1.Effects of Gynostemma Pentaphyllum on serum markers of liver fibrosis in cholesterol-induced non-alcoholic fatty liver disease in rabbits

Objective To investigate the effects of Gynostemma Pentaphyllum on serum markers of liver fibrosis in cholesterol-induced non-alcoholic fatty liver disease (NAFLD) in rabbits. Methods Forty adult male white rabbits were randomly divided into a normal control group, a model group, a treatment group and a simvastatin group, with 10 rabbits in each group. NAFLD was induced with a high-cholesterol diet. After modeling, the rabbits in the treatment group were intragastrically administrated with Gynostemma Pentaphyllum 5 mg/(kg?d), the simvastatin group with simvastatin 5 mg/(kg?d), and the model and normal control groups with the equal volume of distilled water for 9 weeks. The serum levels of total cholesterol (TC) and triglyceride (TG), the serum inflammatory maker C-reactive protein (CRP), the serum markers of liver fibrosis such as hyaluronic acid and laminin (LN), and the TG level in the liver tissue were detected. Results Compared with the normal control group, the serum levels of TC (60.50 ± 9.77 mg/L vs.1.30 ± 0.44 mg/L), TG (1.72 ± 0.61 mmol/L vs. 0.85 ± 0.39 mmol/L), CRP (256.79 ± 30.78 mg/L vs. 8.71 ± 1.41 mg/L), HA (798.8 ± 69.4 ng/ml vs. 121.2 ± 6.8 ng/ml),LN (964.8 ± 62.8 ng/ml vs. 142.4 ± 12.2 ng/ml) in the model group were increased significantly (all P<0.01). Compared with the model group, the serum levels of TC (36.44 ± 6.57 mmol/L vs. 60.50 ± 9.77 mmol/L), TG (1.31 ± 0.39 mmol/L vs. 1.72 ± 0.61 mmol/L), CRP (68.77 ± 10.78 mg/L vs. 256.79 ± 30.78 mg/L), HA (420.8 ± 60.2 ng/ml vs. 798.8 ± 69.4 ng/ml), LN (449.8 ± 56.6 ng/ml vs. 964.8 ± 62.8 ng/ml) and the TG level in the liver tissue (0.52 ± 0.10 mmol/L vs. 0.77 ± 0.08 mmol/L) in the treatment group were decreased significantly (all P<0.01). The TG level in the liver tissue in the treatment group was significantly lower than that in the simvastatin group (0.52 ± 0.10 mmol/L vs. 0.59 ± 0.09 mmol/L;P<0.05). There were no significant difference in the serum levels of TC, TG,CRP,HA and LN between the treatment group and the simvastatin group (all P>0.05). Conclusions Gynostemma Pentaphyllum may regulate lipid metabolism, alleviate inflammation, and decrease serum markers of liver fibrosis, and might protect against liver fibrosis in rabbits with NAFLD.

4.Effect of Gypenosides on Lipid Peroxidation in Rats with Type 2 Diabetes Mellitus Complicated by Nonalcoholic Fatty Liver Disease

Objective To obserVe the effect of gyPenoside on liPid Peroxidation and hePatic lesion in rats with tyPe 2 diabetes mellitus and nonalcohol fatty liVer disease. Methods Totally,65 SPF male SD rats were randomly diVided into blank control grouP (grouP N),NAFLD model grouP (grouP NM),and NAFLD with T2DM model grouP. The NAFLD with T2DM model grouP was further diVided into three subgrouPs:JH grouP,Perfused with 1 g·kg-1 ·d-1 GPS;JL grouP,Perfused with 0. 5 g·kg-1 ·d-1 GPS;model control grouP,Perfused with the same Volume of water. Blood sugar,triglycerides ( TG) ,total cholesterol ( TC) ,alanine aminotransferase ( ALT) ,asPart aminotransferase ( AST) ,adePonectin ( ADP) in the Plasma were measured. TG, malondialdehyde (MDA),and suPeroxide dismutase (SOD) in the liVer tissue were also tested. Results ADP leVel was (7. 46±1. 12),(3. 58±0. 98),(4. 89±1. 02),(4. 79±1. 01) and (4. 13±0. 89) ng·mL-1 in N,M,NM,JH and JL grouPs, resPectiVely. The ADP leVel was significantly higher in grouP JH and JL than in grouP M (P<0. 01),and significantly higher in grouP JH than in grouP JL (P<0. 05). MDA leVel was (2. 98±0. 09),(4. 22±0. 11),(3. 66±0. 10),(3. 72±0. 11),(3. 99±0. 13) nmol·mL_1 in N,M,NM,JH and JL grouPs,resPectiVely. The MDA leVel was significantly lower in grouP JH and JL than in grouP M (P<0. 01),and significantly lower in grouP JH than in grouP JL (P<0. 05). SOD leVel was (240. 8±17. 4), (149. 9±20. 6),(181. 6±19. 4),(209. 8±19. 2),(189. 4±18. 9) U·mL_1 in N,M,NM,JH,and JL grouPs,resPectiVely. SOD leVel was significantly higher in grouP JH and JL than in grouP M (P<0. 01),and significantly higher in grouP JH than in grouP JL (P<0. 05). TG leVel was (28. 98±1. 68),(214. 46±5. 44),(198. 46±6. 98),(142. 87±6. 64) and (164. 92±7. 56) mg·g-1 in N,M,NM,JH and JL grouPs,resPectiVely. TG leVel was significantly lower in grouP JH and JL than in grouP M ( P<0. 01),and significantly lower in grouP JH than in grouP JL (P<0. 05). ALT and AST were significantly lower in grouP JH and JL than in grouP M (P<0. 01),and significantly lower in grouP JH than in grouP JL (P<0. 05). Conclusion The liPid Peroxidation in the liVer of rats with T2DM comPlicated with NAFLD can be reduced by gyPenoside,and hePatic lesion may be alleViated through inhibition of liPid Peroxidation.
5.Anti-inflammatory mechanisms of purslane from Yunnan province: an experimental study

Objective To explore the anti-inflammatory mechanisms of purslane by evaluating its effects on the expression of tumor necrosis factor-α (TNF-α) and intercellular adhesion molecule 1 (ICAM-1).Methods A model of inflammation was developed in 45 mice by painting xylene to the auricle of the right ears,which were then divided into 3 groups to receive no treatment (negative control group),be topically treated with the extraction of purslane from South Korea (positive control group) or Yunnan province (experimental group).Fifteen mice receiving no sensitization nor treatment served as the blank control group.Two hours after the single topical treatment,skin tissue samples were obtained from the site of experimental inflammation and subjected to pathological examination by using hematoxylin and eosin (HE) staining.Immunohistochemistry was performed to quantify the expression of TNF-α and ICAM-1 in the tissue samples.Results Pathological examination showed blood vessels and a small quantity of lymphocytes in murine dermis of the blank control group as well as loose and edematous dermis infiltrated with massive lymphocytes in the negative control group.However,there was only mild edema and perivascular infiltration with some inflammatory cells in the dermis of the positive control group and experimental group.Neither TNF-α nor ICAM-1 was expressed in the skin tissue of the blank control group,but an intense expression was observed for TNF-α in the vascular endothelial cell membrane and for ICAM-1 in the vascular endothelial cell membrane and lymphocyte membrane in the negative control group,which was significantly downregulated by the purslane from South Korea in the positive control group and by the purslane from Yunnan province in the experimental group (all P < 0.01).Rank sum test showed a statistical difference in the expression level of TNF-α and ICAM-1 between the blank control group and experimental group (both P <0.01).Conclusion The purslane from Yunnan province may counteract inflammation by affecting the expression of TNF-α and ICAM-1.
6.Nosocomial Infection Problems Found in Terminal Patients Records Inspection

OBJECTIVE To check the defects of nosocomial infections reports,recording of the patient′s history,and antibiotics usage,so as to improve hospital management and reduce nosocomial infection.METHODS Patient′s records from Jan 1,2008 to Dec 31,2008 were reviewed to find out the defects and feedback to the physicians.RESULTS Some problems of 5995 cases were checked out and corrected,some suggestions were made.CONCLUSIONS Checking the patient′s records may supplement the defects of prospective monitor,direct the antibiotics therapy,and monitor occurrence of drug resistance.
7.Evaluation of serum CA125 in literatures of ovarian neoplasms based on evidence-based medicine

Objective To assess the serum CA125 in literatures of ovarian neoplasms based on evidence-based medicine.Methods The relating reports published in the years of 1996—2003 were collected and evaluated according to the evidence-based medicine principles. Main outcomes include design methods,sample screening,establishment of control group,blind method,criteria of diagnostic trials.Results In the reports collected, gold standard was adopted in 62 reports and independent blind method was adopted in 23 reports, criteria of diagnostic trials such as sensitivity, specificity and likelihood ratio were incomplete and there was no descriptions okf potential bias controlling. As for the literatures sharing the same method ,their conclusion did not totally accord with each other.Conclusion There is no evidence to implicate that serum CA125 was considered an independent predictor of the diagnosis of ovarian cancer.
8.Determination of geniposidic acid,chlorogenic acid and geniposide in Duzhong Pingya Tablet by RP-HPLC

AIM: To estabish a RP-HPLC method for determing geniposidic acid、chlorogenic acid、geniposide in Duzhong Pingya Tablet(Folium Eucommiae).METHODS:Geniposidic acid,chlorogenic acid and geniposide were analysed by using C_(18) column with the mobile phase of methanol-water-aceacid(15∶85∶1.5) at the flow rate of 1 mL/min,the detective wavelength was set at 237 nm. RESULTS: The analytical had good linearity.The precision of geniposidic acid,chlorogenic acid and geniposide were 1.93%、1.50%、1.66%,respectively and the recoveries were 97.3%,97.2%,96.7%,respectively. CONCLUSION: This method is simple,quick and reproducible and could be used for the determination of three constituents in Duzhong Pingya Tablet.
9.STUDIES ON THE CHARACTERISTICS OF A MODERATELY THERMOACIDOPHILIC IRON-OXIDIZING BACTERIVM

A moderately thermoacidophilic iron-oxidizing bacterium,designated as strain MLY,was isolated from a coal spoil heap in China.The optimum of temperature for growth is 50℃~54℃.The optimum of pH is 1.2~1.4.The strain MLY is facultative autotroph and grows heterotrophically on yeast extract.It is able to oxidize ferrous iron(Fe 2+ ),pyrite(FeS 2),and elemental sulfur(S 0) autotrophically and mixotrophically in the presence of yeast extract.Autotrophic oxidation of elemental sulfur is relative weak.The comparison of ferrous iron and pyrite oxidation between strain MLY and A10 Thiobacillus ferrooxidans,strain indicated that MLY is one time faster than A10.
10.A randomized controlled study on the application of parecoxib sodium in laparoscopic cholecystectomy

Objective To observe the effect of parecoxib sodium on relieving acute postoperative pain and inflammatory fac‐tors in laparoscopic cholecystectomy .Methods Totally 96 cases of laparoscopic cholecystectomy were randomly divided into 3 groups :experimental group (group A ,group B) and control group(group C) ,32 cases in each group .Group A received intravenous infusion of parecoxib sodium 40 mg (diluted to 5 mL ) intravenously at the induction of general anesthesia and intravenous injection of 5 mL normal saline 15 min before surgery .In group B ,anesthesia induction ,15 minutes before surgery were given 5 mL of normal saline ,parecoxib sodium 40 mg (diluted to 5 mL) intravenously .In group C ,5 mL of normal saline was given intravenously at the time of general anesthesia induction and 15 min before operation .The total number of PCA and the number of effective compres‐sions and VAS scores at 12 h postoperatively ,the levels of serum TNF‐α,IL‐6 and IL‐12 before operation ,immediately after opera‐tion ,at 2 ,12 ,24 h after operationwere compared .Results The PCA press times and effective press times of patients in group A within 12 hours after the operation were much less than those in group B ,C(all P<0 .05) .The VAS scores of group A at 1 ,2 ,4 ,6 , 8 and 12 h were lower than those in group B and C (P<0 .05) ,but there was no significant difference between group A and group B after 24 h(P>0 .05) .The changes of serum IL‐6 ,TNF‐αlevel in three groups of patients at the end of the operation ,2 ,12 ,24 h after operationwere :group A < group B < group C .While the changes of serum IL‐12 were:group A > group B > group C . Conclusion Parecoxib sodium has a good analgesic effect of laparoscopic cholecystectomy in the induction of general anesthesia than before the end of the operation and application effect is better ,and its analgesic effect can reduce the production of inflammato‐ry factors IL‐6 and TNF‐α,and increase the level of IL‐12 ,which is beneficial to postoperative recovery .
