1.Studg on Adsorption and Separation Properties of Total Flavones From Euphorbia Humifusa、Leaves of Flos Lonicerae and Chrysanthemum Morifolium by Macroporous Resins.

Objective In order to study the application of macroporous resins and so on to the purified active components of Euphorbia humifusa、Leaves of Flos Lonicerae、Chrysanthemum morifolium,adsorption and separation properties for 3 types of macroporous resins and polyamide were investigated.Methods The total flavone was used as the evaluating criteria,we selected suitable macroporous resins and studied optimum technological parameters of the adsorption and elution.Spectrophotometry was used for the determination of total flavone.Results The suitable macroporous resins which were used to the purified active components of traditional Chinese medicine were D101 and DA201 and DM301 for Euphorbia humifusa、DA201 for Leaves of Flos Lonicerae、D101 and DA201 and DM301 for Chrysanthemum morifolium,The concentration of the sample of Euphorbia humifusa for DA201 and D101 were 0.49~1.47 and 0.42~1.31 mg/ml.The concentration of the sample of Leaves of Flos Lonicerae for DA201 was 1.03~2.07 mg/ml.The concentration of the sample of Chrysanthemum morifolium for DA201 and D101 was 0.50~1.00 and 0.71~1.99 mg.ml.In the adsorption course,appeared leaking were 8 and 10、2、2 and 1 BV respectively.In the elution course,when the alcohol concentrations were 20%、30%、40% and 20%、30%、40%;10%、20%、30%;30%、40%、50% and 20%、30%、40%;respectively,the total flavone content in the elution solutions was higher.The influence of temperature to DA201 and D101 adsorpting total flavone for Euphorbia humifusa was not great.But the influence of temperature to DA201 and D101 adsorpting total flavone for Leaves of Flos Lonicerae and Chrysanthemum morifolium were certain degree.Conclusions It is obviously different to refine the total flavone active components of traditional Chinese medicine,while using 3 types of macroporous resins and polyamide.
4.Change of TRAF6 expression in rat cerebral ischemia reperfusion injury

Objective To investigate the potential role of tumor necrosis factor receptor-associated factor-6(TRAF6)in the rat cerebral ischema-reperfusion injury.Methods 40 healthy adult SD rats were divided into 5 groups(n=8)according to the ran-dom control principle:sham operation group,ischemia group,reperfusion 2 h group(R2 h),reperfusion 12 h group(R12 h)and reperfusion 24 h group(R24 h).The rat cerebral ischemia-reperfusion injury model was constructed.The change of TRAF6 expres-sion was examined by RT-PCR and Western-blot.Then,the immunohistochemistry was adopted to locate the TRAF6 protein.Re-sults Compared with the sham group,the expression of TRAF6 in the ischemia group and the R2 h,R12 h and R 24 h groups was obviously increased,but the difference had no statistical significance (P <0.05).TRAF6 was mainly located in the cytoplasm of neuronal cells.Conclusion Activated TRAF6 is involved in the brain cell death induced by cerebral ischemia-reperfusion.
5.Open reduction and internal fixation with anatomical plate to treat complex fractures of distal femur

Objective To investigate the treatment of complex fractures of distal femur with open reduction and internal fixation with anatomical plate. Methods From January 2001 to June 2004, 67 cases of complex fractures of distal femur were treated with open reduction and anatomical plate internal fixation. The allogeneic bone was grafted for 23 cases, ilium was grafted for 35 cases, ilium and fibula were grafted for 9 cases. Bio- resorbable membranes were used between quadriceps femoris and femur. 13 cases were aided with patella traction. All the patients received fumigation and rinsing with Chinese herbs and CPM(continuous passive motion) after operation. Results All the 67 cases were followed up for 10 to 26 months. All the fractures got united. The postoperative knee functions were excellent in 36 cases, good in 23 cases, fair in 5 cases, and poor in 3 cases according to Kolment standards. The excellent and good rate was 88.1% . Conclusions Open reduction, internal fixation with anatomical plate, bone grafting, local use of bio- resorbable membranes, patella traction when necessary, fumigation and rinsing with Chinese herbs and CPM after operation are good measures to treat complex fractures of the distal femur, because they can provide stable fixation, allow early postoperative functional training, and prevent adhesion of quadriceps femoris and stiffness of the knee joint
6.Analysis in stressor of patients with maintained hemodialysis

Objective To observe the stressor characteristic of patients with maintained hemodialy-sis (MHD). Methods The Chinese version of bemodialysis stressor seale(HSS) was adopted among 436 bemodialysis participants and the results underwent analysis. Results Among 436 patients psychological stressors> physiological stressers > social stressors, the most severe stressors included the requirement of long-term dialysis, the rest showed sequence from high to low: medical cost, concerning about the future,changes in family responsibility and decrease in sexuality. The slightest stressors included decreased ability to procreate, being limited in some styles of clothing, arterial and venous stick, reversal in family role with the children and changes in body appearance. Significant differences existed in patients with different age,gender, education background, working status and economic status, it manifested as follows: the more elder-ly, the more severity of stress, male patients had more severe stress than the female, patients with higher e-ducation background showed lower stress level, patients who kept working had lower level of stress, pa-tients who had better economic status showed lower stress level. Conclusions MHD patients often expe-rience high stress levels. Some measures should be taken to help MHD patients to cope better with stress.
7.Clinical features of interstitial lung involvement in primary Sj(o)gren's syndrome

Objective To analyse the clinical features and risk factors of interstitial lung disease in primary SjSgren's syndrome (pSS).Methods A retrospective study was employed.All 130 pSS patients who were hospitalized during 2004 to 2006 were enrolled into this study.Thirty-seven clinical and laboratory variables were used as the research factors and interstitial lung involvement as the related factor.The data were analyzed with the Logistic regression model.Results of 130 patients,22(16.9%) had interstitial lung involvement.Twelve patients were asymptomic when pulmonary involvement was detected.Variables in the univariate analysis which were significantly associated with lung fibrosis were age,exocrine glands(including parotid,sublingual gland and submandibular gland) swelling,oral ulceration and fever.Positive rate of antiU1RNP antibody in group of lung damage was significantly higher than non-lung damage group [5/22(22.7%)vs 6/108 (5.6%),P=0.021].In the Logistic regression model.variables which were significantly associated with pulmonary involvement were exocrine glands swelling (OR=3.739,95%CI 1.069~16.079,P<0.05).oral ulceration (OR=3.739,95%CI 1.069~16.079,P<0.05)and fever (OR=3.067,95%CI 1.198~20.067,P<0.05).Conclusions This study indicates that some of the pulmonary damages of pSS are subclinical.Exocrine glands swelling,oral ulceration and fever are the risk factors for interstitial pulmonary involvements in pSS.

9.hTERT gene antisense oligodeoxynucleotide enhances cisplatin-induced apoptosis in ALL cells

AIM: To explore the effect of human telomerase reverse transcriptase (hTERT) gene antisense phosphorothioate oligodeoxynucleotide (AS PS-ODN) on cisplatin-induced apoptosis in cultured primary acute lymphoblastic leukemia (ALL) cells. METHODS: The expression levels of hTERT protein were detected by immunofluorescence using fluoresce isothiocyanate (FITC) lable. Cell surviving fraction was determined using the trypan blue dye exclusion assay. Apoptosis was detected by morphological observation and flow cytomertric cell cycle analysis. RESULTS: The expression of hTERT protein was inhibited after treated by hTERT AS PS-ODN. Treatment with cisplatin after 24 h of exposure to AS PS-ODN had significantly reduced the number of viable ALL cells. However,there was no difference on ALL cells survival between sense oligodeoxynucleotide (S PS-ODN) /CDDP combination and CDDP-treated cells alone. In morphological observation of apoptotic cells using Hoechst 33258 and PI double staining techniques,cells displayed classic apoptotic changes treated with CDDP or CDDP combined with hTERT AS PS-ODN or S PS-ODN at 48 h. Apoptotic rates of cells added CDDP and AS PS-ODN were higher than that of cells added CDDP only ( P 0.05). CONCLUSION: hTERT AS PS-ODN inhibited the expression of hTERT protein and increased the CDDP-induced apoptosis in primer acute lymphoblastic leukemic cells.
10.Effect of hTERT antisense oligodeoxynucleotide on Cis-diamminedichicloroplatinum-induced apoptosis in Jurkat cells

AIM: To explore the effect of hTERT antisense phosphorothioate oligodeoxynucleotide (ASODN) on apoptosis induced by chemotherapeutic drugs in Jurkat cell lines. METHODS: Cell viability was determined using the trypan blue dye exclusion assay. Apoptosis was detected by morphological observation, DNA gel electrophoresis and flow cytometry analysis. RESULTS: The survival rates of Jurkat cells cultured with daunorubicin, vincristin, and etoposide, respectively were similar with that cultured with those chemotherapic drugs plus hTERT ASODN. The survival rates of Jurkat cells cultured with cis-diamminedichicloroplatinum(DDP) added 24 hours later were higher than that cultured with hTERT ASODN and DDP added 24 hours later. The survival rates of Jurkat cells cultured with DDP were similar with that cultured with hTERT SODN and DDP. In morphological observation of apoptotic cells using Giemsa staining, cells displayed classic apoptotic changes treated with DDP or DDP combined with hTERT ASODN or SODN at 48 hours. Agarose gel electrophoresis of genomic DNA from Jurkat cells treated with ASODN and DDP combination for 48 hours showed typical DNA “ladder”. Neither the DNA from Jurkat cells treated with SODN plus DDP nor the DNA from the cells treated with DDP alone showed ladder pattern. Apoptosis rates of Jurkat cells treated with DDP for 48 hours after 24 hours of exposure to ASODN significantly increased. There was significant difference in the percentage of apoptotic Jurkat cells between hTERT ASODN plus DDP and SODN plus DDP or DDP alone, respectively. CONCLUSION: The hTERT ASODN complementary to the translation initiation region of hTERT mRNA enhanced DDP-induced apoptosis in Jurkat cells.
