1.Value of serum MCP-1 and sICAM-1 in neonates with hypoxic-ischemic encephalopathy

Objective To study the levels of serum MCP-1 and sICAM-1 in neonates with hypoxic-ischemic encephalopathy(HIE) and to explore the clinical value of serum MCP-1 and sICAM 1 for judgment and prognosis of brain damage in neonates with HIE.Methods The levels of serum MCP-1 and sICAM-1 in the 1 st and 7th day after birth of the 53 cases term infant with HIE(29 cases were mild HIE,15 cases were moderate HIE and 9 cases were severe HIE) were measured by enzyme linked immunosorbent assay (ELISA),and compared with 20 cases of normal newborns.Results The levels of serum MCP-1 and sICAM-1 in mild,moderate and severe HIE groups were all significantly higher than those in controls(P < 0.05) ;With increasing of the degree of brain injury,the levels of serum MCP-1 and sICAM-1 increased (P < 0.05) ; Compared with acute period (the 1 st day after birth),the levels of serum MCP-1 and sICAM-1 were clearly decreased in recovery stage(the 7th day after birth) in all HIE groups.Conclusion MCP-1 and sICAM-1 may play important roles in the pathogenesis of neonatal HIE and have clinical value in the diagnosis and treatment.
2.Management of Nosocomial Infections in Fourteen Primary Health Centers: A Survey

OBJECTIVE To understand situation in management of nosocomial infections in primary health centers in order to provide evidence for improving the management quality.METHODS An observatory study was(conducted) in 14 primary health centers using the evaluation criteria based on the requirements of The Guidelines for Management of Nosocomial Infections and the Guidelines for Disinfection in Hospitals.RESULTS Problems were found to a different extent in all of the 14 health centers.Most alarming problems were high incidence(15.7%),high(missing report)(67.7%),frequent use of antibiotics(71.2%),low proportion of nosocomial infection(specimens) submitted for testing(12.9%),lack of surveillance system for nosocomial infections and the data(feedback),and non-qualified hand-washing equipment in wards and clinics.CONCLUSIONS Special attention to management of nosocomial infections,improvement of surveillance system,and capacity-building of professionals in management of nosomial infections is the key issues for sustained and perfect development of management of nosocomial(infections) in primary health centers.
3.Protective effects of vitamin B12, ginseng saponin, and folic acid against murine fetal deformities caused by hyperthermia.

OBJECTIVETo investigate the protective effects of vitamin B(12), ginseng saponin, and folic acid on mouse embryos subjected to high heat.
METHODSMice were used for the experiment.
RESULTSAfter exposure of pregnant mice to high heat, the rates of teratism, stillbirth, and fetal absorption were markedly lower in mice treated with ginseng saponin and folic acid following heat exposure than in untreated mice. There were no significant differences in these rates when comparing mice treated with vitamin B(12) with the untreated mice.
CONCLUSIONSGinseng saponin and folic acid can lessen injuries to murine embryos caused by high heat, while vitamin B(12) has little protective effect against high temperature except for promoting overall embryonic growth.

4.Preparation of IgE high affinity receptor protein and its biological function identification

Objective To prepare human alpha segment of high affinity IgE receptor(FcεRIα)protein by genetic engineering technology and to identify its biological function for laying the foundation for further researching the role of FcεRIα in allergic disea-ses.Methods The human FcεRIα gene was obtained by the PCR based accurate synthesis(PAS)method and the prokaryotic ex-pression vector pET-28a(+)was constructed.The FcεRIα was expressed at low temperature induction and the recombinant protein was purified by His tag.The biological function of recombinant human FcεRIα protein was identified by ELISA.Results The hu-man FcεRIα gene was amplified by PAS with a size of approximately 560 bp.The pET-FcεRIα plasmid was correct through the double enzyme digestion and sequencing identification.The human FcεRIα with a molecular weight of approximately 22 000 was in-duced and purified.The recombinant human FcεRIα could effectively detect human serum anti-FcεRIα autoantibody and could com-bined with serum IgE antibodies with high efficiency.Conclusion Human FcεRIα protein is successfully prepared,which prelimina-rily has the ability for detecting the human serum anti-FcεRIα autoantibodies and IgE antibodies,and provides a favorable practical base for further study.
