1.Correlation of serum cardiac enzymes activities and schizophrenia

Objective To investigate the changes of serum myocardial enzyme in schizophrenia patients spectrum and its clinical significance.Methods 405 patients with schizophrenia hospitalized patients serum myocar-dial enzyme (CK,CK-MB,LDH,α-HBDH level,) on admission and 4 weeks after treatment the serum myocardial enzyme level,analyze its relationship with the disease, and 100 healthy people ( control group) were compared. Results the positive symptoms of schizophrenia patients before treatment,serum CK,CK-MB,LDH,α-HBDH were (1 286.52 ±714.38) U/L,(40.72 ±27.38) U/L,(143.06 ±33.24) U/L,(115.75 ±22.74) U/L,CK,CK-MB were higher than those in the control group (t=10.35,9.29,all P<0.01);after 4 weeks of treatment,CK,CK-MB, LDH,α-HBDH were (98.13 ±38.75)U/L,(12.87 ±5.73)U/L,(125.43 ±25.74)U/L,(102.15 ±26.86)U/L, compared with the control group,the differences were not statistically significant(all P>0.05).Conclusion Serum myocardial enzyme CK and CK-MB increased level was related with diseasein the patients with acute exacerbation of schizophrenia.
2. Interactions between Cannabis Fructus and human gut microflora

Objective To explore the characteristics of the interactions between traditional laxative medicine Cannabis Fructus and human gut microflora. Methods HPLC method was used to determine the content of the main unsaturated fatty acids linoleic acid and linolenic acid in Cannabis Fructus Decoction. At the same time, solid culture and liquid culture in vitro anaerobic culture method were combined with 16 S rRNA technology to analyze the interactions between Cannabis Fructus Decotion and human gut microflora. Moreover, the metabolits of linoleic acid and linolenic acid in Cannabis Fructus Decoction by human intestinal microflora were determined using HPLC method. At the same time, the possible conjugated linolenic acid and linoleic acid were determined. Results Cannabis Fructus Decoction promoted the growth of Proteobacteria significantly, which showed that Escherichia-shigella was significantly increased (P < 0.01), but the growth of Bacteroidetes was decreased (P < 0.01), and the content of unsaturated fatty acids linoleic acid and linolenic acid in Cannabis Fructus Decoction were reduced after being incubated with human intestinal bacteria, and the metabolites were conjugated linoleic acid and conjugated linolenic acid. Conclusion The interactions between Cannabis Fructus Decoction and human intestinal microflora are obvious. The Chinese medicine can change the structure of the gut microflora, and the gut microflora can metabolize the drug components. This analysis method partially restores the pharmacokinetics process of the oral administration drug in the human intestinal tract. It could provide a new insight of the mechanism research of Cannabis Fructus.

Recombinant proteins provide new means for disease treatment, while creating considerable economic benefits. Using commercial crops (mainly tobacco), cereal crops, legumes, and vegetable crops to produce recombinant proteins with medicinal value is a hot-spot for research in "molecular farming". Although many recombinant proteins have been expressed in plants, only a small number have been successfully put into use. To overcome the problems that greatly hamper the development of recombinant protein production in plants, researchers have improved expression systems to increase the yield of recombinant proteins. Starting from analyzing the problems of low yield and/or low biological activity of recombinant proteins produced by plants, the optimization strategies to solve these problems were reviewed, and future research directions for improving the yield of recombinant proteins produced by plants were proposed.

5. Application value of 40Hz auditory event related potential in hearing assessment in noise-exposed workers

OBJECTIVE: To evaluate the application value of 40 Hz auditory event related potential(40 Hz AERP) in hearing assessment in workers exposed to noise by observing the consistency between pure tone audiometry(PTA) and 40 Hz AERP. METHODS: A total of 240 ears of 120 workers who exposed to noise with PTA high-frequency hearing threshold > 25 dB were selected as the research subjects using the convenient sampling method. The thresholds of PTA and 40 Hz AERP at different frequencies were investigated. According to the average hearing threshold of PTA language frequency, the workers were divided into normal hearing group and mild-, medium-, medium-severe-, severe-hearing loss groups, and the difference and correlation between the thresholds of 40 Hz auditory potential and PTA were analyzed. RESULTS: The response thresholds of 40 Hz AERP of 0.5, 1.0, 2.0 kHz in ears of normal hearing group, and mild-and moderate-hearing loss groups were higher than the PTA hearing threshold(P<0.01), while the 40 Hz AERP response thresholds of 0.5 kHz in the ears of medium-severe-and severe-hearing loss groups were lower than the hearing thresholds of PTA(P<0.05). The different value of the response threshold of 40 Hz AERP and PTA of 1.0 and 2.0 kHz in ears of normal hearing group was higher than 0.5 kHz(median: 25.0 vs 15.0 dBHL, 30.0 vs 15.0 dBHL, P<0.01). Except for 0.5 and 1.0 kHz of mild-hearing loss group, the different value of the response threshold of 40 Hz AERP and PTA in ears of the other hearing loss groups were lower than that of the normal hearing group(P<0.01). The 40 Hz AERP response threshold was frequency-specific and correlated well with PTA at the same frequency. The correlation coefficients of 0.5, 1.0 and 2.0 kHz were 0.744, 0.732 and 0.665 respectively(P<0.01). CONCLUSION: It is feasible to evaluate PTA in noise-exposed workers using 40 Hz AERP response threshold, but the 40 Hz AERP cannot completely replace PTA. The measurement frequency and the degree of hearing loss should be considered simultaneously.
6. Mechanism of DCLK1 transcriptional regulation in HCC

AIM: To investigate the molecular mechanism through which DKK1 is transcriptionally regulated in HCC (hepatocellular carcinoma) cells. METHODS: Real time PCR was used to explore whether EGFR was involved in regulating DCLK1 mRNA expression in HCC cells; Western blot assay was used to examine whether EGFR-mediated the up-regulation of DCLK1 protein in HCC cells; Immunohistochemical (IHC) analyses were used to examine the protein expression of EGFR and DCLK1 in 39 human HCC tumor specimens. RESULTS: EGF promoted the expression of DCLK1 mRNA and protein in HepG2 and Huh-7 cells (P<0.05, P<0.01), while knockdown of EGFR with two specific siRNA could reverse EGF-induced the up-regulation of DCLK1 mRNA and protein (P<0.01). IHC analyses revealed that the amount of EGFR correlated significantly with that of DCLK1 (r=0.669 6). CONCLUSION: EGFR promoted DCLK1 transcription in HCC.

8. Effect of different courses of electroacupuncture intervention on recognition memory and proliferation and differentiation of hippocampal neural stem cells in mice with radiation-induced brain injury

OBJECTIVE: To observe the influence of different courses of electroacupuncture (EA) intervention on recognition memory and the proliferation and differentiation of hippocampal neural stem cells in mice with radiation-induced brain injury, so as to explore its mechanisms underlying improving radiation-induced brain injury. METHODS: Se-venty 30-day old C57BL/6J mice were randomly divided into control, model and EA groups, and the latter two groups were further divided into 1 week (W), 2 W and 3 W subgroups (n=10 in the control group and each subgroup). The ra-diation-induced brain injury model was established by radiating the mouse' left head at a dose of 8 Gy for 10 min by using a radiation linear accelerator. EA (1.5 V, 2 Hz/10 Hz) was applied to "Baihui" (GV20), "Fengfu" (GV14) and bilateral "Shenshu" (BL23) for 30 min, once daily for 1, 2 and 3 weeks, respectively. The learning-cognition memory ability was detected by using novel object recognition test in an open test box to record the time for exploring a novel object (TN) and a familiar object and to calculate the recognition index (RI). The neural stem cells' proliferation and differentiation in the hippocampus tissues were evaluated by counting the number of bromodeoxyuridine (BrdU)-labeled cells, neuronal nuclei (NeuN)/BrdU-positive cells and BrdU/glia fibrillary acidic protein (GFAP)-positive cells under microscope after immunofluorescence stain. RESULTS: After modeling, the TN at 90 min and 24 h and RI of the model subgroup 3 W at 90 min and RI of the model subgroup 1, 2 and 3 W at 24 h were significantly decreased in comparison with those of the control group (P<0.01, P<0.05). Moreover, the number of BrdU-positive cells in the model subgroup 1 W and 2 W, the BrdU/NeuN double-labeled cells in the 3 model subgroups and BrdU/GFAP double-labeled cells in the model subgroup 1 W and 3 W were significantly decreased (P<0.01, P<0.05). Following EA interventions, the TN in the 3 EA subgroups at both 90 min and 24 h, and RI of EA subgroup 3 W at 90 min and EA subgroup 2 W and 3 W at 24 h were considerably increased compared with those of the corresponding 3 model subgroups (P<0.05, P<0.01). The numbers of BrdU-positive cells as well as BrdU/NeuN and BrdU/GFAP double-labeled cells were significantly increased in the 3 EA subgroups (P<0.05, P<0.01, P<0.001). CONCLUSION: EA of GV20, GV14 and BL23 can improve the recognition memory ability of mice with radiation-induced brain injury, which may be related to its effect in promoting the proliferation and differentiation of stem cells in the hippocampus.
9. Preparation of hydrogel matrix sustained-release tablets of resveratrol-solid lipid nanoparticles

Objective: To prepare hydrogel matrix sustained-release tablets of resveratrol solid lipid nanoparticles, and the factors that might influence drug release and in vitro release models were also investigated in present study. Methods: The resveratrol-solid lipid nanoparticles (Res-SLN) were prepared by thin-film ultrasonic dispersion method, and then the hydrogel matrix was prepared by dispersed in excipients of hydrogel matrix sustained-release tablets. Single factor analysis was used to study the effects of varieties of adhesives, PEG, HPMC, and the amounts of HPMC K15 on drug release. Then the orthogonal design was used to gain the optimum formulation. Zero-order, first-order, Higuchi, and Ritger-Pappas models were used for the model fitting of drug release. Results: Hydrogel matrix sustained-release tablet of Res-SLN was better accorded with Zero-order kinetics model. The equation was Mt/M∞ = 0.078 7 t + 0.003 6 (r = 0.998 0). In vitro release behavior was in line with Ritger-Peppas equation, and the drug release from the tablets was controlled by degradation of the matrix. Conclusion: Prescription of hydrogel matrix sustained-release tablet of Res-SLN is reasonable; The preparation technology is feasible and exhibits perfect sustained-release characteristics in vitro in 12 h.
10. Experimental Study on Enhancing the Antitussive Efficacy and Reducing the Toxicity of Dioscorea bulbifera L. by the Processing Technology

OBJECTIVE: To investigate the effects of different processing methods on antitussive and hepatotoxicity of Dioscorea bulbifera L.. METHODS The raw and processed products of Dioscorea bulbifera L., fried Dioscorea bulbifera L. (FD), wine-fried Dioscorea bulbifera L. (WD), liquorice liquid-fried Dioscorea bulbifera L. (LD) and angelicae liquid-fried Dioscorea bulbifera L. (AD) were administered to mice at a dose of 1.7 g•kg-1 for 7 d. The effects of processing on the cough relief of Dioscorea bulbifera L. were evaluated by the cough test induced by concentrated ammonia water. The detoxification effect and the preliminary mechanism of processing on Dioscorea bulbifera L. were evaluated by the detection and analysis of the relevant indicators. RESULTS: Compared with the RD group, LD and AD group had longer cough latency (P<0.05 or P<0.01) and fewer cough times (P<0.05 or P<0.01), lower levels of serum alanine aminotransferase (ALT), aspartate aminotransferase (AST) and malondialdehyde (MDA) in liver tissue (P<0.05 or P < 0.01). ) The levels of serum AST and ALT, liver MDA, T-SOD, CAT, GSH, GPx and GST were significantly reversed (P<0.05 or P<0.01), and the levels of serum AST and ALT, liver MDA, T-SOD, CAT, GSH, GPx and GST were compared between the two groups. There was no significant difference (P>0.05). Compared with the AD group, there were more coughing times in LD group (P<0.05). Other processed products (FD or WD) had shorter cough latency (P<0.05 or P<0.01) and more cough times (P<0.05 or P<0.01) but had no effects on serum AST and ALT, liver tissue MDA, T-SOD, CAT, GSH, GPx (P>0.05) in mice induced by concentrated ammonia water. The intervention of WD only had significant reversal effect on the low GST level of liver (P<0.05 or P<0.01). The intervention of FD had no significant reversal effect on the low GST level of liver (P>0.05). CONCLUSION: Processing with Angelica sinensis and licorice could enhance the antitussive effect of Dioscorea bulbifera L. and reduce its hepatotoxicity. The mechanism of attenuation may be related to the inhibition of lipid peroxidation and the enhancement of antioxidant level in liver. The antitussive effect of two kinds of processing, stir-fried and wine-fried, is only effective, but no synergistic effect, no toxic effect and no attenuation effect are found.
