2.Audiologic Comprehensive Evaluation of Sensorineural Deafness in Children

Objective To study the clinical application of the complete audiologic evaluation battery consisting of ABR,ASSR and behavioral audiometry in sound field for the identification of sensorineural losses in children.Methods In the study 48 children were divided as group PTA of the children assessed by pure tone audiometry and ABR and ASSR test,and group BA of the children unable to perform on behavioral evaluations.Results The analysis of the audiometric results of group PTA showed significant correlation between the ASSR thresholds and pure tone thresholds(P<0.01) at various frequencies.The correlation coefficients were 0.75,0.76,0.76,and 0.83 at different frequencies.The ASSR-PTA linear regression was utilized.The comparison of the ABR and ASSR results of group BA indicated that 23 ears responded in ASSR but without responses in ABR.Those without ASSR responses also had no ABR There were 116 tests in total four frequencies for the 29 ears.83 responses were obtained from ASSR,89 responses from behavior audiometry and 96 from the use of the both tests.Conclusion The comprehensive audiologic evaluation battery could be used to test younger children with severe heating loss children as compared to the pure tone audiometry alone.More detailed data can be obtained thus from children to assist in rehabilitation in a more effective mariner.
3.Correlation study on serum homocysteine,folic acid,vitamin B12,ferritin levels And cerebral infarction

Objective To investigate the correlation between serum homocysteine(Hcy) ,folic Acid ,vitamin B12 ,ferritin levels and cerebral infarction ,and provide a new way for prevention and treatment of cerebral infarction .Methods Forty -eight hospital‐ized patients with ischemic cerebrovascular disease who were in the neurology ward department of the hospital from April 2012 to November 2014 were enrolled in the study as cerebral infarction group .Meanwhile ,30 healthy elderly patients were recruited ran‐domly as a healthy control group during the same period .There was no significant difference in sex and age between the two groups .People with other diseases were excluded .Hospitalized patients were diagnosed within 24 -72 hours and received CT and MRI scan and other related auxiliary examination after being admitted to hospital .All these patients underwent plasma Hcy ,folic acid ,vitamin B12 ,ferritin level tests which were determined by using chemiluminescence and radioimmunoassay .SPSS17 .0 statistical software was used for statistical analysis ,and the correlation between Hcy ,folic acid ,vitamin B12 ,ferritin and cerebral infarction were analyzed .Results The plasma Hcy and ferritin levels in cerebral infarction group were higher than those in control group ,and the difference is significant(P<0 .05) .Meanwhile ,folic acid ,vitamin B12 levels in cerebral infarction group were lower than those in control group and the difference was significant(P<0 .05) .There were significant differences in levels of Hcy ,folic acid ,vitamin B12 ,ferritin between groups of different cerebral infarction levels .Conclusion Detections of serum Hcy ,folic acid ,vitamin B12 and ferritin could be of important clinical value in cerebral infarction .
4.Antibacterial components from artificially induced dragon's blood of Dracaena cambodiana.

Ten compounds were isolated from the artificially induced dragon's blood of Dracaena cambodiana by various column chromatographies on silica and sephadex LH-20 gel. Based on spectral analysis of NMR and MS, their structures were identified as 3, 4-dihydroxyallylbenzene (1), 3', 4', 5'-trimethoxycinnamylalcohol (2), pinoresinol (3), (2R)-7, 4'-dihydroxy-8-methylflavane (4), (2R)-7,4'-dihydroxy-5-methoxy-8-methylflavane(5),(2S)-7,3'-dihydroxy-4'-methoxy-8-methylflavane(6) ,(2S)-4',7-dihydroxy-6, 8-dimethylflavane(7), 4,2',4'-trihydroxychalcone(8), 4,4'-dihydroxy-2-methoxydihydrochalcon(9) and Cambodianin E (10). Antibacterial activity assay showed that compounds 1, 4 and 10 have inhibitory effect on Fusarium oxysporum f. sp. cuben, Fusarium oxysporum f. sp. niveum, Fusarium oxysporum f. sp. vasinfectum and Ralstonia solanacearum.

5.Effect of early restrictive fluid resuscitation on inflammatory and immune factors in patients with severe pelvic fracture

Purpose:To study the effect of early restrictive fluid resuscitation (EFR) on inflammatory and immune factors in patients with severe pelvic fracture (SPF).Methods:A total of 174 SPF patients in the Department of Orthopaedics,the First Affiliated Hospital of Chengdu Medical College from July 2015 to June 2018 were involved in this study and divided into EFR group (n =87) and control group (n =87) using the random number table method.Conventional fluid resuscitation (CFR) was performed in control group,and EFR was performed in EFR group.The incidences of acute respiratory distress syndrome (ARDS) and multiple organ dysfunction syndrome (MODS) during rescue,successful rescue rate,blood transfusion volume,fluid input,and resuscitation time were compared between the two groups.The parameters including prothrombin time (PT),hematocrit (HCT),platelet (PLT) and blood lactate (BL) at the 4th hour after fluid resuscitation were recorded.The levels of inflammatory factors (TNF-α,IL-6,CRP) and immune factors (CD3+,CD4+,CD8+,CD4+/CD8+) were compared between the two groups before treatment and 7 days after treatment.The revised acute physiologic and chronic health evaluation system and the sequential organ failure assessment scores were adopted for evaluation before treatment and 7 days after treatment.Results:The incidences of ARDS and MODS during rescue in EFR group were significantly lower than those in control group (p=0.015 and 0.010 respectively),and the successful rescue rate in EFR group was significantly higher than that in control group (p =0.011).The blood transfusion volume,fluid input,resuscitation time in EFR group were significantly lower than those in control group (p =0.016,0.002 and 0.001 respectively).At the 4th hour after fluid resuscitation,PTand BL in EFR group were significantly lower than those in control group (p =0.021 and 0.003 respectively),while HCT and PLT in EFR group were significantly higher than those in control group (p =0.016 and 0.021 respectively).On day 7 after treatment,TNF-α,IL-6,CRP and CD8+ in EFR group were significantly lower than those in control group (p =0.003,0.004,0.007 and 0.003 respectively),while CD3+,CD4+ and CD4+/CD8+ in EFR group were significantly higher than those in control group (p =0.004,0.000,0.007 respectively).On day 7 after treatment,the revised acute physiologic and chronic health evaluation (APACHE) system and the sequential organ failure assessment (SOFA) scores in EFR group were significantly lower than those in control group.Conclusion:EFR can effectively eliminate inflammatory factors,improve immune function,maintain the stability of blood components,reduce the incidences of ARDS and MODS,and elevate the successful rescue rate in patients with SPF.
6.Construction and identification of helper-dependent adenoviral vector encoding hemagglutinin protein of swin-origin influenza virus A H1N1.

Abstract: In order to investigate immune protection against swine-origin influenza virus (S-OIV) A H1N1, the helper-dependent adenovirus vector (HDAd) system was exploited to construct recombinant HDAd encoding hemagglutinin (HA). The HA gene was synthesized and cloned to the HDAd backbone. Then, the HDAd/HA DNA molecules were transfected into 293Cre4 cells with calcium phosphate. The cells were infected by helper virus 16 hours after the transfection. The 293Cre4 cells were coinfected with HDAd/HA and the helper virus for large-scale preparation of HDAd/HA. The HDAd/HA was obtained and purified twice with CsCI density ultracentrifugation and observed morphologically under transmission electron microscope, and the expression of HA protein was analyzed with RTPCR. Recombinant HDAd/HA expressing HA protein was successfully constructed which could pave the way for in vivo investigation on immunogenicity and efficacy against S-OIV A H1N1 infection.

7.Aeromonas hydrophila bloodstream infection in patients with hematologic diseases: a report of 4 cases

Abstract: To report the diagnosis, treatment and outcome of 4 patients with hematological diseases complicated with Aeromonas hydrophila bloodstream infection in the Second Affiliated Hospital of Kunming Medical University, further clarify the importance of blood culture and deepen the clinical understanding of the disease. Four patients with hematological diseases complicated with Aeromonas hydrophila bloodstream infection treated in the Second Affiliated Hospital of Kunming Medical University from 2017 to 2021 were recruited as the study objects. The clinical manifestations, blood culture collection, detection time of Aeromonas hydrophila, laboratory examination, treatment and prognosis of the patients were retrospectively analyzed. In this study, 4 cases were male patients with hematological diseases, who were in myelosuppression after chemotherapy. After fever, blood culture was collected and Aeromonas hydrophila was detected. The positive time of blood culture in 4 cases ranged from 4 to 11 hours. The results of antibiotic sensitivity showed that it was highly sensitive to the second, third and fourth generation cephalosporins, quinolones and carbapenems. Four patients were treated with imipenem cilastatin sodium in the early stage, and one patient recovered after active anti infection and leukocyte raising treatment. One patient did not complete chemotherapy due to a request for discharged, and the follow-up was unknown. Two patients developed rapidly into necrotizing fasciitis and died later. Hematological diseases complicated with Aeromonas hydrophila bloodstream infection are rare, but the mortality rate is high. For patients with repeated fever and considering infection, blood culture should be carried out as soon as possible to confirm the pathogen and drug sensitivity test. During clinical treatment, the treatment should be adjusted in time in combination with the patient's situation. In addition to anti-infection treatment, the patient's immunity should be improved and the development of necrotizing fasciitis should be vigilant.

9.Effects of the Culture Method on the Construction of Dermal Substitutes in vitro

Culture environment is the key factor in the construction of dermal skin.It was investigated that the effects of the culture methods,including the static culture and spinner flask culture,and stir speeds on the cells proliferation,metabolism and distribution within collagenchitosan sponges.A higher cell density and specific growth rate was obtained with spinner flask culture versus static culture,especially,the 80 r/min spinner flask culture.The cell distribution in dermal substitutes from stirred culture system was more uniform than static culture,as well as that with increase of stir speeds in spinner flask.In summary,the spinner flasks culture with proper stir speed shows promise for the construction of dermal substitutes in vitro.
10.Anti-inflammatory Effect of Xinfeng Capsule on Adjuvant Arthritis Model Rats

OBJECTIVE:To study the anti-inflammatory effect of Xinfeng capsule on adjuvant arthritis (AA) in rats. METH-ODS:70 SD rats were randomized into a normal control group(isovolumetric normal saline),a model control group(isovolumet-ric normal saline),a positive control TCM group [Qufeng zhitong capsule 0.4 g(crude drug)/kg],a positive control chemical medi-cine group(Leflunomide tablet 2.1 mg/kg)and groups of low,middle and high doses of Xinfeng capsule,which were respectively marked as groups A,B,C,D,E,F and G. All groups of rats except for Group A were given freund’s complete adjuvant intracu-taneous injection to establishe AA models,and from the 12th day after the inflammation was induced,were given the correspond-ing drug,ig,for 28 consecutive days. The degree of toe swelling and arthritis index were determined and calculated for all groups of rats before and after the administration. The contents of interleukin 1β(IL-1β) and tumor necrosis factor α(TNF-α) in serum and synovial membrane of all groups of rats were determined 24 h after the last administration,and pathomorphological changes of their ankles were observed by light microscope. RESULTS:Compared to group A,group B demonstrated higher degree of toe swelling,arthritis index and contents of IL-1β and TNF-α in serum and synovial membrane,with statistical significance(P<0.01). After the administration,the degree of toe swelling and arthritis index of groups C,D,E,F and G reduced,with statistical signifi-cance(P<0.05). Compared to group B,groups C,D,E,F and G showed lower degree of toe swelling,arthritis index and con-tents of IL-1β and TNF-α in serum and synovial membrane,with statistical significance(P<0.01 or P<0.05). Synovial membrane hyperplasia and inflammatory cell infiltration were noted in the ankle tissue of the rats in group B,and such symptoms were not so serious in groups C,D,F and G. CONCLUSIONS:Xinfeng capsule has anti-inflammatory effect for AA rats by a mechanism which may be related to accelerating synoviocyte apoptosis and inhibiting synovial membrane hyperplasia.
