1.Value of serum MCP-1 and sICAM-1 in neonates with hypoxic-ischemic encephalopathy

Objective To study the levels of serum MCP-1 and sICAM-1 in neonates with hypoxic-ischemic encephalopathy(HIE) and to explore the clinical value of serum MCP-1 and sICAM 1 for judgment and prognosis of brain damage in neonates with HIE.Methods The levels of serum MCP-1 and sICAM-1 in the 1 st and 7th day after birth of the 53 cases term infant with HIE(29 cases were mild HIE,15 cases were moderate HIE and 9 cases were severe HIE) were measured by enzyme linked immunosorbent assay (ELISA),and compared with 20 cases of normal newborns.Results The levels of serum MCP-1 and sICAM-1 in mild,moderate and severe HIE groups were all significantly higher than those in controls(P < 0.05) ;With increasing of the degree of brain injury,the levels of serum MCP-1 and sICAM-1 increased (P < 0.05) ; Compared with acute period (the 1 st day after birth),the levels of serum MCP-1 and sICAM-1 were clearly decreased in recovery stage(the 7th day after birth) in all HIE groups.Conclusion MCP-1 and sICAM-1 may play important roles in the pathogenesis of neonatal HIE and have clinical value in the diagnosis and treatment.
2.The value of serum MMP-9 concentration in the diagnosis of recurrence of endometriosis after operation

Objective:To investigate the value of serum matrix metalloproteinases- 9(MMP-9) level in the diagnosis of recurrence of endometriasis(EMs) after operation.Methods:Sixty-nine patients who had a previous operations for endometriosis were recruited in the study from February 2002 to March 2005. All the patient were examined by laparoscopy 6-12 months later after surgery. The serum were collected from the 69 patients and 25 health women for MMP-9 level measurement. ELISA was used to measure the level of serum MMP-9.Results:Twenty-two recurrent patients were diagnosed by Laparoscopy. The level of serum MMP-9 were 252.21?17.90 ng/ml in patients with recurrent endometriosis, 31.26?1.84 ng/ml in patients without recurrent endometriosis, 12.93?0.57 ng/ml in women without endometriosis. There was a clinical significance between the recurrent endometriosis group and non-recurrent endometriosis group and health women. The sensitivity for detection of recurrent endometriosis was 90.9% and the specificity was 85.1% using serum MMP-9 profile.Conclusion:Serum MMP-9 measurement is a sensitive, specific and non-invasive methods for the diagnosis of recurrent endometriosis.
3.cDNA microarray in gene expression profiles of ovarian cancer

Objective To study the gene expression profiles of ovarian cancer by cDNA microarray,and preliminarily analyze the function of part of differential expression genes.Methods BiostarH-40s gene microarray containing 4 097 genes was used to analyze gene expression patterns in tissue samples from 5 cases of human ovarian serous cystadenocarinoma(cy5-dUTP present ) and 5 cases of normal ovarian tissues(cy3-dUTP present).Results 163 genes of differential expression were found in more than 4 cases of ovarian cancer,the expression of 66 genes increased(up-regulated),the expression of 97 genes decreased(down-regulated). 37 differential genes with difinite gene function classification were found including three protocogene and tumor suppressor genes,one cyclin protein gene,three cytoskeletal and movement protein genes,one DNA binding,transcription and transcription factors gene,two cell receptor genes,five immune-related protein genes,seven metabolism genes,two protein translation and synthesis genes,three growth-related genes and seven other types of genes.Conclusion Gene expression profiles of ovarian cancer can be detected by cDNA microarray,and differential expression genes and their gene function classification of ovarian cancer are found.
4.Clinic curative effects of low-dose mifepristone in treating recurrent endometriosis and changes in ovarian function and serum cortisol level

0.05).After treatment,blood serum E2 level in group M was (74.18?22.32) pmol?L-1,whereas that in group D was (24.96?3.94)pmol?L-1,there was significant difference(P0.05).Conclusion Clinical symptoms of patients with recurrent endometriosis in two groups are improved.Serum E2 level in group M remains in the follicular phase,but group D declines to postmenopausal range.Mifepristone does not result in lack of estrogen,ovulation recovers rapidly after treatment in group M compared with group D.Mifepristone and danazol have not obvious impact on blood serum cortisol.
5.Primary determination for activity and expression of Stx 2a’-LHRH chimeric toxin

AIM: To express chimeric toxin Stx2a’-LHRH a nd to investigate the cytotoxic activity of recombinant toxin Stx2a’-LHRH to huma n carcinoma cells.METHODS: Stx2a’-LHRH sequences that added the res tri ction endonucleases NcoⅠ and EcoRⅠ at the 5' and 3 ends were amplified by PCR a nd digested with appropriate restriction enzymes. The digested fragment was subc loned into the vector obtatined by digestion of plasmid pET-28a(+) with NcoⅠ an d EcoRⅠ. E. coli BL21 (DE3) cells were transformed with plasmids of interst and cultured in LB medium containing ampicillin. Expression of the recombinant protein was induced by the addition of isopropylthio-?-D-galactoside (IPTG). T h e cytotoxity of Stx2a’-LHRH to Hep-2 cells was observed under the microscop y. RESULTS: Recombitant plasmid pET-SL was constructed successfu lly and the clones expressing pET-SL stablely were obtained. A special electroph oretic band in SDS-PAGE (a glycoprotein of 28kD) was noted. Stx2a’-LHRH killed He lp-2 cells clearly. CONCLUSION: In this study, construction of c himeric toxin Stx2a’-LHRH and its expression were described. Moreover, it has o bvious cytotoxity to Hep-2 cell. These finding could open up new vistas in the s tudy of targeted durgs.
6.Protective effects of vitamin B12, ginseng saponin, and folic acid against murine fetal deformities caused by hyperthermia.

OBJECTIVETo investigate the protective effects of vitamin B(12), ginseng saponin, and folic acid on mouse embryos subjected to high heat.
METHODSMice were used for the experiment.
RESULTSAfter exposure of pregnant mice to high heat, the rates of teratism, stillbirth, and fetal absorption were markedly lower in mice treated with ginseng saponin and folic acid following heat exposure than in untreated mice. There were no significant differences in these rates when comparing mice treated with vitamin B(12) with the untreated mice.
CONCLUSIONSGinseng saponin and folic acid can lessen injuries to murine embryos caused by high heat, while vitamin B(12) has little protective effect against high temperature except for promoting overall embryonic growth.
