2.Neovascularization profile in rat allogenic penetrating keratoplasty

AIM: To observe the neovascularization process with no intervention in rat allogenic penetrating keratoplasty. METHODS: Allogenic penetrating keratoplasties were successfully performed in 34 female SPF SD rats with no intervention after operations. Corneal neovascularization(CNV) process was noted on day 4, 7, 15 and 30 with operating microscope. The vascular area surface was calculated using the formula C/12×3.14×[r2-(r-I)2].RESULTS: CNV was noted in 29 out of 34 rats (85%). Firstly, the new vessels distributed around the cornea like a brush then gradually extended towards the center. The vessels were distorted and massive with branched tails, they continued growing to reticulated veins in peak time then gradually atrophied. The average neovascularization area (SE) on day 4, 7, 15 and 30 was 11.8±3.5mm2, 18.5±4.0mm2,14.4±4.3mm2 and 6.0±1.8mm2 respectively and 12.7±1.9mm2 in total. The average percentage that new vessels accounting the whole cornea area(SE) was 30.8%±8.7%, 65.3%±12.8%, 59.4%±14.5%,36.2%±10.9% and 48.7%±6.4% in total.CONCLUSION: In rat allogenic penetrating keratoplasties without intervention, CNV presented on day 4 and reached the maximum area on day 7. Then the vessels gradually atrophied, about 50% of the maximum area still remained on day 30.
3.The relationship between circadian clock gene Per2 and VEGF, cyclin D1 expression in esophageal cancer

Objective To explore the expression of circadian clock gene Per2 in the tissue of esophageal cancer and the relevance between VEGF,cyclin D1.Methods 80 cases with esophageal cancer who received surgical treatment and pathological specimens preserved were selected,and the normal tissues adjacent to carcinoma were used as control.The expression of Per2,VEGF and cyclin D1 were detected by immunohistochemistry,and the data were analyzed.Results Compared the tissue adjacent to carcinoma with carcinoma tissue,Per2,VEGF and cyclin D1 expression had statistically significant differences(x2 =26.86,24.34,23.72,all P <0.01).Per2 expression in different differentiation degree and the TMN stages had statistically significant differences (x2 =6.45,7.77,all P < 0.05) ;cyclin D1 expression in lymph node metastasis and TMN stages had statistically significant differences(x2 =12.99,8.99,all P < 0.01) ;VEGF expression in lymph node metastasis and TMN stages had statistically significant differences(x2 =6.68,6.14,all P < 0.01).Per2 expression was negatively correlated with VEGF,cyclin D1 expression (r =-0.644,-0.523,all P < 0.01).Conclusion Clock gene Per2 and VEGF,cyclin D1 expression in esophageal cancer has certain correlation,it can be a prognostic indicator of esophageal cancer.
4.The roles of C-reactive protein and pleural effusion in prognosis evaluation in the early stage of acute pancreatitis

ObjectiveTo investigate the values of C-reactive protein (CRP) and pleural effusion in predicting the severity in the early stage of severe acute pancreatitis (SAP) . Methods A total of 89patients with acute pancreatitis were collected from October 2008 through October 2010 for retrospective analysis. Patients were divided into two groups, namely mild acute pancreatitis (MAP) group and SAP group as per the Guidelines for Clinical Diagnosis and Classification of Acute Pancreatitis set by the Society of Chinese Medical Association in 2003. The levels of CRP were measured on the 1st, 2nd, 3rd and 7th days after admission. Pleural effusion was also observed on the 1 st day after admission. The data of two groups were analyzed and compared. ResultsThere were significant differences in CRP at all intervals between SAP group and MAP group (P <0.05) . The relative risk of increase in CRP ( > 150 mg/L),pleural effusion and increase in CRP along with pleural effusion were analyzed, and each of these three markers can be used as an independent severity factor of SAP. Particularly, increase in CRP along with pleural effusion could be most sensitive in predicting the severity of SAP with relative risk (RR) to be 4. 8 and specificity of predictive value to be 100%. ConclusionsC-reactive protein and pleural effusion are available, simple and economic biomarkers which can help us predict the risk of acute pancreatitis in the early stage.

6.A STUDY OF THE MECHANISM OF MORPHOLOGICAL CHANGES IN AXONS AFTER SEVERE BRAIN INJURY

To explore the molecular pathological mechanism of severe brain injury, the brain diffuse axon injury (DAI) model and Mamarou free drop model were produced in rats. Sagittal sections of the brain were processed by immunohistochemical ABC method using the mouse serum against NF68 subunit and HSP70. The medulla oblongata was observed under the microscope and electron microscope. Left parietal lobe of the free drop model was examined with HE and HSP70 immunohistochemistry. At 30 min post injury, the axons in medulla oblongata were seen to be crooked, swollen,and deranged. The myelin sheath became slightly separated, and the NFs in axoplasma were abnormal . At 2~24 h post injury,obvious axonal swelling, disconnection and formation of axonal retraction balls were seen. Obvious separation of myelin sheaths, local disconnection, vacuolization,peripheral aggregation of mitochondria and partial dissolution of axoplasma were seen. The NF68 positive axons increased gradually in staining intensity. HSP 70 positive cells of the two groups were detected at 3h after brain injury, reached the peak at 24h, and decreased at 72h. The HSP expression of the two groups were in accord. The research indicated that DAI could lead to a derangement in structure of NFs. Ischemia and anoxia may aggravate the brain injury.
7.Alzheimer-like pathologic changes in hippocampus of middle aged and old aged peole without dementia

Objective To study the relationship among Alzheimer-like neuropathologic changes and their relationship with age and sex. Methods From January 1,1986 to April 1,2000,all autopsy caus of non-demented individuals ≥40 years of age in the General Hospital of PLA were examined for Alzheimer-like neuropathologic changes in hippocampus.There were 41 female and 97 male. Senile plaques(SP),neurofibrillary tangles(NFT) were identified with modified Palmgren-stain method. Hirano bodies(HB) or granulovacuolar degeneration(GVD) were identified with HE stain methods. Results About 40% patients had Alzheimer-like neuropathological changes in hippocampus. There was a highly significant correlation was found between the incidence and severity of SP?NFT?Hirano or GVD and patients' age in the CA1 field of the hippocampus, but no significant difference between male and female. There was a strong correlation among the prevalence and numerical densities of these four types of Alzheimer-like changes. Conclusions Alzheimer-like neuropathologic changes were mainly distributed in CA1 of hippocampus. In CA1 they had relationship with age, but no relationship with sex and they had correlation each other.

Cell proliferation was determined using the MTT assay, and apoptotic cells was determined using the Hoechst 33258 staining.ResultsHPV-18-infected HeLa cells were treated with 10, 20 μg/mlof the alcohol extract ofEuphorbiae ebracteolatae Radixfor 12 h, the cell proliferation inhibition rate were significantly increased compared to the control group (16.36% ± 5.06%, 18.59% ± 5.99%; allP<0.01);HPV-18-infected HeLa cells were treated with 0.625, 1.25, 2.5, 5, 10, 20 μg/ml of the alcohol extract ofEuphorbiae ebracteolatae Radixfor 72 h, the cell proliferation inhibition rate were significantly increased compared to the control group (13.82% ± 3.66%, 30.24% ± 6.58%, 43.29% ± 4.29%, 53.32% ± 1.41%, 54.24% ± 1.97%, 50.68% ±1.74%;allP<0.01). The Hoechst 33258 staining showed that HPV-18-infected HeLa cells were treated with 10 μg/ml of the alcohol extract ofEuphorbiae ebracteolatae Radixfor 24 h, the apoptotic cells were found.ConclusionEuphorbiae ebracteolatae Radixcould inhibit the proliferation and induce apoptosis in HPV-18-infected HeLa cells.
9.The therapeutic effect of Pingyangmycin lipiodol emulsion embolization for hepatic cavernous hemangioma

Objective To evaluate the therapeutic effect of Pingyangmycin lipiodol emulsion (PLE) embolization for cavernous hemangioma of liver (CHL). Methods Seldinger technique was adopted and the catheter was super-selectively sent into the supplying artery of tumors in 18 CHL patients, 5-10 ml of Lidocaine and 5-20 ml of PLE were slowly injected. Before the embolization, 50-100 mg of pethidine was routinely injected. The embolization was conducted in several times in patients with large tumors, with multiple supplying arteries, and whose age was above 60 years. During the 3-48 months'follow-up after the operation,the change of the tumor diameter before and after the embolization was compared by using CT and ultrasound. The clinical symptomatic relief and the complications were also observed. Results The CHL of all the 18 cases were filled up by PLE. The foci were completely vanished in 9 cases, reduced by over 50% in 7 cases,and reduced by 25%-50% in 2 cases,respectively. The follow-up angiographic examination was performed in six cases, the tumor staining was completely vanished and the tumor-supplying artery was closed up. The improving rate of the clinical symtoms reached 89% . There were no serious complications. Conclusion The PLE embolization in hepatic cavernous hemangioma proves to be reliable, safe, minimally invasive, and with little side effect. This therapy, therefore, can be the first method of choice in treating the cavernous hemangioma of the liver.

Based on international framework of rehabilitation, this paper summarized the theories and practice of rehabilitation in Shenzhen. An inclusive rehabilitation model had been established in Shenzhen in light of international concepts of rehabilitation, in which the functions of medical rehabilitation, education, employment, assistive technology, care, right protection, accessible environment and culture had been integrated. The comprehensive services had been provided through this model and the target of inclusive development had been realized.
