1.New biomarkers in diagnosis of acute myocardial infarction
Chinese Journal of General Practitioners 2016;15(9):725-730
With the development of laboratory medicine, more and more biomarkers have been discovered and applied in the diagnosis and treatment of acute myocardial infarction. For patients with suspected myocardial infarction, early diagnosis and timely treatment can save endangered myocardium and reduce fatality rate to a great extent. The proper use of biomarkers is of important value in preventing cardiovascular events and improving prognosis. This paper reviews the recent literature on new biomarkers in diagnosis of acute myocardial infarction, introduces the characteristics of these markers and their clinical diagnostic value.

Objective To evaluate the curative effect, adverse reactions and complications of transcatheter uterine arterial embolization (TUAE) therapy for leiomyoma of uterus with three different embolization agents: silk particles, pingyangmycin-lipiodol emulsion (PLE), polyvinyl alcohol (PVA).Methods One hundred and twenty-six patients of uterus leiomyoma were included, who undergoing TUAE treatment with silk particles(54 cases, silk particles group) ,PLE(40 cases,PLE group) and PVA(32 cases, PVA group) respectively.Adverse reactions and complications were recorded until 6-60 months after TUAE treatment.Measured the volume of the leiomyoma by ultrasonic inspection or enhanced computed tomography (CT).Results All clinical symptoms of three groups were relieved, and the volumes of uterus leiomyoma after TUAE treatment of 6,12,24 months in PVA group [(50.84 ± 30.77), (39.92 ± 25.72), (31.24 ± 30.60) cm~3]were less than those in PLE group[(76.24 ± 31.16), (58.21 ± 32.05), (52.48 ± 31.41 ) cm~3]and silk particles group[(69.22 ± 36.22), (68.31 ± 32.42), (68.01 ± 30.12) cm~3].The inferior belly and perineal region pain were most slight in silk particles group, but no significant difference in complications was observed in three groups. The symptom recurrence was 3 cases, 2 cases in silk particles group and 1 case in PIE group. Conclusions Clinical outcome of TUAE treatment with all the three embolization agents is acceptable.Adverse reactions and complications are associated with volume of the leiomyoma and the area being embolized.PVA is superior in prevention of complications and relapse of uterus leiomyoma after TUAE treatment.

Objective To develop a high performance liquid chromatography(HPLC) method to determine the concentration of ampicillin and probenecid in human plasma. Methods A Symmetry ShieldTM RP18 column(5 ?m,150 mm?3.9 mm)was used. The mobile phases were 0.068 mol/L KH_2PO_4(pH 3.0)∶acetonitre(92∶8,V/V)for ampicillin,H_2O∶acetonitrile∶1 mol/L KH_2PO_4∶1 mol/L acetic acid(59.5∶39.8∶0.62∶0.062,V/V,0.15% triethylamine added to reach pH 2.76) for probenecid. The detective wavelength were at 254 nm for probenecid and 210 nm for ampicillin. Results The calibration curves obtained were linear in the range of 0.625-20 ?g/ml (r=0.999 9) of probenecid and 1.25-40 ?g/ml of ampicillin. Conclusion This analysis method is simple, sensitive and rapid, suitable for studying pharmacokinetics of ampicillin and probenecid.

AIM:To investigate the protective effects of total saponins of panax notoginseng (PNS) on myocardial hypertrophy and fibrosis induced by isoproterenol (ISO) in rats. METHODS: Myocardial hypertrophy and fibrosis model of rats were induced by injection of ISO (5 mg?kg-1?d-1, sc) for 7 days. From day 2, the rats were administered with PNS at dose of 25 and 50 mg?kg-1?d-1, ip for 14 days, the control and ISO model group were received saline injection. Then, the heart-weight (HW), left ventricular weight (LVW), the ratio of HW/BW and LVW/BW (LVI) were measured; the hydroxyproline and malondialdehyde (MDA) and angiotensin (AngII) content of left ventricle. The level of nitric oxide (NO), nitric oxide synthase (NOS), superoxide disrnutase (SOD) and glutathione peroxidase (GSH-Px) activities in left ventricle were determined by spectrophotemetry and radioimmunoassay, respectively. RESULTS: Compared with NS control group, the ratio of HW/BW, LVW/BW and the content of hydroxyproline, AngII, MDA and iNOS activity in the left ventricle were significantly increased. The cNOS, SOD, GSH-Px activities and NO content were obriously decreased in the ISO model group. After treatment with PNS, the left ventricular NO content, cNOS, SOD and GSH-Px activities were markedly higher than those in ISO model group. The content of MDA, AngII and iNOS activities and the ratio of HW/BW, LVI were significantly lower than those in ISO model group. CONCLUSION: PNS reverses the myocardial hypertrophy and fibrosis induced by isoproterenol in rats. This effect may be related to eliminating the oxygen free radicals and raising NO level.

AIM: To study the effect of genistein on activation and proliferation of T cells, and explore the molecular mechanism of genistein. METHODS: Fluorescence conjugated monoclonal antibodies and flow cytometry were used to detect the express of CD69 and CD25 by activated T cells in vitro in response to Concanavalin (ConA )and Phorbol 12,13-dibutyrate(PDB) or T cell proliferation stained by CFSE in response to PDB / Ionomycin or ConA. RESULTS: Genistein inhibited the expression of CD69 and CD25 in activated T cells in response to Con A in a concentration-dependent manner and in response to PDB in a high concentration. Genistein inhibited proliferation of T cells in both groups in a concentration-dependent manner. CONCLUSION: Genistein inhibited activation and proliferation of T cells in vitro in response to polyclonal stimulus, and it may hold potential as a new immunosuppressant.
