The hydroponic culture test method was used to study the physiological and biochemical responses of Paulownia fortunei seedlings under Zn stress, Cd stress, and combined Zn and Cd stress as well as changes in the enrichment and transfer characteristics of heavy metals. Under single and combined heavy metal stress, the biomass, plant height, and peroxidase (POD) activity of P. fortunei decreased as the treatment concentration increased. Combined Zn and Cd affected adversely plant height and biomass. As the concentration of Zn increased when applied alone, the chlorophyll content and catalase (CAT) activity of P. fortunei first increased and then decreased, the superoxide dismutase (SOD) activity increased, and the aboveground malondialdehyde (MDA) content first decreased and then increased. As the concentration of Cd increased when applied alone, chlorophyll content and CAT activity increased, and SOD activity and aboveground MDA content first increased and then decreased. Under both Cd and Zn, the physiological response was more complex. Cd in the seedlings of P. fortunei was concentrated in the root. In contrast, Zn was concentrated in the upper part of the ground, and its transfer coefficient was greater than 1.00. Thus, the addition of Zn promotes the transfer of heavy metals to the above-ground portions of plants. Generally, P. fortunei can effectively promote ecological restoration under complex forms of heavy metal pollution.

Guanosine triphosphate cyclohydrolase (GTP cyclohydrolase,Gch) is a protease with a GTP- cyclohydro domain, which is widely found in vertebrates and invertebrates.Mammals and birds only have Gch 1.In teleost and amphibian, other two paralogs (Gch2 and Gch3) also exists besides Gchl, which also displayed functional differences.Gch is a rate-limiting enzyme that ultimately synthesized the tetrahydrobiopterin (BH4) using guanosine triphosphate as a substrate.BH4 is an essential coenzyme of aromatic amino acid hydroxylase and contributes to the synthesis of various hormones and neurotransmitters.The Gch is an initial step in the catalysis of various pterin biosynthesis and plays important roles in a series of physiological and pathological processes, such as skin pigmentation, ocular pigmentation, methotrexate, folic acid, and tetrahydrobiopterin.The physiological function of Gch is inextricably linked to the biosynthesis of BH4.As the only rate-limiting enzyme in BH4 biosynthesis, the activity of Gch is a useful indicator for the development of neurons and pigment cells.Besides, it is also an important marker of pigment synthesis and neurotransmitter biosynthesis.Nowadays, the functions of Geh in pathogenesis of tumor and cardiovascular diseases have been widely concerned, while the researches on the pigmentation and color formation are mainly concentrated in insects, and rarely in teleost.Therefore, this article summarized the characteristics of Gch genes, protein and the functions of Gch in fish coloration, which has important guiding significance for further illustration the mechanism of Gch in teleost pigmentation and fish color genetic improvements.
4.Treatment of multi-segment fracture of complex femoral shaft with instrument-assisted reduction combined with intramedullary interlocking nail fixation.

OBJECTIVETo investigate the effect of minimally invasive mini-incision and instrumented reduction combined with interlocking intramedullary nailing in the treatment of patients with multi-segment fracture of complex femoral shaft.
METHODSFrom January 2013 to January 2016, 32 patients with multiple fractures segments of femoral shaft were treated with instrumentation-assisted reduction combined with interlocking intramedullary nailing, including 22 males and 10 females with an average age of 45 years old ranging 17 to 68 years old. The time from injured to operation was 5 to 10 days with an average of 7 days. After admission, routine tibial tubercle or supracondylar bone traction was performed. The patient's general condition was evaluated, the operation time and intraoperative blood loss were recorded. According to Thorsen femoral fracture morphology evaluation criteria and Hohl knee function evaluation of postoperative efficacy, postoperative fracture healing, complications and postoperative recovery of limb function were observed.
RESULTSAll patients were followed up for 6 to 24 months with an average of 12 months. The operative time ranged from 48 to 76 minutes with an average of 67 min. The intraoperative blood loss was 150 to 400 ml with an average of 220 ml. The surgical incisions all achieved grade A healing. The fractures reached the clinical standard of healing. The fracture healing time ranged from 4.2 to 10.8 months with an average of 5.7 months. There were no nonunion, incision infection and internal fixation fracture, failure and other complications. According to Thorsen femoral fracture morphology evaluation criteria, the result was excellent in 28 cases, good in 3 cases, fair in 1 case. According to Hohl knee function evaluation criteria, the result was excellent in 30 cases, good in 2 cases.
CONCLUSIONSInstrument-assisted reduction combined with interlocking intramedullary nail fixation is a safe and effective method for the treatment of complex femoral shaft fractures. It has advantages of small trauma, fixed fixation, quick recovery, early postoperative functional exercise.
5.Manual reduction and traction fixation for the treatment of metacarpal neck fractures under ultrasound-guided.

OBJECTIVETo study clinical efficacy of manual reduction and traction fixation for the treatment of metacarpal neck fracture under ultrasound-guided.
METHODSFrom April 2013 to August 2016, 30 patients with metacarpal neck fractures were treated with manual reduction and traction fixation under ultrasound-guided, including 26 males and 4 females aged from 14 to 56 years old with an average of (25.6±1.6) years old, the courses of diseases ranged from 7 h to 5 d with an average of (2.7±0.6) d. Twenty patients were the fifth metacarpal neck fracture, 7 patients were the 4th and 5th metacarpal neck fractures, 3 patients were the second metacarpal neck fracture. Fracture healing, angle of bilateral head shaft angle and active range of metacarpophalangeal joints was measured, and DASH score was applied to evaluate function.
RESULTSTwenty-seven patients were followed up from 6 to 11 months with an average of(7.2±0.8) months. Fracture were healed from 5 to 8 weeks with an average of (5.6±0.4) weeks. The affected shaft angle was (15.1±1.8)°, and health head shaft angle was (13.5±2.8)°, while there was no significant difference (=1.54, >0.05). The affected range motion of metacarpophalangealjoint was(86.3±2.6)°, health active range motion of metacarpophalangeal joint was(91.8±1.6)°, and no significant difference between both side (=1.16, >0.05). DASH score was 4.3±1.5 at 7 months after operation.
CONCLUSIONSManual reduction and traction fixation for the treatment of metacarpal neck fracture under ultrasound-guided could dynamic observe fracture position in time, high patients' acceptability and is a feasible method for the treatment of metacarpal neck fracture.

7.Epidemiologic study on basic TCM syndrome in the subhealth population in the Zhengzhou area.

OBJECTIVETo study the distribution characteristics of clinical manifestation and TCM basic syndromes in the subhealth population.
METHODSThe investigation list for basic syndrome of subhealth used in the study was formulated through retrieving a large amount of literatures, referring to principles of epidemiology, computer science, mathematics and international metrics, and according to the standard denoting figure of GR/T 1.1-2000 and ZY/T001.1-94. The data obtained were analyzed by the statistical methods like multiple factor analysis and cluster analysis to explore the distribution of main symptoms and syndromes in the subhealth condition.
RESULTSThe analysis of data from 2,133 lists showed that in the Zhengzhou area, the main clinical symptoms presented in the subhealth population were aversion to cold, fatigue, spontaneous perspiration, poor memory, bad-temper, short breath, palpitation, etc.; with the syndrome key elements of deficiency, dampness, phlegm, stagnancy, stasis, qi-stagnation, and body fluid loss; the dysfunction of zang-fu organs was mainly observed in Xin, Pi, Shen, Gan, Dan, Wei, Fei, etc.; and the most commonly encountered basic syndromes were Xin-Pi deficiency, Xin-Shen deficiency, Gan-qi stagnation, Pi insufficiency with dampness stagnation, phlegm-qi cementation, Shen-yin deficiency, Shen-yang deficiency, qi insufficiency, Gan-stagnancy transforming to fire, damp-heat accumulation, etc.
CONCLUSIONThe clinical manifestation of the subhealth population in the Zhengzhou area is characterized by a syndrome composed of a certain main symptom with some other accompanied symptoms, the distribution of the basic syndrome is mainly dominated by the deficiency syndrome.

8.Efficacy of Dexamethasone implant injection in the treatment of persistent diabetic macular edema

AIM: To investigate the efficacy of dexamethasone implant injection in the treatment of persistent diabetic macular edema(DME).
METHODS: The clinical data of 30 DME patients(30 eyes)after anti-vascular endothelial growth factor(anti-VEGF)agents from June 2019 to January 2020 were retrospectively analyzed. All patients had underwent intravitreal injection of DEX. Best corrected visual acuity(BCVA), intraocular pressure(IOP), central macular thickness(CMT)and adverse events were recorded.
RESULTS:BCVA(LogMAR)was significantly improved from(0.86±0.47)to(0.57±0.53, 0.42±0.48, 0.43±0.26, 0.45±0.66 and 0.51±0.37)at 1, 2, 3, 4 and 6mo after treatment. CMT improved significantly from baseline(495.64±107.23)μm to(412.57±74.55, 370.21±23.83, 371.53±52.66, 373.28±68.49, 389.35±95.61)μm 1, 2, 3, 4 and 6mo post-injection. The drug reached its peak efficacy 2mo after administration. 4 eyes were given repeated injection at 3mo, 5 eyes were given at 5mo and 2 eyes were given at 6mo following the primary injection. Cataract progression was showed in 1 case. 4 patients demonstrated IOP levels above 22mmHg and were mostly controllable by IOP-lowering medications. None of patients showed any adverse events at 6mo of follow-up.
CONCLUSION: Dexamethasone implant injection showed significant BCVA and CMT improvement. DEX injection may be utilized as an effective DME therapy for patients who are persistent after anti-VEGF treatment.
9.Effect of DR4 Demethylation to the Proliferation and Apoptosis of Myeloid Leukemia K562 Cells.

OBJECTIVE:
To investigate the effect of tumor necrosis factor death receptor (DR) 4 demethylation to the proliferation and apoptosis of myeloid leukemia K562 cells.
METHODS:
The logarithmic phase of K562 cells were treated by desitabine (DCA) at 0, 0.8, 1.6 and 3.2 μmol/L, and the cells were divided into control group, DCA low dose group, DCA medium dose group and DCA high dose group respectively. The cells in control group were treated by tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) 0.5 μg/ml for 24 h, and the cells were divided into TRAIL group. The cells in DCA high dose group were treated by TRAIL 0.5 μg/ml for 24 h, and were divided into DCA high dose + TRAIL group. Methylation-specific polymerase chain reaction (MS-PCR) was used to measure the methylation status of the DR4 gene promoter in the control group and DCA low, medium and high dose groups. Real-time fluorescent quantitative polymerase chain reaction (qRT-PCR) and Western blot were used to determine the relative expression of DR4 mRNA and protein in the control group and DCA low, medium and high dose groups. Dime- thylthiazole (MTT) method was used to determine the inhibition rate of cell proliferation of the cells in control group, DCA high dose group, TRAIL group, DCA high dose + TRAIL group. Flow cytometry was used to determine the apoptotic rate of the cells in control group, DCA high dose group, TRAIL group, DCA high dose + TRAIL group.
RESULTS:
The cells in the control group were methylation-positive, the brightness of the methylation bands of the cells in the DCA low, medium, and high dose groups was gradually decreased to disappear, and the DCA high dose group showed negative for methylation. The relative expression of DR4 mRNA and protein in the control group, DCA low, medium and high dose groups was increased sequentially (r=0.624, 0.704). The inhibition rate of cell proliferation of the cells in the control group, DCA high dose group, TRAIL group, DCA high dose + TRAIL group was increased sequentially (r=0.653, 0.754, 0.709, 0.725) at 24, 48 and 72 h.
CONCLUSION
DCA can reverse the methylation level of DR4 gene promoter in ML K562 cells and up-regulate the expression of DR4, which may enhance the proliferation inhibition and apoptosis promotion effects of TRAIL on K562 cells.

10.Anti-Allergic Rhinitis Mechanism of Bikangfu via Regulation of IL-35/STAT3, Inhibition of Eosinophil Activation and Improvement of Immune Function

ObjectiveTo investigate the mechanism of interleukin-35(IL-35)/signal transducer and activator of transcription 3(STAT3)inhibition of eosinophil activation against allergic rhinitis(AR) by Bifukang. MethodOne hundred patients were randomly divided into a control group and a treatment group,50 cases in each group. The control group was given mometasone furoate nasal spray,and the treatment group was given Bifukang by nasal packing. The course of treatment was 28 days. The clinical efficacy,nasal classification and visual analogue scale(VAS) score of the two groups were observed before and after treatment. Enzyme linked immunosorbent assay(ELISA) was used to detect the expression levels of inflammatory factors [interleukin(IL)-4,IL-10,IL-17,IL-35] and Eotaxin and CC chemokine receptor-3(CCR3)in serum and nasal secretion of the two groups. The expression levels of STAT1,STAT3 and STAT4 were detected by real-time polymerase chain reaction(Real-time PCR).The content of immunoglobulin G(IgG) and the ratio of CD4+/CD8+were detected by ELISA and flow cytometry. ResultAfter treatment, compared with before treatment, the levels of IL-4 and IL-17 in serum and nasal secretion in 2 groups were decreased, while the levels of IL-10 and IL-35 were increased (P<0.05, P<0.01). The expression of STAT1, STAT2 and STAT3 in nasal secretions were significantly decreased (P<0.05). IgG and CD4+/CD8+ were decreased, and the differences were statistically significant (P<0.05, P<0.01).After treatment,compared with the control group,the levels of IL-4 and IL-17 in serum and nasal secretions of the treatment group were decreased,while the levels of IL-10 and IL-35 were increased (P<0.05). The expression of STAT1,STAT3 and STAT4 in the treatment group was significantly inhibited compared with the control group after treatment (P<0.05). In addition, the post-treatment serum CD4+/CD8+ and immunoglobulin G (IgG) levels were reduced in the treatment group compared with those of the control group (P<0.05, P<0.01). During the treatment,there were no abnormal changes in heart,liver,kidney function and routine blood and urine tests in the two groups. ConclusionBifukang has a good effect on allergic rhinitis,and its mechanism may be related to the regulation of IL-35/STAT3 pathway,the inhibition of eosinophil activation and the improvement of related immune function.
