1.Role of β-adrenergic receptor in the occurrence and development of tumor
International Journal of Surgery 2012;39(7):475-478
The β-adrenergic receptor has close relationship with the occurrence and development of tumor. The β-adrenergic receptors were expressed in a variety of tumors. Its activation can stimulate tumor growth by promoting cell proliferation, inhibite apoptosis, promote angiogenesis, and enhance tumor invasion and metastasis. The β-adrenergic receptor blockers can suppress the tumor growth, which may become one of the effective methods of cancer prevention and treatment.
2.Development of six sigma concurrent parameter and tolerance design method based on response surface methodology
Journal of Pharmaceutical Analysis 2008;20(3):198-201,212
Using Response Surface Methodology (RSM), an optimizing model of concurrent parameter and tolerance design is proposed where response mean equals its target in the target being best. The optimizing function of the model is the sum of quality loss and tolerance cost subjecting to the variance confidence region of which six sigma capability can be assured. An example is illustrated in order to compare the differences between the developed model and the parameter design with minimum variance. The results show that the proposed method not only achieves robustness, but also greatly reduces cast. The objectives of high quality and low cost of product and process can be achieved simultaneously by the application of six sigma concurrent parameter and tolerance design.

A low-cost, simple, sensitive detection method of lactate dehydrogense ( LDH) was developed on paper-based microwell arrays microfluidic device. The phenazine methyl sulfate/nitrotetrazolium blue chloride ( PMS/NBT) detection system was used for LDH detection and the colorimetric results were recorded by both Gel Documentation System and a common camera. Under the optimized conditions, the colorimetric intensity showed a linear correlation to the activity of LDH in the range of 10 to 150 U/L with a limit of detection (LOD) of 9. 44 U/L (3σ) by Gel Documentation System;and the linear range was 15-150 U/L by camera with a LOD of 12. 36 U/L (3σ). Foremost, it was found that human serum albumin (HSA) had an effect on the colorimetric enhancement in this detection system. This low-cost, portable paper-based analytical platform could be suitable for the application in the point-of-care with high sensitivity and reproducibility.

This study is to investigate the inhibitory effect of lidamycin (LDM) and its combination with methotrexate (MTX) on lung metastasis of fibrosarcoma by bioluminescence imaging in athymic mice. A stable luciferase transfected HT-1080 cell line was constructed and the capability to establish experimental lung metastasis in athymic mice was confirmed. The optical imaging system was applied to evaluate the formation of lung metastasis in vivo. In addition, metastatic nodules were counted for the evaluation of inhibition rates. As shown, the fluorescent intensity of luciferase-transfected HT-1080 cells was colinear with the cell population and the minimal detected cell population was 100 cells/well. Optical imaging showed that the fluorescent intensity of treated group was apparently lower than that of the control. The inhibition rates of lung metastasis by LDM alone at 0.025 mg x kg(-1) and 0.05 mg x kg(-1) were 53.9% and 75.9%, respectively, while that of MTX alone at 0.5 mg x kg(-1) was 70.2%. The combination of LDM at 0.025 mg x kg(-1) and MTX at 0.5 mg x kg(-1) showed an inhibition rate of 88.7%. The coefficient of drug interaction (CDI) was 0.82. The results herein demonstrated that LDM alone had strong anti-metastasis effect on human fibrosarcoma HT-1080 and the inhibition efficacy is strengthened when combined with MTX.

Objective To investigate the influence of different timing of surgery on patients with poor-grade intracranial aneurysm. Methods The clinical data of 86 patients with aneurysmal subarachnoid hemorrhage (Hunt-Hess gradeⅣ toⅤ)were analyzed retrospectively. The patients were divided into an ultra-early treatment group (the operative time within 24 h after onset;n=40),an early treatment group, (24 to 72 h after onset;n=27),and middle and late treatment group (>72 h after onset;n=19)according to the different timing of surgery. The prognosis of patients at 6 months after procedure was evaluated with the modified Rankin Scale (mRS)scores. The differences of the rate of good prognosis (mRS 0 to 2)and mortality in patients of the 3 groups were compared. Results (1 )The ratios of good prognosis in the ultra-early treatment group, early treatment group, and middle and late treatment group were 55. 0%(n=22),33. 3%(n=9),and 21. 1%(n=4),respectively. There were significant differences (P<0. 05). The mortality rate of the ultra-early treatment group (7. 5%)was lower than that of the early treatment group (25. 9%) or middle and late treatment group (42. 1%). There were significant differences (P<0. 05). (2)There were significant differences in the rate of good prognosis in different timing of surgery in patients with gradeⅣ(P<0. 05);there was no significant difference in the rate of good prognosis in patients with grade Ⅴ(P>0. 05). The mortality ratios in patients of the 3 groups were 2/18,4/11,and 6/11,respectively. There were significant differences (P<0. 05). Conclusion Ultra-early operation may benefit part of the patients with poor-grade aneurysm, particularly in patients with gradeⅣ. As for the patients with grade Ⅴ, ultra-early operation may help to reduce short-term mortality rate, however, the rate of vegetative state is high.
