1.Expression of estrogen and androgen receptors on aneurysm walls and their effects in patients with intracranial aneurysm

Objective To investigate the significance of androgen and estrogen receptor expression levels on aneurysm walls.Methods From November 2007 to June 2016,32 patients received craniotomy for clipping intracranial aneurysms in the West China Hospital,Sichuan University were enrolled prospectively.Nineteen intracranial aneurysm walls and 26 superficial temporal artery branches were obtained (a total of 45 qualified specimens).Immunohistochemical method was used to detect the superficial temporal artery branches and smooth muscle layer of intracranial aneurysm wall and the expression levels of estrogen receptor-α,β and androgen receptors.Image Pro Plus 6.0 software was used to analyze and detect the integral optical density values of the positive cell expression levels.The χ2 test and rank-sum test were used for statistical analysis.Results The median (M) and interquartile range (P25,P75) of the expression levels of estrogen receptor-α,β of the intracranial aneurysm walls were 3 049 (2 112,5 554) and 4 364 (2 314,5 667) respectively.They were lower than 6 544 (3 507,10 103) and 6 972 (5 694,10 024) of the superficial temporal artery branches.The expression level of androgen receptor of aneurysm wall was 3 299 (1 375,4 895),it was higher than 1 130 (794,1 922) of the superficial temporal artery branches.There was significant difference between the two groups (all P<0.05).Conclusion The decreased expression levels of estrogen receptor-α,β and the increased expression level of androgen receptor in the cerebrovascular walls may promote the progress of intracranial aneurysms,however,the specific mechanism needs further study.
2. Effect of combination usage of TSA and CYA on pancreatic cancer PANC-1 cell apoptosis

OBJECTIVE: To evaluate the apoptosis effect of HDAC inhibitor trichostatin A (TSA) combination with Hedgehog (Hh) signaling inhibitor cyclopamine (CYA) on the PANC-1 cells line. METHODS: Hochest 33258 staining was used to measure apoptosis of PANC-1 cells treated with TSA and CYA respectively or cooperatively. The 24 h IC50 of PANC-1 cells line responding to TSA and CYA were detected by CCK8. The mRNA expression of the gene relative with apoptosis and Hh signaling were evaluated by SYBR GREEN based FQ-PCR (fluorescent quantization polymerase chain reaction). The proteins associated with apoptosis (Bcl-2 and activated caspase-3, activated caspase-8, activated caspase-9) and Hh signalling pathway (Gli1, Smo and Ptc-1) were measured by Western blotting (WB). Gli1 was also detected by cell immunofluorescence. RESULTS: TSA and CYA could induce apoptosis of PANC-1 cells by Hochest 33258 stain, respectively or combinatively. The 24 h IC50 of PANC-1 cells line treated with chemical were (0.51±0.07) μmol·L-1 (TSA) and (33.6±2.3) μmol·L-1 (CYA), respectively; the combination index (CI) of TSA and CYA on the PANC-1 is 0.835. Combination usage of TSA and CYA caused more significantly decreased expression of Bcl-2 mRNA and increased Bax mRNA as compared with the TSA or CYA treatment. TSA (0.5 μmol·L-1) inhibited the expression of Gli1 mRNA, TSA promoted the inhibition effect of CYA on Hh pathway, down-regulation of Gli1, Smo mRNA and up-regulation of Gli3 mRNA. Western blotting showed that combination usage of TSA and CYA induced more significant activation of activated caspase-3, activated caspase-8, activated caspase-9 and reduction of Bcl-2, Gli1, Smo and Ptc-1 expression than TSA or CYA treatment. Decreased Gli 1 also confirmed by immunofluorescence. CONCLUSION: Combination usage of TSA and CYA could induce PANC-1 cells line apoptosis and synergistically inhibit Hh pathway.
3.Surgical management of blood blister-like aneurysms of the internal carotid artery:a 10-year single cen-ter experience

Objective To explore the clinical features, surgical techniques and treatment efficacy of blood blis?ter-like aneurysms (BBA) of the internal carotid artery (ICA). Method We retrospectively reviewed 45 BBAs of the ICA treated surgically at our department between Jan. 2006 and Jan. 2016. The data was analyzed. Results Direct clipping of BBA was performed in 38 patients, clipping over wrapping in 3 patients and trapping of ICA in 4 patients. Intraoperative rupture occurred in 12 patients (27%). Postoperative cerebral infarction developed in 10 cases (22%). Seven patients died (16%). During the follow-up time of 47 months (range, 6~126 months), Good results were achieved in 34 patients (76%) with a modified Rankin Scale (mRS) 0~2 and 4 patients were in mRS 3~5. Follow-up angiography revealed a re?currence of BBA in one patient, which was retreated with endovascular coiling. No rebleeding was found in all fol?lowed-up patients. Conclusions BBAs of the ICA are difficult to treat surgically with high risk and complications. Thoughtful pre-operative evaluation of BBA and application of various surgical strategies during operation can signifi?cantly reduce the surgical risk and achieve good outcomes.
4.Expression and significance of alpha5beta1 integrin in abnormal scars.

OBJECTIVETo study the expression of alpha5beta1 integrin in the abnormal scars and its role and significance in the formation and development of abnormal scars.
METHODSThe expression of alpha5beta1 integrin was observed in hypertrophic scar (15 samples), keloid (15 samples) and normal skin (10 samples) with SP immunohistochemical method and colloidal gold immuno-electron microscopic technique. The data were semi-quantitatively analyzed.
RESULTSThe expression levels of alpha5beta1 integrin in the fibroblasts of keloids and hypertrophic scars were higher than normal skin; the expression of alpha5beta1 integrin in the fibroblasts of keloids was higher than hypertrophic scars (P < 0.01).
CONCLUSIONThe alpha5beta1 integrin appears to have close relation to the formation and development of abnormal scars. To find a way to decrease the expression level of alpha5beta1 integrin in fibroblasts may be a new approach to inhibit scar hypertrophy.

5.Clinical efficacy of Yougui capsules and Wuziyanzong pills on oligoasthenospermia.

OBJECTIVETo investigate the clinical effects of the Chinese traditional medicine Yougui Capsules and Wuziyanzong Pills on sperm viability and motility in patients with oligoasthenospermia.
METHODSA total of 80 infertile men oligoasthenospermia were equally randomized into a trial and a control group, the former treated with Yougui Capsules at 1.68 g tid, while the latter with Wuziyanzong Pills at 6 g bid, both for a course of 12 weeks. The sperm viability and motility of the patients were detected and compared before and after medication.
RESULTSAfter 12 weeks of medication, the sperm viability and percentages of grade a and grade a + b sperm were (65.7 +/- 13.1), (22.5 +/- 9.1) and (47.6 +/- 15.8)% in the trial group, significantly higher than (38.1 +/- 11.1), (13.2 +/- 6.8) and (24.1 +/- 10.9)% in the control (P<0.05). What's more, the above three parameters of the two groups were also significantly higher than those before medication, which were (31.9 +/- 16.9), (8.2% +/- 3.7) and (15.7 +/- 13.9)% in the former and (31.7 +/- 17.0), (7.9 +/- 4.5) and (16.9 +/- 13.6)% in the latter (P<0.05).
CONCLUSIONBoth Yougui Capsules and Wuziyanzong Pills can improve sperm viability and motility in patients with oligoasthenospermia, and the former is even more efficacious than the latter.

Objective To compare the difference on health statistical survey system between China and United States of American.Methods Four aspects in statistical agencies,legislation,survey items and health statistics information were compared.Results The system in China has its own characteristics,but it also faced with many challenges,including the relatively slow transfer and exchange of statistical information,the lack of legislation on data dissemination and sharing,the insufficient diversification of survey items and survey content.Conclusion The statistics information center of Ministry of Health should strengthen the management and coordination to ensure the data collection timely and accurately.The policies should be strengthened to protect data privacy and improve data dissemination.

Objective To observe the effect of berberine on visfatin expression in 3T3-L1 adipocytes,and to explore the mechanism underlying berberine improving insulin resistance.Methods Visfatin mRNA and protein expressions were assayed by RT-PCR and Western blot after incubation of berberine at different concentrations and durations with 3T3-L1 adipocytes.Results In 3T3-L1 adipocytes,visfatin mRNA expression was increased at 0- 10?mol/L berberine with the maximum effect at 10?mol/L,being 4.96 folds of control group.While as the berberine concentrations were further increased above 10?mol/L,the visfatin mRNA level decreased.After incubation with berberine for 3 h,the visfatin mRNA level began to increase,and reached the peak when incubation for 12 h(4.57 fold of basal group).Then the mRNA level decreased,though it was still higher than that in control group after 48h intervention.5,10, 20?moL/L berberine could increase visfatin protein expression by 1.13,2.46,2.34 folds respectively as compared with control group(P

OBJECTIVETo observe the effect of naringin of Drynaria Rhizome, a Chinese medical component of Zhuanggu Jianxi Recipe (ZJR) containing serum on caveolin-p38MAPK signal factors (such as caveolin-1, p-p38, p-ATF-2, IL-1β, and TNF-α) in IL-1β induced rabbit degenerated chondrocytes, and further to explore its mechanism for protecting articular cartilages.
METHODSNaringin of Drynaria Rhizome was obtained and analyzed by HPLC-TOF/MS. Four weeks old New Zealand rabbits were killed and their bilateral knee joints were isolated aseptically. CDs were isolated and then cultured in vitro. The second generation of CDs were used for later experiment. The effect of naringin on CDs proliferation was detected by MTT assay. The effect of naringin on the expression of IL-1β-induced collagen II in CDs was detected by immunohistochemical method. The effect of naringin on caveolin-1, p-p38, and p-ATF-2 protein in IL-1β-induced CDs was detected by Western blot. The effect of naringin on mRNA expression of IL-1β and TNF-α in IL-1β-induced CDs was detected by RT-PCR.
RESULTSThe appearance time of naringin in flow graphs of naringin standard solution and ZJR containing serum was 23.5 min, and the molecular weight ranged between 581.0 and 581.5 m/z. Naringin could promote the proliferation of CDs, and inhibit the effect of IL-1β on collagen II in CDs. Compared with the model group, naringin could reduce the expression of caveolin-1, p-p38, p-ATF-2, IL-1β, and TNF-α in IL-1β induced CDs (P < 0.05), which was approximate to the level of the normal group.
CONCLUSIONSNaringin could not only promote the proliferation of CDs, but also protect IL-1β-induced CDs. Its mechanism might be associated with decreasing the expression of caveolin-1, p-p38, and p-ATF-2 proteins, inhibiting caveolin-p38MAPK signal pathway, and further reducing mRNA expression of IL-1β and TNF-α in the downstream of caveolin-p38MAPK signal pathway.

9.Brain arousal dysfunction in severe craniocerebral injury treated with acupuncture.

OBJECTIVETo explore the early rehabilitation effect of acupuncture on brain arousal in severe craniocerebral injury.
METHODSOne hundred and two cases of severe craniocerebral injury were randomly divided into an observation group and a control group, 51 cases in each one. Based on the conventional nursing care in neurological external medicine, in observation group, acupuncture was applied at Shuigou (GV 26), Neiguan (PC 6) and Sanyinjiao (SP 6) mainly. In control group, functional electric stimulation was applied at stimulate the affected muscles of the upper limbs. Thirty days later, the lucid rate from coma, lucid interval and clinical efficacy were compared between two groups. RESULTS; The lucid rate from coma was 82.4% (42/51) in observation group, which was higher than 56.9% (29/51) in control group (P < 0.01). The lucid interval in observation group was shortened remarkably as compared with control group (P < 0.01), and the clinical efficacy was superior apparently to that in control group (P < 0.01).
CONCLUSIONOn the basis of conventional treatment, acupuncture intervention at early stage can accelerate the recovery of brain arousal function in patients with severe craniocerebral injury.

10.Effect of polymorphisms in the β2-adrenergic receptor on the susceptibility and pulmonary function of patients with chronic obstructive pulmonary disease: a meta analysis.

BACKGROUNDChronic obstructive pulmonary disease (COPD) is a heterogeneous and complex disease of which the pathogenesis remains largely unknown. Many factors could influence COPD development and progression. One of them is the genetic risk factor. A severe hereditary deficiency of alpha-1 antitrypsin is the best genetic proof. Four single nucleotide polymorphisms (SNPs) of beta2-adrenergic receptor (β(2)AR) result in single amino acid substitution. Two loci had been extensively studied and found that they could change the function of β(2)AR. Two SNPs consist of substitutions of glycine for arginine at amino acid position 16, glutamic acid for glutamine at position 27. Many studies proved that polymorphisms at position 16 and 27 altered the lung function of COPD patients or the patient's susceptibility to the development of COPD. However, there was no exclusive conclusion. Therefore, a meta analysis was done to investigate the effect of polymorphisms in the β2-adrenergic receptor (ADRB2) gene on the risk of COPD and lung function.
METHODSComprehensive searches of MEDLINE, Embase, Ovid, HighWire, Cochrane Library, and Chinese databases (CBMdisc, VIP, CNKI, and Wanfang data) from January 1980 to September 2011 were performed, using the keywords: COPD OR chronic obstructive pulmonary disease AND adrenoreceptor OR adrenergic receptor AND polymorphism OR mutation OR variation. Case-control research or cross sectional studies in which diagnosis of COPD met the Global Initiative for Chronic Obstructive Lung Disease (GOLD) guidelines; all the studies reported the ADRB2 genotype at position 16 or 27. Outcomes measured were genotype frequency and forced expiratory volume in the first second (FEV(1)%) in both the case and control.
RESULTSTwelve case-control studies and eight cross-sectional studies were included. Compared to the control (n = 1225), neither Gly/Gly (n = 527) nor Arg/Arg (n = 422) homozygotes at position 16 demonstrated increased susceptibility to COPD, with odds ratios (ORs) of 0.95 (95%CI (0.68, 1.31), z = 0.33, P = 0.740) and 0.82 (95%CI (0.52, 1.28), z = 0.88, P = 0.381), respectively. Similar results were obtained for position 27, with ORs of 0.97 (95%CI (0.77, 1.23), z = 0.21, P = 0.833) for Glu/Glu homozygotes (n = 357) and 0.82 (95%CI (0.53, 1.29), z = 0.85, P = 0.393) for Gln/Gln homozygotes (n = 704) (control = 1183). In patients with COPD, Arg/Arg homozygotes (n = 41) had a similar FEV1% compared with Gly/Gly homozygotes (n = 102) (standardized mean difference (SMD) = 0.88, 95%CI (-0.85, 2.62), z = 1.00, P = 0.319). The genotype distribution was different between Caucasian and Asian populations (all P < 0.05 except the genotype Arg/Gly) for both position 16 and 27.
CONCLUSIONSPolymorphisms of ADRB2 at positions 16 and 27 did not change the risk of COPD nor affect lung function or disease severity. The genotype distribution for these polymorphisms was different between Caucasian and Asian populations.
