1.Approach to matter-element analysis method for quality evaluation of Chinese materia medica based on spectrum-activity relationship

To propose a new pattern for quality control and evaluation of Chinese materia medica(CMM).The limitation of quality control pattern for CMM was investigated and a new method for quality evaluation has been explored and designed,combining with the introduction on the conception of matter-element evaluation model and its application to product quality evaluation in value engineering,as well as the research survey on the spectrum-activity relationship of CMM.The new method for quality evaluation referred to the results of research concerning the spectrum-activity relationship of CMM.Taking the quality grade,quality index,and chemical composition of CMM as matter-elements,the evaluated grades and tested data were normalized to establish the matter-element model.With the establishment of the improved quality control method,which is in accordance with the basic characteristics of CMM,it is expected that the safety,efficiency,and quality stability of CMM could be ensured and a new scientific research strategy and technological support could be provided to promote the modernization of CMM.
2.Clinical Curative Effect and Safety of MCT/LCT Fat Emulsions at Different Doses in Preterm and Low Birth Weight Infants

Objective:To analyze the effect of MCT/LCT fat emulsions at different doses on the growth and development of prema-ture and low birth weight infants. Methods: Totally 76 cases of preterm or low birth weight infants were divided into the treatment group (40 cases) and the control group (36 cases), both groups were given MCT/LCT fat emulsions, amino acid and glucose injec-tions in 12-24 h after the birth. The initial dose of MCT/LCT fat emulsions in the treatment group was 2. 0 g·kg-1 ·d-1 and the dose was gradually increased to 3. 0 g·kg-1 ·d-1 with 0. 5 g·kg-1 ·d-1 progressive increase. The initial dose of MCT/LCT fat emulsions in the control group was 0. 5 g·kg-1 ·d-1 , and the dose was increased to 3. 0 g·kg-1 ·d-1 with 0. 5 g·kg-1 ·d-1 progressive in-crease. The treatment course was 7 days. The neonatal weight, serum total bilirubin, direct bilirubin, alanine aminotransferase, total cholesterol, triacylglycerol and blood glucose were monitored in the two groups. Results:The born body mass recovery time of the ob-servation group was (4. 38+0. 93) d, which was significantly shorter than that in the control group [(6. 81+1. 90) d]. After the re-covery, the body weight growth rate of the observation group was (30. 41+1. 81) g·kg-1 ·d-1 , which was significantly higher than that of the control group [(18. 21 +2. 08) g·kg-1 ·d-1, P <0. 05]. After the 7-day treatment, the blood biochemical indices showed no statistically significant difference between the two groups (P>0. 05). Conclusion:Early application of MCT/LCT fat emul-sions at high dose is beneficial to the improvement of nutritional status in premature infants.
3.Study on biosynthesis of silver nanoparticles using fagopyri dibotryis rhizoma extract and optimization of synthesis conditions.

Silver nanoparticles were synthesized from the extract of Fagopyri Dibotryis Rhizoma and the optimization of synthesis was studied. The absorbance of UV-visible spectroscopy was determined under the different influencing factors such as extracting time of Fagopyri Dibotryis Rhizoma powder, reation temperature of synthesis, volume of Fagopyri Dibotryis Rhizoma extract and concentration of AgNO3 to seek the optimization conditions. By means of FT-IR, TEM, DLS and XRD, the silver nanoparticles were characterized. The results showed that when the boiling time of Fagopyri Dibotryis Rhizoma powder was 5 min, resultant temperature was 25 degrees C, the volume ratio of 0.1 g x mL(-1) Fagopyri Dibotryis Rhizoma extract and 1 mmol x L(-1) AgNO3 was 1 to 10, and the reaction time was 3.5 h, the obtained silver nanoparticles had mean size about 27 nm and Zeta potential about -34.3 mV with good uniformity and dispersivity. Therefore, the green synthesis method of silver nanoparticles using extract of traditional Chinese medicine is stable and feasible.

Printed-circuit-board ion trap (PCBIT) is a novel ion trap mass analyzer,which is capable of optimizing its internal electric field distributions by adjusting the radio frequency (RF) voltage-divided ratio to improve its analytical performance.This work introduced odd electric field components into the trapping volume to achieve unidirectional ion ejection by applying asymmetric RF voltages to x electrode pairs of PCBIT.In this case,the center of ion vibration was displaced away from the geometrical center of PCBIT and ions were ejected predominantly through one of x electrode pairs.The relationship between asymmetric voltage-divided ratio (AV) and internal electric field distributions was investigated by simulation software SIMION and AXSIM.At the same time,the ion trajectories and simulated mass spectrum peaks were calculated.The results showed that,for ions with m/z 609 Th,a mass resolution over 2500 and an ion unidirectional ejection efficiency of over 90% were achieved in PCBIT with AV=20% at an appropriate frequency of AC.Using this method,ion unidirectional ejection efficiency of PCBIT can be significantly improved while maintaining a high mass resolution,which makes the PCBIT more suitable for developing miniaturized mass spectrometer.
6.Induced transformation of rat oval cells line WB-F344 and tumor formation in NOD/SCID mice

Purpose To study the malignant transformation after treating rat oval cell line ( WB-F344 ) with chemical carcinogen N-methyl-N′-nitro-N-nitrosoguanidine ( MNNG) . Methods WB-F344 cells were cultured with MNNG for severe times. The biological characteristics of induced cells were detected through the following methods:to check proliferation activity by flow cytometry analysis, to examine malignant transformation degree of induced cells by soft agar assay and tumor formation in NOD/SCID mice, and to investi-gate the transcriptional and protein levels of hepatocellular carcinoma marker GGT, GST-P by real time-PCR. Results Oval cells in-duced by MNNG showed changes in biological characteristics and malignant molecular markers. Conclusion Hepatic oval cells model is successfully established, which can be confirmed by tumor formation in NOD/SCID mice.
7.Study on Adjuvant Immunoactivity of Polysaccharides from Panax Japonicus

Objective To study the adjuvant immunoactivity of polysaccharides from Panax japonicus by alcohol of different concentrations;To discuss its part with the strongest adjuvant immunoactivity. Methods Polysaccharides from Panax japonicus was sunk with alcohol of different concentrations, and 30%alcohol compound, 60%and 90%alcohol polysaccharide were obtained. Different segments of polysaccharide and OVA protein were injected to mice once a week for three times for immunity. Five days after the last immunity, the mice were executed to collect blood, and the antibody titer was determined. The three parts of alcohol compound were scanned by infrared spectrum to determine the type of polysaccharide preliminarily. Results Compared with the control group, the antibody titer of different segments of polysaccharide obviously increased, especially the polysaccharide sunk by 60%alcohol. Infrared spectrum analysis showed that polysaccharides from Panax japonicus contained pyranose ring structure. Conclusion Polysaccharides from Panax japonicus has significant adjuvant immunoactivity, and polysaccharide sunk by 60%alcohol has the strongest adjuvant effects.

9.Impact of basic diseases on the clinical symptoms and prognosis of cryptococcosis

Objective Cryptococcosis is a potential life-threatening systemic mycosis with a heterogeneous susceptible popu-lation which is classified into three groups according to the current guidelines, including AIDS patients, organ transplantation recipients ( OTR) , and non-HIV-infected and non-transplant hosts ( NHNT) .This study aimed to discuss the influence of basic diseases on the clinical features and prognosis of NHNT cryptococcosis patients. Methods Using a retrospective cohort study design, we retrieved the clinical data about 73 NHNT cryptococcosis patients treated in Jinling Hospital.Based on the presence or absence of immunodefi-ciency or infection-increasing complications, we divided the patients into a basic disease group ( n=35) and a non-basic disease group ( n=38) and analyzed their clinical characteristics, chest radiographic features, laboratory results, and clinical outcomes. Results Compared with the non-basic disease group, the basic disease group showed a significantly higher incidence rate of disseminated disea-ses (62.9%vs 21.1%, P<0.01), more cases of patchy consolidation (47.4%vs 16.7%, P<0.05) and mixed lesion (31.6%vs 3.3%, P<0.05) in chest radiography, and a higher mortality (30.0%vs 5.3%, P=0.016). Conclusion Basic diseases have a great impact on the clinical features and prognosis of NHNT cryptococcosis.NHNT patients with basic diseases are susceptible to dis-seminated diseases with severer clinical symptoms and a higher mortality.
10.Separation of Ephedrine and Pseudoephedrine by Capillary Electrophoresis

Objective To establish a capillary electrophoresis method to separate ephedrine and psedudoephedrine. Methods RM-β-CD and HP-β-CD were set as additives. A capillary electrophoresis method was set up. The effects of types and concentrations of additives, the concentrations and pH values of buffered solution, running voltage and organic solvent on the separation of ephedrine and psedudoephedrine were investigated.Results Ephedrine and pseudoephedrine could be successfully separated by using either RM-β-CD or HP-β-CD as additives. When RM-β-CD was used as additive, the best separation conditions were as follows: separation voltage 10 kV, 25 mmol/L Tris-H3PO4 (pH 2.42), 20 mg/mL of RM-β-CD. Under the conditions, the resolution of ephedrine and pseudoephedrine was 1.56 and they were separated successfully within 13 min. When HP-β-CD was used as additive, the best separation conditions were as follows: separation voltage 10 kV, 25 mmol/L Tris-H3PO4 (pH 3.00), 50 mg/mL of HP-β-CD. Under the conditions, the resolution of ephedrine and pseudoephedrine was 2.73 and they were separated successfully within 15 min.ConclusionThis method is reliable, rapid and repeatable. It can be used as separation determination method for ephedrine and pseudoephedrine.
