpose of this article was to evaluate the health risk of calomel in clinical external application on the basis of toxicity data, to ensure safe and rational application of TCM containing calomel. The toxicity data of transdermal administration of calomel or mercurous chloride were collected by searching the literature. The daily maximum exposure dosage of calomel in clinical external application was estimated by following the four procedures of risk assessment, and Margin of Safety (MOS) as an evaluation indicator was then calculated to evaluate the safety of calomel on clinical application. It has been reported that the adult in single transdermal administration of calomel at 1. 5 g was lethal. Based on the LOAEL of calomel for long-term transdermal exposure (1 month) in rats was 0.096 g · kg(-1) · d(-1), the NOAEL of calomel for patients (about 60 kg) by external application within 2 weeks was estimated to be 1.46 mg · kg(-1) · d(-1). When MOS value equals to 1, the daily maximum exposure of calomel in clinical external application within 2 weeks was calculated to be 1.1 g. The results suggest that daily single dose of calomel in clinical external application should be lower than 1.5 g for adults, and more attention should be paid to changes in hepatic and renal function of patients when repeated dose more than 1.1 g within 2 weeks. The approach of risk assessment could be helpful in rational application of TCM containing mercury.

4.Clinical analysis of 224 patients with diabetic ketoacidosis

Objective To summarize the clinical data of patients with diabetic ketoacidosis (DKA), and improve the ability of diagnosis and treatment for patients with DKA. Methods The clinical data of 224 patients with DKA were retrospectively analyzed. Results After the positive diagnosis and comprehensive treatment including liquid-supplementing and small doses of insulin therapy, in 224 patients, 210 cases were cured, and the treatment success rate was 93.75%. Fourteen cases were dead, and the fatality rate was 6.25%. Conclusions Early diagnosis, liquid-supplementing and reasonable application of insulin therapy is the key to the success of DKA treatment.
5.Correlation between hemodynamic change of extracranial cerebral vessels evaluated by ultrasound and intracranial arterial stenosis

Objective By analyzing the correlation between Doppler spectrum of extracranial cerebral vessels ECV and intracranial arterial stenosis IAS to identify the value of hemodynamic change of ECV in predicting IAS Methods One hundred and four patients with moderate to severe IAS or intracranial arterial occlusion were enrolled in the case group while 32 patients without obvious cerebrovascular disease in the same period were enrolled in the control group Ultrasound examinations of ECV were performed for all patients the hemodynamic parameters of bilateral common carotid artery CCA internal carotid artery ICA and vertebral artery VA were recorded and compared between case and control group Results Compared with control group the ECV of patients with IAS had lower velocity and higher resistance index RI Of all the parameters end diastolic velocity EDV of ICA and EDV of VA were most accurate in predicting IAS with best cut off value of 21 3 cm/s 9 6 cm/s and accuracy of 76 1 % 81 0% respectively Conclusions The ECV of IAS patients tend to have lower velocity and higher RI Analysis of hemodynamic parameters of ECV might be useful in predicting IAS.
6.14-3-3 proteins are associated with GLUT4 in rat adipocytes

Objective To investigate the interaction between glucose transporter 4(GLUT4) and the(14-3-3) proteins in rat adipocytes.Methods Isolated primary adipocytes were obtained from the epididymal fat pads of male Sprague-Dawley rats by collagenase Ⅰ digestion.Total cell extract as well as three fractions,referred to as Fraction T,H and L,were obtained from rat adipocytes using hypotonic lysis and glycerol gradient velocity sedimentation.Immunoadsorption of GLUT4 with Trisacryl beads cross-linked with 1F8,a specific monoclonal antibody to GLUT4,was conducted in total cell extract and the three fractions and followed by elution and Western blot analysis.Co-immunoprecipitation of(14-3-3) proteins and GLUT4 was demonstrated by detecting(14-3-3) in the eluates.Results(14-3-3) proteins were co-immunoprecipitated with GLUT4 either in the total adipocyte extract, or in each of the three fractions mentioned above.The co-precipitated(14-3-3)s were identified as two bands in Western blot analysis,the molecular weights of which were approximately 29kDa and 60kDa respectively,indicating that they might be in the state of homo-or hetero-dimers while interacting with GLUT4.Conclusion There is a physiological interaction between(143-3) proteins and GLUT4 in rat adipocyte.
7.Brain Targeting Drug Nano Micelle in Vitro Experimental Study

Objective To develop a novel targeted delivery system for the treatment of brain metastases from breast cancer.Methods Micelles were prepared by membrane hydration method,and the micelles were connected to Angiopep-2.Results The micelles were confirmed to be well targeted in vitro.Conclusion Nano-particles with the targeting molecule Angiopep-2 has good brain-targeting function.
9.Effects of Carisolv caries removal method on dentin bonding strength

Objective: To study the effects of Carisolv caries removal method on dentin bonding strength. Methods:Thirty extracted human third molars with moderate occlusal caries were used. Each molar was split into two parts, one part was prepared by Carisolv method and another was prepared by the low-speed burs. Then samples were randomly divided into three groups. Three adhesive systems were used: Prime&Bond NT +Dyract AP compomer、Uni-Etch +One-step plus+Renew resin composite and Prompt-L-Pop +Z100 resin composite. Microtensile bonding strength(MTBS) of the three adhesive systems were tested. Results:The MTBS of the three adhensive systems were (17.22?7.95) MPa, (25.40?8.44) MPa, (17.66?8.33) MPa in Carisolv group, and were (16.01?7.43) MPa, (23.45?7.55) MPa, (16.26?7.97) MPa in burs group. There was no significant difference between the two caries removal methods.Conclusion:Carisolv caries removal method has no side effects on dentin bonding strength of the three adhesive systems.
10.Cloning and Optimized Prokaryotic Expression of a pbmag-1 cDNA Fragment from Plasmodium berghei ANKA

Objective To clone and express a novel gene cDNA fragment,pbmag-1,from Plasmodium berghei ANKA strain. Methods The cDNA sequence of pbmag-1 was obtained from the GenBank of P.berghei ANKA genomic databases,with which a pair of primers was designed and RT-PCR was used to get a cDNA fragment of the gene from the parasite. The expanded cDNA 3′ fragment of the gene was obtained by 3′-RACE using the oligo dT primer and a set of specific primers. The intact cDNA 3′ fragment was cloned into a prokaryotic expressional vector and transformed into the BL21-(DE3)-RIL strain of Escherichia coli. The recombinant protein of PbMAg-1 was expressed with an optimized strategy and used to immunize mice. Results The pbmag-1 cDNA fragment obtained was 1 341 bp in length,A/T rich (73%) and with a correct 3′ end sequence. By Western blot,the anti-serum of mice immunized with the recombinant protein of PbMAg-1/GST,which was expressed as inclusion bodies,specifically recognized a band with Mr 64 000 molecule from the protein extracts of P. berghei-infected mouse erythrocytes. Conclusion The pbmag-1 cDNA sequence with intact 3′ has been obtained,which will be used for further study on its role in the immune response of P. berghei infection.
