1.THE STUDY OF NON-CYTOTOXIC CHEMOTHERAPY IN GASTRIC CANCER

objective To study the inhibition effect and its mechanism of a non - cytotoxic chemotherapy drug in the growth of gastric cancer. Methods The inhibition effect was observed in the growth of xenotransplanted gastric cancer cells MKN45 with a gastrin receptor(GR) antagonist. On the basis of the determination of expressed GR in 34 patients with gastric cancer, we evaluated the preliminary clinical effect of PGL to gastric cancer with the positive GR. Results It is less tumor area, weight and gastric cancer cells DNA Index, average DNA contents and S phase fraction in PGL group than in control group. It is the least in PGL + SST group. GR is easy to be expressed in advanced gastric cancer of the body, cardia and fundus. With the 16-21 months follow- up of 14 patients with positive GR gastric cancer, we find that 2 patients died in control group of 8 patients, and all patients are alive in PGL group. Conclusion PGL can regulate the growth of gastric cancer cells MKN45. It would be able to improve the prognosis of gastric cancer.
2.Volumetric and functional changes of skeletal muscles following microwave coagulation

BACKGROUND: Microwave had been widely used in medical field, which can lead to local coagulation necrosis and repair the necrosis with fibrous tissue. However, whether microwave coagulation can be used in stomatology is poorly understood.OBJECTIVE: To observe the functional and volumetric changes of skeletal muscles following microwave coagulation, and to explore the possibility of microwave coagulation for volumetric reduction of skeletal muscle.METHODS: Both sides of tibialis anterior muscle of 20 New Zealand rabbits were exposed; one side was coagulated by 2 450 MHz microwave therapeutic instrument at 70 W for 20 seconds. No treatment was performed at the other side. Rabbits were sacrificed at hours 24, 48, weeks 1 and 8 after microwave coagulation. The volumetric changes of the ablated tibialis anterior muscle were measured, and electricitic physiology observations were conducted on the ablated muscle at 8 weeks after microwave coagulation before being sacrificed.RESULTS AND CONCLUSION: The volume of ablated muscle increased at hours 24 and 48, which was (5.82±0.93) and (6.04±0.47) mL, especially greater at hour 48 after microwave coagulation. After 1 week, the muscle volume began to decrease to (4.90±0.80) mL, reduced to (4.27±0.67) mL at week 8, which was 23.6% volumetric loss. However, the electrophysiologic observation showed that the latent periods were (1.765+0.393) and (1.760±0.394) ms, and the wave width was (6.273±0.808)and (6.259±0.773) ms of the control group and experimental group, respectively, without apparent differences (P > 0.05). The volume of the skeleton muscle increased at hour 48 after microwave coagulation, and then decreased, but the muscle function of the skeleton muscle can be preserved.
3.Morphological changes of ablated muscle after MCT

Objective:To observe the morphological changes in skeletal muscles after therapeutic. Methods:Tibialis anterior muscles of 25 New Zealand rabbits were exposed and coagulated with 70 W for 20 s. The rabbits were sacrificed at 1 hour?24 hours?48 hours?1 week and 2 months after MCT. The specimens of coagulated areas were prepared for histological observation. Results:Obvious rims could be found bet ween coagulated and normal tissues .The ablated site showed tissue fixation in the inner zone and coagulative necrosis in found the outer zone.these were four zones can be seen under high powered magnification:the application zone,the central zone,the transition zone and the reference zone. Demarcation zone of necrosis appeared 24h after MCT. Tibrosis encapsulation occurred after 7 days.Replacement with fine cicatrix was demonstrated after 2 months. Conclusion:Tissue destroyed by coagulation can be replaced with a fine cicatrix gradually.
4.Study on Quality Standard of Patch for Hyperosteogeny

OBJECTIVE:To establish the quality standard of Patch for hyperosteogeny.METHODS:Mastic and Myrrh in Patch for hyperosteogeny were qualitatively identified by TLC,and the content of paeoniflorin in preparation were determined by HPLC.RESULTS:The identification of TLC has a good resolution and specificity.The linear range of paeoniflorin were 0.204~1.428 ?g(r=0.999 2) with an average recovery of 99.41%(RSD=0.64%,n=6).CONCLUSION:Established standard can be used for the quality control of Patch for hyperosteogeny.

6.Effects of ZHX3 gene silence on the expression of osteoblast-related factors in BMSCs

Objective To investigate the effects of zinc fingers and homeoboxes 3 (ZHX3) silence on expressions of smad3, smad4 and RUNX2 in bone marrow mesenchymal stem cells (BMSCs). Methods ZHX3 low expression vector (ZHX3 silent group) was constructed and was transfected to rat BMSCs. Empty vector was transfected into BMSCs and was used as vehicle control group, and wild type BMSCs was used as the control group. The cell transfection rate was measured under a fluorescence microscope, and then the successful transfection was identified. The immunocytochemistry and immu?noblotting methods were used to detect the expression levels of smad3, smad4 and RUNX2. Results (1) Cells with BMSCs phenotype can be obtained by recovery culturing. (2) After transfection, the green fluorescent protein was found in ZHX3 si?lence group and vehicle control group. Blank control group showed no significant fluorescence. The expression level of ZHX 3 was significantly lower in ZHX3 silence group than that of vehicle control group. (3) Results of immunofluorescence asssay showed that the positive expressions of smad3 and smad4 were located in nucleus and cytoplasm, the positive expression of RUNX2 was mainly located in nucleus. Positive cells were observed in three groups. There was no significant difference in fluorescence intensity between the control group and the vehicle control group, but the fluorescence intensity was significant?ly lower in ZHX3 gene silence group than that of two control groups. (4) There were no significant differences in expressions of smad3, smad4 and RUNX2 betweem control group and the vehicle control group, but they were significantly higher than those of ZHX3 silence group(P < 0.05). Conclusion ZHX3 gene silence can delay vitro osteogenesis of BMSCs, which may play a role by the down-regulated expression levels of smad3, smad4 and RUNX2.
7.Comparison of different ways to trace BMSCs after cerebral ischemia-reperfusion injury

Objective To compare different ways to trace bone marrow mesenchymal stem cells (BMSCs) after being transplanted in cerebral ischemia-reperfusion injury. Methods Male SD rats of SPF grade were randomly divided into sham group, model group (ischemia-reperfusion,IR), BrdU tracing group, PKH26 tracing group and GFP tracing group. Fo?cal cerebral ischemia-reperfusion model was established by blocking middle cerebral artery. 24 hours after cerebral isch?emia-reperfusion injury, 10μL BMSCs that were labeled respectively by BrdU, PKH26, GFP were added respectively into BrdU, PKH26 and GFP tracing group while equal volum of normal saline was added into sham group and model group. Mod?el and transplanting cells efficacy was determined by neural behavioral score, TTC staining and brain water content;Neurons were counted using tar violet staining;The number of transplant cells in the transplanting site was assessed by fluorescence microscopy. Results Before transplanting, there was no significant difference among BrdU, PKH26 and GFP group in cell labeled efficacy. By contrast, neural behavioral score, brain infarct volume and brain tissue water content were significantly lower in all three tracing groups than that in model group 4 weeks after transplantation while neuron counts were markedly higher. There was no significant difference of above parameters among the three tracing groups. However, the number of traced transplanting cells in damaging area in GFP group is significantly higher than that in BrdU group and PKH26 group. Conclusion In cerebral ischemia-reperfusion injury, the tracing effect of GFP last longer, therefore it is significantly more effective than BrdU and PKH26.

9.THE 3D VISUALIZATION OF THE PULMONARY VENOUS SYSTEM OF CHINESE PEOPLE

Objective To build a digitized 3D model of the pulmonary venous system of Chinese people for the purpose of providing anatomic information of the pulmonary venous system for anatomic teaching,imaging diagnosis and surgical operation of pulmonary diseases. Methods The primal images from the first Chinese visible human data set of chest were converted into image sequence only cantaining the sections of pulmonary veins after manual registration and image segmentation.Then these images were imported into 3DMed software to build a 3D model of the pulmonary venous system through threshold registration algorithm. Results The three dimensional structures of the pulmonary venous system were reconstructed entirely.All reconstructed structures could be displayed individually or jointly,and could be rotated continuously on any plane.Conclusion The branches and the spacial characters of the pulmonary venous system can be clearly shown on the reconstructed 3D model.It will help anatomy teaching and provide morphological data for image diagnosis and lung operations.
