1.Clinical studies of immunofunction and protein metabolism by combined supplementation of glutamine and recombinant human growth hormone in postoperative patients with cardiogenic cachexia of rheumatic heart disease

Objective To study the effects of recombinant human growth hormone(rhGH) on the treatment for valve replacement postoperative in rheumatic heart disease associated cardiac eaehex.Methods Fortytwo patients with rheumatic heart disease associated cardiac eachexia were divided into two groups.Group one(n=20,rhGH group) received standard enteral nutrition (15 kal·kg-1 · d-1)with rhGH 10U injection subeutaneously from postoperation day 7 to day 14 and group two(n = 22,eontrol group) received standard enteral nutrition (15 kal·kg-1· d-1) for the same period.Haemoglobin, serum total protein, serum albumin, blood glucose, handgrip exercise and triceps skin_fold thickness were determined.Meehanieal ventilation, hypostatie pneumonia incidence rate, and length of stay were observed.Results The levels of serum total protein, serum albumin and blood glucose eoneentration in the rhGH group at the 14th day were inereased significantly compared to that in the control group(P <0.01).Haemoglobin, triceps skinfold thiekness and handgrip exercise in rhGH group were significantly different from those in the control group(P <0.05).Postoperative meehanieal ventilation time, intensive care unit time, hospital stay time were signifieantly shorter than those in the eontrol group (P < 0.05), and hypostatie pneumonia was significantly lower than that in the eontroi group(P < 0.01).Conclusions The rhGH can obviously improve anabolie effects of patients with rheumatic heart disease associated cardiac eachexia whieh can reduce hypostatie pneumonia and shorten postoperative hospital stay time.
2.Effect of potassium magnesium aspartate on arrhythmia and heart function in valve replacement postoperative patients of rheumatic heart disease

Objective To evaluate the curative effect of potassium magnesium aspartate on arrhythmia and heart function in valve replacement postoperative patients of rheumatic heart disease. Methods Two hundred and eighty patients with rheumatic heart disease were divided into 2 groups in random,with no statistical significance. Treatment group (n=155) were received i.v. potassium magnesium aspartate 40 ml every day and control group (n=125) were only given conventional therapy. 3.Efficacy of hysteroscopic guiding endometrial resection on dysfunctional uterine bleeding and its influence factors

Objective To investigate the efficacy and influence factors of hysteroscopic guiding endometrial resection on dysfunctional uterine bleeding.Methods One hundred and eighty women with dysfunctional uterine bleeding were selected as our subjects in Maternal and Child Health Hospital of Qinhuangdao from Jan.2009 to Jan.2011.One hundred and fifty which meet with operation indications underwent hysteroscopic guiding endometrial resection.Operation periods,blood loss,adverse effects rate were recorded.Serum follicle stimulating hormone (FSH),luteinizing hormone (LH),estradiol (E2) and progesterone (P) hormone levels were detected at preoperative and postoperative.The efficacy was recorded at 6 and 12 months after the procedure.Results The average operation time and amount of bleeding were (31.2 ± 11.0)min and (32.3 ± 10.5) ml,and no blood transfusion performed during the operation.All cases got the succeed operation.There were no significant difference in terms of serum FSH,LH,E2 and P concentrations before and after treatment(P >0.05).At 6 months after treatment,improvement rate was 64.0% (96/150),and effective rate was 32.0% (48/150).The total effective rate was 96.0% (144/150).At 12 months after treatment,the improvement rate was 58.0% (87/150),effective rate was 36.0% (54/150).The total effective rate was 94.0%(141/150).There was significant different in terms of total effective rate between 6 months and 12 months after treatment(x2 =113.6,P > 0.05).Postoperative complications occurred in 2 cases,including 1 cases of abdominal pain,and 1 cases of dysmenorrhea.Uterine cavity depth,age and disease duration were the main factors affect the clinical efficacy.(OR(95 % CI):-1.14 (-1.17 to-1.07),1.60 (1.31 to 2.08),-1.02(-1.14 to-1.08),P < 0.05).Conclusion Treatment of hysteroscopic guiding endometrial resection on dysfunctional uterine bleeding is proved to be efficacy due to the less adverse reaction.
4.Research Progress in molecular mechanisms between RSUME and invasive pituitary adenoma

Pituitary adenoma, a benign intracranial tumor, is the third most common brain tumor, second only to glioma and me-ningioma. Pituitary adenoma has been defined as a benign tumor, but some pituitary adenomas can invade the surrounding tissue. This tumor is difficult to resect and can easily recur after surgery. RWD-containing sumoylation enhancer (RSUME) can stabilize the activity of hypoxia-inducible factor-1α and inhibitor kappaB by the small ubiquitin-related modifiers. This phenomenon indicates the impor-tance of RSUME in pituitary adenoma because it promotes the invasiveness of the tumor. However, the correlation between RSUME and the invasion of pituitary adenoma remains unclear. In this study, the roles of RSUME on HIF-1α/VEGF signaling pathway and IκB/NF-κB compomers in the invasiveness of pituitary adenoma were reviewed.

Objective To study the effect of in utero exposure of TCDD on synthesis of EGF and EGFR in submandibular gland of rat offspring. Methods SD rats were treated with TCDD 5??g/kg on Day 15 of gestation, the expression of EGF and EGFR in submandibular gland of rat offspring were analyzed by immunohistochemistry method. Results On PND32 and PND49, the expression of EGF and EGFR in TCDD experiment group were more than that of control group, the difference was statistically significant.Conclusion In utero exposure of TCDD can promote the synthesis of EGF and EGFR in submandibular gland of rat offspring from prepuberty stage through puberty stage. 6.Inhibition of LPS-induced expression of myeloid differentiation factor 88 by fenoterol is associated with its anti-inflammatory effect

Objective To explore the molecular mechanism of inhibition of LPS-induced inflammation by fenoterol, a β2 adrenoceptor agonist in monocyte. Methods Concentrations of interleukin 1β (IL-1β), tumor necrosis factor α (TNF-α) and MCP-1 from cell supernatants from THP-1 cells and wild type or MyD88- / - mice peritoneal macrophages stimulated by LPS in the presence or absence of fenoterol were determined by use of an ELISA system. Expression of MyD88 (myeloid differentiation factor 88) stimulated by LPS in the presence or absence of fenoterol were determined by Western blot. Results Fenoterol inhibited LPS-induced activation of MyD88 and secretion of inflammatory cytokines (TNF-α, MCP-1, and IL-1β). The reaction of MyD88- / - mice peritoneal macrophages to LPS was much lower than that of the wild type mice peritoneal macrophages. Conclusions MyD88 plays an important role in inflammation induced by LPS. The inhibition of LPS-induced expression of MyD88 by fenoterol is associated with its anti-inflammatory effect.
7.Role of hypoxia-induced factors in cancer development mediated by cancer stem cells

Hypoxia and hypoxia-induced factors (HIFs) are main regulators for tumor stem cells, metastasis-initiating cells and their differentiated progenies to adapt to the environment which lacks oxygen and nutrient in the process of cancer development. HIFs are up-regulated in many tumors, including leukemia, glioblastoma, melanoma, prostate cancer, breast cancer and pancreatic cancer, in where they are especially highly expressed in hypoxic regions. HIFs activation can induce expression of numerous stem cell related genes and multidrug resistance genes, which may play important roles in tumour and stem cell-mediated self-renewing, energy metabolism alternation, invasion, metastasis, angiogenesis and treatment resis?tance of neoplastic cells. Consequently, it will provide new clues for cancer therapy after investigating the role of HIFs in tar?geted regulation and metabolic pathway modulation in various stem cell-mediated tumor cells.
8.Effects of recombinant immunotoxin hIL-2-LuffinP1 on lymphocyte proliferation in vitro

Objective To observe the biological activity of the recombinant immunotoxin hIL-2-LuffinP1 derived by gene engineering in vitro.Methods To observe the inhibitory effect on lymphocytes in vitro,the splenocytes isolated from C57 and BALB/c mice were used for mixed mouse lymphocyte response(MLR),and the lymphocyte proliferation was observed in vitro with BALB/c mice splenocytes stimulated by concanavalin A(ConA).Different concentrations of hIL-2-LuffinP1 was added into the reaction system,with LuffinP1 adopted as control.3H-TdR incorporation assay was used to detect the effects of hIL-2-LuffinP1 on lymphocyte proliferation,and its effects on CTLL-2 cells(IL-2 dependent cells) were also observed.The experiments consisted of six groups(PBS,IL-2,LuffinP1,LuffinP1+IL-2,hIL-2-LuffinP1 and hIL-2-LuffinP1+IL-2).CTLL-2 cells were cultured for 12h after being treated with different agents,and then the cells were stained by trypan blue to estimate the dead cells in every 100 cells.Results It was showed that CPM declined correspondingly to the gradually increased concentration of hIL-2-LuffinP1,and cell proliferation was inhibited obviously.The inhibition rate was up to 97% at the concentration of 10-6mmol/L of hIL-2-LuffinP1.The activity of inhibiting lymphocyte proliferation was proportional to the dosage.On the contrary,no obvious inhibition to lymphocyte proliferation was found in the control group treated with LuffinP1,the inhibition ratio only reached to 14% with the same concentration of hIL-2-LuffinP1.It was also found that,when hIL-2-LuffinP1 and LuffinP1 in different concentrations added to spleen cells stimulated by ConA,hIL-2-LuffinP1 still exhibited strong ability of inhibition on the splenocyte proliferation,while LuffinP1 has no obvious inhibiting effect on the lymphocyte proliferation in the two reaction systems mentioned above.MLR showed the same results.Furthermore,in the experiment of cytotoxic effects of IL-2-LuffinP1 on CTLL-2 cells,the inhibition rates of hIL-2-LuffinP1+IL-2 group and hIL-2-LuffinP1 group were 29.6% and 47.4% respectively,the difference was significant compared with IL-2 group(P

10.Effect of Tangnaoluotong Combined with Intravenous Low Intensity Laser Irradiation on Blood Rheology and Endothelin of Diabetes Mellitus and Cerebral Infarction Patients

Objective To observe the effect on the patients of diabetes mellitus and cerebral infarction in terms of level of blood rheology and endothelin after using both intravenous low intensity laser irradiation (ILILI) and traditional Chinese medicine Tangnaoluotong in the treatment. Method Ninety patients of diabetes mellitus and cerebral infarction, having the symptom of deficiency in both vital energy and yin with blood stasis, were divided into three treatment groups at random:ILILI group (A), TCM treatment group (B), and a group with compound treatment of ILILI and TCM (C). The level of blood rheology and endothelin were observed after 1 month’s treatment. Result Compared with group A and group B, group C could significantly reduce the level of fibrinogen and endothelin, improve the deformed ability of RBC. Conclusion ILILI with Tangnaoluotong can significantly alter the blood rheology and endothelin on the patients of diabetes mellitus and cerebral infarction, which result in a better curative effect.
